RESUMO

BACKGROUND: Dysregulation of enhancer transcription occurs in multiple cancers. Enhancer RNAs (eRNAs) are transcribed products from enhancers that play critical roles in transcriptional control. Characterizing the genetic basis of eRNA expression may elucidate the molecular mechanisms underlying cancers. METHODS: Initially, a comprehensive analysis of eRNA quantitative trait loci (eRNAQTLs) was performed in The Cancer Genome Atlas (TCGA), and functional features were characterized using multi-omics data. To establish the first eRNAQTL profiles for colorectal cancer (CRC) in China, epigenomic data were used to define active enhancers, which were subsequently integrated with transcription and genotyping data from 154 paired CRC samples. Finally, large-scale case-control studies (34,585 cases and 69,544 controls) were conducted along with multipronged experiments to investigate the potential mechanisms by which candidate eRNAQTLs affect CRC risk. RESULTS: A total of 300,112 eRNAQTLs were identified across 30 different cancer types, which exert their influence on eRNA transcription by modulating chromatin status, binding affinity to transcription factors and RNA-binding proteins. These eRNAQTLs were found to be significantly enriched in cancer risk loci, explaining a substantial proportion of cancer heritability. Additionally, tumor-specific eRNAQTLs exhibited high responsiveness to the development of cancer. Moreover, the target genes of these eRNAs were associated with dysregulated signaling pathways and immune cell infiltration in cancer, highlighting their potential as therapeutic targets. Furthermore, multiple ethnic population studies have confirmed that an eRNAQTL rs3094296-T variant decreases the risk of CRC in populations from China (OR = 0.91, 95%CI 0.88-0.95, P = 2.92 × 10-7) and Europe (OR = 0.92, 95%CI 0.88-0.95, P = 4.61 × 10-6). Mechanistically, rs3094296 had an allele-specific effect on the transcription of the eRNA ENSR00000155786, which functioned as a transcriptional activator promoting the expression of its target gene SENP7. These two genes synergistically suppressed tumor cell proliferation. Our curated list of variants, genes, and drugs has been made available in CancereRNAQTL ( http://canernaqtl.whu.edu.cn/#/ ) to serve as an informative resource for advancing this field. CONCLUSION: Our findings underscore the significance of eRNAQTLs in transcriptional regulation and disease heritability, pinpointing the potential of eRNA-based therapeutic strategies in cancers.

RESUMO

With reduced toxicity and tunable optoelectronic properties, mixed cation halide perovskites (MCHPs) featuring partially substituted Pb with Sn and Ge have emerged as promising candidates for photovoltaic applications. However, the introduction of the disorder through large-scale preparation and alloying strategies leads to a significant challenge in comprehending the disorder's microscopic-level impact. Here, we found that, in addition to compositional variation, a synergy of disorder and cation radii ratio significantly affects optoelectronic properties. For Pb-Ge/Ge-Sn MCHPs, severe octahedral distortion with increasing degree of disorder adjusted their bandgaps in a wide range, giving rise to large effective masses, exciton binding energies, and weak visible absorption coefficients. The synergy of disorder and distortion transforms the Wannier excitons into localized characteristics, whereas the optoelectronic properties of Pb-Sn MCHPs are modulated by the disorder. Our work highlights the role of disorder in the tunability of optoelectronic properties, providing a novel strategy for designing photovoltaic materials.

RESUMO

Objective@#To analyze physical fitness and health status and gender differences of middle school students among 5 minorities (Mongolian, Hui, Uyghur, Zhuang and Korean), and to provide the theoretical basis for the strategy formulation.@*Methods@#The present data came from 3 waves of Chinese National Survey on Students Constitution and Health (2010, 2014, and 2019). According to National Standards for Students Physical Health (2014 Revision), excellent, and excellent good physical fitness and health status were defined. Cochran Armitage test was used to examine the trends of physical fitness and health status. Chi square test and Logistic regression were used to analyze the difference of physical fitness and health status by sex and survey year.@*Results@#From 2010 to 2019, the excellent physical fitness and health status rate of Mongolian, Hui, Uyghur, Zhuang and Korean students increased from 1.8%, 0.7%, 0.3%, 0.5% and 1.3% to 4.3%, 2.8%, 1.2%, 1.3% and 3.5%, respectively. The excellent good physical fitness and health status rate of Mongolian, Hui, Uyghur and Zhuang students increased from 12.9%, 8.0%, 7.2 % and 8.4% to 24.7%, 20.1%, 12.6% and 19.8%( Z =6.15,6.71,4.12,3.06,5.26;11.88,13.42,6.70,11.08, P <0.05), respectively. In 2019, students aged 13 to 15 years showed higher proportion of excellent/excellent good physical fitness and health status than that of students aged 16 to 18. Boys were more likely to be in excellent/excellent good physical fitness and health status than girls from 2010 to 2019. The sex difference in excellent/excellent good physical fitness and health status narrowed during 2010 and 2019.@*Conclusion@#Physical fitness and health status of minority students improved while sex difference narrowed during last decade, but there is still a long way to reach the goal proposed by China. Targeted intervention should be proposed to promote physical fitness and health status in accordance with the developmental characteristics, especially for girls and students aged 16 to 18 years.

RESUMO

Objective:To construct a prediction model for difficult tracheal intubation in the patients with obstructive sleep apnea-hypopnea syndrome (OSAHS).Methods:A total of 324 patients with OSAHS undergoing surgery with general anesthesia, admitted to our hospital from June 2019 to June 2021, were included in model group, and 175 patients with OSAHS undergoing surgery with general anesthesia, admitted from July 2021 to July 2022, were selected and served as validation group. The patients in model group were divided into occurrence group and non-occurrence group according to whether difficult tracheal intubation occurred. Logistic regression was used to construct the prediction model, and R4.2.1 software was used to draw the risk nomogram and calibration curve. The predictive accuracy of the model was evaluated by the area under the receiver operating characteristic curve. Results:Body mass index (BMI), sagittal minimum cross-sectional area, horizontal minimum cross-sectional area and mandibular distance were risk factors for difficult tracheal intubation in OSAHS patients ( P<0.05). A prediction model was developed using the above factors: Logit P=33.726+ 1.411×BMI score-0.014×sagittal airway minimum cross-sectional area-0.013×airway horizontal minimum cross-sectional area-0.312× mandibular distance. The area under the receiver operating characteristic curve was 0.846, Youden index 0.585, sensitivity 0.831, specificity 0.755, and the accuracy 0.889 (Hosmer-Lemeshow test χ2=9.24, P=0.322) in model group. The area under the external validation curve was 0.802, Youden index 0.545, sensitivity 0.636, specificity 0.908, and the accuracy 0.893 (Hosmer-Lemeshow test χ2=10.24, P=0.287) in validation group. Conclusions:The prediction model based on BMI, sagittal minimum cross-sectional area of airway, horizontal minimum cross-sectional area of airway and mandibular distance has a high value in predicting the risk of difficult tracheal intubation in patients with OSAHS.

RESUMO

The purification of hydrogen (H2) has been a vital step in H2 production processes such as steam−methane reforming. By first-principle calculations, we revealed the potential applications of holey TMC6 (TM = Mo and W) membranes in H2 purification. The adsorption and diffusion behaviors of five gas molecules (including H2, N2, CO, CO2, and CH4) were compared on TMC6 membranes with different phases. Though the studied gas molecules show weak physisorption on the TMC6 membranes, the smaller pore size makes the gas molecules much more difficult to permeate into h-TMC6 rather than into s-TMC6. With suitable pore sizes, the s-TMC6 structures not only show an extremely low diffusion barrier (around 0.1 eV) and acceptable permeance capability for the H2 but also exhibit considerably high selectivity for both H2/CH4 and H2/CO2 (>1015), especially under relatively low temperature (150−250 K). Moreover, classical molecular dynamics simulations on the permeation process of a H2, CO2, and CH4 mixture also validated that s-TMC6 could effectively separate H2 from the gas mixture. Hence, the s-MoC6 and s-WC6 are predicted to be qualified H2 purification membranes, especially below room temperature.

RESUMO

Chronic stress affects the reproductive health of mammals; however, the impact of adrenocorticotropin hormone (ACTH) level elevation during chronic stress on the reproduction of weaned sows remains unclear. In this study, nine weaned sows with the same parturition date were randomly divided into control group (n = 4) and ACTH group (n = 5). Each group received intravenous administration of ACTH three times daily for 7 days. Blood samples were collected every 3 h after injection. A radioimmunoassay was used to measure the concentrations of cortisol, luteinizing hormone (LH), follicle-stimulating hormone (FSH), progesterone (P4) and estradiol-17ß (E2) in the blood. Estrus was determined according to changes in the vulva and the boar contact test. The mRNA expressions of glucocorticoid receptor, FSH receptor, LH receptor (LHR) in the corpus luteum (CL) were detected by qRT-PCR. The results showed that ACTH administration substantially delayed the initiation of estrus and the pre-ovulatory LH peak. The sows of control group ovulated within 10 days and the ovulation rate was 100%, while it was 60% in the ACTH group. Two sows of ACTH group showed pseudo-estrus. The E2 concentrations significantly decreased in the ACTH group at 36 h, 42 h and 66 h of the experimental period. The P4 concentrations in the ACTH group significantly decreased at 132, 138, and 147 h of the experimental period. ACTH significantly reduced the LHR mRNA expression in CLs. In conclusion, long-term repeated ACTH administration affects the endocrinology, estrus onset, and ovarian function of weaned sows.

RESUMO

One efficient strategy for addressing the global water shortage is advanced membrane separation, which depends on the precise pore size being close to the hydrated ion size and other surface properties like charge and polarity. However, it is very difficult to fabricate uniform pores with diameters of <1 nm on monolayer membranes. By applying an electric field (bias voltage) perpendicular to the direction of the pressure difference, herein we demonstrate for the first time that a monolayer nanoporous graphene membrane with pores much larger than hydrated ions exhibits high salt rejection and allows a high rate of water transport. This theoretical proposal goes beyond the pore size limitation and shows promise for the design of high-performance reverse osmosis membranes.

RESUMO

Taking the Chinese city of Xiamen as an example, simulation and quantitative analysis were performed on the transmissions of the Coronavirus Disease 2019 (COVID-19) and the influence of intervention combinations to assist policymakers in the preparation of targeted response measures. A machine learning model was built to estimate the effectiveness of interventions and simulate transmission in different scenarios. The comparison was conducted between simulated and real cases in Xiamen. A web interface with adjustable parameters, including choice of intervention measures, intervention weights, vaccination, and viral variants, was designed for users to run the simulation. The total case number was set as the outcome. The cumulative number was 4,614,641 without restrictions and 78 under the strictest intervention set. Simulation with the parameters closest to the real situation of the Xiamen outbreak was performed to verify the accuracy and reliability of the model. The simulation model generated a duration of 52 days before the daily cases dropped to zero and the final cumulative case number of 200, which were 25 more days and 36 fewer cases than the real situation, respectively. Targeted interventions could benefit the prevention and control of COVID-19 outbreak while safeguarding public health and mitigating impacts on people's livelihood.

RESUMO

ObjectiveTo investigate the mechanism of interleukin-35(IL-35)/signal transducer and activator of transcription 3(STAT3)inhibition of eosinophil activation against allergic rhinitis(AR) by Bifukang. MethodOne hundred patients were randomly divided into a control group and a treatment group,50 cases in each group. The control group was given mometasone furoate nasal spray, and the treatment group was given Bifukang by nasal packing. The course of treatment was 28 days. The clinical efficacy, nasal classification and visual analogue scale(VAS) score of the two groups were observed before and after treatment. Enzyme linked immunosorbent assay(ELISA) was used to detect the expression levels of inflammatory factors [interleukin(IL)-4,IL-10,IL-17,IL-35] and Eotaxin and CC chemokine receptor-3(CCR3)in serum and nasal secretion of the two groups. The expression levels of STAT1,STAT3 and STAT4 were detected by real-time polymerase chain reaction(Real-time PCR).The content of immunoglobulin G(IgG) and the ratio of CD4+/CD8+were detected by ELISA and flow cytometry. ResultAfter treatment, compared with before treatment, the levels of IL-4 and IL-17 in serum and nasal secretion in 2 groups were decreased, while the levels of IL-10 and IL-35 were increased (P<0.05, P<0.01). The expression of STAT1, STAT2 and STAT3 in nasal secretions were significantly decreased (P<0.05). IgG and CD4+/CD8+ were decreased, and the differences were statistically significant (P<0.05, P<0.01).After treatment, compared with the control group, the levels of IL-4 and IL-17 in serum and nasal secretions of the treatment group were decreased, while the levels of IL-10 and IL-35 were increased (P<0.05). The expression of STAT1,STAT3 and STAT4 in the treatment group was significantly inhibited compared with the control group after treatment (P<0.05). In addition, the post-treatment serum CD4+/CD8+ and immunoglobulin G (IgG) levels were reduced in the treatment group compared with those of the control group (P<0.05, P<0.01). During the treatment, there were no abnormal changes in heart, liver, kidney function and routine blood and urine tests in the two groups. ConclusionBifukang has a good effect on allergic rhinitis, and its mechanism may be related to the regulation of IL-35/STAT3 pathway, the inhibition of eosinophil activation and the improvement of related immune function.

RESUMO

High-performance cathode catalysts are always desirable for nonaqueous lithium-oxygen (Li-O2) batteries. Using density functional theory calculations, the structural, electronic, and magnetic properties of SSX-Gr with different C/X ratios (X = H or F) are systematically studied. Then, a detailed mechanism on the dissociation of O2 and the migration of Li on the SSX-Gr is revealed, based on which C6X and C8X are confirmed as the potential candidates as cathode catalysts. The studies on reaction pathways suggest that the four-electron pathway is the more possible catalytic pathway for the selected SSX-Gr. The free energy diagrams for discharging and charging processes catalyzed by SSX-Gr reveal that C6F exhibits the highest application potential due to its considerably low oxygen reduction overpotential (0.83 V) and oxygen evolution overpotential (1.47 V). The extra spins induced by single-side functionalization endow graphene with the electrocatalytic activity.

RESUMO

Ginsenoside Rg_3 is widely used in clinical practice as an anti-tumor adjuvant drug, but its application is limited due to its poor oral absorption. In this study, we intended to construct a ginsenoside Rg_3 nanostructured lipid carrier modified by the pullulan(PUL-Rg_3-NLC) to improve the adhesion properties of ginsenoside Rg_3, promote the drug uptake and improve the anti-tumor efficacy. PUL-Rg_3-NLC was characterized by morphology, particle size and Zeta potential. In vivo adhesion characteristics were evaluated by oral gavage tests, and the results were verified from multiple perspectives in combination with in vitro uptake behavior and in vitro pharmacodynamics. The results showed that PUL-Rg_3-NLC, with a particle size of(102±1.89) nm, was characterized by gastric adhesion and could be retained in gastric tissues for a long time, and its uptake by BGC-823 cells was promoted mainly through the pathway mediated by the caveolin-mediated endocytosis. In vitro MTT, cell apoptosis, wound-healing assay and invasion assay all showed some anti-tumor effects. Therefore, PUL-Rg_3-NLC can significantly promote the adhesion of Rg_3 in the stomach, promote the uptake of drugs by gastric cancer cells, and improve the anti-tumor effect. This study can provide some reference for the adjuvant treatment of gastric cancer.

RESUMO

A high-performance gas separation membrane for hydrogen (H2) purification is still highly desirable for the sustainable development of our society. Based on the structure of γ-graphyne, we theoretically designed the two-dimensional nanomaterials γ-C4X (X = O, S or Se) with intrinsic pores that may be suitable for gas separation. By first-principles calculations, we obtained the geometric structures of γ-C4X, and confirmed that γ-C4O and γ-C4S are stable at room temperature. Due to the moderate size of the intrinsic pores, γ-C4O exhibits a lower diffusion barrier and higher permeance for H2 than those of γ-C4S. It is worth noting that at room temperature, the high selectivity (1019) for separating H2 from a H2/CH4 mixture by γ-C4O shows great potential for H2 purification. Moreover, the classic molecular dynamics simulations at 300 K demonstrate that H2 can easily permeate through the intrinsic pores of γ-C4O membranes with high permeability and selectivity, which supports our first-principles calculations.

RESUMO

BACKGROUND@#Mutations in the isocitrate dehydrogenase 1 (IDH1) and IDH2 genes are important for both the integrated diagnosis and the prognosis of diffuse gliomas. The p.R132H mutation of IDH1 is the most frequently observed IDH mutation, while IDH2 mutations were relatively rarely studied. The aim of the study was to determine the pathological and genetic characteristics of lower-grade gliomas that carry IDH2 mutations.@*METHODS@#Data from 238 adult patients with lower-grade gliomas were retrospectively analyzed. The status of IDH1/2 gene mutations, telomerase reverse transcriptase (TERT) promoter mutations, O-methylguanine-DNA-methyltransferase (MGMT) promoter methylation, 1p/19q co-deletion and the expressions of IDH1 R132H, alpha-thalassemia X-linked mental retardation, and p53 were evaluated. Progression-free survival (PFS) and overall survival (OS) were calculated via Kaplan-Meier estimation using the log-rank test.@*RESULTS@#Totally, 71% (169/238) of patients were positive for IDH mutations, including 12 patients harboring mutations in IDH2. Among the 12 patients with IDH2 mutations, ten patients harbored the R172K mutation, one patient harbored the R172S mutation and one harbored the R172W mutation. Of these, 11 tumors occurred in the frontal lobe and showed morphology typical of oligodendroglioma. The proportion of grade II tumors was higher than that of grade III tumors in IDH2 mutant-gliomas. IDH2 mutations were frequently associated with TERT promoter mutations, 1p/19q co-deletion and MGMT promoter methylation. IDH2 mutations were associated with better outcomes compared with IDH wild-type gliomas (P < 0.05). However, the PFS and OS did not differ from that of IDH1 mutant patients (P = 0.95 and P = 0.60, respectively).@*CONCLUSIONS@#IDH2 mutations are more frequent in oligodendrogliomas and associated with a better prognosis. IDH2 mutations may segregate in distinct clinico-pathological and genetic subtypes of gliomas, and therefore may merit routine investigation.

RESUMO

Objective To observe the changes of brain function in patients with trigeminal neuralgia after administration of flurbiprofen axetil by using the resting-state functional magnetic resonance imaging(fMRI)and based on the amplitude of low-frequency fluctuation(ALFF). Methods Resting fMRI data of 20 patients with trigeminal neuralgia before and after treatment with flurbiprofen axetil were collected by 1.5T magnetic resonance imaging system. The resting fMRI data were pretreated by Statistical Parametric Mapping and DPABI(a toolbox for Data Processing and Analysis for Brain Imaging)software, and the difference of low-frequency oscillation amplitude of brain spontaneous activity before and after treatment with flurbiprofen axetil was analyzed by ALFF. Results The Visual Analogue Scale of pain intensity after flurbiprofen axetil injection was significantly lower than that before administration, and the pain relieved significantly(P=0.000).The ALFF values of right dorsolateral prefrontal lobe, bilateral medial prefrontal lobe, and right middle cingulate gyrus in patients treated with flurbiprofen axetil at rest were significantly lower than those before administration(P=0.000). Conclusions The analgesic effect of flurbiprofen axetil is exerted on the central system. This agent can inhibit the abnormal brain function caused by chronic pain stimulation and thus reduce pain. However, the specific mechanism needs further investigations.

To date, most studies of heterogeneous catalysis have focused on metal particles supported on the surface of substrates. However, studies of the catalytic properties of metallic nanoparticles supported on the interior surface of nanotubes are rare. Using first-principles calculations based on density functional theory, we have studied the CO oxidation on a single nickel atom confined in a nitrogen vacancy on the inside surface of boron nitride nanotubes (BNNT). By exploring the Eley-Rideal mechanism, we find that an Ni atom embedded on the interior surface of BNNTs exhibits a much higher catalytic activity for CO oxidation when compared with Ni doped on their outside surface. In addition, the energy barriers of the rate-determining step for CO oxidation on Ni embedded on the inside wall of BNNT(5,5), BNNT(6,6) and BNNT(7,7) are 0.39, 0.29 and 0.33 eV, respectively. The results illustrate the merit of confinement for CO oxidation.

RESUMO

Background The evaluation of hip arthroplasty is a challenge in computed tomography (CT). The virtual monochromatic spectral (VMS) images with metal artifact reduction software (MARs) in spectral CT can reduce the artifacts and improve the image quality. Purpose To evaluate the effects of VMS images and MARs for metal artifact reduction in patients with unilateral hip arthroplasty. Material and Methods Thirty-five patients underwent dual-energy CT. Four sets of VMS images without MARs and four sets of VMS images with MARs were obtained. Artifact index (AI), CT number, and SD value were assessed at the periprosthetic region and the pelvic organs. The scores of two observers for different images and the inter-observer agreement were evaluated. Results The AIs in 120 and 140 keV images were significantly lower than those in 80 and 100 keV images. The AIs of the periprosthetic region in VMS images with MARs were significantly lower than those in VMS images without MARs, while the AIs of pelvic organs were not significantly different. VMS images with MARs improved the accuracy of CT numbers for the periprosthetic region. The inter-observer agreements were good for all the images. VMS images with MARs at 120 and 140 keV had higher subjective scores and could improve the image quality, leading to reliable diagnosis of prosthesis-related problems. Conclusion VMS images with MARs at 120 and 140 keV could significantly reduce the artifacts from hip arthroplasty and improve the image quality at the periprosthetic region but had no obvious advantage for pelvic organs.

RESUMO

OBJECTIVE: To summarize the pathologic and computed tomography (CT) imaging characteristics of myoepithelial carcinoma in the salivary gland. STUDY DESIGN: Ten patients with myoepithelial carcinoma of the salivary gland underwent enhanced CT scanning, and the pathologic and CT imaging features were evaluated, including the pathologic type, size, margin morphology, and enhancement pattern of the lesions. RESULTS: Myoepithelial carcinoma had a multinodular or lobulated outline. The cut surfaces were gray-brown with no or incomplete capsules, and visible bleeding was observed in some cases. The pathologic cell type of the tumor was spindle-shaped, clear, plasmacytoid, or epithelioid cells or a mixture of cell types. Nonenhanced CT imaging showed an irregular lobulated or multinodular lesion with ill-defined margins and inhomogeneous attenuation and punctate calcification in a minority of the lesions. The enhanced CT imaging results revealed moderate and intense inhomogeneous enhancement, including cystic and slit-like regions with no enhancement, intense nodular enhancement, and small tortuous vessels in the arterial phase. CONCLUSION: The pathologic features and CT imaging findings of myoepithelial carcinoma were characteristic, and familiarity with these imaging findings might aid in the diagnosis of this entity.
