RESUMO

The low expression rate of exogenous genes in cyanobacteria is one of the bottlenecks of cyanobacteria genetic engineering. The T7 RNA polymerase expression system has achieved the efficient expression of exogenous genes in Escherichia coli. Cyanobacteria and E. coli are both Gram-negative bacteria with high genetic homology. The construction of T7 RNA polymerase expression system in cyanobacteria may improve the expression of foreign genes. In order to construct the T7 RNA polymerase expression system in Anabaena sp. PCC 7120, methods such as overlapping extension PCR and digestion-ligation technique were used to construct a site-specific integration vector pEASY-T1-F1-TacT7RNAPCmR-F2 and a shuttle expression vector pRL-T7-hG-CSF. The site-specific integration vector is capable of expressing T7 RNA polymerase, and the shuttle expression vector expresses hG-CSF driven by the T7 promoter. Then we introduced the site-specific integration vector into the wild type cyanobacteria by electroporation and transferred the shuttle expression vector into the site-integrated transgenic cyanobacteria by triparental conjugative transfer. In the end, we identified the presence of foreign genes in cyanobacteria by PCR, tested the transcription level of foreign genes in cyanobacteria by RT-PCR, and detected the protein expression of foreign genes in cyanobacteria by Western blotting. The two vectors were successfully constructed, the T7 RNA polymerase gene and hG-CSF gene were transferred into cyanobacteria well, and both genes were also expressed in cyanobacteria. In summary, the T7 RNA polymerase expression system was successfully constructed in cyanobacteria, and the expression rate of hG-CSF gene was doubled than the traditional cyanobacteria expression systems. This expression system will provide a better tool for the application of cyanobacteria genetic engineering and will promote the development of cyanobacteria as a chassis cell in the fields of synthetic biology in the future.

Well-defined novel, linear, biodegradable, amphiphilic thermo-responsive ABA-type triblock copolymers, poly[2-(2-methoxyethoxy) ethyl methacrylate-co-oligo(ethylene glycol) methacrylate]-b-poly(ε-caprolactone)-b-poly[2-(2-methoxyethoxy) ethyl methacrylate-co-oligo(ethylene glycol) methacrylate] [P(MEO2MA-co-OEGMA)-b-PCL-b-P(MEO2MA-co-OEGMA)] (tBPs), were synthesized via a combination of ring-opening polymerization (ROP) of ε-caprolactone (εCL) and reversible addition-fragmentation chain transfer polymerization (RAFT) of MEO2MA and OEGMA comonomers. The chemical structures and compositions of these copolymers were characterized using Fourier transform infrared spectroscopy (FT-IR) and proton nuclear magnetic resonance (¹H NMR). The molecular weights of the copolymers were obtained using gel permeation chromatography (GPC) measurements. Thermo-responsive micelles were obtained by self-assembly of copolymers in aqueous medium. The temperature sensitivity and micelllization behavior of amphiphilic triblock copolymers solutions were studied by transmittance, fluorescence probe, surface tension, dynamic light scattering (DLS) and transmission electron microscopy (TEM). A hydrophobic drug, anethole, was encapsulated in micelles by using the dialysis method. The average particle sizes of drug-loaded micelles were determined by dynamic light scattering measurement. In vitro, the sustained release of the anethole was performed in pH 7.4 phosphate-buffered saline (PBS) at different temperatures. Results showed that the triblock copolymer's micelles were quite effective in the encapsulation and controlled release of anethole. The vial inversion test demonstrated that the triblock copolymers could trigger the sol-gel transition which also depended on the temperature, and its sol-gel transition temperature gradually decreased with increasing concentration. The hydrogel system could also be used as a carrier of hydrophobic drugs in medicine.

Novel temperature- and pH-responsive ABA-type triblock copolymers, P(DEAEMA-co-MEO2MA-co-OEGMA)-b-PEG-b-P(DEAEMA-co-MEO2MA-co-OEGMA), composed of a poly(ethylene glycol) (PEG) middle block and temperature- and pH-sensitive outer blocks, were synthesized by atom transfer radical polymerization (ATRP). The composition and structure of the copolymer were characterized by ¹H NMR and gel permeation chromatography (GPC). The temperature- and pH-sensitivity, micellization, and the sol⁻gel transitions of the triblock copolymers in aqueous solutions were studied using transmittance measurements, surface tension, viscosity, fluorescence probe technique, dynamic light scattering (DLS), zeta-potential measurements, and transmission electron microscopy (TEM). The lower critical solution temperature (LCST) of the triblock copolymer, which contains a small amount of a weak base group, (N,N-diethylamino) ethyl methacrylate (DEAEMA), can be tuned precisely and reversibly by changing the solution pH. When the copolymer concentration was sufficiently high, increasing temperature resulted in the free-flowing solution transformation into a micellar gel. The sol-to-gel transition temperature (Tsol⁻gel) in aqueous solution will continue to decrease as solution concentration increases.

INTRODUCTION: Dengue is an important public health concern in developing countries. As it is increasingly serious in mainland China, its spatiotemporal variations in this region must be further understood. METHODOLOGY: On the basis of the data on dengue cases in 2004-2013 collected from the China Information System for Disease Control and Prevention, examinations of spatiotemporal variations of local, imported, and total dengue cases were conducted to characterize this epidemic at the city scale in China. RESULTS: Local cases in September and October accounted for more than half of the total cases in each year. The cities with more than 50 accumulative local cases were mainly distributed along the southeast coastal areas and southwest border regions of China. In 2004-2013, local dengue transmission (indicated by the number of local cases and the locally infected cities) increased yearly and was closely associated with epidemics (represented by the amount of imported cases and the cities with imported cases). At the city level, local transmission tended to be spatially clustered in the Zhejiang-Fujian coastal area, the Pearl River Delta, and Yunnan-Burma border region, especially in 2005, 2010, 2012, and 2013. CONCLUSIONS: The results showed that China's local dengue transmission is spatially and temporally featured, and that the prevalence of this epidemic is mostly related to imported cases from overseas epidemic areas. This study provides useful support for hygiene authorities of central and local governments to take effective measures to prevent and control this disease.
