RESUMO

Cephalosporins play an indispensable role against bacterial infections. Deacetyloxycephalosporin C synthase (DAOCS), also called expandase, is a key enzyme in cephalosporin biosynthesis that epoxides penicillin to form the hexavalent thiazide ring of cephalosporin. DAOCS in fungus Acremonium chrysogenum was identified as a bifunctional enzyme with both ring expansion and hydroxylation, whereas two separate enzymes in bacteria catalyze these two reactions. In this review, we briefly summarize its source and function, improvement of the conversion rate of penicillin to deacetyloxycephalosporin C through enzyme modification, crystallography features, the prediction of the active site, and application perspective.

Sexual reproduction remains an understudied feature of oomycete biology. To expand our knowledge of this process, we used RNA-seq and quantitative proteomics to examine matings in Phytophthora infestans. Exhibiting significant changes in mRNA abundance in three matings between different A1 and A2 strains compared to nonmating controls were 1170 genes, most being mating-induced. Rising by >10-fold in at least one cross were 455 genes, and 182 in all three crosses. Most genes had elevated expression in a self-fertile strain. Many mating-induced genes were associated with cell wall biosynthesis, which may relate to forming the thick-walled sexual spore (oospore). Several gene families were induced during mating including one encoding histidine, serine, and tyrosine-rich putative wall proteins, and another encoding prolyl hydroxylases which may strengthen the extracellular matrix. The sizes of these families vary >10-fold between Phytophthora species and one exhibits concerted evolution, highlighting two features of genome dynamics within the genus. Proteomic analyses of mature oospores and nonmating hyphae using isobaric tags for quantification identified 835 shared proteins, with 5% showing >2-fold changes in abundance between the tissues. Enriched in oospores were ß-glucanases potentially involved in digesting the oospore wall during germination. Despite being dormant, oospores contained a mostly normal complement of proteins required for core cellular functions. The RNA-seq data generated here and in prior studies were used to identify new housekeeping controls for gene expression studies that are more stable than existing normalization standards. We also observed >2-fold variation in the fraction of polyA+ RNA between life stages, which should be considered when quantifying transcripts and may also be relevant to understanding translational control during development.

An organic solar cell based on a bulk heterojunction of a conjugated polymer and a methanofullerene PC61BM or PC71BM exhibits a complex morphology that controls both its photovoltaic and mechanical compliance (flexibility and stretchability). Here, the donor-acceptor blend of poly(thieno[3,4-b]-thiophene/benzodithiophene) (PTB7) and PC71BM containing a small amount of diiodooctane (DIO) in the spin-casting solution is reported to exhibit elastic deformability. The blend comprises nanometer-size, nanocrystalline grains that are relatively uniformly distributed. Large external deformation is accommodated by relative sliding between the grains. Reorientation of the nanocrystallites and the global reorientation of the PTB7 polymer chain were observed along the stretching direction up to 100% strain, which was reversible as the blend was allowed to relax to 0% strain. The polymer solar cell based on PTB7:PC71BM:DIO with such reversible morphological changes exhibited a rubbery elasticity at room temperature. The device could be stretched up to 100% strain, and the power-conversion efficiency shows a slight increase up to 30% strain and a global increase of power generation as the photoactive area increases with strain. Solar cells were fabricated employing a layer of the PTB7:PC71BM:DIO blend sandwiched between a pair of stretchable transparent electrodes, each comprising a stack of a silver nanowire percolation network and a single-wall carbon nanotube network embedded in the surface of a poly(urethane acylate) elastomer film. The solar cells were semitransparent and could be stretched like a rubbery film by as much as 100% strain. The measured power-conversion efficiency was 3.48%, which was increased to 3.67% after one cycle of stretching to 50% strain and lowered to 2.99% after 100 stretching cycles. The total power generation from the cells was significantly increased, thanks to the expanded active area as the cells were stretched.

BACKGROUND: Many plant pathogen secretory proteins are known to be elicitors or pathogenic factors,which play an important role in the host-pathogen interaction process. Bioinformatics approaches make possible the large scale prediction and analysis of secretory proteins from the Puccinia helianthi transcriptome. The internet-based software SignalP v4.1, TargetP v1.01, Big-PI predictor, TMHMM v2.0 and ProtComp v9.0 were utilized to predict the signal peptides and the signal peptide-dependent secreted proteins among the 35,286 ORFs of the P. helianthi transcriptome. RESULTS: 908 ORFs (accounting for 2.6% of the total proteins) were identified as putative secretory proteins containing signal peptides. The length of the majority of proteins ranged from 51 to 300 amino acids (aa), while the signal peptides were from 18 to 20 aa long. Signal peptidase I (SpI) cleavage sites were found in 463 of these putative secretory signal peptides. 55 proteins contained the lipoprotein signal peptide recognition site of signal peptidase II (SpII). Out of 908 secretory proteins, 581 (63.8%) have functions related to signal recognition and transduction, metabolism, transport and catabolism. Additionally, 143 putative secretory proteins were categorized into 27 functional groups based on Gene Ontology terms, including 14 groups in biological process, seven in cellular component, and six in molecular function. Gene ontology analysis of the secretory proteins revealed an enrichment of hydrolase activity. Pathway associations were established for 82 (9.0%) secretory proteins. A number of cell wall degrading enzymes and three homologous proteins specific to Phytophthora sojae effectors were also identified, which may be involved in the pathogenicity of the sunflower rust pathogen. CONCLUSIONS: This investigation proposes a new approach for identifying elicitors and pathogenic factors. The eventual identification and characterization of 908 extracellularly secreted proteins will advance our understanding of the molecular mechanisms of interactions between sunflower and rust pathogen and will enhance our ability to intervene in disease states.

Highly efficient organic light emitting diodes (OLEDs) based on multiple layers of vapor evaporated small molecules, indium tin oxide transparent electrode, and glass substrate have been extensively investigated and are being commercialized. The light extraction from the exciton radiative decay is limited to less than 30% due to plasmonic quenching on the metallic cathode and the waveguide in the multi-layer sandwich structure. Here we report a flexible nanocomposite electrode comprising single-walled carbon nanotubes and silver nanowires stacked and embedded in the surface of a polymer substrate. Nanoparticles of barium strontium titanate are dispersed within the substrate to enhance light extraction efficiency. Green polymer OLED (PLEDs) fabricated on the nanocomposite electrode exhibit a maximum current efficiency of 118 cd/A at 10,000 cd/m(2) with the calculated external quantum efficiency being 38.9%. The efficiencies of white PLEDs are 46.7 cd/A and 30.5%, respectively. The devices can be bent to 3 mm radius repeatedly without significant loss of electroluminescent performance. The nanocomposite electrode could pave the way to high-efficiency flexible OLEDs with simplified device structure and low fabrication cost.

Transparent conductive electrodes with high surface conductivity, high transmittance in the visible wavelength range, and mechanical compliance are one of the major challenges in the fabrication of stretchable optoelectronic devices. We report the preparation of a transparent conductive electrode (TCE) based on a silver nanowire (AgNW) percolation network modified with graphene oxide (GO). The monatomic thickness, mechanical flexibility, and strong bonding with AgNWs enable the GO sheets to wrap around and solder the AgNW junctions and thus dramatically reduce the inter-nanowire contact resistance without heat treatment or high force pressing. The GO-soldered AgNW network has a figure-of-merit sheet resistance of 14 ohm/sq with 88% transmittance at 550 nm. Its storage stability is improved compared to a conventional high-temperature annealed AgNW network. The GO-soldered AgNW network on polyethylene terephthalate films was processed from solutions using a drawdown machine at room temperature. When bent to 4 mm radius, its sheet resistance was increased by only 2-3% after 12,000 bending cycles. GO solder can also improve the stretchability of the AgNW network. Composite TCE fabricated by inlaying a GO-soldered AgNW network in the surface layer of polyurethane acrylate films is stretchable, by greater than 100% linear strain without losing electrical conductivity. Fully stretchable white polymer light-emitting diodes (PLEDs) were fabricated for the first time, employing the stretchable TCE as both the anode and cathode. The PLED can survive after 100 stretching cycles between 0 and 40% strain and can be stretched up to 130% linear strain at room temperature.

We report the electronic recording of the touch contact and pressure using an active matrix pressure sensor array made of transparent zinc oxide thin-film transistors and tactile feedback display using an array of diaphragm actuators made of an interpenetrating polymer elastomer network. Digital replay, editing and manipulation of the recorded touch events were demonstrated with both spatial and temporal resolutions. Analog reproduction of the force is also shown possible using the polymer actuators, despite of the high driving voltage. The ability to record, store, edit, and replay touch information adds an additional dimension to digital technologies and extends the capabilities of modern information exchange with the potential to revolutionize physical learning, social networking, e-commerce, robotics, gaming, medical and military applications.

A quick recovery: A semitransparent composite conductor comprising a layer of silver nanowire percolation network inlaid in the surface layer of a Diels-Alder-based healable polymer film is fabricated. The composite is flexible and highly conductive, and is capable of both structural and electrical healing via heating. Cut samples that completely lose their conductivity can recover 97% of it within 5 minutes of heating at 110 °C. The cutting and healing can be repeated at the same location for multiple cycles.

Thermoplastics under voltages are used in diverse applications ranging from insulating cables to organic capacitors. Electromechanical instabilities have been proposed as a mechanism that causes electrical breakdown of thermoplastics. However, existing experiments cannot provide direct observations of the instability process, and existing theories for the instabilities generally assume thermoplastics are mechanically unconstrained. Here, we report in situ observations of electromechanical instabilities in various thermoplastics. A theory is formulated for electromechanical instabilities of thermoplastics under different mechanical constraints. We find that the instabilities generally occur in thermoplastics when temperature is above their glass transition temperatures and electric field reaches a critical value. The critical electric field for the instabilities scales with square root of yield stress of the thermoplastic and depends on its Young's modulus and hardening property.

RESUMO

The bistable electroactive polymer is a new smart material capable of large strain, rigid-to-rigid actuation. At the rubbery state of the polymer heated to above its glass transition, stable electrically-induced actuation is obtained at strains as large as 150%. Electromechanical instability can be effectively overcome by the formation of interpenetrating polymer networks. An application as a refreshable braille display is demonstrated.

Stretchable transparent composites have been synthesized consisting of a silver nanowire (AgNW) network embedded in the surface layer of a crosslinked poly(acrylate) matrix. The interpenetrating networks of AgNWs and the crosslinked polymer matrix lead to high surface conductivity, high transparency, and rubbery elasticity. The presence of carboxylic acid groups on the polymer chains enhances the bonding between AgNWs and the polymer matrix, and further increases the stretchability of the composites. The sheet resistance of the composite electrode increases by only 2.3 times at 50% strain. Repeated stretching to 50% strain and relaxation only causes a small increase of the sheet resistance after 600 cycles. The morphology of the composites during reversible stretching and relaxation has been investigated to expound the conductivity changes.

White polymer phosphorescent light-emitting diodes have been fabricated employing silver nanowire (AgNW)-polymer composite substrates. Solution processing is used to form all the layers in the devices, except for the cathode, for potentially low fabrication cost. The current efficiency in the front viewing direction is 20.3 cd A(-1) for devices comprising two complementary dopants (bis[(4,6-difluorophenyl)-pyridinato-N,C(2)] (picolinato)Ir(iii) (FIrpic) with sky blue emission and bis(1-phenylisoquinoline)(acetylacetonate) (Ir(piq)) for red emission). The efficiency is 42.3 cd A(-1) at 4000 cd cm(-2) for devices comprising the three primary colors (FIrpic, Ir(piq), and tris(2-(4-tolyl)pyridinato-N,C(2)) (Ir(mppy)(3)), with green emission). These values are, respectively, 35% and 41% higher than those of control devices fabricated on indium tin oxide (ITO) coated on glass substrates. The enhanced current efficiency is attributed to enhanced light out-coupling thanks to light scattering by the silver nanowires embedded in the transparent composite electrode.

RESUMO

A new compliant electrode-based on silver nanowire-polymer composite has been developed. The composite electrode has low sheet resistance (as low as 10 Ω/sq), remains conductive (10(2) -10(3) Ω/sq) at strains as high as 140%, and can support Joule heating. The combination of the composite and a bistable electroactive polymer produces electrically-induced, large-strain actuation and relaxation, reversibly without the need of mechanical programming.

RESUMO

Shape-memory polymer light-emitting diodes (PLEDs) using a new silver nanowire/polymer electrode are reported. The electrode can be stretched by up to 16% with only a small increase in sheet resistance. Large deformation shape change and recovery of the PLEDs to various bistable curvatures result in minimal loss of electroluminescence performance.
