RESUMO

Subnival glasshouse plants provide a text-book example of high-altitude adaptation with reproductive organs enclosed in specialized semi-translucent bracts, monocarpic reproduction and continuous survival under stress. Here, we present genomic, transcriptomic and metabolomic analyses for one such plant, the Noble rhubarb (Rheum nobile). Comparative genomic analyses show that an expanded number of genes and retained genes from two recent whole-genome duplication events are both relevant to subnival adaptation of this species. Most photosynthesis genes are downregulated within bracts compared to within leaves, and indeed bracts exhibit a sharp reduction in photosynthetic pigments, indicating that the bracts no longer perform photosynthesis. Contrastingly, genes related to flavonol synthesis are upregulated, providing enhanced defense against UV irradiation damage. Additionally, anatomically abnormal mesophyll combined with the downregulation of genes related to mesophyll differentiation in bracts illustrates the innovation and specification of the glass-like bracts. We further detect substantial accumulation of antifreeze proteins (e.g. AFPs, LEAs) and various metabolites (e.g. Proline, Protective sugars, procyanidins) in over-wintering roots. These findings provide new insights into subnival adaptation and the evolution of glasshouse alpine plants.

Rhubarb is the collective name for various perennial plants from the genus Rheum L. and the Polygonaceae family. They are one of the most ancient, commonly used, and important herbs in traditional Chinese medicine. Rhubarb is a major source of anthraquinones, but how they are synthesized remains largely unknown. Here, we generate a genome sequence assembly of one important medicinal rhubarb R. tanguticum at the chromosome level, with 2.76 Gb assembled into 11 chromosomes. The genome is shaped by two recent whole-genome duplication events and recent bursts of retrotransposons. Metabolic analyses show that the major anthraquinones are mainly synthesized in its roots. Transcriptomic analysis reveals a co-expression module with a high correlation to anthraquinone biosynthesis that includes key chalcone synthase genes. One CHS, four CYP450 and two BGL genes involved in secondary metabolism show significantly upregulated expression levels in roots compared with other tissues and clustered in the co-expression module, which implies that they may also act as candidate genes for anthraquinone biosynthesis. This study provides valuable insights into the genetic bases of anthraquinone biosynthesis that will facilitate improved breeding practices and agronomic properties for rhubarb in the future.

BACKGROUND: Reirradiation with stereotactic body radiotherapy (SBRT) for patients with primary or secondary lung malignancies represents an appealing definitive approach, but its feasibility and safety are not well defined. The purpose of this study was to investigate the tumor control probability (TCP) and toxicity for patients receiving reirradiation with SBRT. PATIENTS AND METHODS: Eligible patients with recurrence of primary or secondary lung malignancies from our hospital were subjected to reirradiation with SBRT, and PubMed- and Embase-indexed articles were reviewed. The patient characteristics, pertinent SBRT dosimetric details, local tumor control, and toxicities were extracted. The logistic dose-response models were compared for TCP and overall survival (OS) in terms of the physical dose and three-, four-, and five-fraction equivalent doses. RESULTS: The data of 17 patients from our hospital and 195 patients extracted from 12 articles were summarized. Reirradiation with SBRT yielded 2-year estimates of 80% TCP for doses of 50.10 Gy, 55.85 Gy, and 60.54 Gy in three, four, and five fractions, respectively. The estimated TCP with common fractionation schemes were 50%, 60%, and 70% for 42.04 Gy, 47.44 Gy, and 53.32 Gy in five fractions, respectively. Similarly, the 2-year estimated OS was 50%, 60%, and 70% for 41.62 Gy, 46.88 Gy, and 52.55 Gy in five fractions, respectively. Central tumor localization may be associated with severe toxicity. CONCLUSIONS: Reirradiation with SBRT doses of 50-60 Gy in 3-5 fractions is feasible for appropriately selected patients with recurrence of peripheral primary or secondary lung malignancies, but should be carefully considered for centrally-located tumors due to potentially severe toxicity. Further studies are warranted for optimal dose/fractionation schedules and more accurate selection of patients suitable for reirradiation with SBRT.

Research has demonstrated that some risk and protective factors of gaming disorder (GD) have been identified. However, the interaction of GD with risk and protective factors has rarely been examined from a network analysis perspective. Therefore, the present study examined the relationships between GD, risk factors (i.e., social anxiety and fear of missing out), and protective factors (i.e., resilience, authenticity, and family closeness) utilizing a network analysis approach among a sample of Chinese university students. A cross-sectional study with 1364 participants was performed using the Gaming Disorder Test, the short version of Social Interaction Anxiety Scale (SIAS), the Chinese Trait-State Fear of Missing Out Scale (T-SFoMOSC), the Brief Resilience Scale (BRS), the Authenticity Scale, and a single item assessing family closeness. Social anxiety, FoMO, and two factors of authenticity (i.e., self-alienation and accepting external influence) had positive relationship with GD, while resilience, authentic living, and family closeness had negative relationship with GD. Social anxiety was identified as the core node in the domain-level network. The item "continuation or escalation of gaming" (gd3) and the item "gaming problems" (gd4) constituted the highest edge weight in the facet-level network. The network comparison demonstrated there were no significant differences in relation to network structures and global strengths between gender. The findings suggest bivariate relationships among GD, risk factors (i.e., social anxiety and fear of missing out), and protective factors (i.e., resilience, authenticity, and family closeness). Interventions for GD must be considered through preventing risk factors and increasing protective factors including improving the gaming motivation and maladaptive cognition to decrease negative emotion (e.g., social anxiety and FoMO), enhancing resilience, authentic living and parent-child relationship, as well as decreasing self-alienation and accepting external influence. Background and aims: Studies have shown that gaming disorder (GD) is associated with rumination and poor sleep quality. However, the reciprocal relationship between GD, rumination and sleep quality is unclear. Moreover, the differences between gender and between left-behind experiences in the aforementioned relationship remain unknown. Therefore, the present study examined gender differences and left-behind experiences in the relationship between GD, rumination, and sleep quality among a sample of Chinese university students during the late stage of COVID-19 pandemic using a network analysis approach. Methods: A cross-sectional online survey of 1,872 Chinese university students was conducted comprising demographic information (age, gender, and left-behind experience), gaming experience, gaming frequency, Gaming Disorder Test (GDT), Short Version of Rumination Response Scale (RRS), and Pittsburgh Sleep Quality Index (PSQI). Results: Among Chinese university students, the prevalence of (i) GD was 3.5% and (ii) sleep disturbance was 14%. GD had positive and weak connection with rumination and sleep quality in the domain-level relational network. The network structures and global strengths both showed no significant differences between gender and between left-behind experiences. The nodes gd3 ("continuation or escalation of gaming") and gd4 ("gaming problems") had the strongest edge in the network. Conclusion: The results suggest reciprocal relationships between GD, rumination, and sleep quality. Gender and left-behind experiences did not influence the reciprocal relationship between GD, rumination, and sleep quality during the late stage of COVID-19 pandemic. Using network analysis, the findings provide novel insights that rumination and sleep quality may have interacted with GD among Chinese students during the late stage of COVID-19 pandemic. Reducing or eliminating negative rumination may decrease GD and improve sleep quality. Moreover, good sleep quality contributes to positive rumination which may decrease the risk of GD among Chinese university students.

Salt and drought impair plant osmotic homeostasis and greatly limit plant growth and development. Plants decrease stomatal aperture to reduce water loss and maintain osmotic homeostasis, leading to improved stress tolerance. Herein, we identified the C2 H2 transcription factor gene OSMOTIC STRESS INDUCED C2 H2 1 (OSIC1) from Populus alba var. pyramidalis to be induced by salt, drought, polyethylene glycol 6000 (PEG6000) and abscisic acid (ABA). Overexpression of OSIC1 conferred transgenic poplar more tolerance to high salinity, drought and PEG6000 treatment by reducing stomatal aperture, while its mutant generated by the CRISPR/Cas9 system showed the opposite phenotype. Furthermore, OSIC1 directly up-regulates PalCuAOζ in vitro and in vivo, encoding a copper-containing polyamine oxidase, to enhance H2 O2 accumulation in guard cells and thus modulates stomatal closure when stresses occur. Additionally, ABA-, drought- and salt-induced PalMPK3 phosphorylates OSIC1 to increase its transcriptional activity to PalCuAOζ. This regulation of OSIC1 at the transcriptional and protein levels guarantees rapid stomatal closure when poplar responds to osmotic stress. Our results revealed a novel transcriptional regulatory mechanism of H2 O2 production in guard cells mediated by the OSIC1-PalCuAOζ module. These findings deepen our understanding of how perennial woody plants, like poplar, respond to osmotic stress caused by salt and drought and provide potential targets for breeding.

Background: The risk perception of contracting COVID-19 is an important topic for assessing and predicting COVID-19 infection and health education during the pandemic. However, studies that use latent profiles and network analysis together to measure the risk perception of COVID-19 are rare. Therefore, this study combined latent profile analysis and network analysis to measure risk perception toward COVID-19 among Chinese university students through a cross-sectional and longitudinal study. Methods: A sample of 1,837 Chinese university students (735 males, 40%) completed the cross-sectional study with an eight-item risk perception questionnaire in January 2020, while 334 Chinese university students (111 males, 33.2%) completed the longitudinal study at three time points. Results: A two-class model including a low risk perception class (n = 1,005, 54.7%) and a high risk perception class (n = 832, 45.3%) was selected for the cross-sectional study. Nodes rp6 ("Average people have chances of contracting COVID-19'') and rp7 ("Average people worry about catching COVID-19") had the strongest edge intensity (r = 0.491), while node rp5 ("The COVID-19 outbreak affects the whole country") had the highest strength centrality in the cross-sectional study. The risk perception of contracting COVID-19 decreased continuously at the three time points. Moreover, the network structures and global strengths had no significant differences in the longitudinal study. Conclusions: The risk perception of contracting COVID-19 decreased continually during the COVID-19 pandemic, which indicated the importance of cultural influence and effective government management in China. In addition, university students displayed strong trust and confidence in the government's ability to fight COVID-19. The results indicate that the government should take strong measures to prevent and intervene in various risks and reinforce the public's trust through positive media communications.

Hybrid supercapacitors are considered the next-generation energy storage equipment due to their superior performance. In hybrid supercapacitors, battery electrodes need to have large absolute capacities while displaying high cycling stability. However, enhancing areal capacity via decreasing the size of electrode materials results in reductions in cycling stability. To balance the capacity-stability trade-off, rationally designed proper electrode structures are in urgent need and still of great challenge. Here we report a high-capacity and high cycling stability electrode material by developing a nickel phosphate lamination structure with ultrathin nanosheets as building blocks. The nickel phosphate lamination electrode material exhibits a large specific capacity of 473.9 C g-1 (131.6 mAh g-1, 1053 F g-1) at 2.0 A g-1 and only about 21% capacity loss at 15 A g-1 (375 C g-1, 104.2 mAh g-1, 833.3 F g-1) in 6.0 M KOH. Furthermore, hybrid supercapacitors are constructed with nickel phosphate lamination and activated carbon (AC), possessing high energy density (42.1 Wh kg-1 at 160 W kg-1) as well as long cycle life (almost 100% capacity retention after 1000 cycles and 94% retention after 8000 cycles). The electrochemical performance of the nickel phosphate lamination structure not only is commensurate with the nanostructure or ultrathin materials carefully designed in supercapacitors but also has a longer cycling lifespan than them. The encouraging results show the great potential of this material for energy storage device applications.

The mental health of individuals has become increasingly important during the novel coronavirus-2019 (COVID-19) pandemic. Given the number of healthcare staff that are helping to treat the victims of COVID-19 all over the world, there is a lack of research concerning the mental health of healthcare staff, and of the prior studies carried out, the research has been relatively descriptive and has not used more sophisticated types of analyses (e.g. latent profile analysis [LPA]). The aim of the present study was to investigate profiles of mental health among Chinese healthcare staff during the COVID-19 pandemic. The sample comprised of 456 healthcare staff, and participants completed an online survey including individual information and their working status during the COVID-19 pandemic. The survey included the Chinese Mental Health Scale (MHS-C), Self-Rating Anxiety Scale (SAS), and Self-Rating Depression Scale (SDS). Utilizing the LPA, two profiles of mental health (good mental health and poor mental health) were identified for Chinese healthcare staff during the COVID-19 pandemic. Compared to those with a good mental health profile, those with poor mental health profile had significantly higher scores on SAS and SDS. Female healthcare staff had higher mental health disturbances than males. Taking care and protecting the mental health of healthcare staff is very important in the fight against COVID-19. The need for employers to implement positive and effective measures among mental healthcare staff is likely to help them to cope better with mental health issues and improve mental health, as well as enhance resilience. Healthcare staff with good mental health can dedicate themselves to better nursing practice and nursing education during the COVID-19 epidemic.

Background and Aims: In previous correlational research, the relationship between gaming disorder (GD), compensation motivation, game flow, time spent gaming, and fear of missing out (FoMO) has been examined. However, network analysis has rarely been applied to explore the relationship between GD, self-compensation motivation, game flow, time spent gaming, and FoMO. Therefore, the present study used network analysis to examine the relationship between the aforementioned variables among a sample of gamers. Methods: The present study comprised gamers (N = 1,635) recruited from three Chinese universities, who completed an online survey including the Gaming Disorder Test, Self-Compensation Motivation Questionnaire, Game Flow Questionnaire, and Trait-State Fear of Missing Out Scale, as well as four items related to time spent gaming. Results: Self-compensation motivation, game flow, time spent gaming, and FoMO were all significantly and positively associated with GD. In the domain-level and facet-level networks, weekday gaming hours and weekend gaming hours had the strongest edge intensity. The domain-level, facet-level, and item-level networks analysis also showed that GD was connected with self-compensation motivation, game flow, time spent gaming, and FoMO. The network structure demonstrated a significant difference between males and females (2.33 vs. 2.81, p = 0.001) using the domain-level network comparison test (NCT). Conclusions: The results suggest that GD is closely associated with self-compensation motivation, game flow, time spent gaming, and FoMO. FoMO and gaming motivation (i.e., self-compensation and game flow) may increase time spent gaming and facilitate GD. Therefore, interventions that decrease game immersion and time spent gaming are likely to decrease GD.

Background: The novel coronavirus-2019 (COVID-19) disease has spread quickly throughout China and around the world, endangering human health and life. Individuals' perceptions and attitudes as well as related health education measures may affect disease progression and prognosis during the COVID-19 outbreak. To promote and implement health education, research must focus on the perceptions and attitudes towards COVID-19 among Chinese people. The present study aimed to examine the profiles and predictive factors of the perceptions and attitudes towards COVID-19 in a sample of Chinese people. Methods: A sample of 2,663 Chinese people comprising medical staff and members of the general public completed an online survey on Wenjuanxing. The survey measured demographic variables (e.g., gender, age, education level, and place of residence) and perceptions and attitudes towards COVID-19. Results: Two profiles of perceptions and attitudes towards COVID-19 (positive and negative perceptions and attitudes) were identified in the sample. Place of residence during the COVID-19 pandemic and first response to COVID-19 were found to be independent predictive factors for COVID-19 related perceptions and attitudes. Conclusion: In addition to drug therapy and mental health services, the perceptions and attitudes of Chinese people towards COVID-19 should be considered when promoting health education during the COVID-19 pandemic.

Drought severely limits plant development and growth; accordingly, plants have evolved strategies to prevent water loss and adapt to water deficit conditions. However, experimental cases that corroborate these evolutionary processes are limited. The LACCASEs (LACs) family is involved in various plant development and growth processes. Here, we performed an evolutionary analysis of LACs from Populus euphratica and characterized the functions of LACs in Arabidopsis and poplar. The results showed that in PeuLACs, multiple gene duplications led to apparent functional redundancy as the result of various selective pressures. Among them, PeuLAC2 underwent strong positive selection. Heterologous expression analyses showed that the overexpression of PeuLAC2 alters the xylem structure of plants, including thickening the secondary cell wall (SCW) and increasing the fiber cell length and stem tensile strength. Altogether, these changes improve the water transport capacity of plants. The analysis of the physiological experimental results showed that PeuLAC2-OE lines exhibited a stronger antioxidant response and greater drought tolerance than WT. Three genes screened by transcriptome analysis, NAC025, BG1, and UGT, that are associated with SCW synthesis and drought stress were all upregulated in the PeuLAC2-OE lines, implying that the overexpression of PeuLAC2 thickened the SCW, improved the water transport capacity of the plant, and further enhanced its drought tolerance. Our study highlights that genes that have undergone adaptive evolution may participate in the development of adaptive traits in P. euphratica and that PeuLAC2 could be a candidate gene for molecular genetic breeding in trees.

The role of fear of missing out (FoMO) in addictive behaviors has recently attracted growing attention. In view of negative effects of gaming disorder (GD) among adolescents and emerging adults, research examining the relationship between FoMO and GD is needed, alongside the roles of impulsivity and gender in the relationship between FoMO and GD. This study examined whether impulsivity as a mediator and gender as a moderator impacted on the relationship between FoMO and GD among a sample of Chinese university students. A sample of 1,288 Chinese university students from three universities completed an online survey through the Wenjuanxing platform. The Chinese Trait-State Fear of Missing Out Scale, the Barratt Impulsiveness Scale-Brief, and the Chinese Gaming Disorder Scale were used in this study. The model results indicated that impulsivity partially mediated the relationship between FoMO and GD, and that there was a larger effect size between impulsivity and GD among males. High levels of FoMO among individuals may impact on executive functions leading to more impulsivity, and are associated with GD. Gender may moderate the relationship between impulsivity and GD. This study deepens the understanding of the relationship between FoMO and GD, and provides new perspectives for practitioners to incorporate into health prevention programs to help regulate emotion, control impulsivity, and decrease GD.

Manganese oxides composed of various valence states Mnx+ (x = 2, 3, and 4) have attracted wide attention as promising electrode materials for asymmetric supercapacitor. However, the poor electrical conductivity limited their performance and application. Appropriate regulation content of Mnx+ in mixed-valent manganese oxide can tune the electronic structure and further improve their conductivity and performance. Herein, we prepared manganese oxides with different Mn2+/Mn3+ ratios through an over-reduction (OR) strategy for tuning the internal electron structure of mixed-valent manganese, which could make these material oxides a good platform for researching the structure-property relationships. The Mn2+/Mn3+ ratio of manganese oxide could be precisely tuned from 0.6 to 1.7 by controlling the amount of reducing agent for manipulating the redox processes, where the manganese oxide electrode with the most appropriate Mn2+/Mn3+ ratio, as 1.65 (OR4) exhibits large capacitance (274 F g-1) and the assembling asymmetric supercapacitors by combining OR4 (positive) and the commercial activated carbon (as negative) achieved large 2.0 V voltage window and high energy density of 27.7 Wh kg-1 (power density of 500 W kg-1). The cycle lifespan of the OR4//AC could keep about 92.9% after 10 000-cycle tests owing to the Jahn-Teller distortion of the Mn(III)O6 octahedron, which is more competitive compared to other work. Moreover, a red-light-emitting diode (LED) can easily be lit for 15 min by two all-solid supercapacitor devices in a series.

BACKGROUND AND AIMS: The widespread use of social media on smartphones has lead to the fear of missing out (FoMO) and smartphone addiction among a minority of adolescents and adults. However, few studies have investigated the impact of trait affect on sleep quality via FoMO and smartphone addiction. The present study examined whether FoMO (trait-FoMO and state-FoMO) and smartphone addiction mediated the relationship between positive affect (PA)/negative affect (NA) and sleep quality, and the prevalence of sleep disturbance among Chinese university students. METHODS: The sample comprised 1,164 university students and they completed a survey which included the Chinese Trait-State Fear of Missing Out Scale (T-SFoMOS-C), Mobile Phone Addiction Index (MPAI), International Positive and Negative Affect Scale Short-Form (I-PANAS-SF), and the Pittsburgh Sleep Quality Index (PSQI). RESULTS: The prevalence of sleep disturbance was found to be 15.98% among Chinese university students. The serial multiple mediation effects indicated that PA directly impacted on sleep quality, but the mediation effects of trait-FoMO and state-FoMO were not found. NA impacted on sleep quality via the mediation effects of trait-FoMO/state-FoMO and smartphone addiction. CONCLUSION: Negative affect was positively associated with poor sleep quality, which was partially mediated by FoMO and smartphone addiction among Chinese university students. Individuals with high negative affect were more likely to have high levels of FoMO and were more prone to smartphone addiction as well as experiencing poor sleep quality. These findings provide an evidence base for emotion management, prevention of smartphone addiction, and sleep improvement.

BACKGROUND: Research into the 'fear of missing out' (FoMO) has greatly increased in recent years. In Asia, many university students frequently use social networking sites (SNSs) via their smartphone. There has also been some studies examining problematic social media use, but there are few Chinese studies concerning FoMO. This may be partly due to the lack of standardized measurement tools for assessing FoMO. Therefore, the present study psychometrically validated the Chinese version of the Trait-State Fear of Missing Out Scale (T-SFoMOS-C) and tested its reliability, validity and measurement invariance among Chinese university students. METHODS: A total of 2,017 university students (aged 17 to 25 years) completed an online survey including the Chinese Trait-State Fear of missing Out Scale (T-SFoMOS-C), the Social Network Site Intensity Scale (SNSIS), and the International Positive and Negative Affect Scale short-form (I-PANAS-SF). RESULTS: Item analysis and exploratory factor analysis was carried out on the T-SFoMOS-C. Confirmatory factor analysis (χ2 = 177.49, df = 50, p < .01; TLI = 0.959; CFI = 0.960; SRMR = 0.038; RMSEA = 0.050) and measurement invariance showed that the T-SFoMOS-C for university students had good construct validity among different groups. The internal consistency of the T-SFoMOS-C (0.81), the test-retest reliability (0.81), and the composite reliability of state-FoMO and trait-FoMO (0.76 and 0.80) were also good. The T-SFoMOS-C was significantly correlated with the SNSIS (0.40) and the Negative Affect (NA) (0.26), respectively. CONCLUSIONS: The T-SFoMOS-C is relatively reliable and valid among different groups, supporting its utility among Chinese university students.

BACKGROUND AND AIMS: Research into the fear of missing out (FoMO) has greatly increased in recent years. Given the negative consequences of gaming disorder (GD) among a small minority of individuals, there is an increasing need for research examining the impact of FoMO on GD. However, little is known about the roles of impulsivity and gaming time as mediators in the relationship between FoMO and GD. The present study examined whether impulsivity and gaming time mediated the relationship between FoMO (trait-FoMO and state-FoMO) and GD among Chinese university students, as well as the prevalence of GD. METHODS: A total of 1127 university students completed an online survey including the Chinese Trait-State Fear of Missing Out Scale (T-SFoMOS-C), the Barratt Impulsiveness Scale-Brief (BIS-Brief), gaming time survey, and the Chinese Gaming Disorder Scale (CGDS). RESULTS: The prevalence of GD was 6.4% among Chinese university students. Trait-FoMO was found to indirectly impact GD via impulsivity and gamine time, whereas the direct effect of trait-FoMO on GD and the mediation effects of gaming time were not confirmed. State-FoMO impacted on GD both directly, and indirectly via the mediation effects of impulsivity as well as impulsivity and gaming time. CONCLUSION: Trait-FoMO on GD was fully mediated via impulsivity and gaming time, whereas state-FoMO on GD was partly mediated via impulsivity and gaming time. Individuals with high levels of FoMO were more likely to show impulsivity and spend a longer time gaming, and these factors were associated with GD. These findings provide insights to incorporate into health prevention programs to help regulate emotion, control impulsivity, and decrease GD.

Poplars are worldwidely cultivated with ecologically and economically important value. Populus alba var. pyramidalis (= P. bolleana) is a main tree of the farmland shelter-belt system in the arid region of Northwest China due to its rapid growth, erect stems, and high biomass production. However, the full-length messenger RNA (mRNA) sequences and complete structure of P. alba var. pyramidalis remain unclear. In this study, using single-molecular real-time (SMRT) and next-generation high-throughput sequencing (NGS) platform, we sequenced transcripts from leaf, root, xylem, and phloem of P. alba var. pyramidalis, to obtain the full-length mRNA transcripts and annotate the complete structure. In total, 86,327 mapped full-length non-chimeric (FLNC) reads were identified, with 705 previously unannotated loci and 3,410 long noncoding RNAs (lncRNAs) and 174 fusion genes found. Alternative spicing (AS) events were detected in 7,536 genes, of which 4,652 genes had multiple AS events. A total of 10,213 alternative polyadenylation (APA) sites were identified, with two or more APA sites observed in 2,212 genes. Our transcriptome data provided the full-length sequences and gene isoforms of transcripts for P. alba var. pyramidalis, which will be helpful in improving our understanding for the genome annotation and gene structures of P. alba var. pyramidalis.

Novel genes provide important genetic resource for organism innovation. However, the evidence from genetic experiment is limited. In plants, γ-aminobutyric acid (GABA) transporters (GATs) primarily transport GABA and further involve in plant growth, development, and response to various stresses. In this study, we have identified the GATs family in Populus species and characterized their functional evolution and divergence in a desert poplar species (Populus euphratica). We found that the GATs underwent genus-specific expansion via multiple whole-genome duplications in Populus species. The purifying selection were identified across those GATs evolution and divergence in poplar diversity, except two paralogous PeuGAT2 and PeuGAT3 from P. euphratica. The both genes arose from a tandem duplication event about 49 million years ago and have experienced strong positive selection, suggesting that the divergence in PeuGAT3 protein function/structure might define gene function better than in expression pattern. Both PeuGAT genes were functionally characterized in Arabidopsis and poplar, respectively. The overexpression of PeuGAT3 increased the thickness of xylem cells walls in both Arabidopsis and poplar and enhanced the lignin content of xylem tissues and the proline accumulation in poplar leaves, all of which may improve tolerance of salt/drought stress in desert poplars. Our findings help clarify the genetic mechanisms underpinning high tolerance in desert poplars and suggest that PeuGAT3 could be an attractive candidate gene for engineering trees with improved brown-rot resistance.

As a major abiotic stress, soil salinity limits seed germination and plant growth, development and production. Seed germination is highly related not only to the seedlings survival rate but also subsequent vegetative growth. Populus euphratica and P. pruinosa are closely related species that show a distinguished adaptability to salinity stress. In this study, we performed an integrative transcriptome analyses of three seed germination phases from P. euphratica and P. pruinosa under salt stress. A two-dimensional data set of this study provides a comprehensive view of the dynamic biochemical processes that underpin seed germination and salt tolerance. Our analysis identified 12831 differentially expressed genes (DEGs) for seed germination processes and 8071 DEGs for salt tolerance in the two species. Furthermore, we identified the expression profiles and main pathways in each growth phase. For seed germination, a large number of DEGs, including those involved in energy production and hormonal regulation pathways, were transiently and specifically induced in the late phase. In the comparison of salt tolerance between the two species, the flavonoid and brassinosteroid pathways were significantly enriched. More specifically, in the flavonoid pathway, FLS and F3'5'H exhibited significant differential expression. In the brassinosteroid pathway, the expression levels of DWF4, BR6OX2 and ROT3 were notably higher in P. pruinosa than in P. euphratica. Our results describe transcript dynamics and highlight secondary metabolite pathways involved in the response to salt stress during the seed germination of two desert poplars.
