RESUMO

Rhipicephalus evertsi evertsi feeding on hosts inoculated with sheep red blood cells (SRBC) and bovine serum albumin (BSA) suppressed the primary antibody response to the two antigens. In addition, while the ticks paralysed most hosts in the studies, fatality associated with this toxicosis occurred only in rabbits which had received SRBC, either alone or with BSA. Only those hosts inoculated with BSA developed any resistance against the ticks, manifested by a slight reduction of engorged weights and development of anti-tick antibodies. These results suggest that R. e. evertsi infestation induces a degree of reduced host immune responsiveness to heterologous antigens.

RESUMO

The secondary antibody response to sheep red blood cells and bovine serum albumin (BSA) was measured in rabbits infested with adult Rhipicephalus evertsi evertsi. The response was reduced (particularly for BSA) but still displayed anamnestic characteristics. Resistance against ixodid ticks associated with antibodies detected by gel diffusion and enzyme linked immunosorbent assay techniques early in the primary challenge was acquired by the immunized hosts only. This suggests that a non-specifically activated immune system enables hosts to develop rapid resistance against tick parasitism.

RESUMO

Rabbits treated with goat anti-rabbit thymocyte serum acquired less resistance to Rhipicephalus evertsi evertsi infestation than untreated controls. This inferior resistance was manifested as higher engorged weights of ticks and higher biotic potential, undue persistence of the ticks on the hosts and poor anti-tick antibody and delayed-type hypersensitivity responses. These observations highlight the significance of host T-cells in mediating resistance to ticks.

RESUMO

As many ewes and lambs watered at concrete ponds in well-drained sites acquired fluke infection as those watered at natural ponds in areas suitable for the snail intermediate hosts of Fasciola hepatica. Fasciola transmission, as evidenced by the increasing number of seropositive hosts in an enzyme-linked immunosorbent assay (ELISA), occurred during the heavy rainy period of July-September and declined with the onset of the dry season, being succeeded by the emergence of faecal positive hosts. This indicates the superiority of ELISA over coprological techniques in the early diagnosis and seroepidemiology of this parasitic infection. Host productivity, as reflected in weight gains and haematological parameters, was not significantly affected by the fluke infections.

RESUMO

Anorexia in forty-six (6.1%) out of 753 neonatal lambs was investigated. Thirty-five of these lambs (76.1%) died while eleven (23.9%) recovered after treatment. Coagulated rubber-like milk clots were found in fifteen (42.9%), phytobezoars in nine (25.7%) and trichophytobezoars and coarse straws in eleven (31.4%) of the dead lambs.

Host resistance, accompanied by demonstrable anti-tick antibodies, developed in groups of rabbits that were infested repeatedly with different numbers of Rhipicephalus evertsi evertsi larvae. This resistance was associated with a drastic reduction in the number of ticks that attached but not in the ability to feed and moult by immatures already established on the hosts. Furthermore, resistance reduced to below 50% the proportion of nymphs which emerged from the larvae applied to the three host groups. Nymphs weighing 5-9.9 and 15-19.9 mg moulted to give mainly males or females respectively. The proportion of males and females which moulted from the remaining weight categories was variable. Anti-tick antibodies were detected by enzyme-linked immunosorbent assay as early as 7 days after primary infestation in all hosts. The titres plateaued after the second challenge and declined drastically during the fifth infestation. No appreciable differences were observed in the antibody responses stimulated by different challenge regimens.

Two groups of rabbits infested repeatedly with Rhipicephalus evertsi evertsi and R appendiculatus acquired resistance as evidenced by reduced ectoparasite engorged weights and development of anti-tick antibodies in the hosts. The enzyme-linked immunosorbent assay (ELISA) using salivary gland antigens of R appendiculatus was able to detect antibody responses to both the homologous and heterologous tick challenges. It was concluded that the immune response in hosts infested by these two tick species stimulates the production of antibodies which show cross reactions.

Rabbits infested four times in succession with adult Rhipicephalus appendiculatus developed anti-tick antibodies, demonstrated by the enzyme-linked immunosorbent assay, following primary infestation and increased by subsequent infestations. Maximum antibody activity was detected after the third infestation while lower ixodid engorged weights occurred from the second infestation onwards. The antibody activity stimulated by the fourth application of ticks was slightly less than that of the third infestation. A slight reduction in antibody activity occurred in the hosts during a tick-free period of 24 days after the third challenge.

Twenty-nine rabbits were exposed to 4 infestations with 20, 50, and 100 adult red-legged ticks (Rhipicephalus evertsi evertsi). Upon challenge, 16 rabbits (55%) contracted paralysis, and 14 of these (88%) died. Most deaths occurred in rabbits challenged with 100 ticks. Immunity to paralysis and subsequent resistance to tick feeding developed in the hosts that survived the initial challenge. Antibody titers to a salivary gland antigen increased progressively as determined by the following tests: passive hemagglutination, agar gel double immunodiffusion, and enzyme-linked immunosorbent assay. Rabbits challenged with 20 ticks displayed declining titers with advanced frequency of challenge and few of the rabbits in this group developed paralysis. There was no decline in antibody titers in groups of rabbits exposed to 50- and 100-tick loads. It is suggested that a certain quantum of antigen injected by the ticks is necessary for induction of immunity to paralysis.

In a series of field trials 48 (55%) of 88 field cases of East Coast fever treated with halofuginone lactate recovered and survived and 40 (45%) died while 31 (86%) of 36 untreated control animals died of East Coast fever and five (14%) recovered. For cases diagnosed and treated early a 100% recovery rate was achieved. A single dose at 1.2 mg/kg body weight per os was adequate. Recovery rate was only 36% for cases diagnosed and treated at an advanced stage. Such cases required two doses at 1.2 mg/kg or a combination of 1.2 and 2.4 mg/kg body weight given at about two days interval. The temperature dropped to normal levels within 48 h of treatment and schizonts started degenerating as early as 24 h after treatment and had disappeared by 48 to 72 h. Piroplasms were not affected by the drug. Recovered animals remained free of East Coast fever for periods up to 12 months of observation despite continuous tick challenge. It was concluded that, with early detection and treatment augmented with proper tick control, outbreaks of East Coast fever can be effectively controlled with halofuginone lactate.
