BACKGROUND: Some observations and reports show that people with high consumption of Solanum aethiopicum (African garden egg) have relief in arthritic pains and swelling. We aimed at assessing the effect of methanol extract of Solanum aethiopicum in experimentally induced inflammation using leukocyte mobilization and vascular permeability tests in rats and human red blood cell (HRBC) membrane stabilization as studies. METHODS: Twenty five (25) adult Wistar rats of either sex (120 g - 200 g) divided into five groups of five rats each were used for each of the animal models. Groups 2, 3 and 4 were administered varied doses of the extract (100, 200 and 400 mg/kg), while groups 1 (vehicle control) and 5 (treatment control) received normal saline and indomethacin (50 mg/kg) respectively. Vascular permeability was induced by the intra-peritoneal injection of 1 ml of acetic acid and monitored using 0.5 ml intravenous injection of 1% Evans blue solution. Leukocyte mobilization was induced by the intra-peritoneal injection of 0.5 ml of 3% agar suspension in normal saline. Heat and hypotonicity induced heamolysis of HRBC membrane was used to assess membrane stabilization. RESULTS: The methanol extracts of garden egg significantly and dose dependently reduced (p≤0.05) the acetic acid induced vascular permeability and agar induced leukocyte mobilization in rats. The percentage inhibitions of induced vascular permeability were 21 ± 3.39, 25 ±1.92 and 60 ± 3.81 for the 100, 200 and 400 mg/kg of the extract while the inhibitions of the agar induced leucocyte migration were 23 ± 2.17, 26 ± 1.58 and 32 ± 1.58 for the 100, 200 and 400 mg/kg of the extract respectively. The extract also, at doses of 100, 200, 400, 600 and 800 µg/ml significantly inhibited heat induced lysis of the human red cell membrane with values of 66.46 ± 2.89, 65.14 ± 4.58, 46.53 ± 2.52, 61.88 ± 4.51and 86.67 ± 3.06 respectively. CONCLUSIONS: These results show that methanol extract of Solanum aethiopicum has anti-inflammatory properties and can reduce inflammatory injury and tissue damage.

OBJECTIVE: To evaluate the possible anti-inflammatory effect of garden egg [Solanum aethiopicum (S. aethiopicum)] using experimentally induced inflammatory models in rats. METHODS: Oedema was induced on the rat hind paw by the injection of 0.1 mL undiluted fresh egg albumin (philogistic agent) into the subplantar surface of the rat paw. Tissue granuloma was induced in the rats by the implantation of two autoclaved cotton pellets (30 mg) under the flank of previously shaved back of anaesthetised rats. Garden egg extract doses were administered to the rats for seven consecutive days. On day 8, the animals were killed and the pellets surrounded by granuloma tissue were dissected out and dried. RESULTS: Extracts of garden egg significantly (P<0.05) reduced the fresh egg albumin-induced rat paw oedema and also significantly (P<0.05) reduced the granuloma tissue formation in the treated groups when compared to the control. CONCLUSIONS: This study shows that the fruits of garden egg (S. aethiopicum) have anti-inflammatory activity in the models studied.

To determine the impact of socioeconomic status on plasma trace element status and pregnancy outcomes, 349 pregnant women, aged 15-40 years (mean 27.04 +/- 2.75 years), recruited at < or = 25 weeks (mean 21.76 +/- 3.12 weeks) gestational age, were followed up till delivery during which maternal and foetal outcomes were recorded. Plasma copper, iron, and zinc were determined using atomic absorption spectrophotometer while maternal sociodemographic data were obtained using a questionnaire. Except for copper, lower plasma iron and zinc were significantly (p < 0.05) higher in women from socioeconomically-disadvantaged groups. Both adverse maternal health and foetal outcomes also seemed to be more prevalent in socioeconomically-disadvantaged women, although without a definite trend. This study has shown that, in economically-disadvantaged setting of developing countries, maternal socioeconomic status impacts on maternal trace element (copper, iron, and zinc) status and health and foetal outcomes.

We recruited 59 individuals of known HIV serostatus after informed consent however, 44 were serodiscordant heterosexual partners [serodiscordant seronegative (SSN group) and serodiscordant seropositive (SSP group)] while 15 were seronegative healthy individuals (SNH). In the case-control study we choose to determine Malondialdehyde (MDA) concentration as a marker of lipid peroxidation index (oxidative stress) spectrophotometrically and quantify HIV mRNA by Real Time-nucleic acid sequence based amplification assay (RT-NASBA). Here our result show for the first time a high concentration of lipid peroxidation product (MDA, 116.6%) with a significant (P < 0.05) increase in HIV serodiscordant seropositive subjects over their seronegative partners. However, Spearman rank correlation statistics of SSP group showed a positive correlation value (P < 0.01, r = 0.89) between MDA and mRNA and a negative correlation between MDA and T-cell ratio (P < 0.01, r = 0.96).The study may strongly indicate a possible lipid peroxidation product threshold for predicting HIV infection and progression in serodiscordant heterosexual partners.

Antioxidants significantly inhibit oxidative processes. The study seeks to determine the activity of endogenous antioxidants and CD4+ T-cell expression in HIV-serodiscordant-heterosexual partners. The case-control study had the following groups; A- (13 serodiscordant-seronegative subjects), B- (13 serodiscordant-seropositive subjects) and C/control- (13 healthy volunteers). CD4+ T-cell expression was determined using a FACScan (fluorescent activated cell sorting) flow cytometer. CAT (catalase), superoxide dismutase, glutathione peroxidase (GHPX) and glutathione S-transferase (GST) activities were assayed using spectrophotometer. The activities of SOD, GHPX, GST and CAT were significantly (P < 0.05) increased by 164.7% (0.090 +/- 0.032), 126% (662 +/- 96), 355.2% (22.023 +/- 1.4) and 119.1% (2.76 +/- 0.10), respectively, in group A when compared with B. The mean CD4+ T-cell (1348 +/- 142) showed a significant (P < 0.05) increase by 237% when compared with group B (400 +/- 182). Conversely, group B revealed a significant (P < 0.05) decrease in activity by 86.5% (CAT), 76.5% (SOD), 106.8% (GHPX) and 81.8% (GST) when compared with C. CD4+ T-cells in groups A and C (1390 +/- 190) did not show any significant decrease (3.11%). The antioxidant activity showed a positive correlation (P < 0.01, r = 0.89) with their respective CD4+ T-cells in groups A and C. Group B showed same positive correlation (P < 0.01, r = 0.76). These results show that high activity of endogenous antioxidants may have a protective role on CD4+ T-cells, which limits HIV infection.
