RESUMO

The aim of this study was to examine the effects of Flavobacterium psychrophilum, a pathogen that is economically important in the aquaculture sector, on the neuroendocrine response of Oncorhynchus mykiss during a time course experiment with sampling at 0.5, 1, 2, 6, 10, and 30 days post injection (dpi). In the brain, serotonin (5HT) content increased in the infected group at all the measured time points, a similar pattern was observed for 5-hydroxyindole-3-acetic acid (5HIAA). Infected fish presented an increase in brain dopamine levels on day 0.5 and 1 dpi. A non-significant variation in noradrenaline levels was observed on all treatment days. Foregut 5-HT and 5-HIAA content in the infected group presented the highest 5-HT concentrations with 248.6 and 983.5 ng/g tissue at 0.5 dpi respectively. Midgut 5-HT and 5-HIAA levels presented the highest 5-HT concentrations, 486.9 ng/g tissue and 1006.4 ng/g tissue respectively, at the beginning of the experiment (0.5 dpi). 5-HT levels in the hindgut presented the highest concentrations with 233.9 ng/g tissue at 0.5 dpi, while 5-HIAA presented the highest concentrations, 690.5 ng/g tissue, at the same time point. After injection with F. psychrophilum the neuroendocrine response in rainbow trout was tissue dependent. Brain levels of 5HT and 5HIIA indicate that the neuroendocrine response increased together with dopamine following intramuscular infection. These increases are in line with reports from other authors, indicating an early response of catecholamines as neurotransmitters to stressful stimulus. In addition the intestinal response was also increased, implying that there could be a possible relationship between the serotonergic system at the intestinal level and the immune system.

A técnica de sacos de náilon suspensos no rúmen foi utilizada para estudar a degradabilidade da proteína bruta e do desdobramento da mimosina da Leucaena leucocephala em búfalos submetidos a um experimento em quadrado latino (4 x 4) com diferentes níveis de proteína bruta na raçäo (8,7; 10,5; 12,5 e 14,9 por cento na MS). As raçöes näo promoveram diferenças significativas (P>0,01) na degradabilidade ruminal da proteína bruta da leucaena, que variou entre 30,92 por cento a 71,70 por cento em 3 e 48 horas de suspensäo no rúmen, respectivamente. Houve uma lenta degradaçäo ruminal da proteína e um acentuado desdobramento da mimosina nas amostras de leucaena colocadas dentro dos sacos de náilon no rúmen de búfalos

Some parameters (glycolysis, respiration, levels of glycolytic enzymes) of the lymphoid cells from the Sticker's lymphosarcoma were established in order to better define the biochemical behavior of the venereal tumor of the dog. For comparative purposes lymphocytes from peripheral blood of normal and tumor-bearing dogs were also studied. Lactic acid produced by the tumor cells during aerobic glycolysis is liberated in the reaction medium. Oxygen uptake is enhanced in the presence of succinate, but not with pyruvate, alpha-ketoglutarate, or malate as substrates. Higher levels of some of the enzymes from the glycolytic pathways as well as differences on the physicochemical and kinetic properties of the glycolytic regulatory enzymes are found in Sticker's tumor cells, when compared with the lymphocytes from peripheral blood of normal and tumor-bearing dogs. A fructose-bisphosphate positively modulated pyruvatekinase is found in the tumor cells.

Kinetic studies were carried out on the reaction catalyzed by pyruvate kinase (ATP:pyruvate phosphotransferase, E.C. 2.7.1.40) purified from white striated (epaxial) muscle of two marine fish Mugil lisa (Brazilian mullet) and Chaetoditerus faber (harvest fish). This included the establishment of kinetic parameters. Attention was given to the effect of fructose 1,6-bisphosphate (Fru-P2) on PK activity. Effects of ATP, alanine and the divalent ions, Mg2+, Mn2+, Cu2+, Be2+ and Co2+, on the fish muscle enzyme were also studied.

RESUMO

A comparative study on the glycolytic enzymes from chicken erythrocytes and somatic tissues has been carried out, the results being shown as active units per mg protein in supernatants of 1085, 12,100 and 106,000 g fractionated centrifugation. The profiles of the glycolytic enzymes have been analyzed in terms of their activity relative to hexokinase and as the ratios between pairs of enzymes bearing a product-substrate relationship. Chicken erythrocyte displays a very peculiar profile of glycolytic enzymes. It possesses a FruP2-activated pyruvate kinase of the L isoenzyme type, which does not seem to be the predominant isoenzyme together with the M type, the content in glycolytic enzymes being much lower than in the somatic tissues.
