RESUMO

Lichens play crucial roles in the ecosystems, contributing to soil formation and nutrient cycling, and being used in biomonitoring efforts to assess the sustainability of ecosystems including air quality. Previous studies on heavy metal accumulation in lichens have mostly relied on manipulated environments, such as transplanted lichens, leaving us with a dearth of research on how lichens physiologically respond to heavy metal exposure in their natural habitats. To fill this knowledge gap, we investigated lichens from two of South Korea's geographically distant regions, Gangwon Province and Jeju Island, and examined whether difference in ambient heavy metal concentrations could be detected through physiological variables, including chlorophyll damage, lipid oxidation, and protein content. The physiological variables of lichens in response to heavy metals differed according to the collection area: Arsenic exerted a significant impact on chlorophyll degradation and protein content. The degree of fatty acid oxidation in lichens was associated with increased Cu concentrations. Our research highlights the value of lichens as a bioindicator, as we found that even small variations in ambient heavy metal concentrations can be detected in natural lichens. Furthermore, our study sheds light on which physiology variables that can be used as indicators of specific heavy metals, underscoring the potential of lichens for future ecology studies.

Lecanora is one of the largest genera of lichens worldwide. These lichens can be easily seen, and are commonly found on trees and rocks. Most Korean Lecanora species belong to the Lecanora subfusca group, which has well-defined superficial thallus, red-brown apothecia, and soredia. The new species of L. neobarkmaniana grows on rocks, farinose soredia coalescing, usually covering the whole thallus, and containing atranorin and zeorin. We used internal transcribed spacer (ITS) and mitochondrial small subunit (mtSSU) sequence data to identify the phylogenetic relationship across Lecanora sequence data and found the species to form different clades. In this study, we reported some interesting findings and described the genetic relationship with other sorediate Lecanora species and the characteristics of the new species. An identification key for the Korean sorediate Lecanora species is given.

Acarosporaceae is a crustose lichen and is known as a species that has more than 50 multi-spores, and has hyaline spores. Those taxa are often found in rock and soil in mountain areas or coastal regions in Korea, and very diverse forms and species are known. However, after an overall genetic phylogenetic analysis of carbonized ascomata in 2015, species consisting only of the morphological base are newly divided, and several species of Acarosporaceae in Korea are also being discovered in this situation. As a result of analysis using internal transcribed spacer (ITS) and nuLSU gene analysis, Korean species belonged to Acarospora and Sarcogyne clade, and Acarospora classified as the Acarospora clade was mixed with the Polysporina group and the Sarcogyne clade is mixed with the Acarospora. We identified two new species (Acarospora beangnokdamensis J. S. Park & S. O. Oh, sp. nov., Sarcogyne jejuensis J. S. Park & S. O. Oh, sp. nov.) through morphological, molecular, and secondary metabolite substance and found one new record (Sarcogyne oceanica K. Knudsen & Kocourk). We have made a classification key for Acarospora and Sarcogyne in Korea and reported all information together here.

The new genus and species, Jejulea byssolomoides, is described from Jeju Island, Korea. This lichen is characterized by saxicolous, crustose, pale greenish-gray, partly finely filamentous, matt, smooth thallus, prominent convex brown to dark brown ascomata with a concolorous margin constricted at the dark brown base, 300-800 µm diameter, 200-250 µm high, without a distinct proper margin, adhering to the substratum ending in a minute byssoid white external part of cylindrical cells, fusiform 3-5 septate ascospores (17-23 × 4-5 µm). Phylogenetic analyses using ITS and mtSSU sequences place Jejulea in the Pilocarpaceae (Lecanorales). The new taxon is closely related to Byssoloma, a cosmopolitan group of foliicolous lichens, which is most diverse in the tropics. Like Byssoloma, Jejulea also forms a byssoid apothecial margin.

Two new species, Thelopsis ullungdoensis and Phylloblastia gyeongsangbukensis are described from Ullung-Do (Island), South Korea. The closest relatives from Europe and Korea are epiphytic Thelopsis flaveola which differs by their immersed or semi-immersed yellow ascomata, ascospores without halo and their habitat of smooth bark (mainly Fagus) in humid and cold climates. Thelopsis gangwondoensis differs by its bigger semi-immersed ascomata (600-700 µm in diam.), oblong halonate ascospores (8-12 × 6-8 µm) and its habitat of smooth bark of deciduous trees. P. gyeongsangbukensis differs from its relatives within the genus in having 5-septate ascospores (22-26 × 6-8 µm), semi-immersed, subglobose ascomata with a flattened top (250-400 µm) and a thin, matt, uneven gray-brown to gray-green continuous thallus without isidia.

An algific talus slope is composed of broken rocks with vents connected to an ice cave, releasing cool air in summer and relatively warmer air in winter to maintain a more stable microclimate all year round. Such geological features create a very unusual and delicate ecosystem. Although there are around 25 major algific talus slopes in Korea, lichen ecology of these areas had not been investigated to date. In this study, we report the first exploration of lichen diversity and ecology at an algific talus slope, Jangyeol-ri, in Korea. A total of 37 specimens were collected over 2017-2018. Morphological and sequencing analysis revealed 27 species belonging to 18 genera present in the area. Of particular interest among these species was Solorina saccata, as it has previously not been reported in Korea and most members of genus Solorina are known to inhabit alpine regions of the Northern Hemisphere. We provide here a taxonomic key for S. saccata alongside molecular phylogenetic analyses and prediction of potential habitats in South Korea. Furthermore, regions in South Korea potentially suitable for Solorina spp. were predicted based on climatic features of known habitats around the globe. Our results showed that the suitable areas are mostly at high altitudes in mountainous areas where the annual temperature range does not exceed 26.6 °C. Further survey of other environmental conditions determining the suitability of Solorina spp. should lead to a more precise prediction of suitable habitats and trace the origin of Solorina spp. in Korea.

Arthonia dokdoensis sp. nov., a lichenicolous fungus from the subcosmopolitan Arthonia molendoi complex growing on crustose thalli of species of the genus Orientophila (subfamily Xanthorioideae, Teloschistaceae), as well as the lichen species Rufoplaca toktoana sp. nov. (subfamily Caloplacoideae, Teloschistaceae) similar to Rufoplaca kaernefeltiana, both from Dokdo Islands, Republic of Korea, are described, illustrated, and compared with closely related taxa. In the phylogenetic tree of the Arthoniaceae based on 12S mtSSU and RPB2 gene sequences, the phylogenetic position of the A. dokdoensis and the relationship with the A. molendoi group are illustrated, while the position of the newly described R. toktoana is confirmed by phylogenetic tree based on ITS nrDNA data.

RESUMO

The genus Chrysothrix is very common around the world and easy to be recognized by its bright yellowish granular thallus. In this study, investigations of lichen mycota in South Korea and Chile from 2010 to 2017 have been done, and some Chrysothrix specimens were collected, based on the morphological and chemical study on these specimens, five species were confirmed in this study, including one new species from Chile, Chrysothrix chilensis D. Liu & J.-S Hur, and one new record from South Korea, C. xanthina (Vain.) Kalb. Meanwhile, detail description and illustration for each species were present in this study.

Lichen genus Stereocaulon (Schreb.) Hoffm is distributed throughout the world. Although 15 Stereocaulon species have been recorded in Korea, no detailed taxonomic or revisionary research has been conducted for nearly two decades. In this study, we collected 260 putative Stereocaulon spp. samples and identified the species based on morphological, chemical, and molecular characteristics. From the collected samples, 10 species of Stereocaulon were identified, nine of which had already been reported, although this was the first report for the tenth, S. octomerellum Hue, in Korea. General characteristics of Stereocaulon spp. include coralloid phyllocladia and tubercular cephalodia; however, the specimen first collected in Korea was a rare species with tomentum on the pseudopodetia. The specimen of S. octomerellum is characterized by the presence of a primary thallus, granule to short coralloid phyllocladia, and pseudopodetia up to 1 cm in size, with tubercular cephalodia. To determine the phylogeny of the specimens, we compared the ITS sequences of ribosomal DNA and the ß-tubulin gene sequences. Phylogenetic analysis showed that the Korean Stereocaulon species were monophyletic and placed in the previous phylogenetic classification. Species of S. intermedium and S. exutum, however, were polyphyletic, and are morphologically variable and widespread species. Overall, we present here detailed morphological and chemical descriptions of each species identified and a revised key of all known Stereocaulon species in South Korea.

The crustose lichen genus Pertusaria comprises over ca. 800 species worldwide. In total, 20 Pertusaria species were localized to the Mt. Sorak and Jeju-do in Korea. To date, information regarding the distribution of Pertusaria species in the South Korean peninsula is scarce. In this study, we collected Pertusaria species across South Korea and identified them based on morphological, chemical, and molecular characteristics. Of the 387 samples collected, we identified 24 taxa and 1 variety, of which 17 were previously recorded, and 6 taxa were newly found in South Korea (P. leioplaca, P. leucosora var. violascens, P. texana, P. thiospoda, P. thwaitesii, and P. xanthodes), 2 known species were transferred to Varicellaria (Varicellaria lactea and V. velata), one species was transferred to Variolaria as a new record (Variolaria multipunctoides) and one was a new species (P. jogyeensis J. S. Park & J.-S. Hur, sp. nov.). Characteristics of the newly discovered species, P. jogyeensis, are as follows: smooth to bumpy thallus, scattered to crowded poriform apothecia, blackish ostioles, definitely sunken, thin yellowish green rims around ostioles, 8-spored ascus, and the presence of perlatolic acid and thiophaninic acid (chlorinated xanthone). Phylogenetic studies on P. jogyeensis based on the mitochondrial small subunit sequence revealed proximity to P. flavicans and P. texana, and supported its classification as a new species within the genus Pertusaria. Additionally, we describe the chemical composition and morphology of all listed species in detail and provide an artificial key for identification.

A new species and 2 new records of lichen genus Buellia were discovered from Chuja-do Island in Jeju Province during a recent floristic survey: B. chujana X. Y. Wang, S. Y. Kondr., L. Lokös & J.-S. Hur sp. nov., B. halonia (Ach.) Tuck., and B. mamillana (Tuck.) W. A. Weber. The new species is characterized by a brown, areolate thallus, the presence of perlatolic acid, and a saxicolous habitat. Together with previously recorded species, 10 Buellia species were confirmed from Jeju-do Island. Among these species, 3 growing in the exposed rocky area contained xanthone (yellowish lichen thallus, UV + orange), indicating that production of xanthone in this genus might be a defense strategy against the harm of UV light. Although the genus Buellia has been thoroughly studied in Korea before, novel species have been discovered continuously, and large species diversity has been found in this crustose genus, even from a small rocky island. This study indicates that the coastal area harbors a vast number of crustose lichen species, and there is great potential to discover unknown lichens in the coastal rocky area in Korea.

During a Korean mushroom diversity survey from 2011 to 2014, we found one new Xylaria species (X. ripicola sp. nov.) and one Xylaria species that had not been previously observed in Korea (X. tentaculata). To confirm the phylogenetic placement of the new species, we conducted a phylogenetic investigation based on internal transcribed spacer regions of ribosomal DNA sequences. Additionally, the new species, X. ripicola, was subsequently analyzed for RNA polymerase II subunit sequences. We also evaluated the macroscopic and microscopic features of this species. Herein, X. ripicola is described as a new species that was collected from a natural beach habitat and X. tentaculata is formally reported as newly found in Korea.

Three monophyletic branches are strongly supported in a phylogenetic analysis of the Teloschistaceae based on combined data sets of internal transcribed spacer and large subunit nrDNA and 12S small subunit mtDNA sequences. These are described as new monotypic genera: Jasonhuria S. Y. Kondr., L. Lokös et S. -O. Oh, Loekoesia S. Y. Kondr., S. -O. Oh et J. -S. Hur and Olegblumia S. Y. Kondr., L. Lokös et J. -S. Hur. Three new combinations for the type species of these genera are proposed.

Lichens are symbiotic organisms which produce distinct secondary metabolic products. In the present study, we tested the cytotoxic activity of 17 lichen species against several human cancer cells and further investigated the molecular mechanisms underlying their anti-cancer activity. We found that among 17 lichens species, F. cucullata exhibited the most potent cytotoxicity in several human cancer cells. High performance liquid chromatography analysis revealed that the acetone extract of F. cucullata contains usnic acid, salazinic acid, Squamatic acid, Baeomycesic acid, d-protolichesterinic acid, and lichesterinic acid as subcomponents. MTT assay showed that cancer cell lines were more vulnerable to the cytotoxic effects of the extract than non-cancer cell lines. Furthermore, among the identified subcomponents, usnic acid treatment had a similar cytotoxic effect on cancer cell lines but with lower potency than the extract. At a lethal dose, treatment with the extract or with usnic acid greatly increased the apoptotic cell population and specifically activated the apoptotic signaling pathway; however, using sub-lethal doses, extract and usnic acid treatment decreased cancer cell motility and inhibited in vitro and in vivo tumorigenic potentials. In these cells, we observed significantly reduced levels of epithelial-mesenchymal transition (EMT) markers and phosphor-Akt, while phosphor-c-Jun and phosphor-ERK1/2 levels were only marginally affected. Overall, the anti-cancer activity of the extract is more potent than that of usnic acid alone. Taken together, F. cucullata and its subcomponent, usnic acid together with additional component, exert anti-cancer effects on human cancer cells through the induction of apoptosis and the inhibition of EMT.

On the basis of extensive field investigation and a series of herbarium specimen identifications, we present and discuss the descriptions and distribution of 22 species of Ramalina found in the Hengduan Mountains of southwestern China. In this revisionary study, representatives of the Ramalina genus, including R. americana, R. confirmata, R. dendriscoides, R. obtusata, R. pacifica, R. pentecostii, R. peruviana, R. shinanoana, and R. subcomplanata are found for the first time in this area. In addition, R. holstii is reported for the first time China. Finally, a newly described species identified as Ramalina hengduanshanensis S. O. Oh & L. S. Wang is reported. It is characterized as growing from a narrow holdfast, solid, sparsely or richly and irregularly dichotomously branched, palmate and flattened lobes with distinctly dorsiventral appearance, surface rugose to reticulate, surface rugosely cracked, dense chondroid tissue, helmet shaped soralia at the tip. The species grows on rock and tree at the highest elevations in this area. Although very few lichen species belonging to the genus Ramalina have been collected above 4,000 m, this new species is found at this elevation. We present detailed morphological, anatomical, and chemical descriptions of this species along with molecular phylogenetic analysis of the internal transcribed spacer rDNA sequences.

Sticta (Schreber.) Ach. is one of the common lichen genera in tropical and subtropical regions, but not in the Korean Peninsula. For almost two decades, no detailed taxonomic or revisionary study has been done on this genus. This study was based on the specimens deposited in the lichen herbarium at the Korean Lichen Research Institute, and the samples were identified on the basis of recent literature. In this revisionary study, a total of eight species of Sticta, including a newly recorded one are documented. These species include Sticta fuliginosa (Dicks.) Ach., Sticta gracilis (Müll. Arg.) Zahlbr., Sticta limbata (Sm.) Ach., Sticta nylanderiana Zahlbr., Sticta sublimbata (J. Steiner) Swinscow & Krog, Sticta weigelii (Ach.) Vain., Sticta wrightii Tuck., and Sticta yatabeana Müll. Arg. Detailed descriptions of S. nylanderiana, S. sublimbata, S. weigelii, and S. yatabeana with their morphological, anatomical, and chemical characteristics are provided. A key description of all known Sticta species of the Korean Peninsula is also presented.

Graphis upretii is a new lichen species discovered in Vietnam. The species is characterized by a loosely corticate, rough, whitish grey to greyish green thallus, elongate and irregularly branched lirellae with an apically thin complete thalline margin (negrosina morph), laterally carbonized, entire proper exciple, clear hymenium, hyaline, 16~20 transversely locular ascospores, and about 50~95 × 10~15 µm in size. In addition, members of the taxon produce norstictic and stictic acids. Currently, the lichen flora of Vietnam include Arthonia radiata, Brigantiaea tricolor, Coenogonium implexum, Dirina paradoxa, Herpothallon sipmanii, Pertusaria pertusa, and Sarcographina cyclospora.

Lichen Endocarpon pusillum is a highly desiccation-tolerant and dominant species in biological soil crusts in arid and semi-arid regions. We report the draft genome sequence of a lichen-forming fungus, E. pusillum strain KoLRILF000583. The draft genome assembly has a size of 37,173,200 bp with a GC content of 49.71%, consisting of 40 scaffolds.

RESUMO

Umbilicaria muehlenbergii strain KoLRILF000956 is amenable to Agrobacterium tumefaciens-mediated transformation (ATMT), making it the only known genetically tractable lichen-forming fungus to date. We report another advancement in lichen genetics, a draft genome assembly for U. muehlenbergii with a size of 34,812,353 bp and a GC content of 47.12%, consisting of seven scaffolds.

The lichen-forming fungus Cladonia metacorallifera strain KoLRI002260 is capable of producing a number of secondary metabolites, including usnic, didymic, and squamatic acids, which have antitumor, antioxidant, and antibiotic activities. The draft genome assembly has a size of 36,682,060 bp, with a G+C content of 44.91%, and consists of 30 scaffolds.
