Indoor air pollution is a serious health problem throughout the world. Plants are known to be able to reduce the effect of air pollution and improve indoor air quality (IAQ). The aim of the present study was to compare the effectiveness of four plant species (Tradescantia zebrina hort. ex Bosse, Philodendron scandens K. Koch & Sello, Ficus pumila L. and Chlorophtytum comosum (Thunb.) Jacques) planted in an active living wall (ALW) for capturing particle pollutants. The ALW was introduced in a glass chamber and exposed to large (10-40 µm) and fine (1.2-10 µm) airborne particles containing a fixed concentration of potentially toxic elements (Al, B, Cd, Co, Cr, Cu, Ni and Pb). The surface particle deposition (sPM) was estimated in the leaves from the four species and the potentially toxic element concentration in the particulate matter (PM) was measured in plants, medium culture and in the ALW support system. The distribution of different particle size fractions differed between species. The capacity to trap particles on leaf surfaces was similar among the species (4.7-13 ng cm-2) except when comparing Tradescantia and Chlorophytum with Ficus, being higher in the latter species. Differences in toxic elements accumulation capacity were observed between species depending on the elements considered. The percentage of reduction in indoor pollution using an ALW was in a range of 65-79% being similar between species. Plants were the most important component of the ALW in terms of accumulation of indoor potentially toxic elements. The data presented here could be used to model the effectiveness of ALW systems schemes in improving IAQ.

INTRODUCTION: The microbiological safety and control of the water used in dental practice has a critical importance for avoiding cross-linked infections in the dental office. The aim of this study was to establish coxsackie virus filtration of the water applied to a dental unit. METHODS: A specific water filter-system was used, to verify the viral load in the outgoing water. The statistical analysis was performedusing the Shapiro-Wilk and t-Student test. RESULTS: The outcome of the evaluation of the virologic tests shows an excellent capability of virus filtration that attested 99.9999% in the volume analyzed. A statistical difference was found in the bacterial water contamination parameter before and after filtration. (P = 0.000000). CONCLUSIONS: According to the tests, medical devices applied to a dental unit are able to filter viruses and therefore reduce risk of contamination in the dental office.

In Portugal, many abandoned mines are often close to agricultural areas and might be used for plant food cultivation. Soils in the vicinity of two Mn- and Fe-abandoned mines (Ferragudo and Rosalgar, SW of Portugal) were collected to cultivate two different food species (Brassica rapa subsp. pekinensis (Lour.) Hanelt and Origanum vulgare L.). Chemical characterization of the soil-plant system and potential risk of adverse effects for human health posed by plants associated with soil contamination, based on the estimation of hazard quotient (HQ), were assessed in a microcosm assay under greenhouse conditions. In both soils, the average total concentrations of Fe and Mn were above the normal values for soils in the region and their concentration in shoots of both species was very high. Brassica rapa subsp. pekinensis grew better in Ferragudo than in Rosalgar soils, and it behaved as an excluder of Cu, Mn, Fe, S and Zn in both soils. The HQ for Cu, Fe, Mn and Zn in the studied species grown on both soils was lower than unit indicating that its consumption is safe. The high Mn tolerance found in both species might be due in part to the high contents of Fe in the soil available fraction that might contribute to an antagonism effect in the uptake and translocation of Mn. The obtained results emphasize the need of further studies with different food crops before cultivation in the studied soils to assess health risks associated with high metal intake.

Phytostabilization is a green, cost-effective technique for mine rehabilitation and ecological restoration. In this study, the phytostabilization capacity of Erica australis L. and Nerium oleander L. was assessed in the climatic and geochemical context of the Riotinto mining district, southwestern Spain, where both plant species colonize harsh substrates of mine wastes and contaminated river banks. In addition to tolerating extreme acidic conditions (up to pH 3.36 for E. australis), both species were found to grow on substrates very poor in bioavailable nutrients (e.g., N and P) and highly enriched with potentially phytotoxic elements (e.g., Cu, Cd, Pb, S). The selective root absorption of essential elements and the sequestration of potentially toxic elements in the root cortex are the main adaptations that allow the studied species to cope in very limiting edaphic environments. Being capable of a tight elemental homeostatic control and tolerating extreme acidic conditions, E. australis is the best candidate for use in phytostabilization programs, ideally to promote early stages of colonization, improve physical and chemical conditions of substrates and favor the establishing of less tolerant species, such as N. oleander.

De-regulated T-cell activation and functions are pivotal in the orchestration of immune-mediated tissue damage in IBD. We investigated the role of DNAM-1 (co-activating)/TIGIT (co-inhibitory)/ligand axis in the regulation of T-cell functions and its involvement in IBD pathogenesis. We show that DNAM-1 and TIGIT display a peculiar expression pattern on gut mucosa T-cell populations, in a microenvironment where their shared ligands (PVR and Nectin-2) are physiologically present. Moreover, DNAM-1 family receptor/ligand system is perturbed in IBD lesions, in a disease activity-dependent manner. The expression profile of CCR6 and CD103 mucosa addressins suggests that microenvironment-associated factors, rather than skewed recruitment of circulating T-cell populations, play a more relevant role in supporting the establishment of DNAM-1 and TIGIT expression pattern in mucosal T-cell populations, and may explain its alteration in IBD. Although both co-receptors mark functionally competent T cells, DNAM-1 and TIGIT segregate on T cells endowed with different proliferative potential. Moreover, their opposing role in regulating T-cell proliferation exquisitely depends on ligand availability. All together, our data propose a role for DNAM-1 and TIGIT in regulating mucosal T-cell activation and immune homeostasis, and highlight the involvement of an imbalance of this system in IBD.

We examined the possible effects of host body size and throat gland development on the abundance of blood-feeding nycteribiid and streblid flies parasitizing a Malagasy fruit bat, Rousettus madagascariensis G. Grandidier, 1928. Data were collected in the Parc National d'Ankarana in northern Madagascar during four visits: September 2014, 2015 (dry season), and January 2015, 2016 (wet season). Two bat fly species were identified, Eucampsipoda madagascarensis Theodor, 1955 (Nycteribiidae) and Megastrebla wenzeli (Jobling, 1952) (Streblidae). A positive correlation was found between host body size and abundance of E. madagascarensis during the four visits, suggesting that larger hosts have more parasites, and for M. wenzeli, this relationship was identified only during the wet season visits. In male hosts, body size and throat gland development are correlated with variation in E. madagascarensis abundance during the two seasons; this relationship was not found for M. wenzeli. We present some explanations for the observed patterns of bat fly abundance associated with throat gland development: increased vascularization and easier access to bloodmeals, chemical properties of gland secretions acting as attractants or perhaps being consumed, and modification of hair around the gland providing protection from bat grooming.

In newly diagnosed myeloma patients, upfront autologous transplant (ASCT) prolongs progression-free survival 1 (PFS1) compared with chemotherapy plus lenalidomide (CC+R). Salvage ASCT at first relapse may still effectively rescue patients who did not receive upfront ASCT. To evaluate the long-term benefit of upfront ASCT vs CC+R and the impact of salvage ASCT in patients who received upfront CC+R, we conducted a pooled analysis of 2 phase III trials (RV-MM-209 and EMN-441). Primary endpoints were PFS1, progression-free survival 2 (PFS2), overall survival (OS). A total of 268 patients were randomized to 2 courses of melphalan 200 mg/m2 and ASCT (MEL200-ASCT) and 261 to CC+R. Median follow-up was 46 months. MEL200-ASCT significantly improved PFS1 (median: 42 vs 24 months, HR 0.53; P<0.001), PFS2 (4 years: 71 vs 54%, HR 0.53, P<0.001) and OS (4 years: 84 vs 70%, HR 0.51, P<0.001) compared with CC+R. The advantage was noticed in good and bad prognosis patients. Only 53% of patients relapsing from CC+R received ASCT at first relapse. Upfront ASCT significantly reduced the risk of death (HR 0.51; P=0.007) in comparison with salvage ASCT. In conclusion, these data confirm the role of upfront ASCT as the standard approach for all young myeloma patients.

The effect of the addition (0-10%) of two types of sewage sludge composts (composted sewage sludge [CS] and sewage sludge co-composted with olive prune wastes [CSO]) on a polymetallic acidic soil from the Riotinto mining area was evaluated by i) a soil incubation experiment and ii) a greenhouse pot experiment using tomato (Solanum lycopersicum Mill.), ryegrass (Lolium perenne L.) and ahipa (Pachyrhizus ahipa (Wedd.) Parodi). Compost addition improved the soil organic carbon content, increased the pH and the electrical conductivity and enhanced enzyme activities and soil respiration, more for CSO than for CS. Plant growth was generally enhanced after compost addition, but not proportionally to the dose. Foliar concentrations of some hazardous elements (As, Cr, Fe) in tomato growing in non-amended soil were above the thresholds, questioning the adequacy of using this plant species. However, leaf concentrations of essential and potentially toxic elements (Fe, As, Cr and Pb) in tomato and/or ryegrass were reduced after the amendment with both composts, generally more for CSO than for CS. Conversely, foliar concentrations in ahipa, a plant species which is able to grow without the need of compost addition, were safe except for As and were only slightly affected by compost addition. This plant species would be a suitable candidate due to its low requirements and due to the limited element translocation to the leaves. Concerning the composts, amelioration of plant and soil properties was better accomplished when using CSO, a compost of sewage sludge and plant remains, than when using CS, which only contained the sludge.

RESUMO

A grating spectrometer with an electron multiplying charge-coupled device camera is used to diagnose a 50 kV, 5 A, 20 ms hydrogen diagnostic neutral beam. The ion source density is determined from Stark broadened Hß emission and the spectrum of Doppler-shifted Hα emission is used to quantify the fraction of ions at full, half, and one-third beam energy under a variety of operating conditions including fueling gas pressure and arc discharge current. Beam current is optimized at low-density conditions in the ion source while the energy fractions are found to be steady over most operating conditions.

This phase 2 trial evaluated three low-dose intensity subcutaneous bortezomib-based treatments in patients ⩾75 years with newly diagnosed multiple myeloma (MM). Patients received subcutaneous bortezomib plus oral prednisone (VP, N=51) or VP plus cyclophosphamide (VCP, N=51) or VP plus melphalan (VMP, N=50), followed by bortezomib maintenance, and half of the patients were frail. Response rate was 64% with VP, 67% with VCP and 86% with VMP, and very good partial response rate or better was 26%, 28.5% and 49%, respectively. Median progression-free survival was 14.0, 15.2 and 17.1 months, and 2-year OS was 60%, 70% and 76% in VP, VCP, VMP, respectively. At least one drug-related grade ⩾3 non-hematologic adverse event (AE) occurred in 22% of VP, 37% of VCP and 33% of VMP patients; the discontinuation rate for AEs was 12%, 14% and 20%, and the 6-month rate of toxicity-related deaths was 4%, 4% and 8%, respectively. The most common grade ⩾3 AEs included infections (8-20%), and constitutional (10-14%) and cardiovascular events (4-12%); peripheral neuropathy was limited (4-6%). Bortezomib maintenance was effective and feasible. VP, VCP and VMP regimens demonstrated no substantial difference. Yet, toxicity was higher with VMP, suggesting that a two-drug combination followed by maintenance should be preferred in frail patients.

We previously observed that nicotine, administered to rats (Wistar) during pregnancy and lactation periods, provokes, in the progeny, late morphofunctional alterations in Leydig cell, body weight increase in adulthood (90 days post partum, dpp) as well as seminiferous epithelium injury. Aiming to investigate whether the spermatogenic damage previously observed in adult progenies from pregnant and lactating nicotine-exposed rat dams are maintained or whether it is worsened in older rats, we analyzed the morphological testicular alterations after up to two complete periods of spermatogenesis (53 days each), spermatic parameters, and sperm DNA fragmentation. Pregnant and lactating rats were nicotine-exposed (2 mg/kg/day) through an osmotic minipump implanted on the first day of pregnancy and replaced after birth. Absolute Control (no minipump) and Sham Control (minipump without nicotine) groups were established. The offspring were killed at 90, 143, and 196 dpp. Significant alterations in morphometric and stereological testicular parameters, such as concentration of sperm number, daily sperm production, and plasma and intratesticular levels of cholesterol and testosterone were not observed in nicotine-exposed rats. Testicular histopathological analysis showed small intraepithelial vacuolization and an accentuated germ cell desquamation in exposed rats. However, the offspring from nicotine-exposed dams exhibited higher frequency of morphologically abnormal spermatozoa and lower sperm motility in comparison with control groups. In addition, nicotine-exposed groups showed a significant reduction in sperm mitochondrial activity and an increased sperm DNA fragmentation (Comet assay). These results indicate a late reproductive damage in the male progeny caused by maternal nicotine exposure, related to the decrease in sperm quality.

Polymerase chain reaction (PCR)-based minimal residual disease (MRD) analysis is a useful prognostic tool in multiple myeloma (MM), although its long-term impact still needs to be addressed. This report presents the updated results of the GIMEMA-VEL-03-096 trial. Thirty-nine MM patients receiving bortezomib-thalidomide-dexamethasone after autologous transplantation were monitored for MRD by both nested and real-time quantitative-PCR until relapse. Our data confirm the strong impact of MRD on survival: overall survival was 72% at 8 years median follow-up for patients in major MRD response versus 48% for those experiencing MRD persistence (P=0.041). In addition, MRD kinetics resulted predictive for relapse: indeed median remission duration was not reached for patients in major MRD response, 38 months for those experiencing MRD reappearance and 9 months for patients with MRD persistence (P<0.001). Moreover: (1) 26 patients achieving major MRD response (67%) benefit of excellent disease control (median TNT: 42 months); (2) MRD reappearance heralds relapse, with a TNT comparable to that of MRD persistence (9 versus 10 months, P=0.706); (3) the median lag between MRD reappearance and need for salvage treatment is 9 months. These results suggest the usefulness of a long-term MRD monitoring in MM patients and the need for maintenance or pre-emptive treatments ensuring durable responses.

BACKGROUND: Neuroimmune interactions and inflammation have been proposed as factors involved in sensory-motor dysfunction and symptom generation in adult irritable bowel syndrome (IBS) patients. In children with IBS and healthy controls, we measured ileocolonic mast cell infiltration and fecal calprotectin and evaluated the relationships between these parameters and abdominal pain symptoms and stooling pattern. METHODS: Irritable bowel syndrome patients diagnosed according to Pediatric Rome III criteria and healthy controls kept a 2-week pain/stooling diary. Ileocolonic mucosal mast cells (MC) and MC in close proximity to nerve fibers (MC-NF) were identified immunohistochemically and quantified. Fecal calprotectin concentration was measured. KEY RESULTS: 21 IBS patients and 10 controls were enrolled. The MC-NF count was significantly higher in the ileum (p = 0.01), right colon (p = 0.04), and left colon (p < 0.001) of IBS patients compared with controls. No differences in fecal calprotectin concentration were noted. Abdominal pain intensity score correlated with ileal MC count (r(s) = 0.47, p = 0.030) and right colon MC-NF count (r(s) = 0.52, p = 0.015). In addition, children with IBS with >3 abdominal pain episodes/week had greater ileal (p = 0.002) and right colonic (p = 0.01) MC counts and greater ileal (p = 0.05) and right colonic (p = 0.016) MC-NF counts than children with less frequent pain. No relationship was found between MC and MC-NF and fecal calprotectin or stooling pattern. CONCLUSIONS & INFERENCES: Mast cells-nerve fibers counts are increased in the ileocolonic mucosa of children with IBS. Mast cells and MC-NF counts are related to the intensity and frequency of abdominal pain.

This multicenter phase II trial evaluated the safety and efficacy of lenalidomide-prednisone (RP) induction, followed by lenalidomide-melphalan-prednisone (MPR) consolidation and RP maintenance in elderly unfit newly diagnosed myeloma patients. Patients received four 28-day RP induction courses (lenalidomide 25 mg/day on days 1-21 and prednisone 50 mg three times/week), followed by six 28-day MPR consolidation cycles (melphalan 2 mg, prednisone 50 mg three times/week and lenalidomide 10-15 mg/day on days 1-21), and maintenance with lenalidomide (10 mg/day on days 1-21 every 28 days) plus prednisone (25 mg three times/week). Forty-six patients were enrolled. Median age was 75 years, 59% of patients had at least one comorbidity and 35% at least two. Partial response rate was 80%, including 29% very good partial response. Median time to progression was 19.6 months, median progression-free survival was 18.4 months and 2-year overall survival was 80%. At the tolerated consolidation dose (melphalan 25 mg/month and lenalidomide 10 mg/day), the most frequent grade 3 adverse events were neutropenia (36.4%), anemia (12.1%), cutaneous reactions (18.2%) and infections (12.1%). Grade 4 neutropenia occurred in 12.1% of patients. In conclusion, RP induction followed by MPR consolidation and RP maintenance showed a manageable safety profile, and reduced the risk of severe hematological toxicity in unfit elderly myeloma patients.

The present study attempts to investigate the size, composition and seedling dynamics of the seed bank of a metalliferous and vulnerable species, Erica andevalensis. Samples were taken during spring and autumn from two different sites. We also studied the effects of nutrient solution, irrigation from the river Tinto and irrigation deficit on seeding establishment and survival. Only E. andevalensis and Poaceae species emerged from the seed banks, although the former was dominant (98%). Germination and seedling establishment was totally inhibited by the waters of the river Tinto. Seed density was high in the soils of both seed banks irrigated with water and nutrient solution. We found no seasonal differences in the seed bank and number of germinated seeds and mortality rate and density were similar. From the standpoint of restoration management, the results indicate that the seed bank is a very important factor for successful species establishment.

Introduction: A review of current literature failed to find any detailed description on the surgical technique of open intraperitoneal mesh hernia repair. The aim of the present study was to describe the open surgical technique used at our institution to repair incisional hernias with intraperitoneal composite prosthesis, and the short-term outcomes including recurrence, complications and patient satisfaction with the procedure. Patients and Methods: A transversal, observational and descriptive report on open intraperitoneal hernioplasty was designed. Short-term outcomes were assessed at a transversal cut out point at a 12-month interview during the follow-up. The interview included a physical exam and a specific questionnaire inquiring over the patient's satisfaction with the procedure. Results: According to the inguinal ring location, most hernias originated between the xiphoid appendix and the umbilicus (64 percent). Mean surgical time was 2.6 hours. One or more complications presented in 11 patients (19 percent). One year after surgery there was not any recurrence. According to the patient' satisfaction questionnaire final score, 17 patients (29 percent) reported an excellent satisfaction with the procedure, 33 patients (57 percent) reported a very good satisfaction and 8 patients (14 percent) a good satisfaction. Conclusions: Intraperitoneal hernioplasty constitutes a secure option to repair incisional hernias. At short-term follow-up there were no recurrences.


Introducción: No se encuentra en la literatura ninguna descripción detallada de Ia técnica para instalación de prótesis intraperitoneales por vía abierta. El objetivo del presente estudio es describir la técnica que utilizamos en nuestra institución y Ios resultados a corto plazo de estas operaciones: recurrencia, complicaciones y satisfacción del usuario con el procedimiento. Pacientes y Método: El presente es un reporte transversal, observacional y descriptivo de la técnica de Ia hernioplastía intraperitoneal abierta y de los resultados iniciales de esta técnica tomando como punto de corte transversal el control de los 12 meses de seguimiento. El control incluyó una entrevista, examen físico y un cuestionario específico sobre satisfacción con el procedimiento. Resultados: De acuerdo a la localización del anillo herniario, la mayoría de las hernias se originaron entre la cicatriz umbilical y el apéndice xifoides (60,4 por ciento). El tiempo quirúrgico promedio fue 2,6 horas. Se presentaron una o más complicaciones en 11 pacientes (19 por ciento). Un año después de la cirugía no se encontró ninguna recurrencia. De acuerdo a la puntuación final del cuestionario de satisfacción usuaria, 17 pacientes (29 por ciento) reportaron una satisfacción excelente con el procedimiento, 33 pacientes (57 por ciento) una satisfacción muy buena y 8 pacientes (14 por ciento) una satisfacción buena. Conclusiones: La hernioplastía intraperitoneal constituye una opción segura para la reparación de hernias incisionales. En el seguimiento a corto plazo no presenta recurrencias.

The tolerance to high Zn was studied in the metallophyte Erica andevalensis Cabezudo & Rivera grown in nutrient solutions at different Zn concentrations (5, 500, 1,000, 1,500 and 2,000 µM Zn). Plant growth and nutrient uptake were determined. Metabolic changes were assessed by the analysis of peroxidase activity, organic metabolites related to metal chelation (amino acids, organic acids (malate, citrate) or protection (polyamines). While plants tolerated up to 1,500 µM Zn, despite presenting of low growth rates, the concentration of 2,000 µM Zn was toxic producing high mortality rates. Roots accumulated high Zn concentration (11,971 mg/kg) at 1,500 µM external Zn) apparently avoiding metal transfer into shoots. After 30 days of treatment with high Zn (1,000 and 1,500 µM Zn), the leaves accumulated high levels of glutamine. Short-term treatment with 500 µM Zn, significantly increased the concentration of asparagine and glutamine in roots. Citrate concentration was also considerably increased when exposing roots to Zn excess. Metal immobilization in the root system, low interference with the uptake of nutrients and an increased production of putative organic ligands (amino acids, citrate) might have provided the Zn tolerance displayed by Erica andevalensis.

Although revegetation using native flora is a low cost way to stabilize soil and restore the landscape contaminated with metals, little is known regarding the Pb-tolerance of many of these species. For this purpose, we evaluated the tolerance of Erica andevalensis to Pb by growing plants in nutrient solutions with increasing concentrations of Pb (up to 100 microM). Plant growth and different physiological parameters were determined to ascertain tolerance to metal stress. Additionally, an electron microscopy study coupled with EDX-analysis was performed to get clues on the Pb uptake and translocation from roots into stem and leaves. The LOEC (the lowest observed effect concentration) of Pb was 40 microM while the IC50 (inhibition concentration) was 80 microM Pb. Chemical analysis revealed a root > stem > leaf accumulation pattern. There was a severe reduction in fresh biomass and chlorophyll concentration at the highest Pb dose. The SEM-EDX study indicated that Pb was mostly located in root epidermal tissues. The blockage of Pb on the root probably avoided its toxic effects by limiting Pb transport to other tissues.

Mucosal interleukin (IL)-17A-producing T cells contribute to protective antimicrobial responses and to epithelial barrier integrity; their role in celiac disease (CD) is debated. We analyzed the frequency and developmental dynamics of mucosal (intraepithelial lymphocytes (IEL)) and circulating (peripheral blood (PB)) IL-17A (T17) and/or interferon (IFN)-γ-producing (T1, T1/T17) T-cell populations in 86 pediatric controls and 116 age-matched CD patients upon phorbol myristate acetate/ionomycin or CD3/CD28 stimulation. T17 and T1/17 are physiologically present among IEL and PB populations, and their frequency is selectively and significantly reduced in CD IEL. The physiological age-dependent increase of Th17 IEL is also absent in CD, while IFN-γ-producing PB-T cells significantly accumulate with patient's age. Finally, the amplitude of IL-17A+ and IFN-γ+ T-cell pools are significantly correlated in different individuals; this relationship only applies to CD4+ T cells in controls, while it involves also the CD4- counterpart in CD patients. In conclusion, both size and dynamics of mucosa-associated and circulating IL-17A+ T-cell pools are finely regulated in human pediatric subjects, and severely disturbed in CD. The impaired IL-17A+ IEL-T pool may negatively impact on epithelial barrier efficiency, and contribute to CD mucosa damage; the disturbed dynamics of circulating IL-17A+ and IFN-γ+ T-cell pools may be involved in the extraintestinal autoimmune manifestations associated with CD.
