Uncaria tomentosa is a plant native to the Amazon that has immunomodulatory and antitumor properties due to the alkaloids found in the plant, being able to modify the immune response by potentiating or suspending the action of cytokines secreted by macrophages that induce the immune response, either by the classical route (M1) or through the alternative route (M2). Macrophages activated by M1 convert L-arginine into L-citrulline and nitric oxide (NO), whereas macrophages activated by the M2 pathway use the enzymatic activity of arginase to convert the same substrate into L-ornithine and urea. The aim of this work was to evaluate the immunostimulating activity of the crude hydroalcoholic extract from the bark of the U. tomentosa stem in RAW 264.7 macrophages. Concentrations of 0.2, 0.1 and 0.05 mg/mL of U. tomentosa extract associated with LPS, INF-γ and IL-4 inducers were tested by determining NO production and arginase enzyme activity. Nitric oxide production was enhanced by the extract when associated with LPS and LPS + INF-γ inducers. In the activity of the arginase enzyme, the extract decreased the stimulation of IL-4 on the enzyme, mainly at 0.2 mg/mL concentration. Therefore, it is concluded that the crude hydroalcoholic extract of the stem bark of U. tomentosa in RAW 264.7 cells, at a concentration of 0.2 mg/mL, showed considerable pro-inflammatory activity.

OBJECTIVE: The effect of menopause transition in body composition was evaluated in a cross-sectional study. METHOD: The study was carried out in an outpatient clinic of Brazil enrolling premenopausal (n = 64) and postmenopausal (n = 42) women aged between 44 and 52 years, with weight stability (±2 kg) for at least 6 months before evaluation. Participants answered a sociodemographic semi-structured questionnaire, the Menopause Rating Scale, the International Physical Activity Questionnaire, 24-h dietary recall and a visual analogue scale of appetite. Blood biochemical, anthropometry and densitometry measurements were used for body composition estimation. RESULTS: Most participants were overweight (31.4%) or obese (45.7%) and categorized as 'high active' in physical activity (65.7%). Lean mass and bone mass decreased in the first few years of menopause. A metabolic turn to an increase of lipids was observed, represented by greater total cholesterol and high-density lipoprotein cholesterol levels. Menopause transition did not alter body fat distribution. Total body fat, android fat and gynoid fat were positively related to smoking habit, and android fat was also positively related to waist circumference. CONCLUSION: Taken together, early postmenopause can be considered a time window of opportunity for preventing ailments such as atherogenic profile, obesity, increased cardiovascular risk and osteoporosis.

In most cases, endoparasitic mites were reported to parasitize the respiratory system, particularly the anterior nasal chambers, larynx, trachea, lungs, and the air and conjunctival sacs. However, some mites are only carried by hummingbirds, because flowers serving as a nutrient source for these small mites, such as for Apodiformes, a tropical nectar-feeding in their natural habitat. From August 2012 to May 2014, humminbirds were captured using mist nets, and examined for the presence of nasal mites (identified according to specific keys) in the Pantanal, and Cerrado biomes, from the Mato Grosso state, midwestern Brazil. Overall, 76 Apodiform specimens, from the Trochilidae family were captured, and 20 female nasal mites, identified as being from the Rhinoseius genus, were collected from eight hosts (10.5%), of the following species: Amazilia fimbriata, Amazilia versicolor, Eupetonema macroura, and Thalurania furcata. The Rhinoseius spp. has been reported on, in many ecosystems in the Americas, including Brazil, however, this is the first report on Apodiformes from the Pantanal biome. Phoretic mites are not responsible for direct damage to the population of Apodiformes, as they are not pathogenic. However, they compete for food with hummingbirds and decrease the quantity of food available in the environment. They also affect the reproductive dynamics of plants, feed on pollen, and interfere with the intricacies of the biodiversity where they live.


Resumo Os ácaros nasais são comumente encontrados em beija-flores (Apodiformes). Na maioria dos casos, os ácaros foram relatados como endoparasitas, parasitando o sistema respiratório, particularmente as câmaras nasais anteriores, a laringe, a traqueia, os pulmões e os sacos aéreos e conjuntivais. No entanto, alguns ácaros são transportados apenas por beija-flores, pois flores servem como fonte de nutrientes para esses pequenos ácaros, assim como para Apodiformes tropicais alimentados de néctar em seu habitat natural. De agosto de 2012 a maio de 2014, beija-flores foram capturadas com redes de neblina e examinadas quanto à presença de ácaros nasais (identificados de acordo com chaves específicas) nos biomas Pantanal e Cerrado, no estado do Mato Grosso, Centro-Oeste do Brasil. No total, 76 exemplares Apodiformes da família Trochilidae foram capturados e 20 fêmeas de ácaros nasais, identificados como pertencentes ao gênero Rhinoseius, foram coletados de oito hospedeiros (10,5%), das seguintes espécies: Amazilia fimbriata, Amazilia versicolor, Eupetonema macroura e Thalurania furcata. Rhinoseius spp. foi relatado em muitos ecossistemas nas Américas, incluindo o Brasil, no entanto, este é o primeiro relato em Apodiformes do bioma Pantanal. Os ácaros foréticos não são responsáveis ​​por danos diretos à população de Apodiformes, pois não são patogênicos. No entanto, eles competem por comida com beija-flores e diminuem a quantidade de comida disponível no ambiente. 2,4-D is a selective pre- and postemergence herbicide used for several crops. It is hazardous for the environment and risk for humans; therefore, several studies attempt to evaluate its effects and consequences of its use. The nervous system is supposedly a target for this herbicide, and this comprehensive review gathers the information about animal models that have been used for the study of the neurotoxicity of 2,4-D. The studies used several methods to evaluate the neurotoxicity of this herbicide, most of which used rodents, mainly rats, two used fish, and one used chicken eggs. The main behavioral effect observed concerned alterations in locomotor patterns and reduced motor activity. Biochemical analysis showed decreased levels of serotonin (5-HT) and increased levels of its metabolites and increased or decreased levels of DA and its metabolites depending on the brain area analyzed. Hypomyelination is also a possible effect of 2,4-D when the exposure occurs during the proliferation and development of the oligodendrocytes. The worst neuropathologic effects were observed in fish. Since most studies focused on the neurotoxicity of 2,4-D in rodents, the effect it may have on other species and groups of animals, especially with different physiology, is unclear and it should be researched.

Lutjanid species exhibit multiple spawning behaviour during an extended spawning season in warm months, asynchronous oocyte development and indeterminate fecundity. Although early studies have contributed to knowledge of the reproductive cycle of many species within the group, they have not considered aspects about the number of cortical alveoli oocyte stage throughout maturity phases along spawning season. The latter aspect is also considered very important to confirm indeterminate fecundity hypothesis. In the present study, were analyzed 154 Brazilian snapper Lutjanus alexandrei female gonads obtained from artisanal fisheries in Pernambuco State (Brazil) between October 2010 and March 2011. Were measured oocyte size frequency distribution for maturity phases (developing, spawning capable and actively spawning), and oocyte development stage (unyolked oocytes, cortical alveoli, primary, secondary and tertiary vitellogenic oocytes and hydrated oocytes), and also the oocyte stage frequency during spawning season. The frequency of cortical alveoli oocyte stage was constantly found in the spawning period (>37%), showing slight variation throughout maturity phases. The absence of gap in the oocyte size frequency distribution between primary and secondary oocyte growth stages during spawning season is a strong indicator of continuous oocyte recruitment from reserve stocks. In addition, co-occurrence of tertiary vitellogenic oocytes, hydrated oocytes, post-ovulatory follicles and yellow-brown bodies in the histological sections of ovaries reinforce indeterminate fecundity hypothesis.

Resumo Os lutjanídeos apresentam um comportamento de desovas parceladas ao longo de uma extensa estação reprodutiva nos meses mais quentes do ano, desenvolvimento assincrônico dos ovócitos e padrão de regulação de fecundidade indeterminado. Embora muitos estudos anteriores tenham contribuído no conhecimento do ciclo reprodutivo das espécies do grupo, não foram considerados os aspectos relacionados ao percentual dos ovócitos em estágio de cortical alveoli entre as fases de maturidade sexual durante a estação reprodutiva. Este último aspecto, considerado importante para confirmação da hipótese de fecundidade do tipo indeterminada. Para isso, no presente trabalho foram analisadas 154 gônadas de fêmeas da baúna de fogo Lutjanus alexandrei obtidas da pesca artesanal em Pernambuco entre o período de outubro de 2010 a março de 2011. Foram mensurados os diâmetros dos ovócitos nas fases de maturidade sexual (desenvolvimento, capaz de desovar e ativamente desovando), e para todos os estágios ovocitários (ovócitos pre-vitelogênicos, cortical alveolar, vitelogênese primária, secundária, terciária e ovócitos hidratados), e avaliada a frequência de ocorrência ao longo do período de desova. O percentual de ovócitos em estágio cortical alveoli permaneceu constante durante o período de reprodutivo (> 37%), apresentando pouca variação entre as fases de maturidade sexual. A ausência de um intervalo na frequência de diâmetro dos ovócitos em crescimento primário e secundário entre todas as fases de maturidade sexual é um forte indicador de recrutamento contínuo de ovócitos do estoque de reserva. Além disso, a presença conjunta de ovócitos em vitelogênese terciária, ovócitos hidratados, folículos pós-ovulatórios e corpos amarelados nos cortes histológicos reforçam a hipótese de fecundidade do tipo indeterminada.

OBJECTIVES: The objective of this study was to evaluate the association between estimated human papillomavirus (HPV) viral load and abnormal cytology on anal samples. METHODS: Anal cytological samples of 42 HIV-positive patients were analysed by conventional cytology and Hybrid Capture II. RESULTS: On cytology, 30.95% (13 of 42) anal samples were positive for cytological abnormalities, 47.61% (20 of 42) were negative and 21.42% (nine of 42) were unsatisfactory. High-risk HPV infection was more frequent in anal samples with cytological abnormalities than in negative samples (P = 0.0002, Fisher's exact test), it was detected in all samples with cytological abnormalities and in 35% (seven of 20) of the negative samples. On samples with cytological abnormalities, the median of the relative light unit/cutoff (RLU/CO) value (viral load estimate) was 10.39 (1.02-572.6) and in negative samples it was 0.51 (0.26-51.70). The median of the RLU/CO value was higher in samples with cytological abnormalities when compared with the median in negative samples (P = 0.0001, Mann-Whitney U-test) and only samples with cytological abnormalities showed RLU/CO values > 100. CONCLUSIONS: The estimated high-risk HPV viral load is significantly higher in samples with cytological abnormalities than in negative anal samples and may be useful as an adjunct to anal cytology for triage of patients to high-resolution anoscopy and biopsy.

AIM: This study was carried out to determine how the partners of patients with a permanent colostomy perceive everyday life, particularly its sexual aspects. METHOD: Data were collected prospectively from October 2011 to May 2012 using a three-part questionnaire to obtain demographic, social and sexual information. Both multiple-choice and yes/no questions were used. RESULTS: One hundred and eight questionnaires were given to 36 partners of patients with a permanent colostomy (the ostomy group) and to 72 partners of normal individuals matched for age, sex and social class. The sexual performance of the spouse was significantly better in the control group (P = 0.004), sexual performance of the partner was also better in the control group (P = 0.023) and reduced frequency of sexual activity was higher in the ostomy group (P = 0.042). There was, however, no significant difference in sexual interest between the two groups (P = 0.507). CONCLUSION: The study demonstrated that about half of the partners of patients with a permanent colostomy experienced dissatisfaction with their spouse's performance, with a reduction in sexual interest and frequency of intercourse compared with the control group. Healthcare professionals should pay more attention to the partner of patients having a permanent colostomy.

A terapia celular vem sendo utilizada com resultados promissores no tratamento da tendinite equina, entretanto ainda existem dúvidas quanto à persistência e ao comportamento dessas células quando implantadas no local da lesão, e quanto à sua migração para outros focos inflamatórios. O objetivo deste estudo foi avaliar a marcação das células-tronco mesenquimais (CTMs) com nanocristal antes e após o implante em lesões tendíneas experimentais do tendão flexor digital superficial (TFDS) de equinos, bem como observar a possibilidade de migração das CTMs marcadas para outro foco de lesão, o membro contralateral do mesmo animal. Para isso, foi realizada a indução de lesão experimental no TFDS em ambos os membros torácicos de cinco equinos e, após sete dias, foram implantadas as CTMs autólogas marcadas com o nanocristal Qtracker 655 em um dos membros dos animais. Após sete dias do implante, foi realizada a biópsia tendínea para posterior avaliação histopatológica, utilizando-se microscopia com fluorescência. Também foi realizado o teste de viabilidade celular antes e após a incubação com o nanocristal. As CTMs marcadas e injetadas no tecido tendíneo mantiveram sua fluorescência sete dias após seu implante, e não ocorreu migração para o membro contralateral. O uso do nanocristal para a marcação das CTMs derivadas da medula óssea equina mostrou-se efetivo pelo fato de essa nanopartícula não ter alterado a viabilidade celular e por ela ter permanecido ativa durante o período implantado...


Cell therapy has been used with promising results in the treatment of equine tendinitis. However, there are still doubts about the persistence and behavior of these cells implanted in the injured tissue and their migration to other inflamed sites. The aim of this study was to evaluate the labeling of mesenchymal stem cells (MSCs) with nanocrystals before and after implantation in experimental tendinitis of the superficial digital flexor tendon (SDFT) of horses, observing the migration possibility of MSCs marked to another lesion, performed on the contralateral limb of the same animal. An experimental lesion was induced in SDFT in both forelimbs of five horses, and after seven days autologous MSCs labeled with Qtracker(r) 655 were implanted in one member of the animals. Tendon biopsy was performed for subsequent histopathological evaluation using fluorescence microscopy seven days after the implant. Cell viability test was also performed before and after incubation with the cell labeling kit. MSCs labeled and injected into the tendon tissue maintained their fluorescence seven days after their implantation and there was no migration to the contralateral limb. The use of nanocrystals for labeling MSCs was effective because it does not alter cell viability and remains active during the experimental period...

This work aims to study the female reproductive tract of silky sharks, Carcharhinus falciformis, captured in the South and Equatorial Atlantic Ocean. Samples were collected between January 2008 and March 2010 through oceanic commercial vessels that targeted tuna and swordfish, with a total of 17 females collected. The methodologies followed for analyzing the ovaries of those females included both macroscopic and histological analysis. Macroscopically, it was possible to determine that the ovaries on these sharks is suspended by mesenteries in the anterior section of the body cavity, heavily irrigated by blood vessels, and contains a wide range of oocytes. Ovaries were found in three distinct maturational stages: Stage I (Immature), Stage II (Maturing) and Stage III (Mature). Immature ovaries were small, with widths ranging from 1.0 to 3.1 cm, and had a gelatinous or granulose internal structure; maturing ovaries were slightly larger, ranging in width between 5.2 and 6.0 cm; mature ovaries ranged in width between 6.5 and 7.8 cm, and had a more rounded shape and the presence of large and well developed oocytes. Under microscopic examination, it was observed that the ovaries were covered with simple epithelial tissue during the early development stages and a simple cubic epithelium in the final stages of maturation. During the initial maturation stages the epigonal organ was not differentiated from the ovary. In mature specimens, the ovary showed a simple cubic epithelium and just below this epithelium there was a layer of dense connective tissue and muscle with the presence of vitellogenic oocytes and fat cells. A thin yolk membrane enclosing the oocytes was also evident. Finally, it was possible to distinguish a zona pellucida, separating the oocytes from the follicle wall and a basal lamina between the granular layers and the teak layer.


Este trabalho tem como objetivo estudar o trato reprodutivo feminino de tubarões lombo preto, Carcharhinus falciformis, capturados no Atlântico Sul e Equatorial. As amostras foram coletadas entre janeiro de 2008 e março de 2010, através das embarcações da frota atuneira, que tem como pesca alvo o atum e espadarte, com um total de 17 fêmeas coletadas. A metodologia seguida para analisar os ovários das fêmeas incluiu a análise macroscópica e histológica. Macroscopicamente, foi possível determinar que o ovário destes tubarões, é suspenso por mesentério na seção anterior da cavidade do corpo, muito irrigada por vasos sanguíneos, contendo uma vasta gama de ovócitos. Os ovários foram encontrados em três diferentes estágios de maturação: estádio I (imaturo), estágio II (maturação) e fase III (Maturo). Ovários imaturos foram pequenos, com largura variando de 1,0 a 3,1 centímetros, e tinham uma estrutura gelatinosa ou granulosa interna; ovários em maturação foram ligeiramente maiores, variando de largura entre 5,2 e 6,0 cm; ovários maduros variaram de largura entre 6,5 e 7,8 cm, possuíam uma forma mais arredondada e a presença de ovócitos grandes e bem desenvolvidos. Ao exame microscópico, foi possível observar que os ovários foram cobertos com tecido epitelial simples durante os estágios iniciais de desenvolvimento, e um epitélio cúbico simples na fase final de maturação. Durante os estágios iniciais de maturação, o órgão epigonal não foi diferenciado do ovário. Em espécimes maduros, o ovário mostrou um epitélio cúbico simples, e só abaixo deste epitélio havia uma camada de tecido conjuntivo denso e músculo com a presença de ovócitos vitelogênicos e células de gordura. Também foi evidenciada uma membrana fina juntando os ovócitos. Finalmente, foi possível distinguir uma zona pelúcida, separando os ovócitos da parede do folículo e uma lâmina basal entre as camadas granular e da camada teca.

Estudou-se a eficácia do plasma rico em plaquetas (PRP) no tratamento de lesões condrais articulares, experimentalmente induzidas em equinos. Para isso, foi induzida uma lesão condral, na tróclea medial femoral dos dois membros pélvicos de quatro animais. Após 30 dias da indução, as oito articulações foram divididas em dois grupos. Os animais do grupo 1 receberam o tratamento intralesional e intra-articular com PRP, e os do grupo 2 foram tratados apenas com solução fisiológica. As avaliações clínicas, constituídas de exames de claudicação e análises do líquido sinovial, foram realizadas antes da indução da lesão - tempo zero -, quinzenalmente, até 120 dias e aos 150 dias. Avaliações macroscópicas, histológicas e histoquímicas foram realizadas no tempo zero e aos 150 dias. Os equinos do grupo 1 apresentaram melhora do grau de claudicação em relação aos do grupo 2. Os exames macroscópicos, histológicos e histoquímicos revelaram melhor tecido de reparação igualmente no grupo 1. Concluiu-se que a administração de PRP apresentou efeitos benéficos no tratamento de lesões condrais experimentais de equinos.


The use of platelet-rich plasma (PRP) in the treatment of articular cartilage defects induced experimentally in horses was studied. For this purpose, both patellofemoral joints of four animals were approached through arthroscopic surgery to perform a cartilage defect on the medial femoral trochlea. After 30 days of induction the eight joints were divided into two groups. Group 1 animals received intralesional and intra-articular treatment with PRP and Group 2 animals were treated only with saline solution. The clinical assessments, constituted by lameness signs and synovial fluid analysis, were performed before induction of injury (time zero) and every 15 days in 120 days, with last analysis on day 150. The macroscopic and morphologic analysis were performed at 0 and 150 days. During the experiment Group 1 animals showed improvement in lameness when compared to Group 2 animals. The macroscopic and morphological analysis showed a better tissue repair in the treated horses. Thus, the implantation of autologous PRP showed beneficial effects in the treatment of chondral lesions, experimentally induced in horses.

Foram induzidas lesões no tendão flexor digital superficial (TFDS) de ambos os membros anteriores de seis equinos, seguidas de implante autólogo de células da fração mononuclear de medula óssea em apenas um membro de cada animal. Os animais foram avaliados por parâmetros clínicos, ultrassonográficos, histopatológico e imunoistoquímico. Paralelamente, realizou-se o cultivo de novas amostras para a caracterização das células utilizando-se marcadores CD34 e CD45 por meio da citometria de fluxo, confirmando a presença de células mesenquimais na fração mononuclear. A caracterização das fibras colágenas tipo I e tipo III no tecido neoformado mostrou melhora na qualidade da cicatrização tendínea dos membros tratados. A terapia com implante autólogo das células da fração mononuclear melhorou a organização tecidual e a sua qualidade, apresentando maior expressão significativa para colágeno tipo I.


The present study was developed inducing a lesion in the SDFT of both thoracic limbs of six horses followed by autologous implantation of mononuclear cells from bone marrow in only one affected limb of each horse. The horses were evaluated through clinical and ultrasonography exams, and through histopathology and immunohistochemistry patterns. Concomitantly, new samples were cultivated and characterized using CD34 and CD45 markers, proving the presence of mesenchymal cells in the mononuclear fraction. The characterization of collagen fibers type I and type III in the new tissue has showed an improvement in tendon healing in treated limbs. The therapy with autologous implant of the mononuclear fraction has improved tissue organization and its quality, having a significanlyt higher expression of collagen type I.

RESUMO

This paper describes the case of a horse treated in a Teaching Veterinary Hospital. The sutures of the flexor apparatus associated with cell therapy, the stem cell, provided a rapid response of repair compared to other cases treated in routine of the Hospital. The association of conventional treatments with the mononuclear fraction of bone marrow significantly contributed to the reduction of the hyperextension of the metatarsophalangeal joint. Ultrasound images showed rapid fill cell and decrease in inflammation.

OBJECTIVE: To evaluate the antiinflammatory effects of RC-3095 in 2 experimental models of arthritis, collagen-induced arthritis (CIA) and antigen-induced arthritis (AIA), and to determine the mechanisms of action involved. METHODS: RC-3095 was administered daily to mice with CIA and mice with AIA, after induction of disease with methylated bovine serum albumin. Disease incidence and severity were assessed using a clinical index and evaluation of histologic features, respectively. In mice with CIA, gastrin-releasing peptide receptor (GRPR) was detected by immunohistochemical analysis, while in mice with AIA, migration of neutrophils, presence of glycosaminoglycans, and lymphocyte proliferation, determined using the MTT assay, were assessed. Expression of cytokines interleukin-17 (IL-17), IL-1ß, and tumor necrosis factor α (TNFα) was evaluated in all mouse knees using enzyme-linked immunosorbent assay. Treg cell production was assessed by flow cytometry in the joints of mice with AIA. RESULTS: In mice with AIA, administration of RC-3095 reduced neutrophil migration, mechanical hypernociception, and proteoglycan loss. These findings were associated with inhibition of the levels of all 3 proinflammatory cytokines, decreased lymphocyte proliferation, and increased Treg cell numbers. In the CIA model, treatment with RC-3095 led to a significant reduction in arthritis clinical scores and the severity of disease determined histologically. Synovial inflammation, synovial hyperplasia, pannus formation, and extensive erosive changes were all dramatically reduced in the arthritic mice treated with RC-3095. Furthermore, arthritic mice treated with RC-3095 showed a significant reduction in the concentrations of IL-17, IL-1ß, and TNFα, and showed a diminished expression of GRPR. CONCLUSION: These findings suggest that the GRP pathway has a significant role in chronic arthritis, and its inhibition can be explored as a possible therapeutic strategy in rheumatoid arthritis.

The rainbow runner, Elagatis bipinnulata, which belongs to the Carangidae family, has a circumtropical distribution. It is found throughout the Atlantic Ocean, from Massachusetts (USA) to Bahia (Brazil). The reproductive biology of the rainbow runner was studied, using specimens captured off the São Pedro and São Paulo Archipelago. From July 1999 to November 2003, a total of 352 fishes were analysed (201 females and 151 males). Fork length (FL) was measured and specimens were gutted for gonads collection. In the laboratory, gonads length, width and weight were measured, and sexes identified macroscopically. Through histological analysis, five different maturation stages were identified for females: immature, maturing, mature, spent and resting. A predominance of maturing and mature females was observed from January to May. The highest gonad index (GI) values were also observed during this period, ranging from 7.7 to 55. Mean sexual maturity size (L50) for females was estimated at 64.6 cm (FL). In the studied area the species exhibited total spawning, with two synchronous groups. Testicles were histologically very similar making them impossible to differentiate sexual maturation stages. Considerable variation was observed in the male gonads weight, hampering the assessment of size at maturity. However, GI values for males were also higher from January to May. These results suggest that the spawning period of the rainbow runner in the São Pedro and São Paulo Archipelago occurs during the first semester of the year.

OBJECTIVE: To determine the effects of RC-3095 in clinical and histopathologic parameters and inflammatory mediators on complete Freund's adjuvant-induced arthritis (CFA). METHODS: The arthritis was induced by injection of CFA into the left hind footpad. The animals were divided into control, vehicle injected control, placebo group (saline subcutaneously 50ml/kg, once daily for 8 days after modeling), treatment group (0.3mg/kg of RC-3095 subcutaneously, once daily for 8 days after induction). Clinical evaluation was accomplished daily, through scoring of the paw edema. The animals were sacrificed 15 days after induction for collection of hind foot joints for histology. We used a histological scoring system which was previously described, and interferon (INF)-gamma, interleukin (IL)-1beta, tumor necrosis factor (TNF), interleukin (IL)-6 and interleukin (IL)-10 were measured by ELISA. RESULTS: There was a significant inhibition of joint histological findings in the RC-3095 treated group, including synovial inflammatory infiltration and hyperplasia, cartilage and bone erosion. IFN-gamma, IL-1beta, TNF, IL-6 and IL-10 serum levels were significantly lower in the treated group. Paw swelling and subcutaneous inflammation, evaluated clinically, were not different between CFA-induced groups. CONCLUSIONS: RC-3095 was able to improve experimental arthritis, attenuate joint damage and decrease serum levels of IFN-gamma, IL-1beta, TNF, IL-6 and IL-10. These data indicate that interference with GRP pathway is a potential new strategy for the treatment of RA that needs further investigational studies.
