RESUMO

Toll-like receptors are involved in the recognition of bacterial toxins, which cause infection in the respiratory system. This study aimed to evaluate microanatomical and histological alterations in the lungs of 24 healthy Akkaraman and Romanov lambs after the administration of lipoteichoic acid (LTA), lipopolysaccharide (LPS), and LTA + LPS and investigate the gene, protein, and immune expression levels of TLR4, MyD88, and TNF-α molecules, known to have immune functions. Microanatomical examinations showed thickened peribronchial and alveolar walls in the lungs of groups LTA, LPS, and LTA + LPS of both breeds due to immune cell infiltration. TLR4, MyD88, and TNF-α immunoexpressions were positive to varying degrees in the cytoplasm and nucleus of the bronchial and bronchiolar luminal epithelial cells, alveolar epithelial cells, and alveolar macrophages. TLR4 and TNF-α protein expressions were statistically different in the LPS-treated Romanov lambs, compared to the other groups. Among the Akkaraman lambs, TLR4 gene expression was significantly higher in group LPS, and among the Romanov lambs, TLR4, MyD88, and TNF-α gene expressions were significantly higher in group LTA + LPS. Therefore, TLR4, MyD88, and TNF-α molecules, involved in the immune response, were found to be expressed at different levels against LTA and LPS in the lungs of two different sheep breeds.

The conjunctions of the cornea and sclera in the eyes of donkeys, cattle, dogs, sheep, pigs and rabbits, regardless of gender, were examined in this study. No animals were specifically sacrificed for this investigation. Scanning electron microscopy, light microscopy, and dissecting microscopy were used in this research. In the limbus of all the animals investigated, the cornea and sclera fused in accordance with a pattern. At the corneo-scleral junction, the sclera was situated anteriorly and the cornea posteriorly in the dorsal and ventral sections of the bulbus oculi. In the medial and lateral parts of the eyeball, the cornea and sclera were facing each other and interlaced. Pigmentation and the sulcus scleralis externus could be used to identify the macro-and micro-anatomical boundaries of the limbus. In addition, the cytoplasm of basal epithelial cells shrank, signaling the end of the corneal epithelium and the start of the conjunctival epithelium. The presence of Bowman's membrane in cattle and sheep eyes was definitely determined in histological examinations. Bowman's membrane in these animals came to an end at the limbus, which is where the conjunctival epithelium starts and the corneal epithelium ends. In all areas of the cornea, Bowman's membrane revealed irregular, abrupt thickening and thinning. The corneal epithelium was thick in the vertex and thinner towards the limbus, whereas Descemet's membrane was thin in the center (vertex) and thick in the periphery (near the limbus). In this study, pictures and diagrams were used to illustrate the general anatomical, histological, and morphometric characteristics of the limbus in the species under investigation. The data from our study showed that the limbus region of the bulbus oculi was narrow in the lateral and medial parts and wide in the dorsal and ventral parts. This was confirmed in the studied animals as a general rule. The width value will undoubtedly affect the number of cells covered by the regions. It is conceivable that these cells will play a significant role in the decision of where to perform surgical procedures in order to promote wound healing, giving doctors an advantage. In this circumstances, we think that the limbus should be studied in terms of clinical methods because it has different shapes depending on the species and the position of the bulbus.

This study represents the first morphological description of the lymphatic drainage of the ciliary body in vivo by comparative hyperspectral fluorescence imaging techniques of Quantum Qdot655 (QD) nanoparticles and indocyanine green (ICG). A volume of 1.25 µl of QD was injected into the left anterior camera of all rats. Similarly,1.25 µl of ICG diluted at a ratio of one-fourth with physiological saline solution was injected into the right anterior camera of all rats. The thickness of the skin in the mandibular area, connective tissue, and the depth of the mandibular lymph node (MLN) made image retrieval difficult. For QD, 302 nm UV excitation and 605 nm fluorescence peak emission were applied. The detection of QD and ICG used in this study in the MLNs is definitive evidence that aqueous humor (AH) follows a uveolymphatic pathway. Scanning electron microscope and the energy dispersive X-ray analyzer spectrum were used to examine both the Schlemm's canal and the MLN. For the first time, the QD was detected in the cortex of MLN. The QCM analysis of both QD-AH and ICG-AH was used to determine whether there was any interaction between them. This comparative study shows the importance of experimental animal modeling in pharmacological studies regarding eye research and drugs. In a female rat, the signal was taken from the parotid lymph node with QD injections.
