RESUMO

Young adult male Wistar rats (eight controls and ten experimental animals per group) were injected parenterally for 28 days: 1) cortisol or 2) cortisol and salmon calcitonin. Morphological changes of the stomach wall and enterochromaffin cells, stained immunocytochemically for serotonin, were assessed. In the group treated with cortisol superficial desquamation and erosions of the gastral mucosa were found. In this group the number of enterochromaffin cells was significantly decreased and low intensity of the immunocytochemical reaction of these cells was observed. In the cortisol-calcitonin-treated group there were no apparent changes of the gastral mucosa and the enterochromaffin cells presented a normal picture comparable to that of controls.

Male young adult Wistar rats were treated parenterally with cortisol and cortisol combined with salmon calcitonin for 28 and 56 days and the stomach was investigated histologically as well as the G cells using immunohistochemistry. After 28 days of cortisol administration desquamation of the superficial layers of gastric mucosa was found and after 56 days ulcer-like changes developed and increased gastrin immunoreactivity was observed. Administration of high doses of salmon calcitonin together with cortisol resulted in a significant hypoplasia of G cells and prevented the pathological changes in the gastric mucosa.

RESUMO

In the experimental model of ARDS in rats there was investigated the influence of Intal (Fisons, GB) on the changes in the rat's lungs. Intal was inhaled through the respiratory system. Morphological examinations were carried out by hematoxylin and eosin staining. The results of experiment show that Intal seems to be a protective remedy in ARDS.

RESUMO

2-(6-methoxy-2-naphthyl) propionic acid is introduced to treatment as non-steroid antiinflammatory drug (NSAID) under the trade-mark of Naprosyn. Inhibition of prostaglandin synthesis is regarded as the most likely mechanism of its action. In some patients, its side-effects include gastritis, reactivation of ulcerous niche, and upper gastrointestinal haemorrhage. The absence of complex studies addressed to the question of morphological and histochemical changes in gastric mucosa after oral administration of Naprosyn prompted our undertaking. In experimental animals, with varying doses (10 mg/kg and 50 mg/kg) and variously long administrations (1 week and 3 weeks), a trial has been reported here upon. In the frozen-sectioned preparations, the histochemical reaction for acid phosphatase activity, according to Gomori, was made. The paraffin sections were subjected to the HE staining, PAS staining according to McManus and with the Masson's method. Our results of the morphological and histochemical studies in rats support the clinical observations of mucosal destruction in stomach in patients after oral administration of Naprosyn.

In rats subjected to acute or protracted stress of immobilization, gastric and duodenal mucosae were monitored for presence of ulcers and their enterochromaffin cells were examined, identifying them with the use of anti-serotonin antibodies and PAP technique. In parallel, serotonin and 5-hydroxyindolacetic acid levels in the stomach and duodenum were estimated. Ulcers developed only in the stomach and exclusively in stress-unadapted animals. Development of ulcers was paralleled by enterochromaffin cell degranulation, decrease in serotonin levels, and increase in 5-hydroxyindolacetic levels in both the stomach and the duodenum. Significance of the findings for contemporary hypothesis of gastric ulcers' pathogenesis was discussed.
