RESUMO

For the first time in skeletal muscle, the sphingolipid profile and key enzymes involved in the generation of ceramide in cells were investigated in simulated microgravity. It was found that, in C57B1/6 mice, the 4-day hindlimb unloading, in addition to reducing the mass of m. soleus, leads to the ceramide accumulation (3-fold) and the decrease of sphingomyelin content in this muscle (7.2-fold), as well as to the increase (2.7-fold) of protein level of acid sphingomyelinase. In a loaded m. biceps brachii the amount of ceramide is also enhanced, but both the amount of sphingomyelin and sphingomyelinase, as well as the muscle mass do not change, while the level of serine palmitoyltranspherase becomes significantly lower than in control mice. Taking into account the negative effects of ceramide in skeletal muscle (insulin resistance, inhibition of protein synthesis and increase of its decay) we can assume that sphingolipid mechanisms may be involved in the development of structural and functional abnormalities of skeletal muscle under conditions of weightlessness.

RESUMO

Immunological biochips (immunobiochips) based on antibodies--test-systems, allowing simultaneously to determine an existence of different antigens in the material. Biochip of this class is usually a plate (substrate), on which antibody molecules with known specificity are immobilized within well-defined areas. The advantage of the use of biochips is the ability to define a large number of antigens at very low flow rates of antibodies. Biochips can be used for many tasks, including immunophenotyping for tumor cells. There are presented the results of testing of biochips made in laboratory conditions to determine surface antigens of cells in patients with chronic lymphocytic leukemia. It is showed a good agreement between the results of immunophenotyping of cells in chronic lymphocytic leukemia patients with the use of biochips with data of flow cytometry.

RESUMO

The authors present their experience gained in the first clinical use of immunological biochips for detection of cellular surface antigens for immunomorphological diagnosis of chronic lymphocytic leukemia and compare the results of this method and flow cytofluorometry.

RESUMO

y We report preliminary results of efficacious clinical application of a new immunological method with the use of biochips for diagnosis of chronic lymphocytic leukemia. This method has been developed by the authors to detect a wide range of cell surface antigens. We compared the amount of cells expressing certain antigens with clinical properties of chronic lymphocytic leukemia. Higher amounts of CD9, CD21, CD31, CD71, CD38 positive lymphocytes were shown to occur in case of favorable course of the disease.
