Knowledge about the basal level of DNA damage in leucocytes of healthy control populations is essential before estimation of the effects of exposure to external agents in biomonitoring studies. The aim of this study was to analyse the effects of some lifestyle factors on baseline DNA damage in leucocytes of humans. The material consisted of the peripheral blood from 276 healthy volunteer blood donors. In addition to the standard blood donation questionnaire, they were asked about age, gender, occupation, radiological history, smoking habit, alcohol consumption, medicine use and pet ownership. The results showed marked intra-individual variability. Significant differences in DNA damage levels were observed between individuals in different age and sex groups, between smokers and non-smokers and between samples taken in different seasons of the year, with the highest DNA damage in those obtained in the summer. Significantly higher levels of DNA damage were noted in leucocytes of donors older than 29 years, in men compared with women and in male smokers. Significantly higher DNA strand breaks were observed in heavy smokers. A non-significantly higher level of DNA damage was observed in individuals subjected to radiological investigation and in those drinking alcohol, whereas lower levels were observed in leucocytes of pet owners and in donors taking medicines. Pet ownership influences the level of DNA damage and there is an interaction between this effect and that of smoking. The smoker/pet owners showed almost half the level of DNA damage of smokers without pets. The current results confirmed high intra-individual variability between the levels of DNA damage of individuals. The significant factors that influence the DNA damage in leucocytes are age, sex and smoking habit, especially in men and in heavy smokers. The finding of reduced DNA damage in the leucocytes of pet owners suggests the tendency towards a beneficial effect of such company.

The study assessed the incidence of HBV markers (HBsAg, anti-HBc, anti-HBs) important for determination of the risk of reactivation of infection, with particular interest of occult infection (presence of HBV DNA in the absence of HBsAg) in patients treated at the Institute of Hematology and Transfusion Medicine. Anti-HBc frequency was correlated with the age and sex of patients. HBsAg was detected in 16/468 examined patients, 98/468 (21%) were anti-HBc positive. HBV DNA was detected in 41/98 anti-HBc positives; in 13 simultaneously with HBsAg. 28 patients had occult HBV infection (HBV DNA+/HBsAg). Antibody to HBsAg was detected in 163/430 (38%) patients, 81 out of them on protective level (> 100 IU/l). It was shown that occult HBV infection occurs in approximately 6% of patients. In most of them the protective levels of anti-HBs are detected.

INTRODUCTION: Recent studies suggest that multiple myeloma (MM) triggers osteoclastogenesis by disrupting the balance between the receptor activator of NF-kappaB ligand (RANKL) and osteoprotegerin (OPG), its natural antagonist. MATERIAL/METHODS: Determinations of bone marrow (BM) and serum OPG and sRANKL concentrations were performed in 133 MM patients and 42 healthy subjects by the ELISA method using Osteoprotegerin ELISA and sRANKL ELISA kits. RESULTS: MM patients had elevated serum levels of OPG compared with controls (p<0.0001) and OPG levels were higher in patients with renal failure and patients with hipercalcemia (p<0.001 and p=0.04, respectively). Serum OPG levels correlated with age, serum beta 2-microglobulin, and BM OPG concentrations and did not correlate with the presence of osteolysis or with stage of disease. sRANKL serum levels in MM patients and in controls were not statistically different (p=0.42). In MM patients, serum OPG and sRANKL levels were similar at diagnosis and in the plateau phase of disease. There was a correlation between BM and serum sRANKL concentrations (p<0.001). Median values of the sRANKL/OPG ratio for BM and serum of MM patients were 0.14 and 0.11, respectively. The median value of the sRANKL/OPG ratio for the serum of controls was 0.11. CONCLUSIONS: In 20% of MM patients, serum OPG levels are elevated, and this may be a compensative reaction related to increased bone destruction. There is not statistically significant relationship between sRANKL serum and BM levels and the main clinical and laboratory parameters of the disease. Determination of BM and the serum sRANKL/OPG ratio seems to have no clinical value.
