RESUMO

Evidences for an involvement of the bacterial type IV secretion system (T4SS) in the symbiotic relationship between rhizobia and legumes have been pointed out by several recent studies. However, information regarding this secretion system in Mesorhizobium is still very scarce. The aim of the present study was to investigate the phylogeny and expression of the traG gene, which encodes a substrate receptor of the T4SS. In addition, the occurrence and genomic context of this and other T4SS genes, namely, genes from tra/trb and virB/virD4 complexes, were also analyzed in order to unveil the structural and functional organization of T4SS in mesorhizobia. The location of the T4SS genes in the symbiotic region of the analyzed rhizobial genomes, along with the traG phylogeny, suggests that T4SS genes could be horizontally transferred together with the symbiosis genes. Regarding the T4SS structural organization in Mesorhizobium, the virB/virD4 genes were absent in all chickpea (Cicer arietinum L.) microsymbionts and in the Lotus symbiont Mesorhizobium japonicum MAFF303099T. Interestingly, the presence of genes belonging to another secretion system (T3SS) was restricted to these strains lacking the virB/virD4 genes. The traG gene expression was detected in M. mediterraneum Ca36T and M. ciceri LMS-1 strains when exposed to chickpea root exudates and also in the early nodules formed by M. mediterraneum Ca36T, but not in older nodules. This study contributes to a better understanding of the importance of T4SS in mutualistic symbiotic bacteria.

BACKGROUND AND AIM: The hazardous health effects of smoking and second-hand smoke are well known and have been confirmed in several studies. We wondered whether a school based programme involving media models such as those represented by famous soccer players and TV characters, was effective in prevention of smoking habit in secondary school adolescents. METHODS: Since October 2006 to May 2007 an anonymous survey was submitted to 1382 secondary schools pupils. After completing the questionnaire all students of 42 out of 70 classes selected by the school principals underwent a prevention programme consisting of 1 hour lecture on smoke healthy hazard with educational material (slides, video, leaflets). Furthermore each pupil was given card games with significant pictures. Since October 2007 to May 2008 and Since October 2008 to May 2009 pupils underwent a 1 hour interactive lesson on smoke related health hazards respectively. On December 2007 pupils in study attended a theatre event with show business characters acting to smoke dissuasion. No intervention was performed on the 568 pupils of the other classes along all the same 2 school- year period (controls). RESULTS: Among other results at the end of the 2-year program 4% pupils of study group and 14% of controls reported smoking habit (p = 0.001) whereas 7% and 27% (p = 0.001) of study and control pupils respectively ignored smoking induced dependence. CONCLUSION: A school based programme involving media models such as those represented by famous soccer players, TV characters, was effective in prevention of smoking habit in secondary school adolescents.

In this work we characterize the Phodopus roborovskii and Phodopus sungorus karyotypes, describing the constitutive heterochromatin and the telomeric repeats distribution in these chromosomes. In the two species, (peri)centromeric, interstitial and (sub)telomeric C-bands were revealed, presenting a very high colocalization with evolutionary breakpoint regions identified in these karyotypes. Also both species present telomeric sequences located interstitially (ITS), as short ITS blocks or as large ITS blocks, mainly at the (peri)centromeric heterochromatic regions. The number and degree of ITSs amplification varies greatly in the two hamsters, indicating independent evolutionary events of these repeats in each genome. The combination of the data provided interesting insights about the genome organization of these hamster species, also allowing establishing evolutionary considerations on their chromosomes. The obtained results clearly suggest an involvement of the repetitive genomic fraction in the reshaping of P. roborovskii and P. sungorus karyotypes.

This study evaluated the resistance of cattle of different genetic groups to the tick Rhipicephalus microplus and the relationship with traits of the animals' hair and coat. Cows of the Senepol×Nelore (SN), Angus×Nelore (AN) and Nelore (NX) genetic groups were submitted to four consecutive artificial infestations, at 14-day intervals, each one with approximately 20,000 tick larvae placed on the animals' lumbar region. From the 19th to 23rd day of each infestation five counts of the number of ticks were performed on each animal's left body side. The tick count data (TTC) were transformed into log(10) (n+1), and also into percentage of return (PR), where n is the total number of ticks counted at each infestation. Hair samples were collected 24h after the last infestation with flat-nosed pliers. Measures of the average hair length (HL), coat thickness (CT), number of hairs per cm(2) (NHCM2) and weight of the samples (SW) were obtained. Pearson's correlation coefficients were calculated within genetic group to measure association between PR and the hair and coat data. There was a significant difference among genetic groups for the number of ticks, with the AN group having higher counts than the SN and NX groups. For the hair and coat traits, the NX and SN groups had lower values of HL and SW than did the AN group. The SN genetic group had lower NHCM2 counts than the NX and AN groups. There were positive correlations between TTC and CT (P<0.05) and SW (P<0.05) in the SN group. No significant correlation was found for the AN genetic group (P>0.05).

Caring for patients with Global Initiative for Chronic Obstructive Lung Disease (GOLD) stages III and IV with chronic respiratory failure is difficult independent of whether the target is survival or quality of life (QOL). The role of inhaled drug therapy in this specific set of very severe COPD patients has not previously been assessed. The only drug able to prolong survival in these patients is long term oxygen therapy, whereas there is little evidence to indicate long-term domiciliary mechanical ventilation in the routine management of stable hypercapnic patients. Supplemental oxygen during exercise reduces exercise breathlessness and improves exercise capacity of the hypoxaemic patient. Pulmonary rehabilitation including nutritional supplementation is a significant component of therapy, even in these severe patients. Relief of dyspnoea with drugs such as morphine should not be denied to severely disabled patients who share poor QOL with cancer patients. Non-invasive ventilation has been used as a palliative treatment to reduce dyspnoea. Lung Volume Reduction Surgery may improve mortality, exercise capacity, and QOL in selected patients, but is associated with significant morbidity and an early mortality rate in the most severe patients. Lung transplantation is a final step in end-stage patients, but short- and long-term outcomes remain significantly inferior in relation to other "solid" organs recipients.
