Host and tissue-specificity of endophytes are important attributes that limit the endophyte application on multiple crops. Therefore, understanding the endophytic composition of the targeted crop is essential, especially for the dioecious plants where the male and female plants are different. Here, efforts were made to understand the endophytic bacterial composition of the dioecious Siraitia grosvenorii plant using 16S rRNA amplicon sequencing. The present study revealed the association of distinct endophytic bacterial communities with different parts of male and female plants. Roots of male and female plants had a higher bacterial diversity than other parts of plants, and the roots of male plants had more bacterial diversity than the roots of female plants. Endophytes belonging to the phylum Proteobacteria were abundant in all parts of male and female plants except male stems and fruit pulp, where the Firmicutes were most abundant. Class Gammaproteobacteria predominated in both male and female plants, with the genus Acinetobacter as the most dominant and part of the core microbiome of the plant (present in all parts of both, male and female plants). The presence of distinct taxa specific to male and female plants was also identified. Macrococcus, Facklamia, and Propionibacterium were the distinct genera found only in fruit pulp, the edible part of S. grosvenorii. Predictive functional analysis revealed the abundance of enzymes of secondary metabolite (especially mogroside) biosynthesis in the associated endophytic community with predominance in roots. The present study revealed bacterial endophytic communities of male and female S. grosvenorii plants that can be further explored for monk fruit cultivation, mogroside production, and early-stage identification of male and female plants. KEY POINTS: • Male and female Siraitia grosvenorii plants had distinct endophytic communities • The diversity of endophytic communities was specific to different parts of plants • S. grosvenorii-associated endophytes may be valuable for mogroside biosynthesis and monk fruit cultivation.

Damask rose (Rosa damascena Mill.) is one of the most high-value essential oil-bearing plants in the world. However, the flower yield and quality of essential oil of R. damascena are largely influenced by the pruning practices and balanced supply of plant nutrition. The objective of this study was to test the hypothesis whether the pruning system and foliar fertilization of MgSO4 would influence the flower yield, growth and secondary metabolites profile of R. damascena. A field experiment of 10 treatment combinations comprising two pruning systems (complete and partial) and five levels of MgSO4 (water spray, MgSO4 @ 5.0g L-1, 10.0g L-1,15.0g L-1, and 20.0g L-1) was conducted. The experiment was conducted in randomized block design with factorial arrangement. Overall, the flower yield ranged from 503.66 to 1114.47 g bush-1, while oil content varied from 0.039 to 0.046% of the fresh flower. Irrespective of foliar spray, partial pruning produced significantly (P ≤ 0.05) higher flower yield (893.02 and 503.66 g bush-1) compared with complete pruning system in both the years. Regardless of pruning system, the foliar application of MgSO4 @ 15.0g L-1 registered about 26-38% higher flower yield compared with water spray control. The major constituents of essential oil were citronellol (19.75-48.88%), E-geraniol (9.63-29.6%), Z-citral (0.07-5.97%), nonadecane (6.76-22.32%), and heneicosane (2.87-10.21%). The principal component analysis revealed that the major hydrocarbons such as nonadecene, nonadecane, and heptadecane are positively and highly correlated with each others. The results suggest that higher yield and quality of R. damascena can be achieved through partial pruning system in combination with foliar application MgSO4 under rainfed acidic conditions.

The essential oil extracted by hydrodistillation from the flowering twigs of Stevia rebaudiana Bertoni (Asteraceae) was fractioned by chromatography. Forty-three constituents were characterized with the help of GC, GC-MS and other spectroscopic techniques. The essential oil was found to be a complex mixture of mono- and sesqui-terpenes. The cytotoxicity of the essential oil and its fractions was evaluated by sulforhodamine B (SRB) based assay against two cancer cell types viz. C-6 (rat glioma cells) and CHOK1 (Chinese hamster ovary cells). The essential oil and its fractions showed promising cytotoxicity against both cell lines. The highest activity (95.6+/-0.6%) was show by the essential oil on the C-6 cell line at a concentration of 400 microg/mL, which was comparable with that of the standard drug vinblastin.
