RESUMO

This paper describes a novel free-flow electrophoresis (FFE), which is joined with gratis gravity, gas cushion injector (GCI) and self-balance collector instead of multiple channel pump, for the purpose of preparative purification. The FFE was evaluated by systemic experiments. The results manifest that (i) even though one-channel peristaltic pump is used for the driving of background buffer, there is still stable flow in the FFE chamber; (ii) the stable flow is induced by the gravity-induced pressure due to the difference of buffer surfaces in the GCI and self-balance collector; (iii) the pulse flow of background buffer induced by the peristaltic pump is greatly reduced by the GCI with good compressibility of included air; (iv) the FFE can be well used for zone electrophoretic separation of amino acids; (v) up to 20 inlets simultaneous sample injection and up to five to tenfold condensation of amino acid can be achieved by combining the FFE device with the method of moving reaction boundary. To the best of authors' knowledge, FFE has not been used for such separation and condensation of amino acids. The relevant results achieved in the paper have evident significance for the development of preparative FFE.

The scenic effects of clearcutting have been a volatile issue for the American public for much of the past century. A better understanding of the scenic perceptions associated with the cumulative visible effects of clearcutting should contribute to better decisions about its use. This study evaluates the scenic impacts of simulated alternative clearcutting management systems for the White Mountain National Forest. Alternatives represented an unharvested view, and views with removal of 1-5% of the timber every 12 years using either scattered or concentrated clearcut units of 5, 15 and 30 acres in two viewsheds. A random sample of local citizens, four groups of opinion leaders, and US Forest Service employees evaluated these alternatives. Each factor accounts for significant differences in scenic value. All groups rated the scenic value of unharvested scenes very high. All groups find a large drop in scenic value with the introduction of even the least intense harvesting activity. All groups indicated a statistically significant decrease in visual quality as the intensity of harvesting increases. This effect continues throughout the range of harvesting intensities until the level of sustainable yield is reached. In addition, patches 12-15 acres in size were preferred to larger and smaller ones.

We demonstrate a novel strategy for mixing solutions and initiating chemical reactions in microfluidic systems. This method utilizes highly focused nanosecond laser pulses from a Q-switched Nd:YAG laser at lambda = 532 nm to generate cavitation bubbles within 100- and 200-microm-wide microfluidic channels containing the parallel laminar flow of two fluids. The bubble expansion and subsequent collapse within the channel disrupts the laminar flow of the parallel fluid streams and produces a localized region of mixed fluid. We use time-resolved imaging and fluorescence detection methods to visualize the mixing process and to estimate both the volume of mixed fluid and the time scale for the re-establishment of laminar flow. The results show that mixing is initiated by liquid jets that form upon cavitation bubble collapse and occurs approximately 20 micros following the delivery of the laser pulse. The images also reveal that mixing occurs on the millisecond time scale and that laminar flow is re-established on a 50-ms time scale. This process results in a locally mixed fluid volume in the range of 0.5-1.5 nL that is convected downstream with the main flow in the microchannel. We demonstrate the use of this mixing technique by initiating the horseradish peroxidase-catalyzed reaction between hydrogen peroxide and nonfluorescent N-acetyl-3,7-dihydroxyphenoxazine (Amplex Red) to yield fluorescent resorufin. This approach to generate the mixing of adjacent fluids may prove advantageous in many microfluidic applications as it requires neither tailored channel geometries nor the fabrication of specialized on-chip instrumentation.

High-salt stacking in electrokinetic chromatography (EKC) is defined and contrasted to the sweeping method. A recent paper argued the two methods are identical, where high concentrations of micelle in the sample were intended to mimic the effect of high-salt stacking. However, high micelle concentration in the sample matrix in EKC is analogous to using a high-conductivity sample instead of a low-conductivity sample in field amplified stacking. High-salt stacking does not require a sample free of pseuostationary phase, only a sample with a high-mobility co-ion compared to the separation buffer electrokinetic vector. High-salt stacking uses a discontinuous buffer system and should not be confused with continuous buffer stacking systems such as sweeping.
