RESUMO

The stimulating effect of a dye of the triphenylmethane group (fast green) applied at low concentrations (10(-4)%) in the presence of toluene on the specific growth rate of bacteria was found. The cell size did not change during batch cultivation. However, the cell dry weight and zeta-potential increased sharply.

RESUMO

Kinetics of TBARS accumulation, Fe2+ oxidation, and chemiluminescence were measured in nuclear and mitochondrial membrane suspensions during Fe(2+)-induced lipid peroxidation development. Above certain critical Fe2+ concentrations ([Fe2+].) the latent period (t) of lipid peroxidation development were observed. By using the dependence of t on [Fe2+] the [Fe2+]. values were measured in nuclear and mitochondrial membrane suspensions. At the same concentrations of nuclear and mitochondrial membranes (1 mg/ml) the [Fe2+] value changed from 10 to 30 mu kM. In the case of nuclear membrane suspension the [Fe2+] value changed from 20 to 30 mu kM with the change of membrane concentrations from 0.5 to 1.5 mg/ml. For mitochondrial membrane suspension [Fe2+]. value increased from 10 to 40 mu kM with increasing membrane concentrations from 1 to 4 mg/ml. Apparently, [Fe2+]. value is the main parameter determining the kinetics of Fe(2+)-induced lipid peroxidation in native membrane preparations; the [Fe2+]. value in different membrane systems, in turn, is dependent on the ability of the membrane to bind Fe2+ ions.

RESUMO

There were investigated changes of electrokinetic potential (EKP) of hepatocyte nuclei during hydrocortisone induction and activation induced with amino acids mixture. An increase of RNA-polymerase activity was shown to correlate with EKP value. Actinomycin D essentially decreased this potential. Injection of hydrocortisone or amino acids mixture under the suppression of transcription with actinomycin D removed the inhibitor action, but the potential value did not reach that under the action of the activators only. Induction with hydrocortisone increased the potential by various degrees for the nuclei of different size. The results obtained suggest that EKP of the nuclei carries the information about functional condition of the genetic apparatus and can be a valuable and handy instrument of definition of its physiological activity.

RESUMO

It is shown that over 80% of nuclear neutral lipids are found in the nuclear membrane preparations, whereas only 7%--in chromatin. The injection of hydrocortisone to animals significantly decreases the neutral lipids content in both fractions. These results and the data obtained previously by the authors indicate the possible participation of nuclear membrane and chromatin lipids in the process of hormonal regulation of the genetic activity.

It has been found that the rat liver nuclear matrix contains a small amount of phospholipids (2%) and neutral lipids (1.6%). The injection of hydrocortisone increased the nuclear matrix phospholipid content and reduced its neutral lipid levels. A marked increase in sphingomyelin content observed was accompanied by the reduction in phosphatidylcholine level. The decrease in neutral lipid content took place mainly at the expense of a sharp lowering of triglyceride and cholesterol ether levels. The latter may turn into free cholesterol, thus increasing its content. The data obtained testify to steroid hormone influence on lipid metabolism in intranuclear structures. The observed changes in lipid composition may be related to specific hydrocortisone-induced activation of genome.

RESUMO

The authors have demonstrated the estradiol induction of the key glycolytic enzymes of the brain (hexokinase, phosphofructokinase and pyruvate kinase) which is "switched off" by injection of actinomycin-D--an inhibitor of protein biosynthesis. Another steroid hormone, progesterone, has an inhibitory action on the above enzymes. Administration of estradiol to rats significantly elevates the amplitude and frequency of contractility induced in them, while the same doses of progesterone decrease them. Thus, there is a definite correlation between the induction of glycolytic enzymes and sensitivity toward contractility on the one hand and the estradiol/progesterone ratio on the other. It is possible that activation of glycolysis may reduce the threshold of epileptic attacks providing an additional energy for epileptic manifestations.

RESUMO

Preparations of chromatin and labile-bound nonhistone proteins were isolated from cockerel liver cells after treatment of birds with estradiol as well as from controls. Six protein fractions were obtained by means of column chromatography on DEAE cellulose: two of these fractions were quantitatively altered due to the hormonal effect. Electrophoretic separation of both total labile-bound nonhistone proteins and their hormone-dependent fractions showed that protein composition was altered after the estradiol treatment as compared with controls.

RESUMO

When isolating protein components of chromatin from the rat liver phospholipids are found in the fraction of nonhistone proteins but not in that of histone. Administration of hydrocortisone to animals evokes a reliable increase of the phospholipids of hydrocortisone to animals evokes a reliable increase of the phospholipids amount in the preparations of chromatin and labile-bound nonhistone proteins. In this case the fraction of phosphatidylcholine lowers considerably with a simultaneous increase of sphingomyelin and phosphatidylethanolamine. It is supposed that phospholipid metabolism in chromatin is controlled by hormones, which may be significant for mechanisms of the genetic apparatus activation by steroid hormones.

It was shown that there existed a correlative connection between total clip activity and circular dichroism relative ellipticity. It is supposed that the parameters of total clip activity also may carry information about DNA conformation in the complexes with histones.

RESUMO

A method for determining the nucleotide content of DNA blocks from areas under the melting curve is suggested. It is shown that this method is more simple and the total melting curve is decomposed not to the Gauss components, but it is considered as as superposition of the melting curves of individual blocks. Application of the method for the determination of calf thymus DNA is discussed.

RESUMO

After hydrocortisone administration some properties of rat liver chromatin were studied by means of melting technique and circular dichroism. Hydrocortisone caused a more than 2.5-fold increase in amount of the chromatin active fraction. Distinct differences were detected between circular dichroism spectra of total chromatin and of its active fraction as well as between differential curves of the chromatin active fraction melting in control and test samples. The difference was expressed as a decrease in temperature of melting in a low-point zone and as appearance of the secondary maxima in the region of nucleosomes melting. These data suggest that the decondensation of nucleosome-containing sites takes place which is considered as an indirect evidence of the chromatin activation.

RESUMO

It has been shown that specific proteins and peptides are synthetized in uterus cells during 1-4 h after estradiol administration into rabbits. These compounds side by side with peroxidase may serve as biological marker of estradiol action in target-tissue.

RESUMO

Effect of estradiol propionate on activity of seven enzymes from rabbit uterus was studied. Simultaneously with the known estrogen-induced enzymes, activity of some other enzymes from uterus cells (tyrosine transaminase, acetylcholinesterase, butyrylcholinesterase) was also studied. The hormone induced all the enzymes studied except of butyrylcholinesterase. After induction with the estrogen a new isoenzyme fraction was found in peroxidase: at the same time, content of isoenzymes of lactate dehydrogenase, tyrosine transaminase and acetylcholinesterase was increased.

RESUMO

Previous experimental results are discussed. It was shown that two H+--K+-exchange systems operate in the E. coli membranes and that they can be stopped by N,N'-dicyclohexylcarbodiimide. Either of the two systems is assumed to include the ATPase complex F1 x F0 that has relation to the exchange of 2H+ from the cell for K+ of the medium, another system includes as a portion the ionic channel F0. The aim of the paper is to present the current evidence about intramembrane interplay between the fluxes of H+ and K+ during the operation of F1 x F0 and about the lack of the relationship between these fluxes via electrochemical potential differences.
