RESUMO

Rare earth elements (REEs) are essential components of modern technologies and are often challenging to acquire from natural resources. The demand for REEs is so high that there is a clear need to develop efficient and environmentally-friendly recycling methods. In the present study, freeze-dried cells of the extremophile Galdieria sulphuraria were employed to recover yttrium, cerium, europium, and terbium from quaternary-metal aqueous solutions. The biosorption capacity of G. sulphuraria freeze-dried algal biomass was tested at different pHs, contact times, and biosorbent dosages. All rare earths were biosorbed in a more efficient way by the lowest dose of biosorbent, at pH 4.5, within 30 min; the highest removal rate of cerium was recorded at acidic pH (2.5) and after a longer contact time, i.e., 360 min. This study confirms the potential of freeze-dried cells of G. sulphuraria as innovative ecological biosorbents in technological applications for sustainable recycling of metals from e-waste and wastewater.

Chickpea (Cicer arietinum L.) from Valle Agricola is a legume cultivated in Southern Italy whose intake is strictly linked to rural traditions. In order to get new biochemical insight on this landrace and to promote its consumption and marketing, nutritional values (moisture content, total proteins, lipids, total and free amino acids) and metabolic traits are deeply investigated. Valle Agricola chickpea is nutritionally rich in proteins (19.70 g/100 g) and essential amino acids (7.12 g/100 g; ~40% of total). Carbohydrates, whose identity was unraveled by means of UHPLC-HR MS/MS analysis, were almost 60% of chemicals. In particular, a di-galactosylglycerol, a pinitol digalactoside, and a galactosylciceritol were found as constitutive, together with different raffinose-series oligosaccharides. Although lipids were the less constitutive compounds, glycerophospholipids were identified, while among free fatty acids linoleic acid (C18:2) was the most abundant, followed by oleic (C18:1) and palmitic (C16:0) acids. Isoflavones and hydroxybenzoic acid derivatives were also detected. Valle Agricola chickpeas showed very good levels of several mineral nutrients, especially magnesium (164 mg/100 g), potassium (748 mg/100 g), calcium (200 mg/100 g), zinc (4.20 mg/100 g) and manganese (0.45 mg/100 g). The boiling process favorably decreases anti-trypsin and anti-chymotrypsin activities, depleting this precious seed of its intrinsic antinutritional factors.

Walnut husk washing waters (WHWW), a by-product of walnut production, are indiscriminately used for irrigation without preliminary risk assessment. Basing on previous in vitro results on the toxicity of this by-product, we have followed the morphophysiological development of Zea mays, Lactuca sativa cv. Gentilina and L. sativa cv. Canasta under diluted and undiluted WHWW irrigation. Significant development alterations have been observed in root and shoot elongations for all crops as well as in total biomass and chlorophyll content. The genotoxic potential of WHWW has been concurrently verified; acridine orange/ethidium bromide staining evidenced chromatin modifications and DNA degradation and also was confirmed by DNA laddering. The DNA instability was also assessed through RAPD, thus suggesting the danger of the by-product of walnut processing and focusing the attention on the necessity of an efficient treatment of WHWWs. The findings obtained by PCA of agronomic and physiological traits suggested that establishing guidelines for the administration of WHWW for irrigation is of great importance, and it is necessary to supervise their use in agricultural soils.

Buffalo dung is a low-cost substrate with plenty of carbohydrates, an optimal carbon/nitrogen ratio, and a rich microbial flora, and could become a valuable source of biogas. Therefore, in the present study we compared the type and amount of specific eubacteria to the different configurations of pH, temperature and thermal pretreatment after fermentation in batch reactors in order to understand the suitability of buffalo manure for hydrogen production. The phylogenetic structure of the microbial community in fermentation samples was studied using denaturing gradient gel electrophoresis to generate fingerprints of 16S rRNA genes. The sequences analysis revealed abundance of the phyla Bacteroidetes and Firmicutes, and in particular of the order Clostridiales. Very active hydrogen producing bacteria belonging to Clostridium cellulosi species were identified demonstrating the suitability of this substrate to produce hydrogen. Moreover, a large fraction of 16S-rDNA amplicons could not be assigned to lower taxonomic ranks, demonstrating that numerous microorganisms involved in anaerobic fermentation in digesters or bioreactors are still unclassified or unknown.

Three hundred and ninety-seven randomly selected households were interviewed by telephone to determine the numbers and management of owned cats and dogs in the Teramo Province of Italy. The households were selected using stratified random sampling for each municipality; municipalities were combined into coastal, central hills and mountain regions for analysis. The interviews were completed during May and June of 2004 with a response rate of 74% (397/536). Forty-six percent of households (n=181) owned pets; 15% of all households (n=60) owned cats and 33% (n=130) owned dogs. Twenty-seven of these households (7%) owned both cats and dogs. Data were provided on 91 cats evenly divided between males and females. The median age was 3 years (range 0.2-10 years). Forty-one percent of cats (36/87) entered the household as strays. Nearly half lived entirely outside. Seventy percent (62/88) had visited a veterinarian at least once; 43% (39/91) were sterilized. About 1/3 had had a litter and all litters were considered accidental rather than planned. Age, indoor/outdoor status, veterinarian visit and region were all associated with sterilization. Age, confined to a yard, veterinary visit and region were associated with allowing the cat to roam freely. Data were provided on 182 dogs. Sixty-two percent (113/181) were male, with a median age of 4 years (range newborn to 17 years) and 40% (72/181) were purebred. Almost half were acquired as a gift. Sixty-two percent (112/180) were kept entirely outside despite the fact that 82% (147/180) were considered companions rather than working dogs. Almost all of the dogs had been to a veterinarian at least once; only 20% (n=29) were sterilized. Male dogs were significantly less likely to be sterilized than females. Almost half the dogs had had at least one litter. Seventy-six percent (137/180) of dogs knew some basic commands. Sex, source and training to sit/stay/come were significantly associated with whether the dog was sterilized. Dog size, confinement to a yard, trained to leash walk, veterinary visit and region were predictors of roaming freely. Source and trained to leash walk were associated with dog registration. For cats and dogs, education about sterilization seemed to be critically important. For dogs, factors relating to training, which may reflect the strength of the human-animal bond, were also important.

The principal tools currently used in Italy to limit stray dog populations are dog registration and identification, birth control and increasing public awareness. Since 1991, national legislation does not permit euthanasia of unwanted roaming dogs unless they have an incurable condition or are proven to be dangerous. Unattended dogs are placed in long-term shelters in questionable conditions where they often remain for most of their lives. Kennel management is one of the most critical aspects of animal welfare, particularly as the number of stray dogs is rising faster than the current rate of adoption. The Istituto Zooprofilattico Sperimentale dell'Abruzzo e del Molise 'G. Caporale' (IZS A&M) manages two kennels in Teramo and uses standard management procedures that include regular behavioural and physical examinations. All results recorded in a computer database were analysed to identify production goals and improve welfare, in line with a population medicine model. Prevalence and incidence of different pathologies were recorded and analysed to improve veterinary management and organisation. No new dogs were admitted from 2005 onwards, thereby producing an ideal 'closed system' where the effects of rational management can be studied in the absence of unpredictable risks from introduced pathologies. Statistical evaluations revealed major differences between 2006 and 2007. The use of the 'population medicine' management system resulted in improved dog health and welfare, as indicated by the significant reduction in both the prevalence and incidence of major pathologies between 2006 and 2007. A significant improvement was also seen in the control of the incidence of leishmaniasis which reduced sharply during this period.

A cross-sectional telephone survey of randomly selected households examined the extent and types of problems associated with free-roaming dogs and cats in the Teramo province of Italy. The households were sampled randomly within each municipality; municipalities were combined into coastal, central hills and mountain regions for analysis. The survey was conducted in May and June of 2004 with a response rate of 74% (397/536). Ninety percent of respondents (N=356) believed that free-roaming dogs and cats were a problem. They were most commonly concerned about personal safety, followed by animal welfare, public health and environmental sanitation. Sixty-nine percent of respondents (274) actually saw free-roaming dogs or cats where they live. While dogs were most commonly seen, cats were seen in greater numbers. Overall, 10% (39/297) and 5% (21/397) of respondents cared for free-roaming cats and dogs, respectively. Two-thirds of the respondents (251/397) believed that animals were abandoned because the owners lost interest. About 2/3 of respondents (251/397) reported that the community government should have the responsibility for dealing with free-roaming dogs and cats. The respondents supported the idea of building more shelters and controlling the birth rate as control measures rather than euthanizing dogs and cats. The results suggest that free-roaming dogs and cats are a very common sight in this part of Italy with substantial concerns by the residents. However, concerns about the animals' welfare were clearly raised, supporting the laws that make it illegal to euthanize a healthy dog or cat in Italy. Using the information from this study, research on the underlying causes of abandonment of dogs and cats or failing to sterilize them should be undertaken to begin to address this problem. Further, national and regional funding must be provided to support existing laws which should help protect and eventually decrease the numbers of free-roaming and homeless dogs and cats.

The translationally controlled tumor protein (TPT1, also known as TCTP) is a highly conserved, abundantly expressed protein found in mammals as well as in a wide range of other organisms of both the animal and plant kingdom. Initially considered as a growth-related protein, later studies showed TPT1 is endowed with multiple biological activities, including calcium binding. The present study aimed to evaluate the expression of TPT1 in the human placenta and to examine the functional role of the protein in the calcium binding and homeostasis of trophoblast cells. Samples were analyzed by Western blot, reverse transcription-polymerase chain reaction and immunohistochemistry. The effect of TPT1 knockdown by small interfering RNA (siRNA) on calcium uptake and buffering was assessed in the HTR-8/SVneo cell line. TPT1 protein and mRNA were detected in first-trimester and term placenta. In the tissue, TPT1 was localized in the villous trophoblast. TPT1 expression significantly increased during gestation, with the higher protein and mRNA levels reached at term. Recombinant placental TPT1 bound calcium in vitro, while downregulation of the protein levels by siRNA in HTR-8/SVneo cells was associated with a reduced cellular calcium-uptake activity and buffering capacity. These data demonstrate, for the first time, the expression of TPT1 in the human placenta and support a direct role of the protein in placental calcium transport.

BACKGROUND: The translationally controlled tumor protein (TCTP) is an abundantly expressed protein found in a wide range of organisms from both the animal and plant kingdom. Initially described as a growth-related protein, knowledge of the biological actions of TCTP has been recently extended to include calcium binding, regulation of apoptosis, and microtubules stabilization. This report describes expression, distribution, and characterization of TCTP in human prostatic tissues and cell lines. METHODS: Samples were analyzed by Western blot, RT-PCR, immunohistochemistry, and confocal microscopy. Calcium binding activity of the recombinant human prostatic protein was evaluated on a calcium overlay assay. A public SAGE database was analyzed to determine TCTP expression levels in normal and cancer tissues. RESULTS: TCTP protein and mRNA were detected in all the specimens and cell lines analyzed. The protein was mainly expressed by the secretory luminal epithelial and basal layer cells. A significant amount of protein was present in the prostatic fluids. Subcellular distribution studies in prostate epithelial cells detected the protein in the cytoplasm in interphase and colocalized with tubulin during mitosis. The calcium binding capacity of prostatic TCTP was shown in vitro. Finally, SAGE data indicated TCTP as the calcium binding protein with the highest expression levels among those examined. CONCLUSIONS: The results of the present study demonstrate, for the first time, the expression of TCTP in the human prostate and in prostate cancer cells, and suggest the involvement of the protein in key-processes such as apoptosis, cellular differentiation, and in the control of sperm functions.
