RESUMO

Streptococcus (S.) species are important pathogens that cause mastitis in sheep. The study aimed to examine Streptococcus species in sheep milk with subclinical mastitis, assessing their prevalence, antimicrobial resistance, and virulence genes. A total of 200 milk samples were collected from sheep farms in Izmir's five districts. Out of 32 (28.6%) Streptococcus isolates identified by phenotypic methods, 25 were genotypically identified as S. uberis, 5 as S. agalactiae, and 2 as S. dysgalactiae. Disk diffusion was used to determine the antimicrobial resistance of the isolates. PCR was employed to identify antimicrobial resistance and virulence genes in the isolates. The highest resistance was found for cloxacillin (100%), and the highest sensitivity was found for florfenicol (84%). The most common resistance gene combination was tetM+tetS (3/32) for S. uberis in 9.4%. A total of five virulence genes were detected. GapC+sua (56.2%) constituted the most common gene pattern. The highest virulence gene gapC was detected in 78.1% (25/32) of the isolates. The cylE gene was not detected (0%) in the isolates. Streptococcus species may play a role in mastitis in sheep, emphasising the need for meticulous hygienic milking practices.

The fermented liquid sector is developing all over the world due to its contribution to health. Our study has contributed to the debate about whether industrially manufactured fermented liquids live up to their claims by analyzing pathogens and beneficial bacteria using a 16S rRNA sequencing technique called metagenomic analysis. Paenibacillus, Lentibacillus, Bacillus, Enterococcus, Levilactobacillus, and Oenococcus were the most abundant bacterial genera observed as potential probiotics. Pseudomonas stutzeri, Acinetobacter, and Collimonas, which have plant-growth-promoting traits, were also detected. The fact that we encounter biocontroller bacteria that promote plant growth demonstrates that these organisms are widely used in foods and emphasizes the necessity of evaluating them in terms of public health. Their potential applications in agriculture may pose a danger to food hygiene and human health in the long term, so our data suggest that this should be evaluated.

In this study, sustainable water-based films were produced via the solvent-casting method. Petroleum-free-based polyvinyl alcohol (PVA) and carbohydrate-based inulin (INL) were used as matrices. Vegetable-waste pumpkin powder was used in the study because of its sustainability and antibacterial properties. Pickering emulsions were prepared using ß-cyclodextrin. The influence of the different ratios of the ß-cyclodextrin/niaouli essential oil (ß-CD/NEO) inclusion complex (such as 1:1, 1:3, and 1:5) on the morphological (SEM), thermal (TGA), physical (FT-IR), wettability (contact angle), and mechanical (tensile test) characteristics of PVA/inulin films were investigated. Moreover, the antibacterial activities against the Gram (-) (Escherichia coli and Pseudomonas aeruginosa) and Gram (+) (Staphylococcus aureus) bacteria of the obtained films were studied. From the morphological analysis, good emulsion stability and porosity were obtained in the Pickering films with the highest oil content, while instability was observed in the Pickering films with the lowest concentration of oil content. Thermal and spectroscopic analysis indicated there was no significant difference between the Pickering emulsion films and neat films. With the addition of Pickering emulsions, the tensile stress values decreased from 7.3 ± 1.9 MPa to 3.3 ± 0.2. According to the antibacterial efficiency results, films containing pumpkin powder and Pickering emulsion films containing both pumpkin powder and a ratio of 1:1 (ß-CD/NEO) did not have an antibacterial effect, while Pickering emulsion films with a ratio of (ß-CD/NEO) 1:3 and 1:5 showed an antibacterial effect against Escherichia coli, with a zone diameter of 12 cm and 17 cm, respectively. Among the samples, the films with ratio of (ß-CD/NEO) 1:5 had the highest antioxidant capacity, as assessed by DPPH radical scavenging at 12 h intervals. Further, none of the samples showed any cytotoxic effects the according to LDH and WST-1 cytotoxicity analysis for the NIH3T3 cell line. Ultimately, it is expected that these films are completely bio-based and may be potential candidates for use in wound healing applications.

In this study, polyvinyl alcohol (PVA) and psyllium husk (PSH)/D-limonene electrospun meshes were produced by emulsion electrospinning for use as substrates to prevent the growth of bacteria. D-limonene and modified microcrystalline cellulose (mMCC) were preferred as antibacterial agents. SEM micrographs showed that PVA-PSH electrospun mesh with a 4% amount of D-limonene has the best average fiber distribution with 298.38 ± 62.8 nm. Moreover, the fiber morphology disrupts with the addition of 6% D-limonene. FT-IR spectroscopy was used to analyze the chemical structure between matrix-antibacterial agents (mMCC and D-limonene). Although there were some partial physical interactions in the FT-IR spectrum, no chemical reactions were seen between the matrixes and the antibacterial agents. The thermal properties of the meshes were determined using thermal gravimetric analysis (TGA). The thermal stability of the samples increased with the addition of mMCC. Further, the PVA-PSH-mMCC mesh had the highest value of contact angle (81° ± 4.05). The antibacterial activity of functional meshes against Gram (-) (Escherichia coli, Pseudomonas aeruginosa) and Gram (+) bacteria (Staphylococcus aureus) was specified based on a zone inhibition test. PPMD6 meshes had the highest antibacterial results with 21 mm, 16 mm, and 15 mm against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa, respectively. While increasing the amount of D-limonene enhanced the antibacterial activity, it significantly decreased the amount of release in cases of excess D-limonene amount. Due to good fiber morphology, the highest D-limonene release value (83.1%) was observed in PPMD4 functional meshes. The developed functional meshes can be utilized as wound dressing material based on our data.

Periodontal disease is the most common infectious disease of cats and dogs which are strongly associated with periodontal pathogens. The primary etiologic factor in the formation of periodontal disease is microbial dental plaque accumulation on teeth. In our research, we aimed to investigate the presence of periodontal disease-related bacterial species in dental plaques of cats and dogs. Specimens collected from 50 cats and 51 dogs with periodontal disease examined in terms of periodontal pathogens by polymerase chain reaction (PCR) using primers directed to 16S rRNA and tdpA genes. Our findings indicate the presence of periodontal disease-related pathogens, especially Porphyromonas gingivalis (cats 96%, dogs 88%), Prevotella nigrescens (cats 90%, dogs 57%) and, Porphyromonas gulae (cats 70%, dogs 39%). In addition, the prevalence of Tannerella forthysia (cats 2%, dogs 4%) well-known pathogen in cats and dogs were isolated with an extremely low percentage. Furthermore, our results suggest that the feline oral cavity microbiota has considerably more diversity than dogs. Consequently, daily oral hygiene practices may become essential for controlling the pathogenic bacteria which have clinical importance and in preventing the propagation of microorganisms in the oral cavity of cats and dogs.

Systemic fungal diseases are the infections caused by false treatment protocols and generally are not taken into consideration especially in the veterinary field. One-humped camels are found in the western side of the Aegean region of our country and bred for wrestling. The aim of this study is the application of diagnosing systemic fungi infection from camel blood samples by the PCR method. In this study, specific primers for DNA topoisomerase II gene sequences were used. As a result, a systemic fungal infection was detected by the nested PCR method from 10 (20%) out of 50 DNA samples taken from camels located on the western side of the Aegean region. In this study, 3 (30%) samples were identified as Candida albicans, 3 (30%) samples were identified as C. glabrata, and 4 (40%) samples were identified as C. parapsilosis. In conclusion, the 20% positive systemic fungal infection rate in one-humped camels observed in the present study showed that the systemic fungal infections are not taken into considerations in veterinary medicine. Further studies are suggested in order to obtain and to maintain extensive data for systemic fungal diseases in our country for one-humped camels.

The aim of this study was to record sacral bone morphometry that may help in selection of the implant type and proper size in sacroiliac separation. For this reason, sacral lengths and width, the length of each sacral vertebrae, distances between cranial and caudal articular processes, vertical and transversal diameters of the cranial endplate, sacral tuberositas and articular surface areas were obtained from 11 dogs. Additionally, the transverse and vertical diameters of the bony structure and sacral canal were measured from six cross-sections. The data of the study were determined to be representative of the sacral values for average-sized dogs, which was confirmed statistically. The highest value was the sacral width among the linear measurements. The ventral sacral length was longer than the dorsal sacral length. The total lateral area of the sacral wing was measured as 677.46 (142.1)mm2. The transverse diameters of the first sacral vertebra important for screw implantation were 46.02 (4.33)mm and 44.18 (5.29)mm in the first and second cross-sections, respectively.

The bones of 64 digits from eight Holstein male cattle were studied quantitatively to determine whether any differences existed respective on the corresponding bones of the different digits. For this purpose, the greatest and abaxial greatest lengths, the smallest diaphysial breadths, the breadths of proximal and distal ends of the proximal and middle phalanges, the lengths of dorsal surfaces, the heights of extensor processes, the greatest diagonal lengths and the middle breadths of the soles from the distal phalanges were measured. Comparison showed that measurements between the right and left sides did not differ significantly. However, differences were found for almost all measurements between the forelimb and hindlimb. The proximal and middle phalanges were found to be shorter and broader in the forelimb and the broadness was more noticeable than the length. The distal phalanx of the medial forefoot had the greatest value in four measurements while the smallest values were found in the lateral hind foot. The results indicated that the morphometry of the digital bones could be important from both phylogenetic and clinical aspects.
