RESUMO

Antibiotics are used in the treatment of bacterial diseases in commercial aquaculture. In this study, we the biological responses of Oreochromis niloticus juveniles upon dietary florfenicol (FFC) administration at 15 mg (1×) and 45 mg kg biomass-1 day-1 (3×) for 10 days in terms of feed intake, survival, biomass, hematological, erythro-morphological, serum biochemical, and histopathological aberrations as compared with controls. FFC caused a dose-dependent reduction in feed intake, survival, and biomass, with marked variations in hematology, hematological indices, and erythrocytic cellular and nuclear abnormalities, suggesting its apparent cytotoxic and nucleotoxic effects. The serum biomarkers increased significantly in a dose-dependent manner, except for calcium and chloride, which decreased significantly. The therapeutic dose (1×) group exhibited marked histopathological aberrations, such as renal tubular epithelial degeneration and a widened lumen in the kidney, as well as glycogen-type vacuolation and cytoplasmic degeneration in the liver during the dosing period. The extent of kidney and liver tissue damage was more prominent in the 3× group. The 1× serum biomarker levels became normal, with the exception of alkaline phosphatase, within 3 weeks of suspension of dosing. The recovery of the measured parameters and histopathological and erythro-morphological changes suggested that the therapeutic dietary biological responses induced by FFC are reversible and safe for O. niloticus.

Nitrogen species such as ammonia and nitrite are considered as major stressors in modern aquaculture practices. We developed enrichments of ammonia oxidising bacteria (AOB) and nitrite oxidising bacteria (NOB) for effective mitigation of nitrogenous wastes in the shrimp culture operations. The objective of this study was to understand the microbial community composition of AOB and NOB enrichments using the V3-V4 region of the 16S rDNA gene by Illumina MiSeq sequencing. The analysis revealed 2948 and 1069 OTUs at 97% similarity index and Shannon alpha diversity index of 7.64 and 4.85 for AOB and NOB enrichments, respectively. Comparative analysis showed that a total of 887 OTUs were common among AOB and NOB enrichments. The AOB and NOB enrichment were dominated by Eubacteria at 96% and 99.7% respectively. Proteobacterial phylum constituted 31.46% (AOB) and 39.75% (NOB) and dominated by α-Proteobacteria (20%) in AOB and γ-Proteobacteria (16%) in NOB. Among the species in AOB enrichment (2,948) two sequences were assigned to ammonia oxidising bacterial group belonging to Nitrosomonas, and Nitrosococcus genera and two belonged to archaeon group comprising Nitrosopumilus and Candidatus Nitrososphaeraea genera. The NOB enrichment was predominated by Nitrospiraceae and Thermodesulfovibrionaceae. Further, the data revealed the presence of heterotrophic bacteria contributing to the process of nitrification and form microcosm with the AOB and NOB. PICRUSt analysis predicted the presence of 24 different nitrogen cycling genes involved in nitrification, denitrification, ammonia and nitrogen transporter family, nitrate reduction and ammonia assimilation. The study confirms the presence of many lesser known nitrifying bacteria along with well characterised nitrifiers.

The presence of maternal antibodies is a major obstacle for eliciting protective immune responses to foot-and-mouth disease (FMD) vaccines in young, growing animals. In this report, we analyzed the ability of inactivated quadrivalent oil emulsified and aluminium hydroxide adjuvanted FMD vaccines to elicit neutralizing antibody responses in growing calves that had maternal antibodies. Our results demonstrate that oil emulsified vaccines but not aluminium hydroxide adjuvanted FMD vaccines could surmount maternal antibodies to elicit strong and significant levels of neutralizing antibody responses in growing claves.

Despite representing the majority of the world's foot-and-mouth disease (FMD)-susceptible livestock, sheep and goats have generally been neglected with regard to their epidemiological role in the spread of FMD. In the present investigations, FMD virus quadrivalent double emulsion (Montanide ISA 206) vaccines were tested in sheep. The oil adjuvant elicited a better immune response at any time than did aluminum hydroxide gel vaccine, and the response developed quicker. The animals maintained their neutralizing antibody titers at >3 log(10) for the duration of the trial (90 days). Sheep were found to be late responders to serotypes A, C, and Asia-1; a clear upward shift in titer was observed at 60 days postvaccination. However, development of the immune response to serotype O in sheep was superior to that in cattle and goats.
