RESUMO

Gac is a carotenoid-rich, healthful tropical fruit; however, its productivity is limited by soil salinity, a growing environmental stress. This study aimed to evaluate the effects of salinity stress on key physiological traits and metabolites in 30-day-old gac seedling leaves, treated with 0, 25-, 50-, 100-, and 150-mM sodium chloride (NaCl) for four weeks to identify potential alarm, acclimatory, and exhaustion responses. Electrolyte leakage increased with increasing NaCl concentrations (p < 0.05) indicating loss of membrane permeability and conditions that lead to reactive oxygen species production. At 25 and 50 mM NaCl, superoxide dismutase (SOD) activity, starch content, and total soluble sugar increased. Chlorophyll a, and total chlorophyll increased at 25 mM NaCl but decreased at higher NaCl concentrations indicating salinity-induced thylakoid membrane degradation and chlorophyllase activity. Catalase (CAT) activity decreased (p < 0.05) at all NaCl treatments, while ascorbate peroxidase (APX) and guaiacol peroxidase (GPX) activities were highest at 150 mM NaCl. GC-MS-metabolite profiling showed that 150 mM NaCl induced the largest changes in metabolites and was thus distinct. Thirteen pathways and 7.73% of metabolites differed between the control and all the salt-treated seedlings. Salinity decreased TCA cycle intermediates, and there were less sugars for growth but more for osmoprotection, with the latter augmented by increased amino acids. Although 150 mM NaCl level decreased SOD activity, the APX and GPX enzymes were still active, and some carbohydrates and metabolites also accumulated to promote salinity resistance via multiple mechanisms.

Salinity tolerance levels and physiological changes were evaluated for twelve rice cultivars, including four white rice and eight black glutinous rice cultivars, during their seedling stage in response to salinity stress at 100 mM NaCl. All the rice cultivars evaluated showed an apparent decrease in growth characteristics and chlorophyll accumulation under salinity stress. By contrast an increase in proline, hydrogen peroxide, peroxidase (POX) activity and anthocyanins were observed for all cultivars. The K(+)/Na(+) ratios evaluated for all rice cultivars were noted to be highly correlated with the salinity scores thus indicating that the K(+)/Na(+) ratio serves as a reliable indicator of salt stress tolerance in rice. Principal component analysis (PCA) based on physiological salt tolerance indexes could clearly distinguish rice cultivars into 4 salt tolerance clusters. Noteworthy, in comparison to the salt-sensitive ones, rice cultivars that possessed higher degrees of salt tolerance displayed more enhanced activity of catalase (CAT), a smaller increase in anthocyanin, hydrogen peroxide and proline content but a smaller drop in the K(+)/Na(+) ratio and chlorophyll accumulation.

Rice seedlings cv. Khao Dawk Mali 105 (salt-sensitive), Luang Anan (moderately salt-tolerant) and Pokkali (salt-tolerant) were exposed to 0, 50, 100 and 150 mM NaCI for 9 d. Salinity stress caused reduction in leaf relative water contents in all cultivars. Shoot length of cv. Pokkali was least affected by salinity stress whereas increased root length in response to salinity stress was apparent in cvs. Khao Dawk Mali 105 and Luang Anan. Increased salinity level also caused reduction in fresh and dry weights in cvs. Khao Dawk Mali 105 and Luang Anan, but had no effect in cv. Pokkali except at 150 mM. Accumulation of total soluble sugars and sucrose in mature leaves were observed in cv. Khao Dawk Mali 105 exposed to high level of salinity whereas their concentrations in cvs. Luang Anan and Pokkali remained the same as control plants. Accumulation of sucrose in cv. Khao Dawk Mali 105 was suggested to be resulted from the alteration of photosynthate partitioning since the activities of sucrose phosphate synthase were not affected by salinity in this cultivar. On the contrary, salinity stress induced an accumulation of starch in cv. Pokkali. It is suggested that partitioning sugars into starch may involve in salinity tolerance by avoiding metabolic alterations.
