The influences of landscape pattern on water quality are dependent on spatial-temporal scales. However, the effects of landscape composition, landscape configuration, and landscape slope metrics on seasonal water quality at different spatial scales remain unclear. Based on the total nitrogen, total phosphorus, nitrate-N, and ammonium-N data from 26 sampling sites in the Qingshan Lake watershed, this study coupled landscape pattern analysis, redundancy analysis, and partial redundancy analysis to quantify the spatiotemporal scale effects of landscape pattern on riverine nitrogen (N) and phosphorus (P) concentrations. The results showed that: ① The explanatory ability of landscape pattern at the sub-watershed scale on riverine N and P concentrations was 6.8%-8.4% higher than that at the buffer scale, and this effect was more obvious in the dry season. ② At the sub-watershed scale, the percentage of forestland and the interspersion and juxtaposition degree of residential land had a greater influence on riverine N and P concentrations. At the buffer scale, the slope of farmland and residential land and the aggregation degree of forestland patches were the key factors affecting riverine N and P concentrations. ③ The contribution rate of landscape configuration to riverine N and P concentration variations (20.1%-36.5%) was the highest. The sensitivity of the effect of landscape configuration on riverine N and P concentrations to seasonal changes was the highest, and the effect of landscape slope on riverine N and P concentrations had the highest sensitivity to spatial scale changes. Therefore, landscape pattern-regulated non-point source pollution should be considered from a multi-scale perspective. These results can provide scientific basis for the formulation of landscape pattern optimization measures aiming at non-point source pollution control.

OBJECTIVE: To summarize the preliminary application experience of intraoperative ultrasound with burr hole probe in minimally invasive neurosurgery and to explore its application value. METHODS: Thirty-one patients who underwent intraoperative ultrasound guided puncture with burr hole probe in our center from August 2018 to May 2024 were collected, including 16 cases of ventriculoperitoneal shunt operation, 6 cases of assisted stereotactic needle biopsy, 3 cases of intracranial pressure probe implantation in lateral ventricle, 3 cases of brain abscess puncture for external drainage, and 3 cases of intracranial cyst puncture and peritoneal drainage. During the procedures, the burr hole probe was used to locate the intracranial targets and guide the puncture. The postoperative computed tomography (CT) scans or combined postoperative pathological results could verify the accuracy of puncture. In addition, the intervention effect and recovery status of patients were also recorded. RESULTS: The intraoperative ultrasound with burr hole probe could clearly display all the purposed targets and accurately guide the puncture procedures in all cases. All patients achieved satisfactory diagnostic and therapeutic results without new neurological dysfunction and serious complications. CONCLUSIONS: The intraoperative ultrasound with burr hole probe is an effective device for demonstrating intracranial structures. It not only enables minimally invasive and precise diagnosis or treatment of many neurosurgical diseases, but also is simple and safe to operate, which has important promotional value in the neurosurgery.

Based on the continuous data of O3, NO, NO2, and NOx and the meteorological data from March 2019 to February 2020 at six atmospheric composition observation stations in Shanxi Province, the characteristics and influence factors of O3 volume fractions were studied using statistical analysis and backward trajectory analysis. The results showed that O3 volume fractions were generally higher from April to September and lower from October to the following March. During the study period, O3 pollution represented by φ(MDA8O3), i.e., the maximum daily 8-h average of O3 volume fractions, was the most serious at the Jincheng and Linfen stations in the south of Shanxi, followed by that in the Wutaishan, Shuozhou, and Datong stations in the north, with the least pollution occurring at the Taiyuan station in the middle. There were differences between the urban and alpine stations, although their seasonal O3 volume fractions were both summer > spring > autumn > winter. O3 volume fractions at the urban station were usually lower than those at the alpine station; O3 at the urban station might have been influenced by photochemical reactions with precursor NOx; however, this was not the main source of high O3 at the alpine station. The peak and valley values appeared at 15:00 and 06:00, respectively, at the urban station, whereas they appeared at 20:00 and 10:00, respectively, at the alpine station, representing diametrically opposite diurnal variation patterns. Further, the daily amplitude of O3 at the urban station was much larger than that at the alpine station. For urban stations specifically, temperature was the most important meteorological factor affecting O3 volume fraction, compared with sunlight hours, precipitation, and total cloud cover. The NO2 volume fraction in the daytime affected the daily amplitude of O3; although the photochemical generation potential of O3 at the Taiyuan station was good, the O3 volume fractions were the lowest among urban stations due to strong NO titration. The higher O3 corresponded to lower NOx in which NO2 was dominant, and the higher NOx was largely composed of NO, under which conditions O3 would be depleted completely. The surface wind that affected O3 volume fractions of all stations primarily came from the southeast, south, and southwest, and specific wind speed led to the increase in O3 volume fraction. The geographical situation of the station would cause the difference in the transport of atmospheric pollutants, whereas the horizontal transmissions of high O3 from the North China Plain and Fenwei Plain were likely to be the common reason for the increase in O3 volume fraction in Shanxi.

Accurate quantification of non-point source pollution is an important step for non-point source pollution control and management at the watershed scale. Considering the non-point source pollution from baseflow, an improved export coefficient model (IECM) on a weekly scale was established based on the traditional export coefficient model (ECM), which was then used to estimate the surface flow non-point source total nitrogen (TN) loads contributed by different land use types of the Shangwu River watershed in the Qiandao Lake Region. The results showed that IECM performed well for the predictions of TN loads in the studied watershed, with the Nash-Sutcliffe efficiency coefficient (NSE) and R2values of 0.82 and 0.77 (P<0.01) for the calibration period and 0.87 and 0.84 (P<0.01) for the validation period, respectively. The IECM estimated TN exports through surface flow and baseflow were 5.74 kg·(hm2·a)-1and 9.85 kg·(hm2·a)-1 from the Shangwu River watershed in the period of Nov. 2020 to Oct. 2021, which accounted for 36.80% and 63.20% of the corresponding streamflow TN load, respectively. Without consideration of the baseflow non-point source TN pollution, the ECM-estimated surface flow TN loading was 54.21% higher than that estimated by IECM. Obviously, attributing baseflow non-point source pollution to surface flow directly would lead to a serious load overestimation of surface flow. According to IECM, the estimated TN export intensity through surface flow from paddy fields, grasslands, woodlands, rainfed croplands, and residential lands was 10.95, 5.42, 5.20, 12.34, and 2.77 kg·(hm2·a)-1, respectively, which accounted for 5.80%, 4.00%, 26.55%, 0.38%, and 0.03% of the corresponding total streamflow TN loads. Therefore, the future management of non-point source nitrogen pollution in the studied watershed should focus mainly on the prevention and management of groundwater non-point source pollution and control of load export from surface flow on cultivated land (paddy fields and rainfed croplands).

Methane (CH4) and nitrous oxide (N2O) are two extremely important greenhouse gases in the atmosphere. Nitrogen fertilizer is an important factor affecting CH4 and N2O emissions in rice fields. Rational application of nitrogen fertilizer can not only promote high yields of rice but also reduce greenhouse gas emissions. Existing studies have shown that nitrogen reduction and optimal application can effectively improve the nitrogen use efficiency of rice on the basis of ensuring the yield and reduce the loss of N2O caused by nitrification and denitrification of excessive nitrogen in soil. Fertilization times and fertilizer types have significant effects on CH4 and N2O emissions in paddy fields. In this study, a field experiment was conducted for two consecutive years (2019-2020) to study the effects of fertilizer application on CH4 and N2O emissions from rice fields by setting up four treatments consisting of no fertilizer (CK), customary fertilizer application by farmers (CF), twice fertilizer (TT), and 20% replacement of chemical fertilizer by organic fertilizer (OF) using static chamber-gas chromatography. Additionally, the effect of integrating rice yield and integrated global warming potential (GWP) on the greenhouse gas emission intensity (GHGI) per unit of rice yield was analyzed to explore fertilizer application for yield increase and emission reduction in a typical rice growing area in the middle and lower reaches of Yangtze River. The results showed that:① compared with those of CK, the fertilizer treatments reduced CH4 emissions by 14.6%-25.1% and increased N2O emissions by 610%-1836% in both years; ② compared with those of CF, both the TT and OF treatments showed a trend of increasing CH4 emissions and reducing N2O emissions. CH4 emissions increased by 1.8% (P>0.05) and 14.0% (P<0.05), respectively. The annual average of N2O emissions decreased by 63.3% (P<0.05) and 49.2% (P<0.05) in both the TT and OF treatments, respectively. ③ Compared with that of CK, both fertilizer applications increased rice yield and reduced GHGI; compared with that of CF, the OF and TT treatments increased the average annual rice yield by 17.0% and 10.7%, respectively, and reduced GHGI by 6.8% and 13.7%, respectively. The OF treatment had a better yield increase than that of the TT treatment, and the TT treatment had a slightly better emission reduction than that of the OF treatment. In terms of combined yield and GHG emission reduction, both twice fertilizer (TT) and 20% replacement of chemical fertilizer by organic fertilizer (OF) could reduce the intensity of GHG emission per unit of rice yield and achieve yield increase and emission reduction while ensuring rice yield.

The development of a standardized eDNA detection process is the primary step in improving the accuracy and efficiency of eDNA detection. In this study, primers and probes with high specificity were selected to identify the COI gene of Acanthopagrus latus. Through experiments on the influence of different water quantities, methods of water sample preservation and water bathing times on the result of eDNA detection, the accuracy of this method for extracted water samples was improved. Specifically, a water bathing time of 6 h provided an optimal eDNA concentration from the water sample. After 6 h, the concentration began to decrease, so 6 h was determined to be the best water bathing time for A. latus. Five water extraction volumes (250 mL, 500 mL, 1 L, 2 L, and 3 L) were tested, and there was a positive correlation between water extraction volume and the DNA concentration in the water sample. Different water sample preservation methods were also compared, and it was found that at ≤7 d, the concentration obtained with the cryopreservation method for different water extraction volumes was higher than that obtained with the ethanol preservation method. In this study, we established and optimized a technical procedure for eDNA-based detection of A. latus in aquatic environments. We hope to apply this method in field investigations and provide a reference for the study of eDNA in other fishes.

Adsorption is the primary mechanism of antibiotic removal in wastewater treatment plants, wherein the extracellular polymeric substances (EPS) of the activated sludge play an important role. Due to their complex characteristics, the effect of EPS components on antibiotic adsorption is unknown. Therefore, in this study, the role of main components of EPS in antibiotic adsorption was explored using enzymatic treatment. The results revealed that proteinase K and α-amylase can efficiently hydrolyse the proteins and polysaccharides of EPS. The protein content of EPS reduced from 31.25 mg/g VSS to 21.53, 18.75, and 10.76 mg/g VSS, after treatment with proteinase K, α-amylase and their combination, respectively; the polysaccharides content also observed a similar decrease from 15.20 mg/g VSS to 8.22, 7.83, and 6.03 mg/g VSS, respectively. The humic substance in EPS was stable during enzymatic treatment. The equilibrium adsorption capacity of activated sludge treated by enzyme for trimethoprim (TMP)- a typical antibiotic, was significantly increased from 2.19 µg/g VSS to 4.68, 5.34, and 8.36 µg/g VSS after treatment with proteinase K, α-amylase and their mixture. The adsorption process was adequately described by pseudo-second-order kinetic model. A multivariable linear regression model was subsequently used to quantify the adsorption capacity of activated sludge for TMP considering the concentration of EPS components. The modelling and validated results showed that the model could satisfactorily predict the TMP adsorption capacity. The results of this study can provide new insights into the role of EPS on antibiotic transformation in biological wastewater treatment systems.

BACKGROUND: Cognitive decline is one of the greatest concerns for patients with Parkinson's disease (PD) and their care partners. Repetitive transcranial magnetic stimulation (rTMS) is a nonpharmacological treatment option used to improve cognitive function in PD, but its efficacy is unclear. We performed a meta-analysis to determine whether rTMS improves cognition in PD patients. METHODS: Eligibility criteria (PICOS) were as follows: (1) 'P': The patients participating were diagnosed with idiopathic PD; (2) 'I': Intervention using rTMS; (3) 'C': Sham stimulation as control; (4) 'O': The outcome of the study included cognitive evaluations; (5) 'S': The study adopted randomized controlled design. The standardized mean difference (SMD) of change of score was applied to measure efficacy, and we used Version 2 of the Cochrane tool to assess risk of bias. RESULTS: Twelve studies met the inclusion criteria. Compared with sham-controlled group, the pooled result showed a non-significant short-term effect of rTMS on global cognition (SMD: -0.15, 95% CI: -0.59 to 0.29, I2 = 36.7%), executive function (SMD: 0.03, 95% CI: -0.21 to 0.26, I2 = 0.0%), and attention and working memory (SMD: 0.05, 95% CI: -0.25 to 0.35, I2 = 0.0%). Long-term outcomes were either shown to be statistically nonsignificant. CONCLUSIONS: Based on a limited number of studies, rTMS fails to improve cognition in PD. We call for additional high-quality randomized controlled trials with adequate sample sizes to determine the efficacy of rTMS.

BACKGROUND: Erdosteine is a mucolytic drug and has antioxidant activity. OBJECTIVE: The study aimed to develop an HPLC method for determination of erdosteine and its impurities in erdosteine bulk drug and to identify the main impurities to help improve the quality of erdosteine bulk drug. METHOD: The chromatographic separations were performed on a CAPCELL PAK C18 column (250 mm × 4.6 mm id, 5 µm). Acetonitrile-0.01 mol/L citric acid solution (13 + 87, by volume) pumped at a flow rate of 1.0 mL/min was used as the mobile phase. The detection wavelength was 254 nm. Two main impurities in erdosteine bulk drug were enriched by an ODS column chromatography and oxidative degradation, respectively, and then both were purified by semipreparative HPLC. Finally, their structures were identified by a variety of spectral data (MS, 1H NMR and 13C NMR). RESULTS: Good separations of erdosteine and its related impurities were observed. A new impurity was confirmed as ethyl ({2-oxo-2-[(2-oxotetrahydro-3-thiophenyl) amino] ethyl} sulfanyl)acetate, which was erdosteine ethyl ester, and was produced in the refining process of erdosteine bulk drug when using ethanol as a refining solvent. Another impurity was confirmed as ({2-oxo-2-[(2-oxotetrahydro-3-thiophenyl)amino]ethyl}sulfinyl) acetic acid, which was an erdosteine oxide. CONCLUSIONS: An HPLC method for determination of erdosteine and its related impurities was developed and validated. Two main impurities in erdosteine bulk drug were isolated and identified. Avoiding ethanol as the refining solvent can improve the purity of erdosteine bulk drug. HIGHLIGHTS: A new process-related impurity and an oxidative degradation impurity in erdosteine bulk drug were isolated and identified.

BACKGROUND: Parkinson's disease (PD) is the second most common neurodegenerative disease worldwide, which currently lacks disease-modifying therapy to slow down its progression. Idebenone, a coenzyme Q10 (CQ10) analogue, is a well-known antioxidant and has been used to treat neurological disorders. However, the mechanism of Idebenone on PD has not been fully elucidated. This study aims to predict the potential targets of Idebenone and explore its therapeutic mechanism against PD. METHOD: We obtained potential therapeutic targets through database prediction, followed by Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis. Next, we constructed and analyzed a protein-protein interaction network (PPI) and a drug-target-pathway-disease network. A molecular docking test was conducted to identify the interactions between Idebenone and potential targets. Lastly, a PD cell line of SH-SY5Y overexpressing mutant α-synuclein was used to validate the molecular mechanism. RESULT: A total of 87 targets were identified based on network pharmacology. The enrichment analysis highlighted manipulation of MAP kinase activity and the PI3K-AKT signaling pathway as potential pharmacological targets for Idebenone against PD. Additionally, molecular docking showed that AKT and MAPK could bind tightly with Idebenone. In the cell model of PD, Idebenone activated autophagy and promoted α-synuclein degradation by suppressing the AKT/mTOR pathway. Pretreating cells with chloroquine (CQ) to block autophagic flux could diminish the pharmacological effect of Idebenone to clear α-synuclein. CONCLUSION: This study demonstrated that Idebenone exerts its anti-PD effects by enhancing autophagy and clearance of α-synuclein, thus providing a theoretical and experimental basis for Idebenone therapy against PD.

Based on hourly concentration data of PM10 and PM2.5 from 2017 to 2019 and wind speed and direction data at the corresponding times in Yangquan, an urban valley of the Taihang Mountains, the characteristics of air transport in a cross-valley urban area and their influence on aerosols in Yangquan were analyzed using the HYSPLIT model, cluster analysis, and the potential source contribution factor and concentration weight trajectory methods. The pollutant transport characteristics on the east and west sides of the Taihang Mountains were evaluated. The diurnal variation of the aerosols showed a single peak and single valley, with continuous improvements in aerosol concentration from evening to the morning of the next day. The maximum aerosol concentration occurs at 10:00-11:00 in winter and at 09:00 in other seasons, whereas the minimum value occurs at 15:00-16:00. The monthly mean aerosol concentration in Yangquan was highest in January and lowest in August, and PM10 was also high in March and April. The most frequent surface wind directions in Yangquan are easterly and westerly due to the topography of the cross valley. The average concentrations of aerosols were highest in the calm wind weather. Furthermore, the dust weather caused by the westerly wind in spring and autumn and the easterly transport in winter also causes an increase in aerosol concentration in Yangquan. The backward trajectories in combination with the pollution characteristics showed that 26.2% of the pollution trajectories are in spring, 36.4% are in autumn, and 33.7% are in winter, and that these are mainly distributed in the southwest and southeast of Yangquan and also in the northeast in winter. There was significant fine particle transport on both sides of the mountains, and the trajectories originating from or passing through the west side of the Taihang Mountains also transport coarse particles. As indicated by the pollution trajectory, the westerly air transport has an impact on PM10 in excess of the standard rate. The easterly transport has larger noise and mainly affects the exceedance rate of PM2.5. The main potential source areas of aerosol pollution in Yangquan differ in different seasons. The potential source areas are to the southwest and southeast of Yangquan in spring, the southwest and south of Yangquan in autumn, and the south and east of Yangquan in winter. The areas with high PSCF and CWT in PM2.5 were in the southeast region, but those for PM10 were in the southwest region. The area southeast of Shanxi and the border area north of Henan are the main source areas. Based on the influence of aerosol pollution transmission in Yangquan, the cross-valley urban area is affected simultaneously by the areas east and west of the Taihang Mountains, especially by PM2.5 transport from east to west.

The extracellular polymeric substances (EPS) of activated sludge are a mixture of high molecular weight polymers secreted by microorganisms, which are mainly composed of proteins, polysaccharides and humic substances. It is widely accepted that EPS have a good adsorption ability for pollutants with different functional groups. However, recent studies showed the EPS had an inhibitory effect on pollutant sorption, which is contradictory to previous viewpoint. Therefore, in this study, three types of activated sludge with different EPS contents and compositions were used to investigate the role of EPS in an antibiotic-trimethoprim (TMP) sorption process at environmentally relevant concentration. The in situ experiments results showed the adsorption capacity of activated sludge for TMP were increased from 2.98, 5.37 and 28.33 µg/g VSS to 7.87, 12.93 and 150.24 µg/g VSS in nitrifying activated sludge, wastewater treatment plant activated sludge and anaerobic ammonia-oxidized activated sludge, respectively after EPS extracted. The adsorption process can be well described by the pseudo-second-order kinetic model. Results of zeta potential, contact angles and infrared spectrum showed TMP replacing proteins embedded into the cell membrane enhancing the TMP adsorption capacity of activated sludge after EPS extraction. Our results demonstrated that less proteins in EPS of activated sludge is more beneficial for TMP adsorption removal.

Macrophage autophagy defect is closely related to the progression of atherosclerosis (AS) and is regulated by the triggering receptor expressed on myeloid cell 2 (TREM2). TREM2 is a key factor in the development of Alzheimer's disease (AD), the deficiency of which leads to anomalous autophagy in microglia. However, the role of TREM2 in the autophagy of plaque macrophages is still unclear. Geniposide (GP) can inhibit AS progression and enhance macrophage autophagy, although the underlying mechanisms remain unknown. We found that high-fat diet (HFD) feeding significantly increased TREM2 levels and inhibited autophagy in the macrophages of ApoE[Formula: see text] mice. TREM2 overexpression in RAW264.7 macrophages decreased autophagy via activation of mTOR signaling. GP inhibited the progression of AS in ApoE[Formula: see text] mice, reinforced macrophage autophagy, and downregulated TREM2 by inhibiting mTOR signaling. Taken together, augmenting the autophagy levels in plaque macrophages by inhibiting the TREM2/mTOR axis can potentially impede atherosclerotic progression. The promising therapeutic effects of GP seen in this study should be validated in future trials, and the underlying mechanisms have to be elucidated in greater detail.

RESUMO

The phylogenetic tree of Chinese Nemobiinae species was reconstructed based on one mitochondrial COI fragment and two nuclear ribosomal loci (18S and 28S). According to both of concatenated and coalescent trees, Dianemobius, Polionemobius and Pteronemobius (except P. jianfenglingensis Liu Shi) were shown to be monophyletic with highly nodal supports. As to P. jianfenglingensis, it should be moved into a newly established genus, Giganemobius Shen He gen. nov. In addition, a new mute species named as Qionemobius hsterus Shen He gen. nov. sp. nov. was reported. It differs from other genera by having six apical spurs on hind tibia, but lacking forewings. The type specimens are deposited in Museum of Biology, East China Normal University (ECNU).

Marek's disease (MD) is an infectious malignant T-cell lymphoma proliferative disease caused by Marek's disease virus (MDV). In recent years, the emergence of very virulent (vv) and/or very virulent plus (vv +) strains of MDV in the field has been suggested as one of the causes of vaccination failure. The pathogenicity of the MDV strain GX18NNM4, isolated from a clinical outbreak in a broiler breeder flock that was vaccinated with CVI988/Rispens, was investigated. In the vaccination-challenge test, GX18NNM4 was able to break through the protections provided by the vaccines CVI988 and 814. It also significantly reduced body weight gain and caused marked gross lesions and a large area of infiltration of neoplastic lymphocyte cells in the heart, liver, pancreas, etc. of the infected birds. In addition, the expressions of programmed death 1 (PD-1) and its ligand, programmed death ligand 1 (PD-L1), in the spleens and cecal tonsils (CTs) of the unvaccinated challenged birds were significantly increased compared to those in the vaccinated challenged birds, indicating that the PD-1/PD-L1 pathway is related to immune evasion mechanisms. The results showed that the GX18NNM4 strain could cause severe immunosuppression and significantly decrease the protections provided by the current commercial vaccines, thus showing GX18NNM4 to be a vv + MDV strain.

OBJECTIVE: The cystic gliomas are the special type of malignant tumors in the brain and often lead to unsatisfied prognosis, but the microsurgical resection is still the most important treatment. However, they are difficult to be totally removed with craniotomy, especially for those who have flimsy cyst walls. Recently, we attempted to resect them via an innovative surgical technique, "Gelfoam Padding", in order to improve the total resection rate of the tumors safely. PATIENTS AND METHODS: Fifteen patients suffering intracerebral cystic gliomas underwent surgical intervention via "Gelfoam Padding" technique between 2015 and 2018, and the different histopathological results and their features of cyst walls were recorded. Then, the total resection rate of tumors as well as the complications after surgeries were analyzed to assess the applied value of this technique. RESULTS: All the patients were improved in the clinical symptoms after the operations. According to the intraoperative assessment and MRI examinations performed within 72 h after surgery, total resection of the tumor was achieved in all patients. Besides, there were no serious postoperative complications in these cases with this technique. CONCLUSION: The cystic glioma with the flimsy wall was the best applied indication of "Gelfoam Padding" technique, which could not only improve the total resection rate of tumors, but also be safe for the patients.

Sepsis-induced liver injury is very common in intensive care units. Here, we investigated the effects of 6-gingerol on sepsis-induced liver injury and the role of the Nrf2 pathway in this process. 6-Gingerol is the principal ingredient of ginger that exerts anti-inflammatory and antioxidant effects. Using cecal ligation and puncture (CLP) to induce polymicrobial sepsis and related liver injury, we found that mice pre-treated with 6-Gingerol showed less incidences of severe liver inflammation and death than untreated CLP groups. 6-Gingerol administration also inhibited the expression of pyroptosis-related proteins, including NOD-like receptor protein 3 (NLRP3), IL-1ß, and caspase-1. Consistent with these findings, 6-gingerol reduced the effects of pyroptosis induced by lipopolysaccharide (LPS) and adenosine 5'-triphosphate (ATP) in RAW 264.7 cells, as evidenced by IL-1ß and caspase-1 protein levels in the supernatant and propidium iodide (PI) staining. 6-Gingerol was shown to activate the Nrf2 pathway in vivo and in vitro. Notably, Nrf2 siRNA transfection nullified the inhibitory effects of 6-gingerol on pyroptosis in vitro. In summary, these findings suggested that 6-gingerol alleviated sepsis-induced liver injury by inhibiting pyroptosis through the Nrf2 pathway.

Lactic acid bacteria (LAB) are a group of gut commensals increasingly recognized for their potential to deliver bioactive molecules in vivo. The delivery of therapeutic proteins, in particular, can be achieved by anchoring them to the bacterial surface, and various anchoring domains have been described for this application. Here, we investigated a new cell anchoring domain (CAD4a) isolated from a Lactobacillus protein, containing repeats of a SH3_5 motif that binds non-covalently to peptidoglycan in the LAB cell wall. Using a fluorescent reporter, we showed that C-terminal CAD4a bound Lactobacillus fermentum selectively out of a panel of LAB strains, and cell anchoring was uniform across the cell surface. Conditions affecting CAD4a anchoring were studied, including temperature, pH, salt concentration, and bacterial growth phase. Quantitative analysis showed that CAD4a allowed display of 105 molecules of monomeric protein per cell. We demonstrated the surface display of a functional protein with superoxide dismutase (SOD), an antioxidant enzyme potentially useful for treating gut inflammation. SOD displayed on cells could be protected from gastric digestion using a polymer matrix. Taken together, our results show the feasibility of using CAD4a as a novel cell anchor for protein surface display on LAB.

Objective::To establish differential metabolites between different varieties of Angelica sinensis, and provide reference for breeding, introduction, regional cultivation and ecological cultivation of new varieties of A. sinensis. Method::Comprehensive non-target metabonomics analysis was conducted for five new varieties of A. sinensis collected at the same time from the same origin: Mingui No. 1 (MG1), Mingui No. 2 (MG2), Mingui No. 4 (MG4), Mingui No. 5 (MG5), and Mingui No. 6 (MG6). The 50% methanol extract of each variety was taken, and then the differential metabolites among varieties were found by using ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS), software Progenesis QI, principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA), and non-targeted metabonomics analysis. Differential metabolites were identified based on precise molecular weight, secondary fragments, KEGG database, HMDE database and related literature information. Result::The results of UPLC-Q-TOF-MS and multivariate statistical analysis showed that there were significant differences in the metabolites of five Angelica varieties. As compared with MG1, the contents of chlorogenic acid, ferulic acid, tryptophan and ferulic aldehyde were significantly lower in MG2, MG4, MG5 and MG6, while the contents of ligustilide, coumarin, bovine keratin, palmitin, protocatechualdehyde and linolenic acid were significantly higher (P<0.05). The results of multivariate statistical analysis showed that the metabolites of MG2 and MG5 were similar with those of MG6, but were significantly different from those of MG4.In addition, 38 distinct metabolites were identified, involving 7 potential targeted metabolic pathways. Different varieties of A. sinensis could regulate the synthesis of their metabolites through phenylpropanoid biosynthesis and sesquiterpene-like compounds metabolism, lipid metabolism, amino acid metabolism, carbohydrate metabolism, nucleotide metabolism, carotenoids, linolenic acid, linoleic acid and some other metabolic pathways. Conclusion::UPLC-Q-TOF-MS and Progenesis QI metabonomics techniques were used to compare the chemical constituents of different varieties of A. sinensis from the overall level. The differences and their regularities were found, which could provide reference for quality control, variety sorting, identification, breeding and ecological planting of A. sinensis.

Monthly phytolith-occluded organic carbon (PhytOC) content in living leaves and litterfall of Moso bamboo (Phyllostachys edulis) were measured for a year. The PhytOC sequestration rate in living leaves of different months were compared with that in annual litterfall to determine the optimum sampling time of living leaves for estimating PhytOC sequestration rate of Moso bamboo. The contents of phytoliths and PhytOC in living leaves of Moso bamboos were 23.45-101.07 g·kg-1 and 0.73-1.98 g·kg-1, respectively, with significant difference among different months. The monthly PhytOC sequestration rates of living leaves of Moso bamboo in different months ranged from 0.75 to 7.68 kg·hm-2·a-1. The maximum and minimum rates of the PhytOC sequestration occurred in December and April respectively, with significant difference between them. There was no difference between the PhytOC sequestration rate in living leaves of Moso bamboos in February or December and that of litterfall in the whole year. Therefore, February or December should be the optimal month of sampling living leaves for estimating the PhytOC sequestration rate of Moso bamboo stands.
