OBJECTIVE: To explore curative effect of conservative treatment of supination-lateral rotation (SER) with type Ⅲ and Ⅳ ankle fracture by bone setting technique. METHODS: From January 2017 to December 2019, 64 patients diagnosed with SER with type Ⅲ and Ⅳ ankle fracture were treated with manipulative reduction and conservative treatment (manipulation group) and surgical treatment with open reduction and internal fixation (operation group), 32 patients in each group. In manipulation group, there were 17 males and 15 females, aged from 15 to 79 years old with an average of (51.42±13.68) years old;according to Lauge-Hansen classification, there were 8 patients with supination external rotation type Ⅲ and 24 patients with type Ⅳ. In operation group, there were 13 males and 19 females, aged from 18 to 76 years old with an average of (47.36±15.02) years old;7 patients with type Ⅲ and 25 patients with type Ⅳ. Displacement of ankle fracture was measured by Digimizer software, and compared before treatment, 3 and 12 months after treatment between two groups. Lateral medial malleolus displacement, lateral medial malleolus displacement, lateral malleolus displacement, lateral malleolus displacement, lateral malleolus contraction displacement and posterior malleolus displacement were measured and compared between two groups. Mazur score was used to evaluate ankle joint function. RESULTS: All patients were followed up from 12 to 36 months with an average of (17.16±9.36) months. There were statistical differences in lateral medial malleolus displacement, lateral medial malleolus displacement, lateral malleolus displacement, lateral malleolus displacement, lateral malleolus contraction displacement and posterior malleolus displacement in manipulation group before and after reduction(P<0.05). Compared with operation group, there were no statistically significant differences in lateral malleolus shift, lateral malleolus shift, lateral malleolus contraction shift(P>0.05), while there were statistically significant differences in lateral malleolus shift, posterior malleolus shift up and down (P<0.05). Mazur scores of ankle joint at 3 months after treatment in manipulation group and operation group were 68.84±13.08 and 82.53±7.31, respectively, and had statistical differences(P<0.05), while there was no difference in evaluation of clnical effect(P>0.05). There were no differences in Mazur score and evaluation of clnical effect between two groups at 12 months after treatment (P>0.05). CONCLUSION: Bone setting technique could effectively correct lateral displacement of medial malleolus, lateral displacement of medial malleolus, lateral displacement of lateral malleolus and lateral contraction displacement of lateral malleolus in supination lateral rotation type Ⅲ and Ⅳ ankle fracture, and has good long-term clinical effect, which could avoid operation for some patients and restore ankle function after fracture.

OBJECTIVE: To observe the radiological change and curative effect of cervical spondylosis treated with cervical vertebra pulling therapy in young people. METHODS: A total of 65 patients(shedding in 5 patients and final inclusion in 60 patients) with cervical spondylosis who were treated from January 2018 to September 2019 were randomly divided into treatment group and control group according to the digital table method. There were 30 patients in treatment group, including 14 males and 18 females, aged from 20 to 44 years old with an average of(29.83±6.99) years, who were treated with cervical vertebra pulling therapy(once a week, 4 times in total). The control group consisted of 30 cases, 12 males and 18 females, aged from 18 to 43 years old with an average of (31.77±5.93) years, who received sitting traction therapy(once a week, 4 times in total). The changes of C2-C7 Cobb angle, arc-chord distance and T1 slope (T1S) in two groups were observed before treatment and 1 month after treatment, for intra-group and inter-group comparison;and the changes of numerical rating scale(NRS) in two groups were observed before treatment and 1, 3 months after treatment, for intra-group and inter-group comparison. RESULTS: Sixty patients were followed up for (3.2±0.3) months. There were no significant differences in NRS, C2-C7 Cobb angle, arc-chord distance and T1S between two groups before treatment (P>0.05). One month after treatment, the NRS, C2-C7 Cobb angle, arc-chord distance and T1S were(1.67±0.76) scores, (16.55±6.01)°, (10.95±4.04)mm, (18.95±4.19)° in treatment group and(1.40±0.86) scores, (10.23±5.94) °, (6.11±4.17) mm, (13.34±4.25)° in control group respectively. C2-C7 Cobb angle, arc-chord distance and T1S in treatment group were better than those in control group (P<0.05); there was no significant difference in NRS between two groups (P>0.05). Compared with before treatment, there were statistically significant differences in C2-C7 Cobb angle, arc-chord distance and T1S in treatment group(P<0.05), but no statistically significant differences in control group(P>0.05). Three months after operation, NRS of treatment group was (1.60±0.62) scores and that of control group was (4.17±0.70) scores. The treatment group was better than the control group(P<0.05). The scores of treatment group after treatment were lower than those before treatment(P<0.05), and there was no significant difference in control group before and after treatment(P>0.05). CONCLUSION: Cervical vertebra pulling method and cervical vertebra sitting traction can relieve the pain symptoms of patients with cervical spondylosis, but the effect of cervical vertebra pulling method is more durable;cervical vertebra pulling method can correct abnormal cervical sagittal parameters.

BACKGROUND: In recent years, the development of high-throughput sequencing technology has promoted the rapid development of circRNA-related research. Studies have found that circRNA plays a key role in a variety of tumors, but few people study the role of circRNA in nasopharyngeal carcinoma. Under comprehensive treatments, the 5-year survival rate can reach about 70%, but some patients still have distant metastases or recurrences after treatment. Therefore, it is very important to study the molecular mechanisms of the proliferation and invasion of nasopharyngeal carcinoma. METHODS: QRT-PCR was applied to detect the relative expression level of circFOXM1 in NPC and nasopharyngeal epithelial cell lines. We knocked down circFOXM1 and studied the influence of circFOXM1 on NPC cells. Nuclear and cytoplasmic RNA isolation experiments, fluorescence in situ hybridization (FISH), bioinformatics analysis, the dual-luciferase reporter experiment, Western Blot, and other experiments were conducted to verify the relationships among circFOXM1, miR-136-5p, and SMAD2. We collected clinical NPC samples to prove the effect of circFOXM1 on the prognosis and treatment of NPC. RESULTS: In this study, we found that circFOXM1 is highly expressed in nasopharyngeal carcinoma tissue cells compared with adjacent normal tissues and is related to the staging of nasopharyngeal carcinoma. High expression of circFOXM1 indicates a poor prognosis for nasopharyngeal carcinoma. Knockdown of CircFOXM1 inhibited the proliferation and invasion of nasopharyngeal carcinoma cells. CONCLUSION: CircFOXM1 promotes the malignant proliferation of nasopharyngeal carcinoma cells by regulating the miR-136-5p-SMAD2 axis.

Accumulating evidences indicate that circular RNAs (circRNAs), a subclass of noncoding RNAs, play important role in regulating gene expression in eukaryotes. Hsa_circ_0046263 (circ-0046263) was found aberrantly expressed in nasopharyngeal carcinoma (NPC), but its role in tumor growth and metastasis remains largely unclear. Sanger sequencing, RNase R assay, and nucleic acid electrophoresis were conducted to verify the identification of circ-0046263. Nuclear separation and fluorescence in situ hybridization (FISH) assays were used to determine the localization of circ-004263. Dual luciferase reporter and RNA immunoprecipitation (RIP) were employed to confirm the binding of circ-0046263 with miR-133a-5p. Colony formation, proliferation, wound healing, transwell, western blot, and in vivo tumor growth and metastasis assays were performed to assess the roles of circ-0046263, miR-133a-5p, IGFBP3 and their interactions in NPC cells. Circ-0046263 was upregulated in both NPC cell lines and tissues. The in vitro functional studies revealed that knockdown of circ-0046263 inhibited the proliferation, invasion, and migration of NPC cells, whereas its overexpression produced the opposite result. In vivo experiments indicated that knockdown or overexpression of circ-0046263 attenuated or promoted tumor growth and metastasis, respectively. Mechanistically, circ-0046263 could act as a miRNA sponge to absorb miR-133a-5p and upregulate the expression of miRNA downstream target IGFBP3. In addition, miR-133a-5p inhibition or IGFBP3 overexpression could rescue the malignant behavior induced by circ-0046263 silencing. Finally, circ-0046263 plays a tumor-promoting role in NPC to enhance malignant behavior through the miR-133a-5p/IGFBP3 axis, which could be a potential target for NPC therapy.

BACKGROUND: Mounting evidence has reported that microRNA-154-5p (miR-154-5p) is involved in the development of multiple cancers, but its function in nasopharyngeal carcinoma (NPC) remains not well investigated. METHODS: Real-time quantitative PCR (qRT-PCR) was used to detect miR-154-5p expression in NPC tissues and cells. CCK8, colony formation, wound healing and transwell assays were performed to assess cell proliferation, migration and invasion. Dual-luciferase reporter assays and Western blots were performed to confirm the target gene of miR-154-5p. Rescue experiments were conducted to explore the influence of target gene KIF14 on the functions of miR-154-5p. Xenograft tumor model was conducted to detect the effect of miR-154-5p in vivo. RESULTS: qRT-PCR results revealed that the expression of miR-154-5p was down-regulated in NPC tissues and cell lines compared to normal nasopharyngeal tissues and cell line. Overexpression of miR-154-5p inhibited cell migration and invasion. However, miR-154-5p had no influence on the proliferation of NPC cells. MiR-154-5p overexpression suppressed xenograft tumor metastasis in vivo. Dual-luciferase reporter analysis identified KIF14 as a target gene of miR-154-5p. Rescue experiments showed that knockdown of KIF14 reversed the effect of inhibiting miR-154-5p expression on NPC cell migration and invasion. CONCLUSION: Taken together, miR-154-5p suppresses tumor migration and invasion by targeting KIF14 in NPC. The newly identified miR-154-5p/KIF14 interaction offers further insights into the progression of NPC, which may represent a novel target for NPC diagnosis and treatment.

Objectives: To evaluate the pharmacodynamical effects and pharmacological mechanism of Ginsenoside H dripping pills (GH) in chronic unpredictable mild stress (CUMS) model rats. Methods: First, the CUMS-induced rat model was established to assess the anti-depressant effects of GH (28, 56, and 112 mg/kg) by the changes of the behavioral indexes (sucrose preference, crossing score, rearing score) and biochemical indexes (serotonin, dopamine, norepinephrine) in Hippocampus. Then, the components of GH were identified by ultra-performance liquid chromatography-iron trap-time of flight-mass spectrometry (UPLC/IT-TOF MS). After network pharmacology analysis, the active ingredients of GH were further screened out based on OB and DL, and the PPI network of putative targets of active ingredients of GH and depression candidate targets was established based on STRING database. The PPI network was analyzed topologically to obtain key targets, so as to predict the potential pharmacological mechanism of GH acting on depression. Finally, some major target proteins involved in the predictive signaling pathway were validated experimentally. Results: The establishment of CUMS depression model was successful and GH has antidepressant effects, and the middle dose of GH (56 mg/kg) showed the best inhibitory effects on rats with depressant-like behavior induced by CUMS. Twenty-eight chemical components of GH were identified by UPLC/IT-TOF MS. Subsequently, 20(S)-ginsenoside Rh2 was selected as active ingredient and the PPI network of the 43 putative targets of 20(S)-ginsenoside Rh2 containing in GH and the 230 depression candidate targets, was established based on STRING database, and 47 major targets were extracted. Further network pharmacological analysis indicated that the cAMP signaling pathway may be potential pharmacological mechanism regulated by GH acting on depression. Among the cAMP signaling pathway, the major target proteins, namely, cAMP, PKA, CREB, p-CREB, BDNF, were used to verify in the CUMS model rats. The results showed that GH could activate the cAMP-PKA-CREB-BDNF signaling pathway to exert antidepressant effects. Conclusions: An integrative pharmacology-based pattern was used to uncover that GH could increase the contents of DA, NE and 5-HT, activate cAMP-PKA-CREB-BDNF signaling pathway exert antidepressant effects.

In this paper, a novel superhydrophobic magnetic sawdust (SMSD) was fabricated as an oil sorbent. The SMSD was functionalized with Fe3O4 nanoparticles using melamine formaldehyde resin (MFR) as a coupling agent and subsequently hydrophobically-treated with hexadecyltrimethoxysilane (HDTMS). The SMSD showed excellent superhydrophobicity with the water contact angle of 155.3 ± 0.9°. Meanwhile it had remarkable environmental durability, long-term stability, and mechanical durable properties. Taking advantage of its magnetic characteristics, the SMSD could be easily controlled to absorb oil to separate oil-water mixtures with high oil absorption capacity and good reusability. Moreover, the emulsion was successfully separated by SMSD, including water-in-oil and oil-in-water emulsions. This study developed an effective oil absorbent, which was low cost and environmentally-friendly.

A new electrochemical sensor was fabricated for the sensitive and selective detection of acetaminophen (AP), based on a bare glassy carbon electrode (GCE) modified with poly caffeic acid (PCA) and Zn/Ni-ZIF-8-800 using an electro-polymerization method. The carbon material of Zn/Ni-ZIF-8-800 derived from bimetal-organic framework was synthesized by one-step pyrolysis method. Then, the as-prepared materials and the corresponding fabricated sensors were fully characterized by XRD, FTIR, BET, SEM, TEM and XPS analyses. In addition, the electron transfer abilities of the modified sensors were investigated by CV and EIS. Subsequently, the parameters of polymerization time, pH value and scan rate were systematically studied and optimized. Under the optimum conditions, the desirable linear relationships were obtained between the peak current and the AP concentration at the PCA@ Zn/Ni-ZIF-8-800/GCE sensor with a detection limit of 0.0291 µM. The outstanding electrochemical performance of the proposed sensor can be attributed to the good electrocatalytic activity of PCA, the large surface area and high conductivity of Zn/Ni-ZIF-8-800 as well as the synergistic effect of PCA and Zn/Ni-ZIF-8-800. Moreover, the fabricated sensor was also successfully applied for the determination of AP in pharmaceutical dosage forms and human urine sample, and satisfactory recoveries were obtained.

Sensitivity and transparency are critical properties for flexible and wearable electronic devices, and how to engineer both these properties simultaneously is dramatically essential. Here, for the first time, we report the assembly of ordered array structures of silver nanowires (AgNWs) via a simple water-bath pulling method to align the AgNWs embedded on polydimethylsiloxane (PDMS). Compared with sensors prepared by direct drop-casting or transfer-printing methods, our developed sensor represents a considerable breakthrough in both sensitivity and transparency. The maximum transmittance was 86.3% at a wavelength of 550 nm, and the maximum gauge factor was as high as 84.6 at a strain of 30%. This remarkably sensitive and transparent flexible sensor has strictly stable and reliable responses to motion capture and human body signals; it is also expected to be able to help monitor disabled physical conditions or assist medical therapy while ensuring privacy protection.

Block copolymer vesicles have been widely used in the field of drug delivery, microreactors, and cell membrane mimetics. Introducing terminal groups to the block copolymer can control the structures of vesicles, which is important for improving the application of vesicles. In this paper, the effects of terminal groups on the structure of vesicles were studied by dissipative particle dynamics simulation. We considered different locations, hydrophobicity, and numbers of terminal groups. When the terminal group located at the end of a hydrophobic block, the increase of wall thickness and the decrease of cavity size of vesicles were observed with the hydrophobicity of the terminal group increasing. In contrast, when the terminal group located at the end of the hydrophilic block, with the hydrophobicity of terminal groups increasing, the vesicular cavity size increased but the wall thickness of vesicles remained nearly unchanged. Finally, increasing the number of terminal groups lead to a decrease of cavity size and an increase of wall thickness of vesicles. The hydrophobic changes of polymer molecules are regarded as the origin of the structural changes of vesicles. This simulation study supplies a potential approach that controls the structures of vesicles and is expected to facilitate its further applications.

Gemini surfactants cause a lot of concerns owing to their unusual aggregation morphologies and superior physicochemical properties over the conventional surfactants. Research shows that the unique structure of Gemini surfactants, especially the spacer group, has a great impact on the self-assembly behaviors. To understand the determinants of this behavior on the molecular level, we carried out coarse-grained molecular dynamics (CGMD) simulations on aqueous solutions of alkanediyl-a,w-bis (dimethylcetylammonium bromide)-based surfactants with different spacer group lengths. Our simulation results demonstrated that the self-assembled morphologies of Gemini changed from spherical micelles, wormlike micelles to vesicles with the decrease in the spacer length, which were qualitatively consistent with the experimental observations. Both the microscopic dynamics processes and the self-assembly mechanisms for the formation of spherical micelles, wormlike micelles and vesicles were systematically studied through the CGMD simulations. In addition, based on the microscopic analysis, a strategy was proposed to predict the self-assembled morphology of surfactant-based systems based on simulation. This work shed light on new views in the understanding of the self-assembly of Gemini surfactants at a molecular-level and the proposed predicting strategy showed promise for practical applications.

Transformation from linear to branched wormlike micelle generally results in the dramatic loss of viscoelasticity. In this study, adopting Coarse-grained molecular dynamics simulation, the conformational transformation was studied in a binary system of cetyltrimethylammonium chloride (CTAC) and sodium salicylate (Nasal). A gross conformation diagram of micelle at different CTAC concentrations and molar ratios of CTAC/Nasal indicates that the branched structure could form at zeta potential lower than 32mV. Further, the dynamic process displays that four sequential stages, approaching, forming salt bridge, yielding stalk, and coalescing together, involve in this transformation. And a fascinating process, an organic salt bridge first form between two approaching micelles and then induces the surfactant jump out to form stalk, was observed. In addition, some rational strategies were proposed to reduce the probability of this conformational transformation. Our study provides a molecular-level understanding of transformation behaviour from linear to branched wormlike micelles.

Self-assembly of block copolymers (BCPs) in solution is a powerful technology to achieve a broad range of structures, such as spheres, cylinders, vesicles, and other hierarchical structures. However, the BCP self-assembly library is limited, especially with respect to tubular structures. Here we show a versatile strategy to expand the morphology library of block copolymer solution self-assemblies with tubular structures (including tubular dumbbells and tubules) via self-assembly of the most common diblock copolymers P4VP-b-PS BCPs in methanol. No special chemistry is needed in this strategy, which proves the universality of this method. The novelty of the strategy is to keep the BCPs both highly asymmetric and with very high molecular weight. The underlying formation mechanism and kinetics of these tubular structures were elucidated. The prepared tubular structures expand the structure library of BCP solution self-assemblies and open up a new avenue for the further applications of a variety of tubular materials.

The influence of mixed liquor suspended solids (MLSS), soluble microbial product (SMP), extracellular polymeric substance (EPS), colloidal particles and other factors contributed to membrane fouling was analyzed in this pilot test by membrane bioreactor (MBR) process for the leather printing and dyeing industrial park mixed wastewater treatment. The results showed that slight membrane fouling occurred after 120-day experiment with an observable increase in membrane resistance R20 from 1.5 x 10(12) m(-1) to 1.8 x 10(12) m(-1). Also, a linear correlation was found between the proportion of colloidal particles concentration in TOC of MBR former solution and membrane filtration resistance change. However, the change of MLSS, SMP, EPS and other factors was not correlated with the membrane filtration resistance change. Therefore, the colloidal particle was considered to be the main factor causing membrane fouling, which attached to the membrane surface and deposited to block the membrane pore.
