RESUMO

Tissue infection typically results from blood transmission or the direct inoculation of bacteria following trauma. The pathogen-induced destruction of tissue prevents antibiotics from penetrating the infected site, and severe inflammation further impairs the efficacy of conventional treatment. The current study describes the size-dependent induction of macrophage polarization using gold nanoparticles. Gold nanoparticles with a diameter of 50 â€‹nm (Au50) can induce M2 polarization in macrophages by inhibiting the NF-κB signaling pathway and stimulate an inflammatory response in the environment by inhibiting the MAPK signaling pathway LPS. Furthermore, the induced polarization and anti-inflammatory effects of the Au50 nanoparticles promoted the osteogenic differentiation of BMSCs in vitro. In addition, the overexpression of TREM2 in macrophage induced by Au50 nanoparticles was found to promote macrophage phagocytosis of Staphylococcus aureus, enhance the fusion of autophagosomes and lysosomes, accelerate the intracellular degradation of S. aureus, in addition to achieving an effective local treatment of osteomyelitis and infectious skin defects in conjunction with inflammatory regulation and accelerating bone regeneration. The findings, therefore, demonstrate that Au50 nanoparticles can be utilized as a promising nanomaterial for in vivo treatment of infections.

In this research, a novel Ag3PO4 NPs@MoS2 nanosheet-based electrochemiluminescence (ECL) sensing system was developed to provide an effective method for tumor gene detection. At first, fluorine, sulfur-doped BN quantum dot (F, S-BN QD) were prepared as ECL emitter. Sulfur dopant can provide more reactive sites in the ECL reaction. Fluorine atoms in the QD structure further improved the stability of the crystal. Furthermore, Ag3PO4 NP@MoS2 nanosheets were fabricated via a hydrothermal route as ECL reaction catalyst. On the one hand, Ag3PO4 NP@MoS2 nanosheets promoted the generation of more oxidant of coreactant in the F, S-BN QD/H2O2 coreactant ECL pathway. On the other hand, the excellent conductivity of Ag3PO4 NP@MoS2 nanosheets facilitated the electron transfer and effectively reduce the damage of F, S-BN QD by excessive hot electrons. Finally, the proposed biosensor was designed to accurately quantify K-ras tumor gene from 10 fM to 100 pM with a limit of detection (LOD) of 0.2 fM. The sensing system was used to detect K-ras gene in human colorectal cancer tumor and tumor-adjacent tissues samples with satisfactory results. The amplified ECL sensing strategy with Ag3PO4 NPs@MoS2 nanosheet has significant potential value in the clinical detection.

Objective@#To understand the characteristics of rapid naming in exotropia children, and to analyze the influence of clinical indicators related to exotropia on the rapid naming.@*Methods@#A total of 45 exotropia children were recruited according to the diagnostic criteria of consensus of strabismus classification experts (2015) from the Zhongshan ophthalmic center as the case group, and 45 children of the same age, gender and parental educational status were recruited as the control group without any ocular diseases. All children were evaluated the ability of the rapid naming by classical rapid naming test.@*Results@#The letter rapid naming time of children with exotropia was longer than that in control group [(26.87±10.18)(21.98±7.29)s], and the difference was statistically significant (t=2.73, P=0.01), however there was no significant correlation between strabismus degree, symptom duration, AC/A ratio, disease classification, simultaneous vision, the near stereopsis, the far stereopsis and the letter rapid naming in the clinical indicators of exotropia (r=-0.16, 0.23, 0.20, 0.06, 0.09, 0.05, 0.20, P>0.05).@*Conclusion@#Rapid naming might be impaired among children with exotropia, with no significant correlation between this defect and its clinical indicators.

In recent years, the role of inflammation in depression has received people's attention.Studies have suggested that immune disorder may play an important role in depression, and patients with depression exhibit characteristic immunophenotypes.Inflammation seems to interact with a variety of pathogenesis of depression.Therefore, immunoregulation is becoming an adjuvant therapy for depression.Clinically, not only antidepressants show anti-inflammatory effects, but also anti-inflammatory drugs show antidepressant effects, and they mainly including non-steroidal anti-inflammatory drugs, statins, and cytokine inhibitors.In addition, some non-drug treatment methods are also given immunomodulatory effects, such as electric shock therapy, vagus nerve stimulation, acupuncture and exercise therapy.However, there are still some problems in immunomodulation therapy, such as immunomodulation therapy may be only effective for some subgroups of patients, and its efficacy and safety need to be evaluated.In the future, looking for more effective biomarkers and identifying immune-inflammation-related subtype of depression, will serve to explore new diagnosis and treatment strategies.

Aging drives the accumulation of senescent cells (SnCs) by secreting factors that cause the senescence-associated secretory phenotype (SASP), including stem cells in the bone marrow, which contribute to aging-related bone degradation. Osteoarthritis (OA) is a serious chronic injury disease, and increasing age is a major risk factor. The accumulation of SnCs may accelerate the development of OA, and the accumulation of SnCs may benefit from its resistance to apoptotic stimuli. Therefore, local elimination of SnCs could be a promising treatment for OA. Apoptosis inhibitor protein (IAP) is an important antiapoptotic protein in vivo. AT-406 is a small molecule inhibitor of the IAP genes and also regulates the transcription of several genes. Here, we show that SnCs upregulate the antiapoptotic proteins c-IAP1, c-IAP2 and XIAP.The combined inhibition of c-IAP1, c-IAP2 and XIAP using siRNA or AT-406 specifically induce the apoptosis of SnCs.In addition, XIAP and STX17 bind to each other to regulate the fusion of autophagosomes and lysosomes in SnCs, which in turn, affects the fate of SnCs. It is worth noting that the clearance of SnCs attenuated the secretion of SASP and created a proregenerative environment. Most importantly, local clearance of SnCs significantly attenuated the progression of osteoarthritis in rats without significant toxic effects. Thus, local elimination of SnCs may be a potential treatment for OA. This is the first report of inhibition of IAPs for clearing SnCs and suggests that eradication of SnCs may be a new strategy for the treatment of age-related diseases.

The Eplus-260 primate PET is an animal PET imaging system developed by the Institute of High Energy Physics, Chinese Academy of Sciences, which is designed to image non-human primates, especially the brain of large non-human primates. The system consists of 48 block detectors arranged in two 24-sided rings with a ring diameter of 263 mm and an axial extent of 64 mm. Each block detector is composed of a 16 × 16 cerium-doped lutetium-yttrium orthosilicate crystal array with a pixel size of 1.9 × 1.9 × 10 mm3. This article presents a performance evaluation of the PET scanner according to the National Electrical Manufacturers Association NU-4 2008 standards. All measurements were made for an energy window of 360-660 keV and a coincidence timing window of 2 ns. In terms of the FWHM, the FBP reconstructed spatial resolution results in all three directions at the radial position of 5 mm were better than or approached to 2 mm, and remained below 3.0 mm within the central 5 cm diameter of the FOV. The peak absolute sensitivity of the scanner was measured 1.80%. For a monkey-sized phantom, the scatter fraction was 34.2% and the peak noise equivalent count rate (NECR) was 26.5 kcps at 64.3 kBq/cc. The overall imaging capabilities of the scanner were also assessed using in vivo imaging study of a rhesus macaque. The performance measurements demonstrate that the Eplus-260 primate PET scanner has the potential ability to obtain good quality and high-contrast images for non-human primates, especially the brain of large non-human primates and could be considered as one technologically advanced dedicated non-human primate PET scanner available today.

BACKGROUND:Rapid prototyping technique has been recently applied in the medical reconstruction and al ows the production of individual implant for patients with tissue defects, achieving an accurate repair. OBJECTIVE:To repair discontinuous mandibular defects in dogs using rapid prototyped titanium plate in combination with autologous cancellous bone graft. METHODS:Nine hybrid canines were used, and the skul was scanned using spiral CT. Then CT data were used to construct three-dimensional digital model, in which virtual partial mandibulectomy was performed, and an individualized bone-grafting plate was designed. A titanium plate was manufactured using rapid prototyping and titanium casting. Animal experiment was then performed. A 40-mm discontinuous defect in the right mandibular body was created in the involved dogs. The defect was restored immediately using the customized plate in combination with autologous cancellous iliac blocks. Sequential radionuclide bone imaging, biomechanical testing, three-dimensional microcomputed tomographic scanning, radiology and histological examination were used to evaluate the turnover of the grafts. RESULTS AND CONCLUSION:A symmetric mandible was reconstructed using the rapid prototyped grafting plate. The grafted bone survived and got corticalized, while a fibrous intermedium was found between the bone graft and the plate. In the reconstruction of mandibular defects, optimal functional and aesthetic outcomes could be achieved using the rapid prototyped grafting plates.

Objective: To survey the content of serum total cholesterol, LDL and HDL in pa- tients with gastric cancer and to investigate the correlation between serum total cholesterol, LDL, HDL and the differentiation of gastric cancer. Methods: 143 patients with gastric cancer were selected and assigned into 3 groups according to pathology grouping after operation: 1) the well-differentiated group, consisting of 16 patients with papillary adenocarcinoma, and 43 patients with tubi-form well-differentiated adenocarcinoma; 2) the moderately differentiated group, including 39 patients with tubiform moderately differentiated adenocarcinoma, 3) the poorly differentiated group, including 20 patients with poorly differentiated adenocarcinoma, and 14 patients with mucinous adenocarcinoma, and 11 patients with signetring cell carcinoma. In addition, 30 patients with gastric ulcer and 30 health adults were selected as control. Veinous blood of every member was collected, and then the content of serum total cholesterol was surveyed by enzymic method, and the contents of serum LDL and HDL were measured by precipitation method. Results: The level of serum total cholesterol, LDL and HDL before operation in patients with gastric cancer decreased significantly than those with gastric ulcer and normal people (P0.05). There was statistically significance among different differentiation in patients with gastric cancer. The differences of levels of serum total cholesterol, LDL and HDL were statistically significant in gastric cancer of different differentiated groups. Objective To investigate the results of laparoscopy and small incision splenectomy in the treatment of Idiopathic Thrombocytopenic Purpura (ITP). Methods Under CO2 pneumoperitoneum, laparoscopy and small incision splenectomy was performed in 13 patients . After freeing of the spleen with laparoscopy , we performed a small incision under the left costal margin. The splenic pedicle was 1-igated under direct vision, and the spleen was excised and extracted. Analysis was made of the pre-and post-operative platelet counts, and the complication rate and recovery condition of the patients. Results No complications occurred in the 13 palients, and the effect-iveness of ITP treatment was 100%.Conclusions Laparoscopy and small incision splenectomy is a safe and effective method to treat ITP.

Objective:To understand the levels of serum cholesterol in patients with gastric cancer and gastric ulcer before and after operation,and to study the relationship between the level of serum cholesterol and the tissues differential degree of gastric cancer.Methods:To survey the content of serum cholesterol in patients with gastric cancer and patients with gastric ulcer confirmed clinically before and after operation,looking through the medical records of the patients and taking notes about the tissues differentiated degree of the gastric cancer,then making the statistical analysis.Results:Comparing with the normal control group,the level of serum cholesterol in gastric cancer decreased obviously before operation,there was obvious statistical difference(P0.01),and increased after operation,comparing with themselves before operation was statistical difference(P0.05),and no statistical difference comparing with the normal control group after operation.There was statistical difference between the higher(group A)and the lower(group B)differentiated degree of gastric cancer tissues(P0.05)before operation and no statistical difference after operation.After operation the levels of serum cholesterol in group A and group B were all increased.Comparing with the level of serum cholesterol before operation each other,there was obvious statistical difference(P0.01)in group B and no statistical difference(P0.05) in group A.Conclusion:The level of serum cholesterol in patients with gastric cancer decreased before operation and increased after operation,especially in progressive gastric cancer.There was correlation between the level of serum cholesterol and the tissues differentiated degree of gastric cancer.The lower the tissues differentiated degree of gastric cancer was,the lower before operation and the higher after operation the level of serum cholesterol were.
