RESUMO

RESUMO Ricinus communisé uma planta pertence à família Euforbiaceae. A partir de seus frutos é extraído o óleo de rícino e outra fração altamente tóxica (fração não-oleosa). Neste estudo, foi avaliado o efeito da fração não oleosa de R. communis(FNORC) na biodistribuição do Tc-99m em camundongos sadios e em animais transplantados com Sarcoma-180 (S180). Para avaliação da biodistribuição do Tc-99m, o percentual de atividade radioativa total injetada (ATI%) foi mensurada utilizando um contador gama. Os resultados mostraram que, em animais sadios, houve aumento da captação do Tc-99m após o tratamento com FNORC pelos rins, cérebro e estômago. Apenas houve diminuição da absorção deste radioisótopo nos músculos. Em animais com tumor, houve redução significativa da captação do Tc-99m no coração, pulmões e tumor, e foi observado o aumento da captação apenas nos rins. Foi observada também a diminuição da concentração das proteínas plasmáticas totais em animais com tumor após tratamento com FNORC e atividade antitumoral significativa. Diante desses dados, conclui-se que fração não oleosa de R. communis possui atividade antitumoral significativa, porém altera a biodistribuição do Tc-99m, fato este que pode resultar em falhas nos processos de diagnóstico por imagem que utilizem este radioisótopo como marcador.


ABSTRACT Ricinus communis is a plant belonging to the Euforbiaceae family. From its fruits, it is extracted the castor oil and another highly toxic fraction (non-oily fraction). In this study, we evaluated the effect of the non-oily fraction of R. communis (NOFRC) on the bio distribution of Tc-99m in healthy mice and in animals transplanted with Sarcoma-180 (S180). To evaluate the bio distribution of theTc-99m, the percentage of the total injected radioactivity (% TIR) was measured through a gamma counter. There was an increase in the capitation of Tc-99m after the treatment with NOFRC in the kidneys, brain, and stomach of healthy animals. A decrease in the reception of this radionuclide was only found in the muscles. In animals with tumour, there was a significant reduction in the uptake of Tc-99m in the heart, lungs and tumour. An increase in the capitation was only observed in the kidneys. It was also observed a reduction in the total plasma protein concentration in animals with tumours after the treatment with NOFRC, besides significant antitumor activity. We concluded that there is antitumor activity in the NOFRC, but that it alters the biodistribution of Tc-99m, a fact which may result in failures in the diagnostic imaging process using this radioisotope as a marker.

This study evaluated the effects of 2 different types of acute aerobic exercise on the osmotic stability of human erythrocyte membrane and on different hematological and biochemical variables that are associated with this membrane property. The study population consisted of 20 healthy and active men. Participants performed single sessions of 2 types of exercise. The first session consisted of 60 min of moderate-intensity continuous exercise (MICE). The second session, executed a week later, consisted of high-intensity interval exercise (HIIE) until exhaustion. The osmotic stability of the erythrocyte membrane was represented by the inverse of the salt concentration (1/H50) at the midpoint of the sigmoidal curve of dependence between the absorbance of hemoglobin and the NaCl concentration. The values of 1/H50 changed from 2.29±0.1 to 2.33±0.09 after MICE and from 2.30±0.08 to 2.23±0.12 after HIIE. During MICE mean corpuscular volume increased, probably due to in vivo lysis of older erythrocytes, with preservation of cells that were larger and more resistant to in vitro lysis. The study showed that a single bout of acute exercise affected erythrocyte stability, which increased after MICE and decreased after HIIE.

Salmonella enterica serovar Gallinarum (SG) is an intracellular pathogen of chickens. To survive, to invade and to multiply in the intestinal tract and intracellularly it depends on its ability to produce energy in anaerobic conditions. The fumarate reductase (frdABCD), dimethyl sulfoxide (DMSO)-trimethylamine N-oxide (TMAO) reductase (dmsABC), and nitrate reductase (narGHIJ) operons in Salmonella Typhimurium (STM) encode enzymes involved in anaerobic respiration to the electron acceptors fumarate, DMSO, TMAO, and nitrate, respectively. They are regulated in response to nitrate and oxygen availability and changes in cell growth rate. In this study mortality rates of chickens challenged with mutants of Salmonella Gallinarum, which were defective in utilising anaerobic electron acceptors, were assessed in comparison to group of bird challenged with wild strain. The greatest degree of attenuation was observed with mutations affecting nitrate reductase (napA, narG) with additional attenuations induced by a mutation affecting fumarate reductase (frdA) and a double mutant (dmsA torC) affecting DMSO and TMAO reductase.

ABSTRACT The frequency of group A rotavirus and the molecular G and P characterization of strains in beef cattle were determined from July 2003 to July 2004, in herds of the state of São Paulo, Brazil. Samples came from 649 calves, aged between 1 and 60 days, from 14 beef cattle herds, regardless of clinical manifestation of diarrhea. A 64.3% (9/14) frequency of rotavirus among herds and 6.2% (40/649) among animals were detected by ELISA and polyacrylamide gel electrophoresis (PAGE). The higher frequency infection was detected in animals aged between 16 and 30 days (10.5%). Rotavirus infection was diagnosed in animals with (25.8%; 22/85) or without (3.2%; 18/564) clinical signs of diarrhea. A correlation was found between the presence of infection and the clinical manifestation of diarrhea (p 0.05). PAGE analysis of the rotaviruss genome profile identified 7 distinct electropherotypes which indicated extensive genomic diversity of the rotavirus in the region under analysis. Genotyping by polymerase chain reaction (RT-PCR) of the rotavirus strains revealed the genotypes G6P[5], G6P[11], G6P[5]P[11] and G10P[11].


RESUMO Foi determinada a ocorrência de rotavírus do grupo A e a caracterização molecular G e P de estirpes detectadas em bezerros de rebanhos de corte em propriedades rurais do Estado de São Paulo, Brasil. Foram analisadas amostras de fezes de 649 bezerros de 14 rebanhos de corte com idade entre 1 e 60 dias, independentemente da manifestação clínica de diarréia, colhidas de julho de 2003 a julho de 2004. Por meio das técnicas de ELISA e de eletroforese em gel de poliacrilamida (PAGE), determinou-se a ocorrência de rotavírus do grupo A em 64,3% (09/14) dos rebanhos e em 6,2% (40/649) dos animais. A maior freqüência de infecção foi detectada em animais com idade entre 16 e 30 dias (10,5%). Foram diagnosticados bezerros infectados por rotavírus tanto em animais com sinais clínicos de diarréia (25,8%; 22/85) quanto naqueles assintomáticos (3,2%; 18/ 564), existindo, porém, uma correlação entre a presença da infecção e a manifestação clínica da diarréia (p 0,01). A análise do perfil do genoma do rotavírus por PAGE identificou sete eletroferótipos distintos, indicando grande diversidade genômica dos rotavírus na região estudada. A genotipagem pela reação em cadeia da polimerase (RT-PCR) das amostras de rotavírus revelou que as estirpes circulantes nos rebanhos eram G6P[5], G6P[11], G6P[5]P[11] e G10P[11].

ABSTRACT This work was carried out to assess the preenrichment (PE) and enrichment (DE) steps for isolating Salmonella serotypes Enteritidis (SE) and Typhimurium (STM) from chicken feces kept at 4° C for 24 and 96h. The samples were artificially contaminated and kept in 1% peptone water at 4° C for 24 or 96h. After that, part of them was incubated at 37° C/24h and part was inoculated into enrichment broth, selenite broth plus novobiocin (SN) and tetrathionate broth plus novobiocin (TN) incubated at 37°C/24h. The PE culture was inoculated in SN, TN and Rapapport-Vassiliadis novobiocin (RVN), also incubated at 37° C/24h. The enrichment broth was plated on brilliant green agar (BGA), MacConkey agar (MCA), Hektoen agar (HEA), Salmonella-Shigella agar (SSA), xylose-lysine desoxicholate agar (XLDA) and xylose-lysine tergitol 4 (XLT4), which were incubated at 37° C/24h. Salmonella-like colonies were submitted to TSI agar and LIA agar, and incubated at 37° C/24h, as well as to slide agglutination tested with poly O and poly H Salmonella antiserum. When the samples were stored for 24h there was no difference between PE and DE (p > 0.05). However after 96h the PE was superior to DE (p 0.05). RESUMO Este trabalho foi desenvolvido para avaliar comparativamente o isolamento de Salmonella sorotipos Enteritidis (SE) e Typhimurium (STM) a partir do enriquecimento direto (ED) ou processamento com pré-enriquecimento (PE) de amostras de fezes de aves adultas, armazenadas em água peptonada tamponada a 1% (APT) por 24 ou 96h a 4º C. Utilizou-se os caldos de enriquecimento Rapapport-Vassiliadis novobiocina (RVN), tetrationato-novobiocina (TN) e selenitonovobiocina (SN) e os meios para plaqueamento ágar verde brilhante (VB), ágar de MacConkey (MC), ágar de Hektoen (HE), ágar Salmonella-Shigella (SS), ágar xilose lisina desoxicolato (XLD) e ágar xilose lisina tergitol 4 (XLT4). O procedimento bacteriológico incluiu as etapas de pré-enriquecimento, enriquecimento em caldo seletivo, plaqueamento, testes bioquímicos presuntivos e confirmação sorológica com utilização de soros polivalentes anti-antígenos somáticos e anti-antígenos flagelares de Salmonella. Não houve diferença estatisticamente significativa (p > 0,05) para as amostras armazenadas por 24h submetidas tanto ao PE quanto ao ED. Entretanto, em armazenagem por 96h o número de isolamentos nas amostras submetidas ao PE foi estatisticamente superior às submetidas ao ED (p 0,05). Quanto aos caldos enriquecedores, não houve diferença estatística de número de isolamentos (p > 0,05) entre os caldos SN e TN, mas o caldo RVN mostrou-se estatisticamente superior aos demais (p 0,05). Para os meios de plaqueamento, o XLD destacou-se por promover maior número de recuperações, embora sem significado estatístico (p > 0,05) para as amostras estocadas por 24h. Entre os dois sorotipos de Salmonella (SE e STM) não houve diferença estatística no número de recuperações (p > 0,05).

BACKGROUND: Allergens from house dust mites (HDMs), Dermatophagoides pteronyssinus and Blomia tropicalis are clinically relevant in atopic respiratory diseases in tropical countries. AIMS OF THE STUDY: To evaluate immunoglobulin (Ig)E, IgG1, and IgG4 antibody responses to B. tropicalis in Brazilian atopic patients. METHODS: About 110 patients with allergic rhinitis with/without asthma and 33 control subjects underwent skin prick testing (SPT) with HDM extracts, and their sera were tested for IgE and IgG subclass antibodies to D. pteronyssinus and B. tropicalis by enzyme-linked immunosorbent assay (ELISA) and immunoblotting. RESULTS: Most patients (56%) had positive SPT to B. tropicalis extract (B. tropicalis+ group), although 51% were reactive to both B. tropicalis and D. pteronyssinus and 6% were sensitized to B. tropicalis only. IgE-ELISA detected 43%B. tropicalis positivity with high-specific IgE levels in B. tropicalis+ patients. Specific IgG4 levels were higher in B. tropicalis+ than B. tropicalis- groups and correlated with specific IgE levels. The IgG1 levels to B. tropicalis were higher in patients than controls. The major allergenic B. tropicalis components recognized by B. tropicalis+ patient sera were the 54, 66, and 68 kDa proteins. The IgG4-binding protein profiles closely resembled that of IgE. The IgG1 antibodies recognizing multiple B. tropicalis protein species were detected in sera of all three patient groups. CONCLUSIONS: A large percentage of our allergic patients are B. tropicalis+. They are more frequently sensitized to high-molecular weight (HMW) B. tropicalis components than the major low-molecular weight (11-15 kDa) allergens detected in other studies. The results suggest that HMW B. tropicalis antigenic components are potential candidates for evaluating allergen exposure and sensitization, and for immunotherapy treatment.

8-Methoxy psoralen (8-MOP) exerts a short-term (24 h) mitogenic action, and a long-term (48-72 h) anti-proliferative and melanogenic action on two human melanoma cell lines, SK-Mel 28 and C32TG. An increase of intracellular calcium concentration was observed by spectrofluorometry immediately after the addition of 0.1 mM 8-MOP to both cell lines, previously incubated with calcium probe fluo-3 AM (5 micro M). The intracellular Ca2+ chelator BAPTA/AM (1 micro M) blocked both early (mitogenic) and late (anti-proliferative and melanogenic) 8-MOP effects on both cell lines, thus revealing the importance of the calcium signal in both short- and long-term 8-MOP-evoked responses. Long-term biological assays with 5 and 10 mM tetraethylammonium chloride (TEA, an inhibitor of Ca2+-dependent K+ channels) did not affect the responses to psoralen; however, in 24-h assays 10 mM TEA blocked the proliferative peak, indicating a modulation of Ca2+-dependent K+ channels by 8-MOP. No alteration of cAMP basal levels or forskolin-stimulated cAMP levels was promoted by 8-MOP in SK-Mel 28 cells, as determined by radioimmunoassay. However, in C32TG cells forskolin-stimulated cAMP levels were further increased in the presence of 8-MOP. In addition, assays with 1 micro M protein kinase C and calcium/calmodulin-dependent kinase inhibitors, Ro 31-8220 and KN-93, respectively, excluded the participation of these kinases in the responses evoked by 8-MOP. Western blot with antibodies anti-phosphotyrosine indicated a 92% increase of the phosphorylated state of a 43-kDa band, suggesting that the phosphorylation of this protein is a component of the cascade that leads to the increase of tyrosinase activity.

The aim of this paper was to verify hydrological and hydrochemical changes in the Iquiparí Lagoon water column, during three months, as related to an artificial sand bar opening. A drastic reduction in water volume occurred 28 hours after the sand bar opening, exposing the bottom sediment. This was densely colonized by submerged aquatic plants, which were dominant in the ecosystem metabolism, specially near the sand bar. An increase in salinity values, and dissolved and total nutrient concentrations, was observed during the sand bar opening period. In contrast, chlorophyll a concentration decreased. These changes were associated with: 1) mix of remained brackish and marine waters; 2) nutrient release from sediments by physical processes; and 3) submerged plant death. After the sand bar closure, lagoon metabolism was completely regulated by a dense phytoplankton community. We observed that the sand bar opening resulted in an extensile replacement of the lagoon's water and a change in the community mainly responsible for the ecosystem metabolism.

The authors performed a prospective study of 86 patients with septic arthritis of the knee between 1990 e 1994. The age, sex, predisposing factors, time between onset of symptoms and treatment, etiologic organism and their route of entrance in the joint, previous use of antibiotic the data from of treatment, stay at the hospital and complications were evaluated and compared with the literature.

To evaluate non-conventional phosphorus sources, a group of 400 Nellore cows mantained under pasture received a mineral mixture with different P sources (group I: superphosphate - 500 ppm P; group II: superphosphate - 340 ppm P plus rock phosphate Patos 160 ppm P; group III: superphosphate 340 ppm P plus dicalcium phosphate 160 ppm P; group IV - dicalcium phosphate 500 ppm P). There were no differences in pregnancy rate, calving rate and calving interval. A fluorine deposition in bone was observed for the treatments with superphosphate and rock phosphate (66.92 ± 15.53; 69.97 ± 6.5 and 64.05 ± 3.35% respectively for group I, II and III). Superphosphate was almost as good dicalcium phosphate to provide phosphorus for grazing cows and there was a potentially significant economic advantage over dicalcium phosphate.


Quatrocentas vacas Nelore, mantidas em pastagem, receberam mistura mineral contendo fontes não convencionais de fósforo (grupo I - 500 ppm P-superfosfato triplo; grupo II - 340 ppm P como superfosfato e 160 ppm P como rocha Patos; grupo III - 340 ppm P como superfosfato e 160 ppm P como fosfato bicálcico; grupo IV -500 ppm P como fosfato bicálcico). Não se verificou diferenças na taxa de prenhez, porcentagem de nascimento e intervalo entre partos. Observou-se que houve maior concentração de flúor nos ossos para o tratamento com fosfato de rocha. O superfosfato triplo apresentou-se como fonte adequada de P e mostrou vantagem econômica em relação ao bicálcico.

RESUMO

Seventy nine elderly patients with acute abdominal conditions were operated on in the Emergency Service of the Hospital das Clinicas of the University of São Paulo from 1981 to 1985. Total mortality rate was 58.23%. In patients with acute vascular abdominal conditions it was 100%, in inflammatory conditions 55.17% and in intestinal obstruction 47.83%. The lowest mortality was observed in traumatic conditions (27.27%). Procrastination of operations was followed by higher mortality. In operations lasting more than three hours the mortality was significantly higher than in those lasting less than three hours. Associated diseases as well as reoperations increased the postoperative mortality.
