RESUMO

Mass rearing of mosquitoes as required to fulfil research studies is a technically challenging endeavor. Blood meal source has been recognized as a key consideration in mass rearing of mosquitoes that affects colony health and fecundity. Four species of laboratory-colonized mosquitoes from the Department of Entomology, US Army Medical Directorate - Armed Forces Research Institute of Medical Sciences (USAMD-AFRIMS); Anopheles cracens, An. dirus, An. minimus and An. sawadwongporni were fed blood meals from human and rhesus macaque sources using an artificial membrane feeder. The effects of different blood meal sources were evaluated concerning blood-feeding, survival and reproduction (fecundity and hatching rates). Adult survival was monitored at days 7, 14 and 21 post blood-feeding. Although the mosquitoes fed on human blood exhibited higher rates of engorgement, there were no significant differences in blood-feeding rates in An. cracens (P = 0.08) and An. dirus (P = 0.91) between rhesus macaque and human blood sources. Twenty-one days post-feeding, no significant differences were observed in the survival rates of mosquitoes fed on human versus rhesus macaque blood. Except for An. dirus, which had better survival rates with human blood (97.5%) than after feeding on rhesus macaque blood (95.4%). All mosquito species fed on human blood produced significantly more eggs when compared to those fed on rhesus macaque blood. However, there was no statistical difference in hatching rates between blood sources, except for An. dirus, which had better hatching rates with human blood. These results indicate that human and rhesus macaque blood may be a viable alternative for maintaining Anopheles mosquitoes in colony.

Ivermectin mass drug administration to humans or livestock is a potential vector control tool for malaria elimination. The mosquito-lethal effect of ivermectin in clinical trials exceeds that predicted from in vitro laboratory experiments, suggesting that ivermectin metabolites have mosquito-lethal effect. The three primary ivermectin metabolites in humans (i.e., M1 (3″-O-demethyl ivermectin), M3 (4-hydroxymethyl ivermectin), and M6 (3″-O-demethyl, 4-hydroxymethyl ivermectin) were obtained by chemical synthesis or bacterial modification/metabolism. Ivermectin and its metabolites were mixed in human blood at various concentrations, blood-fed to Anopheles dirus and Anopheles minimus mosquitoes, and mortality was observed daily for fourteen days. Ivermectin and metabolite concentrations were quantified by liquid chromatography linked with tandem mass spectrometry to confirm the concentrations in the blood matrix. Results revealed that neither the LC50 nor LC90 values differed between ivermectin and its major metabolites for An. dirus or An. minimus., Additionally, there was no substantial differences in the time to median mosquito mortality when comparing ivermectin and its metabolites, demonstrating an equal rate of mosquito killing between the compounds evaluated. These results demonstrate that ivermectin metabolites have a mosquito-lethal effect equal to the parent compound, contributing to Anopheles mortality after treatment of humans.

Oviposition preference by Toxorhynchites mosquito species is an important aspect of their biological control potential. When ovipositing, the females deposit individual eggs by hovering above the water, indicating that they may be capable of detecting chemical cues from above the water surface. These chemical cues may help the females to determine whether the water is suitable for their larvae. We investigated whether female Tx. splendens has the ability to discriminate against oviposition sites in habitats containing turbid larval/pupal rearing water and predator-prey treatments. In this study, the number of eggs in the test and control cups were compared to assess oviposition preference. According to the results, female Tx. splendens preferred ovipositing in cups with turbid immature water (prey absence) and predator-prey turbid water (prey and predator presences) over ovipositing in filtered water as a control. Results from the third experiment revealed that cups with alive and dead Ae. aegypti larvae proved strongly attractive to Tx. splendens females, while those from conspecific Toxorhynchites larvae caused oviposition deterrence when compared to other treatments in a dual choice test. Our results indicated that both turbid immature water and predator-prey turbid water can efficiently attract Tx. splendens females to deposit eggs. These findings will aid in the development of Toxorhynchites mosquito as a biocontrol agent for the control of Aedes spp. larvae.

There are eight Anopheles spp. present in the Republic of Korea (ROK), including five members of the Anopheles Hyrcanus Group that cannot be identified using current morphological methods. The vector competence of only Anopheles sinensis s.s., An. lesteri, and An. kleini have been investigated. As the geographical distribution of Anopheles spp. varies in the ROK, determining the relative vector competence of the Anopheles spp. provides a basis for delineating malaria risks to Korean populations and U.S. military/civilian populations deployed to the ROK. Anopheles belenrae and An. pullus, collected from a malaria high-risk area in the ROK, were evaluated for vector competence of P. vivax. A total of 1,000 each of An. dirus (Thai strain), and Korean strains of An. pullus and An. belenrae were fed on P. vivax infected blood collected from Thai patients via artificial membrane feeding. The overall oocyst infection rates for An. dirus, An. pullus, and An. belenrae dissected on days 8-9 postfeed were 64.1, 12.0, and 11.6%, respectively. The overall sporozoite infection rates for those species dissected on days 14-15 postfeed were 84.5, 3.4, and 5.1% respectively. The salivary gland sporozoite indices for positive females with +4 (>1,000 sporozoites) were observed in An. dirus (72.8%), but not observed for either An. pullus or An. belenrae. Most sporozoite-positive An. pullus (83.3%) and An. belenrae (71.4%) females were observed with only +1 (1-10 sporozoites) salivary glands. These data indicate that both An. belenrae and An. pullus are very poor vectors of P. vivax.

Mass administration of antimalarial drugs and ivermectin are being considered as potential accelerators of malaria elimination. The safety, tolerability, pharmacokinetics, and mosquito-lethal effects of combinations of ivermectin, dihydroartemisinin-piperaquine, and primaquine were evaluated. Coadministration of ivermectin and dihydroartemisinin-piperaquine resulted in increased ivermectin concentrations with corresponding increases in mosquito-lethal effect across all subjects. Exposure to piperaquine was also increased when coadministered with ivermectin, but electrocardiograph QT-interval prolongation was not increased. One subject had transiently impaired liver function. Ivermectin mosquito-lethal effect was greater than predicted previously against the major Southeast Asian malaria vectors. Both Anopheles dirus and Anopheles minimus mosquito mortality was increased substantially (20-fold and 35-fold increase, respectively) when feeding on volunteer blood after ivermectin administration compared with in vitro ivermectin-spiked blood. This suggests the presence of ivermectin metabolites that impart mosquito-lethal effects. Further studies of this combined approach to accelerate malaria elimination are warranted.

The Anopheles Hyrcanus Group in the Republic of Korea (ROK) consists of 5 morphologically indistinct species that can only be identified with certainty by polymerase chain reaction (PCR). A total of 86 bloodfed Anopheles spp. were collected from a cow barn located in the village of Tongilchon near the demilitarized zone in the ROK on June 13, 2016, and sent to the Armed Forces Research Institute of Medical Sciences in Bangkok, Thailand, where they were identified to species by PCR. The 1st shipment contained 15 An. belenrae and 37 An. pullus females that were used to start the colonies. Parent females that oviposited were identified by PCR for colonization. A higher proportion of F1-F4 females of An. belenrae than An. pullus bloodfed when provided both blood meals on human arms and using a membrane feeding system with human blood. Following blood meals, the females were forced mated for colony maintenance. The mean numbers of eggs oviposited per female for An. belenrae was 127.7 ± 19.3 and for An. pullus was 136 ± 23.6. On average, at 25°C (±2°C) An. belenrae and An. pullus took 15.1 and 16.1 days to develop from egg to adult, respectively. A 2nd group of bloodfed Anopheles spp. was collected at the same location in the ROK on June 24, 2017. This group contained 13 An. belenrae and 27 An. pullus. Similarly, eggs were obtained and adults identified by PCR and then reared to adults and subsequent generations forced mated to members of each of the existing colonies to increase genetic diversity. The colonies were established to evaluate their susceptibility to vector vivax malaria, which is essential to better understand the epidemiology of malaria transmission in Korea. This is the 1st report of colonization of both An. belenrae and An. pullus.

Successful mating by male mosquitoes is dependent on several factors, with sugar feeding being particularly important. The effect of ingested vitamins on adult male mosquitoes is poorly understood. This laboratory study used 3 anopheline species, Anopheles campestris, An. dirus, and An. sawadwongporni, to study the effect of sugar and vitamins on male longevity, copulation, and fecundity. Males were fed 1 of 5 diets containing different combinations of sugar and vitamins: 10% glucose, 10% sucrose, 10% multivitamin syrup, 10% multivitamin syrup + 10% glucose, and 10% multivitamin syrup + 10% sucrose. The longevity of males was measured for a period of 15 days. Forced mating was used to simulate copulation, and fecundity was measured by counting the number of eggs oviposited and the hatch rate of larvae. The longevity of An. campestris and An. dirus was greatest when fed a diet of 10% multivitamin syrup + 10% glucose, and the longevity of An. sawadwongporni was greatest when fed a diet of 10% multivitamin syrup + 10% sucrose. The 1st mating routinely produced the most viable eggs when males were mated with several females. The diet of 10% multivitamin syrup + 10% sucrose produced numerically greater egg production and larval emergence for all 3 species, although this was not always statistically significant due to variability and small sample size. These results indicate that the addition of multivitamin syrup to sucrose may produce healthier and more fit male anophelines. This has potential implications for increasing insectary operations and improving the fitness of laboratory-reared male mosquitoes that will be released for mosquito and disease-pathogen control studies.

Objective:To evaluate adulticidal,larvicidal and oviposition deterrent response of the essential oil from dried Zanthoxylum limonella (Z.limonella) fruit against Aedes aegypti (Ae.aegypti) and Culex quinquefasciatus (Cx.quinquefasciatus).Methods:Z.limonella oil was tested by biological assays at 1%,5% and 10% concentrations in ethanol.Adulticidal efficacy was tested against the 2-3 day old adult females.Larvicidal activity was tested against immature stage of mosquitoes.Oviposition deterrence of the oil was evaluated on gravid females.Results:The adult mortality was observed after 24 h with the LC5o of 6.0% for Ae.aegypti,and 5.7% for Cx.quinquefasciatus.Larvicidal bioassay was carried out with the 10% Z.limonella oil against immature stages of Ae.aegypti and Cx.quinquefasciatus,which caused 100% mortality after 12 h and 24 h.In the larvicidal experiment,Z.limonella showed effective result at 1%,5% and 10% concentrations with the values of LT5o Ae.aegypti =9.78,5.61,0.24 h for larvae and LT5o =64.08,21.23 h for pupae;Cx.quinquefasciatus had LT50 =28.46,20.25,1.01 h for larvae and LT50 =67.52,27.96,4.11 h for pupae,respectively.Oviposition deterrence of the oil was evaluated on gravid females.In the study,10% Z.limonella showed 100% repellency for Ae.aegypti and 99.53% for Cx.quinquefasciatus.Likewise,oviposition activity indexes of these oil concentrations were all negative values ranging from-0.89 to-1.00 for Ae.aegypti and-0.64 to-0.99 for Cx.quinquefasciatus.The oviposition activity indexes values revealed that Z.limonella oil has deterrent effect,and it caused a remarkable negative response resulting in very few eggs.Conclusions:This result indicates that Z.limonella oil can be used as an effective adulticide,larvicide and oviposition deterrent against Ae.aegypti and Cx.quinquefasciatus.

In total, 1,300 each of Anopheles kleini Rueda and Anopheles sinensis Wiedemann sensu stricto (s.s.) females (colonized from the Republic of Korea) and Anopheles dirus Peyton & Harrison (Thai strain) were allowed to feed on blood from Thai malaria patients naturally infected with Plasmodium vivax The overall oocyst infection rates for An. dirus, An. kleini, and An. sinensis s.s. were 77.4, 46.1, and 45.9%, respectively. The mean number of oocysts was significantly higher for An. dirus (82.7) compared with An. kleini (6.1) and An. sinensis s.s. (8.6), whereas the mean number of oocysts for An. kleini and An. sinensis s.s. was similar. The overall sporozoite infection rates for An. dirus, An. kleini, and An. sinensis s.s. dissected on days 14-15, 21, and 28 days post-feed were significantly higher for An. dirus (90.0%) than An. kleini (5.4%), whereas An. kleini sporozoite rates were significantly higher than An. sinensis s.s. (<0.1%). The overall sporozoite indices for positive females with +3 (100-1,000 sporozoites) and +4 (>1,000 sporozoites) salivary gland indices were significantly higher for An. dirus (85.7%), compared with An. kleini (47.1%). Only one An. sinensis s.s. had sporozoites (+2; >10-100 sporozoites). These results indicate that An. kleini is a competent vector of vivax malaria. Although An. sinensis s.s. develops relatively high numbers of oocysts, it is considered a very poor vector of vivax malaria due to a salivary gland barrier.

Cymbopogon citratus (lemongrass) and Syzygium aromaticum (clove) oils were evaluated to determine mortality rates, morphological aberrations, and persistence when used against third and fourth larval instars of Aedes aegypti and Anopheles dirus. The oils were evaluated at 1, 5, and 10 % concentrations in mixtures with soybean oil. Persistence of higher concentrations was measured over a period of 10 days. For Ae. aegypti, both plant oils caused various morphological aberrations to include deformed larvae, incomplete eclosion, white pupae, deformed pupae, dead normal pupae, and incomplete pupal eclosion. All of these aberrations led to larval mortality. In Ae. aegypti larvae, there were no significant differences in mortality at days 1, 5, and 10 or between third and fourth larval instar exposure. In An. dirus, morphological aberrations were rare and S. aromaticum oil was more effective in causing mortality among all larval stages. Both oils were equally effective at producing mortality on days 1, 5, and 10. Both oils had slightly increased LT50 rates from day 1 to day 10. In conclusion, both lemongrass and clove oils have significant effects on the immature stages of Ae. aegypti and An. dirus and could potentially be developed for use as larvicides.

Repellency of essential oil extracted from Curcuma longa, Eucalyptus globulus, and Citrus aurantium at various concentrations (5, 10, 15, 20, and 25 %) with and without 5 % vanillin was evaluated against female mosquitoes: Aedes aegypti and Anopheles dirus. The comparisons were made with a commercial chemical repellent (N,N-diethyl-3-methylbenzamide (DEET) 25 % w/w; KOR YOR 15) by arm in cage method. It was found that the essential oils with 5 % vanillin gave the longest lasting period against two mosquitoes as follows: Curcuma longa gave 150 min for Ae. aegypti, 480 min for An. dirus; Eucalyptus globulus gave 144 min for Ae. aegypti, 390 min for An. dirus; and Citrus aurantium gave 120 min for Ae. aegypti, 360 min for An. dirus. The 25 % Curcuma longa essential oil exhibited the best efficiency as equal as a commercial repellent (480 min against An. dirus). Vanillin can extend the period of time in protection against the two mosquitoes. This study indicates the potential uses of the essential oils (Curcuma longa, Eucalyptus globulus, and Citrus aurantium) with vanillin as natural mosquito repellents.

Established colonies of Anopheles campestris, Anopheles cracens, Anopheles dirus, Anopheles kleini, Anopheles minimus, Anopheles sawadwongporni, and Anopheles sinensis are maintained at the Armed Forces Research Institute of Medical Sciences (AFRIMS). Females were provided blood meals on human blood containing citrate as an anticoagulant using an artificial membrane feeder. The mean wing length, used as an estimate of body size, for each species was compared to blood-feeding duration (time), blood meal volume, and numbers of eggs oviposited. Except for An. campestris and An. cracens, there were significant interspecies differences in wing length. The mean blood meal volumes (mm(3)) of An. kleini and An. sinensis were significantly higher than the other 5 species. For all species, the ratios of unfed females weights/blood meal volumes were similar (range: 0.76-0.88), except for An. kleini (1.08) and An. cracens (0.52), that were significantly higher and lower, respectively. Adult females were allowed to feed undisturbed for 1, 3, and 5min intervals before blood feeding was interrupted. Except for An. campestris and An. sawadwongporni, the number of eggs oviposited were significantly higher for females that fed for 3min when compared to those that only fed for 1min. This information is critical to better understand the biology of colonized Anopheles spp. and their role in the transmission of malaria parasites as they relate to the relative size of adult females, mean volumes of blood of engorged females for each of the anopheline species, and the effect of blood feeding duration on specific blood meal volumes and fecundity.

Repellency activity of Thai essential oils derived from ylang ylang (Cananga odorata (Lamk.) Hook.f. & Thomson: Annonaceae) and lemongrass (Cymbopogon citratus (DC.) Stapf: Poaceae) were tested against two mosquito vectors, Aedes aegypti (L.) and Culex quinquefasciatus (Say). There were compared with two chemical repellents (DEET 20% w/w; Sketolene Shield(®) and IR3535, ethyl butylacetylaminopropionate 12.5% w/w; Johnson's Baby Clear Lotion Anti-Mosquito(®)). Each herbal repellent was applied in three diluents; coconut oil, soybean oil and olive oil at 0.33 µl/cm(2) on the forearm of volunteers. All herbal repellent exhibited higher repellent activity than IR3535 12.5% w/w, but lower repellent activity than DEET 20% w/w. The C. odorata oil in coconut oil exhibited excellent activity with 98.9% protection from bites of A. aegypti for 88.7±10.4 min. In addition, C. citratus in olive oil showed excellent activity with 98.8% protection from bites of C. quinquefasciatus for 170.0±9.0 min. While, DEET 20% w/w gave protection for 155.0±7.1-182.0±12.2 min and 98.5% protection from bites of two mosquito species. However, all herbal repellent provided lower repellency activity (97.4-98.9% protection for 10.5-88.7 min) against A. aegypti than C. quinquefasciatus (98.3-99.2% protection for 60-170 min). Our data exhibited that C. odorata oil and C. citratus oil are suitable to be used as green repellents for mosquito control, which are safe for humans, domestic animals and environmental friendly.

We evaluated the effects of membrane surface area (cm2), female density, and container/cage size on feeding response in laboratory-reared Aedes aegypti. Female density did not affect feeding rates at low surface areas, but higher density did significantly increase feeding as surface area increased. Females in large, cloth cages fed less compared to those in large, plastic cups. The rate of feeding was higher when using live, anesthetized mice versus a membrane feeding system. The AFRIMS Insectary will continue to look for innovative ways to improve our membrane feeding system.

A total of 158 resting blood-fed wild caught Anopheles species were collected on August 26, 2011 at Tongilchon, near the demilitarized zone, in the ROK and shipped on August 28, 2011 to the Entomology Department, Armed Forces Research Institute of Medical Sciences (AFRIMS), Bangkok, Thailand. Four of the 5 species of the Anopheles Hyrcanus Group (Anopheles kleini, An. sinensis, An. pullus, and An. belenrae) were identified by polymerase chain reaction. Eggs were obtained from parent populations, and F, progeny provided blood meals using membrane feeding techniques. The mean numbers of eggs/female for An. sinensis and An. kleini were 157 and 146, respectively. Membrane blood feeding rates for An. sinensis (90.6%) and An. kleini (89.0%) were similar for F1-F30 generations. Colonies of An. kleini and An. sinensis were established by force mating techniques and maintained by the Insectary Section, Entomology Department, AFRIMS, to date. This is the 1st report of successful laboratory colonization of An. kleini collected from a vivax malaria high-risk area in the ROK.

Artificial membrane-feeding techniques have replaced direct feeding on animals for the maintenance of malaria and arbovirus vectors in many laboratories. Membrane feeding facilitates controlled experimentation of pathogen transmission during mosquito feeding. Sheep blood is commonly used due to its availability and low cost. We evaluated the impact of blood source (human, guinea pig, sheep, and hamster via direct feeding) on feeding rates, adult survival, fecundity, hatching rates, and developmental times for five species of laboratory-colonized mosquitoes (Anopheles dirus, An. cracens, An. minimus, An. sawadwongporni, and Ae. aegypti). We found that feeding rates differ among blood sources within mosquito species. Survival, fecundity, and hatching rates were lower in all Anopheles species and Ae. aegypti after membrane feeding on sheep blood. Survival rates seven days post-feeding on sheep blood were significantly lower (P<0.05) for An. dirus (84.2%), An. minimus (67.2%), An. sawadwongporni (51.5%), and An. cracens (35.5%) relative to other blood sources. An. minimus and An. sawadwongporni laid no eggs by seven days post-feeding with sheep blood, while An. dirus and An. cracens produced significantly fewer numbers of eggs and demonstrated significantly lower hatching rates relative to what was observed with the other blood sources. These findings support the conclusion that sheep blood is not a suitable blood source for laboratory rearing of Anopheles spp.

Leptotrombidium chiangraiensis Tanskul & Linthicum, and Leptotrombidium imphalum Vercammen-Grandjean are important vectors of scrub typhus in rice field habitats in northern Thailand. The developmental biology of all stages of the life cycle of two generations of these species of mites infected with Orientia tsutsugamushi (Hayashi) and uninfected mites is reported. The development of the infected lines of both F1 and F2 L. chiangraiensis were significantly longer than their respective uninfected lines (P < 0.05). The developmental times of uninfected and infected F1 lines of L. imphalum were not significantly different; however, F2 infected lines took significantly longer to develop (P < 0.05). Both F1 and F2 generations of infected L. imphalum and L. chiangraiensis oviposited on average >150 fewer eggs than uninfected mites.

Investigations have shown that female mosquitoes with a larger body size (determined by wing length) exhibit higher feeding rates and greater fecundity relative to smaller mosquitoes. In this study, Anopheles dirus and An. sawadwongporni were reared in the laboratory at two different temperatures (23 degrees C and 30 degrees C). Effects of the rearing temperature on body size, fecundity, and larval development period were examined by measuring wing length, adult body weight at emergence, the number of eggs produced and the length of time from the first to the fourth instar. Rearing temperature had a direct effect on body size, fecundity and larval development period for both species. Mosquitoes of both species reared at 23 degrees C were larger in body size, experienced prolonged development and produced a larger clutch of eggs relative to mosquitoes reared at 30 degrees C. However, there was no temperature effect on egg hatching rate and sex ratio.

The essential oils of Cananga odorata (ylang ylang), Citrus sinensis (orange), Cymbopogon citratus (lemongrass), Cymbopogon nardus (citronella grass), Eucalyptus citriodora (eucalyptus), Ocimum basilicum (sweet basil) and Syzygium aromaticum (clove), were tested for their insecticide activity against Aedes aegypti, Culex quinquefasciatus and Anopheles dirus using the WHO standard susceptibility test. These were applied in soybean oil at dose of 1%, 5% and 10% (w/v). C. citratus had the KT, values against the three mosquito species tested but the knockdown rates (at 10, 30 and 60 minutes) were lower than some essential oils. C. citratus oil had high insecticidal activity against Ae. aegypti, Cx. quinquefasciatus and An. dirus, with LC50 values of < 0.1, 2.22 and < 0.1%, respectively. Ten percent C. citratus gave the highest mortality rates (100%) 24 hours after application. This study demonstrates the potential for the essential oil of C. citratus to be used as an insecticide against 3 species of mosquitoes.

This study investigated insect bite protection and length of the protection with 30 repellents which were divided into 3 categories: plant oil, essential oil and essential oil with ethyl alcohol, tested against three mosquito species, Aedes aegypti, Anopheles minimus and Culex quinquefasciatus, under laboratory conditions. The plant oil group was comprised of Phlai (Zingiber cassumunar) and Sweet basil (Ocimum basilicum). Both substances were effective as repellents and feeding deterrents against An. minimus (205 minutes protection time and a biting rate of 0.9%), Cx. quinquefasciatus (165 minutes protection time and 0.9% biting rate) and Ae. aegypti (90 minutes protection time and 0.8% biting rate). Essential oil from citronella grass (Cymbopogon nardus) exhibited protection against biting from all 3 mosquito species: for An. minimus, Cx. quinquefasciatus and Ae. aegypti, the results were 130 minutes and 0.9%, 140 minutes and 0.8%, and 115 minutes and 0.8%, respectively. The period of protection time against Ae. aegypti for all repellent candidates tested was lower than the Thai Industrial Standards Institute (TISI) determined time of greater than 2 hours.
