RESUMO

The realization of multifunctional nanoparticle systems is essential to achieve highly efficient catalytic materials for specific applications; however, their production remains quite challenging. They are typically achieved through the incorporation of multiple inorganic components; however, incorporation of functionality could also be achieved at the organic ligand layer. In this work, we demonstrate the generation of multifunctional nanoparticle catalysts using peptide-based ligands for tandem catalytic functionality. To this end, chimeric peptides were designed that incorporated a Au binding sequence and a catalytic sequence that can drive ester hydrolysis. Using this chimera, Au nanoparticles were prepared, which sufficiently presented the catalytic domain of the peptide to drive tandem catalytic processes occurring at the peptide ligand layer and the Au nanoparticle surface. This work represents unique pathways to achieve multifunctionality from nanoparticle systems tuned by both the inorganic and bio/organic components, which could be highly important for applications beyond catalysis, including theranostics, sensing, and energy technologies.

RESUMO

Artificial hair sensors consisting of a piezoresistive carbon-nanotube-coated glass fiber embedded in a microcapillary are assembled and characterized. Individual sensors resemble a hair plug that may be integrated in a wide range of host materials. The sensors demonstrate an air-flow detection threshold of less than 1 m/s with a piezoresistive sensitivity of 1.3% per m/s air-flow change.

Peptide-capped AYSSGAPPMPPF gold nanoparticles were demonstrated for highly selective chemical vapor sensing using individual multivariable inductor-capacitor-resistor (LCR) resonators. Their multivariable response was achieved by measuring their resonance impedance spectra followed by multivariate spectral analysis. Detection of model toxic vapors and chemical agent simulants, such as acetonitrile, dichloromethane and methyl salicylate, was performed. Dichloromethane (dielectric constant εr = 9.1) and methyl salicylate (εr = 9.0) were discriminated using a single sensor. These sensing materials coupled to multivariable transducers can provide numerous opportunities for tailoring the vapor response selectivity based on the diversity of the amino acid composition of the peptides, and by the modulation of the nature of peptide-nanoparticle interactions through designed combinations of hydrophobic and hydrophilic amino acids.

INTRODUCTION: Participant's adherence to use of study product is a major concern in microbicide clinical trials, which can impact on proving product efficacy. In a previously described assay, single-use microbicide applicators exposed to the vagina were tested by spraying the applicator with trypan blue dye, resulting in vaginal mucus staining on inserted applicators. As subjects in our Phase 3 trials return applicators only at quarterly visits, often mixing inserted and not-inserted applicators together in the same bag, cross-contamination could confound results. In addition, trypan blue is carcinogenic and thus potentially hazardous to technicians spraying daily. METHODS: Applicators that were exposed to the vagina were placed in the same bag as unexposed applicators and shaken daily for up to 4 months. Validation was carried out in three clinical sites in South Africa. RESULTS: Trypan blue was replaced with FD&C Blue #1 granular food dye. Cross-contamination did not occur, nor did the length of time affect reaction to dye. In South Africa, the assay was validated with an accuracy of over 95%. CONCLUSION: Applicator assay modifications render the test safe and suitable for use in clinical trials.

Silk is an attractive biomaterial for use in tissue engineering applications because of its slow degradation, excellent mechanical properties, and biocompatibility. In this report, we demonstrate a simple method to cast patterned films directly from silk fibroin dissolved in an ionic liquid. The films cast from the silk ionic liquid solution were found to support normal cell proliferation and differentiation. The versatility of the silk ionic liquid solutions and the ability to process large amounts of silk into materials with controlled surface topography directly from the dissolved silk ionic liquid solution could enhance the desirability of biomaterials such as silk for a variety of applications.

OBJECTIVE: The objective of this article is to study the effect of PC-815, a novel combination microbicide containing carrageenan and the nonnucleoside reverse transcriptase inhibitor (NNRTI) MIV-150, in blocking HIV-1 and HIV-2 infections in vitro as compared with Carraguard alone. GOAL: The goal of this study was to develop a combination microbicide that is more efficacious than Carraguard against HIV-1 and HIV-2. STUDY DESIGN: The microtiter syncytial assay was used to evaluate: 1) the antiviral and virucidal activity of MIV-150 against HIV-1MN; 2) the additive effect of MIV-150 when combined with carrageenan; and 3) a possible interference of seminal fluid in the antiviral activity of these compounds. RESULTS: MIV-150 effectively inactivated free virus. Combination of MIV-150 and Carraguard demonstrated an additive antiviral effect. Seminal fluid had no effect on the antiviral activity of MIV-150 or Carraguard. The average concentration that blocks 50% of infection (EC50) for PC-815 was approximately 10 times stronger than Carraguard for the different clinical isolates used in the study. CONCLUSION: Theoretically, PC-815 is likely to be a more efficacious microbicide than Carraguard.

The structure of thin films cast from regenerated solutions of Bombyx mori cocoon silk in hexafluoroisopropyl alcohol (HFIP) was studied by synchrotron X-ray diffraction during heating. A solid-state conformational transition from an alpha-helical structure to the well-known beta-sheet silk II structure occurred at a temperature of approximately 140 degrees C. The transition appeared to be homogeneous, as both phases do not coexist within the resolution of the current study. Modulated differential scanning calorimetry (DSC) of the films showed an endothermic melting peak followed by an exothermic crystallization peak, both occurring near 140 degrees C. Oriented fibers were also produced that displayed this helical molecular conformation. Subsequent heating above the structural transition temperature produced oriented beta-sheet fibers very similar in structure to B. mori cocoon fibers. Heat treatment of silk films at temperatures well below their degradation temperature offers a controllable route to materials with well-defined structures and mechanical behavior.

In this work, the suitability of imidazolium-based ionic liquid solvents is investigated for the dissolution and regeneration of silkworm (Bombyx mori) silk. Within an ionic liquid the anion plays a larger role in dictating the ultimate solubility of the silk. The dissolution of the silk in the ionic liquid is confirmed using wide-angle X-ray scattering. The dissolved silk is also processed into 100 mum-thick, two-dimensional films, and the structure of these films is examined. The rinse solvent, acetonitrile or methanol, has a profound impact on both the topography of the films and the secondary structure of the silk protein. The image depicts a silkworm cocoon dissolved in 1-butyl-3-methylimidazolium chloride and then regenerated as a film with birefringence.

It has been shown that men who have sex with men actively seek lubricants that contain nonoxynol-9 (N-9) because they believe that N-9 may help to prevent infection by HIV. However, indirect evidence suggests that N-9 may actually enhance infection. Microscopic examination of rectal lavage and biopsy specimens collected at different time points following rectal application of a lubricant containing 2% N-9 showed rapid exfoliation of the rectal epithelium. Because the rectal epithelium protects target cells in the submucosa from HIV, we conclude that lubricants containing N-9 should be avoided during rectal sex.

OBJECTIVES: To develop an accurate, rapid, and inexpensive method for verifying vaginal applicator use. GOAL: To develop a method for assessing compliance in microbicide clinical trials. STUDY DESIGN: Single use Microlax applicators containing a placebo formulation either were or were not exposed to the vagina. Three assays were developed to determine whether the applicators had been used vaginally. RESULTS: Blinded examiners were able to discern 63% of the time whether or not applicator tips had been exposed to the vagina. Optical density (to measure lactobacilli), increased in media exposed to used applicators but not in media exposed to unused applicators. When tips of applicators were stained with trypan blue, used applicators could be distinguished easily from unused applicators. CONCLUSION: Staining of applicator is accurate, simple, rapid, and inexpensive. This method could be be used in clinical settings in the developing world. Dying applicator tips could prove useful in excluding non-compliant subjects, analyzing data, or developing social intervention strategies to improve compliance.

OBJECTIVE: The current study was conducted to determine the relative safety of sexual lubricants for rectal use. GOAL: Our goal was to identify a sexual lubricant that would not damage the rectal epithelium. STUDY DESIGN: We describe a mouse assay to determine the degree to which lubricants cause rectal sloughing. We also tested lubricants for cytotoxicity and their effect on Herpes simplex virus (HSV-2) infection after rectal challenge. RESULTS: Of the products tested, Delube and KY-Plus are most likely to damage the rectal epithelium, whereas Viamor, Vagisil, and Astroglide would cause some degree of rectal damage. PBS, Carraguard, and methylcellulose were not toxic in any of the assays. CONCLUSION: We have developed a quantitative method to assay the degree of sloughing of the rectal epithelium. Using this and other techniques, we are able to predict the degree to which lubricants could be safe for rectal use.

Sexual transmission of human immunodeficiency virus (HIV) accounts for the majority of new infections worldwide. However, the mechanism of viral transmission across the mucosal barrier is poorly understood. By use of an ectocervical epithelium-derived cell line, we found that the cells are capable of sequestering large amounts of HIV particles but are refractory to cell-free viral infection. The sequestered virus particles remained infectious for >/=6 days and resisted treatment with trypsin. Upon coculture with CD4(+)-susceptible cells, epithelial cells can effectively transmit the virus to these cells, which can result in robust infection of the target cells. Inhibitory studies have shown that heparan sulfate moiety of cell-surface proteoglycans is involved in the viral attachment to these CD4-negative epithelial cells. Genital epithelial cells may play active roles in sequestering, protecting, and transferring virus during sexual transmission of HIV.

Considerable evidence suggests that human immunodeficiency virus (HIV)-infected macrophages and/or lymphocytes may mediate sexual transmission of HIV. We and others have previously demonstrated that when vitally stained donor mouse lymphocytes or macrophages are placed in the vaginas of mice, some of the stained cells can later be found in the iliac lymph nodes. The aim of this study was to assess the extent of mononuclear cell trafficking from the vagina and to test the possibility that carrageenan formulation, a sulfated polysaccharide formulation containing 3% PDR98-15 carrageenan (PC-515; FMC Biopolymer, Rockland, ME), a vaginal microbicide, would prevent vaginal transmigration of macrophages. When supravitally stained mouse macrophages and T cells were inoculated into the vagina of recipient mice, discrete numbers of donor cells migrated to the recipient iliac and inguinal lymph nodes and spleen. When recipient mice were preinoculated with the carrageenan formulation, the number of macrophages in lymph nodes and spleen was reduced by >90%. In contrast, a methylcellulose formulation, which is believed to be inactive, did not significantly reduce migration to the lymphoid organs. Our findings suggest that the carrageenan formulation blocks cell trafficking of macrophages from vagina and that blocking does not result from cytotoxicity. Blocking cell trafficking may help to prevent sexual transmission of HIV.

RESUMO

Approximately 60 vaginal microbicides are under development for the prevention of HIV/AIDS and other sexually transmitted pathogens. The history and current status of the field are discussed with emphasis on the lessons learned from recent clinical trials, along with an emphasis on the mechanisms involved in the sexual transmission of HIV and how this information influences microbicide development. Additionally, the current status of in vitro and animal systems used for evaluating microbicide efficacy, as well as the challenges involved in developing more appropriate and practical assays, are discussed. Also discussed are the challenges that face the microbicide product development field in meeting US Food and Drug Administration requirements regarding product safety and stability.

Scanning electron microscopy was employed to examine the manner of association between in vitro capacitated spermatozoa and zona-free eggs of the hamster. Spermatozoa with intact acrosomes, which were unable to fuse with eggs, were seen in general associated with egg microvilli in the region of the acrosomal cap. Acrosome-reacting spermatozoa were seen associated with egg microvilli with the dissociating acrosomal caps. Acrosome-reacted spermatozoa, which were able to fuse with eggs, generally associated with egg microvilli by the equatorial segment and the anterior portion of the postacrosomal region. It is inferred that the completion of the acrosome reaction signals changes in the plasma membrane over the equatorial segment of the acrosome and the anterior area of the postacrosomal region which give it a greater affinity to and fusibility with the oolemma.
