RESUMO

Biotransformations are reactions mediated by microorganisms, such as fungi. These bioreactions have high chemo- and stereoselectivity on organic substrates and can be applied in the search for new bioactive compounds. In this study, acanthoic acid (AA) was biotransformed using the fungus Xylaria sp., giving the novel compound 3ß,7ß-dihydroxyacanthoic acid (S1). Both the AA and the product S1 were tested against Gram-positive and Gram-negative bacteria. To identify and validate possible biological targets as enzymes or proteins involved in the activity observed in vitro, we used the molecular docking method. Hydroxylation at the C-3 and C-7 positions of the biotransformation product enhanced its activity against Escherichia coli as well as its binding affinity and interactions with superoxide dismutase 1 (SOD1; PDB ID 4A7G). Based on our results, the SOD1 enzyme was suggested to be a possible target for the antioxidant activity of product S1.

INTRODUCTION: Leishmaniasis, a disease caused by a parasite endemic to large areas of tropical and subtropical countries, is a growing public health problem. METHODS: Male BALB/c mice were infected with Leishmania amazonensis and treated with extracts isolated from Annona mucosa. RESULTS: Treated groups had significantly reduced footpad swelling. The group treated intraperitoneally with hexane extract showed footpad swelling similar to groups treated with Pentamidine® and Glucantime®. Groups treated with dichloromethane extract and hexane extract presented the recovering phenotype associated with reduced parasite levels. CONCLUSIONS: Extracts of A. mucosa are promising sources of novel antileishmanial compounds.

RESUMO

The essential oil obtained by hydrodistillation from leaves of Anaxagorea brevipes was analysed by gas chromatography fitted with a flame ionisation detector (GC-FID) and coupled to mass spectrometry (GC-MS). Thirty one components were identified, representing around 75.7% of total oil. The major components were ß-eudesmol (13.16%), α-eudesmol (13.05%), γ-eudesmol (7.54%), guaiol (5.12%), caryophyllene oxide (4.18%) and ß-bisabolene (4.10%). The essential oil showed antimicrobial activity against Gram-positive bacteria and yeast with the MIC values between 25.0 and 100 µg/mL. The highest antiproliferative activity was observed for the oil against MCF-7 (breast, TGI = 12.8 µg/mL), NCI-H460 (lung, TGI = 13.0 µg/mL) and PC-3 (prostate, TGI = 9.6 µg/mL) cell lines, while against no cancer cell line HaCat (keratinocyte) the TGI was 38.8 µg/mL. The oil exhibited a small antioxidant activity assessed through ORAC-FL assay (517 µmol TE/g). This is the first report regarding the chemical composition and bioactivity of A. brevipes essential oil.

In vitro evaluation of alkaloidal fractions of twigs, barks and leaves from two Unonopsis species, Unonopsis guatterioides R.E. Fr. and Unonopsis duckei R.E. Fr., Annonaceae, against promastigote forms of Leishmania amazonensis revealed these species as sources of substances with promising leishmanicidal potential. All alkaloidal fractions from twigs, barks and leaves of U. guatterioides were classified as highly active, with IC50 1.07, 1.90, and 2.79 mg/mL, respectively. Only the alkaloidal fraction from the twigs of U. duckei was classified as inactive.

RESUMO

Phytochemical investigation of the branches of Annona foetida Mart. led to isolation from the CH(2)Cl(2) extract of four alkaloids: Atherospermidine (1), described for the first time in this species, liriodenine (2), O-methylmoschatoline (3), and annomontine (4). Their chemical structures were established on the basis of spectroscopic data from IR, MS, NMR (1D and 2D), and comparison with the literature. Compounds 2-4 showed potent trypanocidal effect when evaluated against epimastigote and trypomastigote forms of Trypanosoma cruzi.

RESUMO

Using modern NMR techniques, including 1H--13C and 1H--15N heteronuclear correlation experiments, the complete and unambiguous 1H, 13C, and 15N NMR chemical shift assignments of annomontine, methoxyannomontine, and N-hydroxyannomontine pyrimidine-beta-carboline alkaloids were performed. All 1H--1H scalar coupling constants and signal multiplicities were determined, and all nOe observations were also included.

RESUMO

Das plantas tóxicas da Amazônia Palicourea longiflora e Strychnos cogens foram isolados 571 fungos endofíticos e 74 bactérias endofíticas. Palicourea longiflora (Rubiaceae) e outras espécies desse gênero estão relacionadas a 90 por cento das mortes de gado na região Amazônica. Strychnos cogens (Loganiaceae) e outras espécies de Strychnos são utilizadas pelos indígenas na confecção de "curares". Entre os endófitos isolados de P. longiflora foram identificados os fungos: Colletotrichum sp. e seu telemorfo Glomerella sp., Guignardia sp., Aspergillus niger, Phomopsis sp. e Xylaria sp., além da bactéria Burkholderia gladioli, pertencente a um grupo de fixadoras de nitrogênio. Dos isolados de S. cogens foram identificados os fungos: Colletotrichum sp., Guignardia sp., Aspergillus niger e Trichoderma sp. Uma amostra de 79 isolados fúngicos teve seus metabólitos extracelulares bioensaiados contra microrganismos patogênicos e fitopatogênicos: 19 isolados inibiram um ou mais microrganismos-teste. Dos oito isolados com melhores resultados de inibição, as móleculas bioativas eram menores que 12.000 daltons, fato verificado pela diálise dos metabólitos.
