RESUMO

T follicular helper (Tfh) and T follicular regulatory (Tfr) cells are the two T cell subsets able to interact with B cells driving germinal center (GC) reactions. These T-B interactions are important for protective immune responses within secondary lymphoid tissue. However, the pathological emergence of ectopic lymphoid structures (ELS) that characterize several autoimmune diseases also involves Tfh and Tfr cells. ELS, often with ectopic GCs, can be identified through biopsies. Sjögren's syndrome (SS) is an example of an autoimmune disease where minor salivary gland (MSG) biopsies are often performed for diagnosis and where ELS can be found. Here, we describe a protocol to identify and isolate T follicular cells from MSGs by flow cytometry and immunohistochemistry.

The ability to recognize the emotions of others is a crucial skill. In the visual modality, sensorimotor mechanisms provide an important route for emotion recognition. Perceiving facial expressions often evokes activity in facial muscles and in motor and somatosensory systems, and this activity relates to performance in emotion tasks. It remains unclear whether and how similar mechanisms extend to audition. Here we examined facial electromyographic and electrodermal responses to nonverbal vocalizations that varied in emotional authenticity. Participants (N = 100) passively listened to laughs and cries that could reflect an authentic or a posed emotion. Bayesian mixed models indicated that listening to laughter evoked stronger facial responses than listening to crying. These responses were sensitive to emotional authenticity. Authentic laughs evoked more activity than posed laughs in the zygomaticus and orbicularis, muscles typically associated with positive affect. We also found that activity in the orbicularis and corrugator related to subjective evaluations in a subsequent authenticity perception task. Stronger responses in the orbicularis predicted higher perceived laughter authenticity. Stronger responses in the corrugator, a muscle associated with negative affect, predicted lower perceived laughter authenticity. Moreover, authentic laughs elicited stronger skin conductance responses than posed laughs. This arousal effect did not predict task performance, however. For crying, physiological responses were not associated with authenticity judgments. Altogether, these findings indicate that emotional authenticity affects peripheral nervous system responses to vocalizations. They also point to a role of sensorimotor mechanisms in the evaluation of authenticity in the auditory modality.

Germinal center (GC) responses are controlled by T follicular helper (Tfh) and T follicular regulatory (Tfr) cells and are crucial for the generation of high-affinity antibodies. Although the biology of human circulating and tissue Tfh cells has been established, the relationship between blood and tissue Tfr cells defined as CXCR5+Foxp3+ T cells remains elusive. We found that blood Tfr cells are increased in Sjögren syndrome, an autoimmune disease with ongoing GC reactions, especially in patients with high autoantibody titers, as well as in healthy individuals upon influenza vaccination. Although blood Tfr cells correlated with humoral responses, they lack full B cell-suppressive capacity, despite being able to suppress T cell proliferation. Blood Tfr cells have a naïve-like phenotype, although they are absent from human thymus or cord blood. We found that these cells were generated in peripheral lymphoid tissues before T-B interaction, as they are maintained in B cell-deficient patients. Therefore, blood CXCR5+Foxp3+ T cells in human pathology indicate ongoing humoral activity but are not fully competent circulating Tfr cells.

The construction, evaluation and analytical application of potentiometric sensors sensitive to vitamin B1 and vitamin B6 are reported. The solid contact electrodes were produced using beta-cyclodextrins as ionophores in a carboxylated poly(vinyl chloride) support matrix. Near Nernstian slopes (mV/decade) of 51.7+/-0.8, 60.6+/-0.6 and 61.1+/-1.4, within the intervals (M) of 1.0 x 10(-4) to 1.0 x 10(-1), 5.8 x 10(-5) to 1.0 x 10(-1) and 4.3 x 10(-5) to 1.0 x 10(-1) were obtained, for thiamine and pyridoxine I and II prepared membranes, respectively. A pH operational range of 6.5-8.5 for thiamine and 2-4.5 for pyridoxine electrodes was found. Assessment of selectivity coefficients toward a large number of inorganic cations and organic cations usually present in multivitamin formulations revealed good performance. Analysis of vitamins B1 and B6 in complex multivitamin drugs was achieved with recoveries within the intervals of 95.1-99.6% for thiamine and 95.1-102% for pyridoxine. Furthermore, the results enabled by the proposed procedure revealed good agreement with those provided by HPLC.
