RESUMO

Despite their ability to mitigate climate change by efficiently absorbing atmospheric carbon dioxide (CO2) and acting as natural long-term carbon sinks, mangrove ecosystems have faced several anthropogenic threats over the past century, resulting in a decline in the global mangrove cover. By using standardized methods and the most recent Bayesian tracer mixing models MixSIAR, this study aimed to quantify source contributions, burial rates, and stocks of organic carbon (Corg) and explore their temporal changes (∼100 years) in seven lead-210 dated sediment cores collected from three contrasting Mexican mangrove areas. The spatial variation in Corg burial rates and stocks in these blue carbon ecosystems primarily depended on the influence of local rivers, which controlled Corg sources and fluxes within the mangrove areas. The Corg burial rates in the cores ranged from 66 ± 16 to 400 ± 40 g m-2 yr-1. The Corg stocks ranged from 84.9 ± 0.7 to 255 ± 2 Mg ha-1 at 50 cm depth and from 137 ± 2 to 241 ± 4 Mg ha-1 at 1 m depth. The highest Corg burial rates and stocks were observed in cores from the carbonate platform of Yucatan and in cores with reduced river influence and high mangrove detritus inputs, in contrast to patterns identified from global databases. Over the past century, the rising trends in Corg burial rates and stocks in the study sites were primarily driven by enhanced inputs of fluvial-derived Corg and, in some cores, mangrove-derived Corg. Despite their decreasing extension, mangrove areas remained highly effective producers and sinks of Corg. Ongoing efforts to enhance the global database should continue, including mangrove area characteristics and reliable timescales to facilitate cross-comparison among studies.

RESUMO

The influence of manganese modification on the spectroscopic features of manganese-doped CeO2 systems synthesized by the microwave-assisted hydrothermal route and their correlation with the presence of O defective structures were verified, focusing on their interaction with poisonous atmospheres. Raman and electron paramagnetic resonance studies confirmed the presence of defective clusters formed by dipoles and/or quadrupoles. The number of paramagnetic species was found to be inversely proportional to the doping concentration, resulting in an increase in the Mn2+ signal, likely due to the reduction of Mn3+ species after the interaction with CO. X-ray photoelectron spectroscopy data showed the pure system with 33% of its cerium species in the Ce3+ configuration, with an abrupt decrease to 19%, after the first modification with Mn, suggesting that 14% of the Ce3+ species are donating one electron to the Mn2+ ions, thus becoming nonparamagnetic Ce4+ species. On the contrary, 58% of the manganese species remain in the Mn2+ configuration with five unpaired electrons, corroborating the paramagnetic feature of the samples seen in the electron paramagnetic resonance study.

RESUMO

This study reports the synthesis of hybrid nanostructures composed of cerium dioxide and microcrystalline cellulose prepared by the microwave-assisted hydrothermal route under distinct temperature and pH values. Their structural, morphological and spectroscopic behaviors were investigated by X-Rays Diffraction, Field Emission Gun Scanning Electron Microscopy, High-Resolution Transmission Electron Microscopy, and Fourier-Transform Infrared, Ultraviolet-Visible, Raman and Positron Annihilation Lifetime spectroscopies to evaluate the presence of structural defects and their correlation with the underlying mechanism regarding the biocide activity of the studied material. The samples showed mean crystallite sizes around 10 nm, characterizing the formation of quantum dots unevenly distributed along the cellulose surface with a certain agglomeration degree. The samples presented the characteristic Ce-O vibration close to 450 cm-1 and a second-order mode around 1050 cm-1, which is indicative of distribution of localized energetic levels originated from defective species, essential in the scavenging of reactive oxygen species. Positron spectroscopic studies showed first and second lifetime components ranging between 202-223 ps and 360-373 ps, respectively, revealing the presence of two distinct defective oxygen species, in addition to an increment in the concentration of Ce3+-oxygen vacancy associates as a function of temperature. Therefore, we have successfully synthesized hybrid nanoceria structures with potential multifunctional therapeutic properties to be further evaluated against the COVID-19.

RESUMO

Globally, oxygen concentration in many coastal areas is depleting. River nutrient discharges may produce hypoxia events. The Southern Gulf of Mexico receives the discharges of the Grijalva-Usumacinta River System, the second largest in the Gulf of Mexico. To evaluate the influence of river discharges on dissolved oxygen concentrations in the receiving coastal ecosystem, we studied the variation of physicochemical variables in the water column. During the dry season, the influence of the river waters to the coastal area is scarce, but during the rainy season the river plume reached ~9 km offshore. The lowest concentration of dissolved oxygen (3.6 mg L-1) was observed within the river plume. We concluded that, in the studied area, hypoxia events (oxygen concentrations ≤ 2 mg L-1) would occur during the rainy season, low winds and in deeper waters (>80 m depth).

RESUMO

The development of the Grijalva-Usumacinta river basin exerts modifications on its discharge area. A sediment core was studied to reconstruct environmental changes and trace element contamination status during the past 45 years. 210Pb-derived mass accumulation rates indicate higher sediment input to the area since 1995, related to increased precipitation and floodings in the catchment area. Sediments show finer particles from the late 1970s on, likely related to dams construction upriver and/or land use changes. Heavy metal enrichment factors (EF < 2) suggest minimum contamination. Benthic foraminifera and redox-sensitive - elements (As, Ba, Co, Cr, Cu, Ni, Pb, V and Zn) indicate the sediments before 2000 were deposited under oxygenated conditions. Afterwards, environmental conditions changed and benthic foraminifera and dinocysts assemblages changed suggesting eutrophication and lower oxygen conditions during the last 20 years. Monitoring should be continued to assess eutrophication/hypoxic/pollution trends that could become deleterious to the marine biota.

RESUMO

In the present study, we describe the molecular and biochemical characterization of sunflower (Helianthus annuus L.) enolase (ENO, EC 4.2.1.11) proteins, which catalyze the formation of phosphoenolpyruvate, the penultimate intermediate in the glycolytic pathway. We cloned and characterized three cDNAs encoding different ENO isoforms from developing sunflower seeds. Studies using fluorescently tagged ENOs confirmed the predicted subcellular localization of ENO isoforms: HaENO1 in the plastid while HaENO2 and HaENO3 were found in the cytosol. The cDNAs were used to express the corresponding 6(His)-tagged proteins in Escherichia coli. The proteins were purified to electrophoretic homogeneity, using immobilized metal ion affinity chromatography, and biochemically characterized. Recombinant HaENO1 and HaENO2, but not HaENO3 were shown to have enolase activity, in agreement with data obtained with the Arabidopsis homolog proteins. Site directed mutagenesis of several critical amino acids was used to attempt to recover enolase activity in recombinant HaENO3, resulting in very small increases that were not additive. A kinetic characterization of the two active isoforms showed that pH had similar effect on their velocity, that they had similar affinity for 2-phosphoglycerate, but that the kcat/Km of the plastidial enzyme was higher than that of the cytosolic isoform. Even though HaENO2 was always the most highly expressed transcript, the levels of expression of the three ENO genes were remarkably distinct in all the vegetative and reproductive tissues studied. This indicates that in seeds the conversion of 2-phosphoglycerate to phosphoenolpyruvate takes place through the cytosolic and the plastidial pathways therefore both routes could contribute to the supply of carbon for lipid synthesis. The identity of the main source of carbon during the period of stored products synthesis is discussed.

RESUMO

Three cDNAs encoding different phosphoglycerate kinase (PGK, EC 2.7.2.3) isoforms, two cytosolic (HacPGK1 and HacPGK2) and one plastidic (HapPGK), were cloned and characterized from developing sunflower (Helianthus annuus L.) seeds. The expression profiles of these genes showed differences in heterotrophic tissues, such as developing seeds and roots, where HacPGK1 was predominant, while HapPGK was highly expressed in photosynthetic tissues. The cDNAs were expressed in Escherichia coli, and the corresponding proteins purified to electrophoretic homogeneity, using immobilized metal ion affinity chromatography, and biochemically characterized. Despite the high level of identity between sequences, the HacPGK1 isoform showed strong differences in terms of specific activity, temperature stability and pH sensitivity in comparison to HacPGK2 and HapPGK. A polyclonal immune serum was raised against the purified HacPGK1 isoform, which showed cross-immunoreactivity with the other PGK isoforms. This serum allowed the localization of high expression levels of PGK isozymes in embryo tissues.

RESUMO

Studies on age and growth in fishes are essential to establish models on population dynamics. The previous issues were determined in this study by scale growth analyses. Between September 2003 and May 2004, 382 organisms were captured using gill and atarraya nets. Eight samples were developed every month a long this study. Standard length in centimeters (L(e)), total weight in grams (P9t)), scale length in centimeters (L(esc)) and growth rings were determined. The size and weight ranged from 38 to 232 mm (L(e)) and 8.7 to 311.9 g (P(t)), respectively. The growth rings analyses reveal four age groups, being the second group the most representative with 34.3% of the total sampled. The scale ring analyses showed two growth rings, one develops between November-January and the other between April-May, in good correlation with low temperature and gonad development, respectively. The growth parameters were established as: L infinity = 281.1mm, W infinity = 877.1g, K = 0.33 and T(0) = 0.88 year(-1). The growth curves based on length and weight were described by using the von Bertalanffy model: L(e) = 28.11[1 -e (-0.33(t+0.88))] and P(t) = 877.17[1-e (-0.33(t+0.88))]3. Finally, it is concluded that commercial catches in this dam are between a year and year and half-old when exploited.

A full-length hexokinase cDNA, HaHXK1, was cloned and characterized from Helianthus annuus L. developing seeds. Based on its sequence and phylogenetic relationships, HaHXK1 is a membrane-associated (type-B) hexokinase. The predicted structural model resembles known hexokinase structures, folding into two domains of unequal size: a large and a small one separated by a deep cleft containing the residues involved in the enzyme active site. A truncated version, without the 24 N-terminal residues, was heterologously expressed in Escherichia coli, purified to electrophoretic homogeneity using immobilized metal ion affinity chromatography and biochemically characterized. The purified enzyme behaved as a monomer on size exclusion chromatography and had a specific activity of 19.3 µmol/min/mg protein, the highest specific activity ever reported for a plant hexokinase. The enzyme had higher affinity for glucose and mannose relative to fructose, but the enzymatic efficiency was higher with glucose. Recombinant HaHXK1 was inhibited by ADP and was insensitive either to glucose-6-phosphate or to trehalose-6-phosphate. Its expression profile showed higher levels in heterotrophic tissues, developing seeds and roots, than in photosynthetic ones. A time course of HXK activity and expression in seeds showed that the highest HXK levels are found at the early stages of reserve compounds, lipids and proteins accumulation.

RESUMO

Lipid biosynthesis in developing sunflower (Helianthus annuus L.) seeds requires reducing power. One of the main sources of cellular NADPH is the oxidative pentose phosphate pathway (OPPP), generated from the oxidation of glucose-6-phosphate. This glycolytic intermediate, which can be imported to the plastid and enter in the OPPP, is the substrate and product of cytosolic phosphoglucose isomerase (cPGI, EC 5.3.1.9). In this report, we describe the cloning of a full-length cDNA encoding cPGI from developing sunflower seeds. The sequence was predicted to code for a protein of 566 residues characterised by the presence of two sugar isomerase domains. This cDNA was heterologously expressed in Escherichia coli as a His-tagged protein. The recombinant protein was purified using immobilised metal ion affinity chromatography and biochemically characterised. The enzyme had a specific activity of 1,436 micromol min(-1) mg(-1) and 1,011 micromol min(-1) mg(-1) protein when the reaction was initiated with glucose-6-phosphate and fructose-6-phosphate, respectively. Activity was not affected by erythrose-4-phosphate, but was inhibited by 6-P gluconate and glyceraldehyde-3-phosphate. A polyclonal immune serum was raised against the purified enzyme, allowing the study of protein levels during the period of active lipid synthesis in seeds. These results were compared with PGI activity profiles and mRNA expression levels obtained from Q-PCR studies. Our results point to the existence of a possible post-translational regulatory mechanism during seed development. Immunolocalisation of the protein in seed tissues further indicated that cPGI is highly expressed in the procambial ring.

RESUMO

INTRODUCTION AND OBJECTIVES: Different neuroimaging findings have been described in association with progressive supranuclear palsy (PSP), but their use as diagnostic support in this condition has been the subject of much discussion. PATIENTS AND METHODS: Three radiologists, non-specialists in neuroradiology and with no specific information regarding neuroimaging in Parkinson syndromes, analyzed (without knowing any clinical details) seven cranial CT in five patients diagnosed as probable PSP according to NINDS-SPSS criteria, nine with idiopathic Parkinson's disease, and six persons aged over 55 years who acted as controls. The radiologists were asked to assess 17 variables as absent, moderate or severe. The results were analyzed using the chi squared test for qualitative variables and Fisher's exact test when necessary. RESULTS: The identification of the variables antero-posterior and transversal atrophy of the mid-brain, atrophy of the pons, enlargement of the perimesencephalic cisterns and the quadrigeminal plate and increased size of the third ventricle were considered to be statistically significant in the cases of PSP as compared with other observations. The small number of patients did not permit the establishment of correlation of statistical importance between the radiological parameters and clinical condition. CONCLUSIONS: We present the point of view of non-specialist neuroimaging workers, in these patients. We found that there were six parameters of interest on cranial CT which permitted differentiation of cases of PSP, idiopathic Parkinson's disease and a control population. Four of these parameters did not appear to a severe degree in patients who did not have PSP. These results are partly comparable to those published in the literature.

RESUMO

OBJECTIVES: To study demographic data about the first time consultations for headache in our neurologic clinic, its diagnostic distribution, its diagnostic and therapeutic management by the neurologist, its source, and the influence of the primary care doctors formation in their search of neurological consultation and therapeutic management in patients with headache. PATIENTS AND METHODS: A protocol with several demographic clinical and medical care data of the all first time visits for headache in our province was picked up, during 3 consecutive months. RESULTS: Headaches represent 21.5% of the first time consultations. The majority of the patients were women. The primary care doctors sent 93.6% of patients. The two more common diagnoses were tension-type headache (60%) and migraine (25.4%), reflecting their prevalences in the population. The more common treatments were the associations of two drugs. The general doctors sent more patients with headache (3.36 per thousand and year) than family doctors (1.84 per thousand and year). The approximation between initial diagnosis (primary care doctor) and final diagnosis (neurologist) was 26.4%, greater for family doctors (38.1%) than for general doctors (23.9%). CONCLUSIONS: Headaches represent the first cause of neurological consultation. The diagnostic distribution of the patients sent to our province shows the prevalences in the population. The family medicine formation is useful for a better selection of the patients sent to neurologists and a better diagnostic approach.
