The toxicity of particulate matter (PM) is strictly associated with its physical-chemical characteristics, such as size or chemical composition. While these properties depend on the origin of the particles, the study of the toxicological profile of PM from single sources has rarely been highlighted. Hence, the focus of this research was to investigate the biological effects of PM from five relevant sources of atmospheric PM: diesel exhaust particles, coke dust, pellet ashes, incinerator ashes, and brake dust. Cytotoxicity, genotoxicity, oxidative, and inflammatory response were assessed in a bronchial cell line (BEAS-2B). BEAS-2B cells were exposed to different concentrations (25, 50, 100, and 150 µg/mL medium) of particles suspended in water. The exposure lasted 24 h for all the assays performed, except for reactive oxygen species, which were evaluated after 30 min, 1 h, and 4 h of treatment. The results showed a different action of the five types of PM. All the tested samples showed a genotoxic action on BEAS-2B, even in the absence of oxidative stress induction. Pellet ashes seemed to be the only ones able to induce oxidative stress by boosting the formation of reactive oxygen species, while brake dust resulted in the most cytotoxic. In conclusion, the study elucidated the differential response of bronchial cells to PM samples generated by different sources. The comparison could be a starting point for a regulatory intervention since it highlighted the toxic potential of each type of PM tested.

Knowledge of the refractive index of water in the deeply supercooled metastable liquid state is important, for example, for an accurate description of optical reflection and refraction processes occurring in clouds. However, a measurement of both the temperature and wavelength dependence of the refractive index under such extreme conditions is challenging. Here, we employ Raman spectroscopy in combination with microscopic water jets in vacuum to obtain the refractive index of supercooled water to a lowest temperature of 230.3 K. While our approach is based on the analysis of Mie resonances in Raman spectra measured by using a single excitation wavelength at 532 nm, it allows us to obtain the refractive index in a wide visible wavelength range from 534 to 675 nm. Because of a direct link between the refractive index and density of water, our results provide a promising approach to help improve our understanding of water's anomalous behavior.

The future of halide perovskites (HaPs) is beclouded by limited understanding of their long-term stability. While HaPs can be altered by radiation that induces multiple processes, they can also return to their original state by "self-healing." Here two-photon (2P) absorption is used to effect light-induced modifications within MAPbI3 single crystals. Then the changes in the photodamaged region are followed by measuring the photoluminescence, from 2P absorption with 2.5 orders of magnitude lower intensity than that used for photodamaging the MAPbI3 . After photodamage, two brightening and one darkening process are found, all of which recover but on different timescales. The first two are attributed to trap-filling (the fastest) and to proton-amine-related chemistry (the slowest), while photodamage is attributed to the lead-iodide sublattice. Surprisingly, while after 2P-irradiation of crystals that are stored in dry, inert ambient, photobrightening (or "light-soaking") occurs, mostly photodarkening is seen after photodamage in humid ambient, showing an important connection between the self-healing of a HaP and the presence of H2 O, for long-term steady-state illumination, practically no difference remains between samples kept in dry or humid environments. This result suggests that photobrightening requires a chemical-reservoir that is sensitive to the presence of H2 O, or possibly other proton-related, particularly amine, chemistry.

We describe a light scattering technique for characterizing colloidal samples under constant flow. It exploits the properties of speckles in the deep Fresnel region-the so-called near field speckles-providing absolute scattering measurements of the static form factor of the sample, as described extensively by Mazzoni et al. [Rev. Sci. Instrum. 84, 043704 (2013)] for static samples. We exploit a strongly astigmatic beam for illuminating the scattering volume with a light sheet a few microns thick. This largely improves the sensitivity of the method to small signals. Moreover, by flowing the sample in the direction perpendicular to the light sheet, the transit times are reduced to a minimum, allowing for fast measurements. We tested the instrument with suspensions of calibrated colloidal polystyrene spheres with a size comparable to the light wavelength. In particular, we recovered the static form factors of suspensions of spherical particles and the phase lag of the zero-angle scattering amplitude, which both compare well to Mie theory predictions. We then applied the method to colloidal fractal aggregates of sub-wavelength particles and measured their fractal dimension. The instrument is designed to be operational in continuous flow analysis systems.

Traffic-related emissions include gas and particles that can alter air quality and affect human and environmental health. Limited studies have demonstrated that particulate debris thrown off from brakes are toxic to higher plants. The acute phytotoxicity of brake pad wear debris (BPWD) investigated using cress seeds grown in soil contaminated with increasing concentrations of debris. Two types of pads were used: a commercially available phenol based pad and an innovative cement-based pad developed within of the LIFE+ COBRA project. The results suggested that even through the BPWD generated by the two pads were similar in and morphology, debris from traditional pads were more phytotoxic than that from cementitious pads, causing significant alterations in terms of root elongation and loss of plasma membrane integrity.

Self-healing, where a modification in some parameter is reversed with time without any external intervention, is one of the particularly interesting properties of halide perovskites. While there are a number of studies showing such self-healing in perovskites, they all are carried out on thin films, where the interface between the perovskite and another phase (including the ambient) is often a dominating and interfering factor in the process. Here, self-healing in perovskite (methylammonium, formamidinium, and cesium lead bromide (MAPbBr3 , FAPbBr3 , and CsPbBr3 )) single crystals is reported, using two-photon microscopy to create damage (photobleaching) ≈110 µm inside the crystals and to monitor the recovery of photoluminescence after the damage. Self-healing occurs in all three perovskites with FAPbBr3 the fastest (≈1 h) and CsPbBr3 the slowest (tens of hours) to recover. This behavior, different from surface-dominated stability trends, is typical of the bulk and is strongly dependent on the localization of degradation products not far from the site of the damage. The mechanism of self-healing is discussed with the possible participation of polybromide species. It provides a closed chemical cycle and does not necessarily involve defect or ion migration phenomena that are often proposed to explain reversible phenomena in halide perovskites.

This study aimed to characterize unwanted immune effects of nanoparticles (NP) using THP-1 cells, human whole blood and enriched peripheral blood monocytes. Commercially available silver NP (AgNP < 100 nm, also confirmed by Single Particle Extinction and Scattering) were used as prototypical NP. Cells were treated with AgNP alone or in combination with classical immune stimuli (i.e. LPS, PHA, PWM) and cytokine assessed; in addition, CD54 and CD86 expression was evaluated in THP-1 cells. AgNP alone induced dose-related IL-8 production in all models, with higher response observed in THP-1 cells, possibly connected to different protein corona formation in bovine versus human serum. AgNP potentiated LPS-induced IL-8 and TNF-α, but not LPS-induced IL-10. AgNP alone induced slight increase in IL-4, and no change in IFN-γ production. While responses to PHA in term of IL-4 and IFN-γ production were not affected, increased PWM-induced IL-4 and IFN-γ production were observed, suggesting potentiation of humoral response. Reduction in PHA-induced IL-10 was observed. Overall, results indicate immunostimulatory effects. THP-1 cells work as well as primary cells, representing a useful and practical alternative, with the awareness that from a physiological point of view the whole blood assay is the one that comes closest to reality.

RESUMO

The fast evaporative cooling of micrometer-sized water droplets in a vacuum offers the appealing possibility to investigate supercooled water-below the melting point but still a liquid-at temperatures far beyond the state of the art. However, it is challenging to obtain a reliable value of the droplet temperature under such extreme experimental conditions. Here, the observation of morphology-dependent resonances in the Raman scattering from a train of perfectly uniform water droplets allows us to measure the variation in droplet size resulting from evaporative mass losses with an absolute precision of better than 0.2%. This finding proves crucial to an unambiguous determination of the droplet temperature. In particular, we find that a fraction of water droplets with an initial diameter of 6379±12 nm remain liquid down to 230.6±0.6 K. Our results question temperature estimates reported recently for larger supercooled water droplets and provide valuable information on the hydrogen-bond network in liquid water in the hard-to-access deeply supercooled regime.

We report the fabrication and characterization of a simple and compact hyperspectral imaging setup based on a stretchable diffraction grating made with a metal-polymer nanocomposite. The nanocomposite is produced by implanting Ag clusters in a poly(dimethylsiloxane) film by supersonic cluster beam implantation. The deformable grating has curved grooves and is imposed on a concave cylindrical surface, thus obtaining optical power in two orthogonal directions. Both diffractive and optical powers are obtained by reflection, thus realizing a diffractive-catoptric optical device. This makes it easier to minimize aberrations. We prove that, despite the extended spectral range and the simplified optical scheme, it is actually possible to work with a traditional CCD sensor and achieve a good spectral and spatial resolution.

Electrical applications require the development of electric field sensors that can reproduce vector electric field waveforms with a very large spectral width ranging from 50 Hz to at least 70 MHz. This makes it possible to measure both the normal operation modes of electrical components and abnormal behaviors such as the corona emission and partial discharges. In this work, we aim to develop a fully dielectric sensor capable of measuring two components of the electric field using a wide class of optical crystals including anisotropic ones, whereas most of the efforts in this field have been devoted to isotropic crystals. We report the results of the measurements performed at 50 Hz and with a lightning impulse, to validate the sensor.

Here we report the quantitative in situ characterization of size distribution evolution of polymeric nanoparticles incubated in murine serum, filtered and unfiltered murine blood. We used an analytical optical approach, named Single Particle Extinction and Scattering (SPES), which relies on the measurements of two independent parameters of single particles. SPES is based on a robust self-reference interference optical scheme which allows a rejection of the spurious signals coming from the background caused by the medium. We employed polystyrene nanoparticles as reference system and polydisperse poly(lactic-co-glycolic acid) nanoparticles. Our results demonstrate that SPES can be used for carrying out ex vivo analysis of nanoparticles to evaluate the modifications that NPs undergo in vivo following different routes of entry. Conversely, Dynamic Light Scattering is not able to provide reliable results for these systems due to the presence of the biological components in solution.

Protein-dense liquid clusters are regions of high protein concentration that have been observed in solutions of several proteins. The typical cluster size varies from several tens to several hundreds of nanometres and their volume fraction remains below 10(-3) of the solution. According to the two-step mechanism of nucleation, the protein-rich clusters serve as locations for and precursors to the nucleation of protein crystals. While the two-step mechanism explained several unusual features of protein crystal nucleation kinetics, a direct observation of its validity for protein crystals has been lacking. Here, two independent observations of crystal nucleation with the proteins lysozyme and glucose isomerase are discussed. Firstly, the evolutions of the protein-rich clusters and nucleating crystals were characterized simultaneously by dynamic light scattering (DLS) and confocal depolarized dynamic light scattering (cDDLS), respectively. It is demonstrated that protein crystals appear following a significant delay after cluster formation. The cDDLS correlation functions follow a Gaussian decay, indicative of nondiffusive motion. A possible explanation is that the crystals are contained inside large clusters and are driven by the elasticity of the cluster surface. Secondly, depolarized oblique illumination dark-field microscopy reveals the evolution from liquid clusters without crystals to newly nucleated crystals contained in the clusters to grown crystals freely diffusing in the solution. Collectively, the observations indicate that the protein-rich clusters in lysozyme and glucose isomerase solutions are locations for crystal nucleation.

RESUMO

The two-dimensional single shot transverse coherence of the Self-Amplified Spontaneous Emission of the SPARC_LAB Free-Electron Laser was measured through the statistical analysis of a speckle field produced by heterodyning the radiation beam with a huge number of reference waves, scattered by a suspension of particles. In this paper we report the measurements and the evaluation of the transverse coherence along the SPARC_LAB undulator modules. The measure method was demonstrated to be precise and robust, it does not require any a priori assumptions and can be implemented over a wide range of wavelengths, from the optical radiation to the x-rays.

We describe a method for measuring small fluctuations in the intensity of a laser source with a resolution of 10⁻4. The current signal generated by a PIN diode is passed to a front-end electronics that discriminates the AC from the DC components, which are physically separated and propagated along circuit paths with different gains. The gain long the AC signal path is set one order of magnitude larger than that along the DC signal path in such a way to optimize the measurement dynamic range. We then derive the relative fluctuation signal by normalizing the input-referred AC signal component to its input-referred DC counterpart. In this way the fluctuation of the optical signal waveform relative to the mean power of the laser is obtained. A "Noise-Scattering-Pattern method" and a "Signal-Power-Spectrum method" are then used to analyze the intensity fluctuations from three different solid-state lasers. This is a powerful tool for the characterization of the intensity stability of lasers. Applications are discussed.

RESUMO

By using the critical Casimir force, we study the attractive strength dependent aggregation of colloids with and without gravity by means of near field scattering. Significant differences were seen between microgravity and ground experiments, both in the structure of the formed fractal aggregates as well as in the kinetics of growth. In microgravity purely diffusive aggregation is observed. By using the continuously variable particle interaction potential we can for the first time experimentally relate the strength of attraction between the particles and the structure of the aggregates.
