BACKGROUND: Dihydrocorynantheol (DHC) is an alkaloid compound isolated from Esenbeckia leiocarpa Engl. that has demonstrated anti-inflammatory properties in experimental models. The aim of this study was to investigate whether the modification of the chemical structure of DHC could alter its anti-inflammatory effect in a mouse model of pleurisy induced by carrageenan. METHODS: DHC was isolated from Esenbeckia leiocarpa Engl. Capillary electrophoresis, physical characteristics, spectral data produced by infrared analysis and nuclearmagnetic resonance ((1)H and (13)C), and mass spectrometry analysis were used to identify and elucidate DHC structure. The DHC compound was subjected to chemical structural modifications by nucleophilic substitution reactions, yielding five analogous compounds: acetyl (1), p-methylbenzoyl (2), benzoyl (3), p-methoxybenzoyl (4) and p-chlorobenzoyl (5). Swiss mice were used throughout the experiments. Pro-inflammatory parameters leukocyte migration, exudate concentrations and myeloperoxidase (MPO) activity were quantified in the fluid leakage from the mouse pleural cavities at 4 h after pleurisy induction. RESULTS: DHC and its analogues acetyl, p-methylbenzoyl, benzoyl, p-methoxybenzoyl and p-chlorobenzoyl inhibited total and differential leukocyte migration and MPO activity (p < 0.05). Only DHC significantly decreased the exudate concentrations (p < 0.01). CONCLUSIONS: DHC was more effective than its analogues as an anti-inflammatory agent in the mouse model of pleurisy induced by carrageenan. We did not determine what physicochemical modifications altered the anti-inflammatory effect of DHC, but this effect may be due to the modifications on the hydroxyl group at carbon 17 of the DHC.

OBJECTIVES: The aim of this study was to investigate the anti-inflammatory effects of the crude hydroalcoholic extract (CHE) isolated from Esenbeckia leiocarpa Engl., and fractions and subfractions derived from it. METHODS: Dried E. leiocarpa Engl. bark was macerated and extracted with ethanol to obtain the CHE. The n-hexane, ethyl acetate, aqueous and alkaloid fractions, as well as two alkaloid subfractions (polar and nonpolar) were obtained from the CHE. A preliminary analysis using thin-layer chromatography was performed. Capillary electrophoresis, physical characteristics and spectral data produced by IR analysis and nuclear magnetic resonance (¹H and ¹³C NMR), and mass spectrometry analysis were used to identify and elucidate the structure of the major compounds. Swiss mice were used in a carrageenan-induced pleurisy model. Pro-inflammatory parameters (leukocyte and exudate concentrations, myeloperoxidase and adenosine-deaminase activity, and nitrate/nitrite, interleukin 1ß and tumour necrosis factor α levels) were quantified in exudates at 4 h after carrageenan-induced pleurisy in mice. KEY FINDINGS: The dihydrocorynantheol alkaloid was isolated as the majority compound in the CHE, ethyl acetate and alkaloid fractions, and in the polar and nonpolar alkaloid subfractions. The CHE, fractions and subfractions inhibited the increases in leukocyte and exudate concentrations, myeloperoxidase and adenosine-deaminase activity, and nitrite/nitrate, interleukin 1ß, and tumour necrosis factor α levels (P<0.05) in the fluid secreted from the pleural cavity of the carrageenan-treated mice. CONCLUSIONS: E. leiocarpa Engl. showed significant in vivo anti-inflammatory action by inhibiting the inflammation caused by carrageenan. This effect may be, in part, due to the dihydrocorynantheol alkaloid, which was identified as the majority compound isolated from E. leiocarpa bark.

Das folhas de Senecio bonariensis Hook. & Arn. foram isolados três sesquiterpenóides. β-Cariofileno (1), óxido de β-cariofileno (2) e germacreno D (3) foram identificados por CG-IE-EM, IV, ¹H and 13C-RMN. A importância ecológica e quimiotaxionômica para o gênero Senecio é discutida, assim como a correlação biossintética entre as substâncias isoladas. As substâncias 1 e 2 tiveram sua atividade antimicrobiana avaliada pelo método de microdiluição em caldo contra cepas bacterianas e fúngicas. Ambas não inibiram o crescimento microbiano nas concentrações testadas.


From the leaves of Senecio bonariensis Hook. & Arn. three sesquiterpenoids were isolated. β-caryophyllene (1), β-caryophyllene oxide (2) and germacrene D (3) were characterized by GC-EI-MS, IR, ¹H and 13C-NMR data. Their ecological and chemotaxonomical significance for the genus Senecio are discussed, as well as the biosynthetic correlation between the isolated substances. The antimicrobial activity of substances 1 and 2 were evaluated by the microdilution method against bacterial and fungal strains. Both compounds did not inhibit the microorganism growth at the tested concentrations.

Here, a microdilution technique based on the M27-A2 protocol (NCCLS, 2002) was employed to compare the susceptibilities of Candida albicans and Candida dubliniensis to essential oils extracted from plants used as spices. The chemical compositions of the essential oils were defined based on the analysis of retention indices obtained by gas chromatography-mass spectroscopy. Taken together, the results showed that the activity of the compounds against the two species was similar.

In this study, the susceptibility to amphotericin B of Candida spp. isolates obtained from patients with candidemia was related to their respective clinical outcomes. The susceptibility tests were carried out in three culture media: RPMI 1640, Antibiotic medium 3 and Yeast Nitrogen Base dextrose. We have found that minimal inhibitory concentrations and minimal fungicidal concentrations obtained using AM3 and YNBd media were significantly higher for Candida spp. from patients who died than for those from patients who survived the candidemia (P < 0.05). The assays with RPMI 1640 medium did not show these differences.

In the present study, the antifungal activity of selected essential oils obtained from plants used as spices was evaluated against both fluconazole-resistant and fluconazole-susceptible Candida spp. The Candida species studied were Candida albicans, Candida dubliniensis, Candida tropicalis, Candida glabrata, and Candida krusei. For comparison purposes, they were arranged in groups as C. albicans, C. dubliniensis, and Candida non-albicans. The essential oils were obtained from Cinnamomum zeylanicum Breyn, Lippia graveolens HBK, Ocimum basilicum L., Origanum vulgare L., Rosmarinus officinalis L., Salvia officinalis L., Thymus vulgaris L., and Zingiber officinale. The susceptibility tests were based on the M27-A2 methodology. The chemical composition of the essential oils was obtained by gas chromatography-mass spectroscopy and by retention indices. The results showed that cinnamon, Mexican oregano, oregano, thyme, and ginger essential oils have different levels of antifungal activity. Oregano and ginger essential oils were found to be the most and the least efficient, respectively. The main finding was that the susceptibilities of fluconazole-resistant C. albicans, C. dubliniensis, and Candida non-albicans to Mexican oregano, oregano, thyme, and ginger essential oils were higher than those of the fluconazole-susceptible yeasts (P<0.05). In contrast, fluconazole-resistant C. albicans and Candida non-albicans were less susceptible to cinnamon essential oil than their fluconazole-susceptible counterparts (P<0.05). A relationship between the yeasts' susceptibilities and the chemical composition of the essential oils studied was apparent when these 2 parameters were compared. Finally, basil, rosemary, and sage essential oils did not show antifungal activity against Candida isolates at the tested concentrations.

Candida dubliniensis is a recently described pathogenic species which shares many phenotypic features with Candida albicans and therefore, may be misidentified in microbiological laboratories. Because molecular methods can be onerous and unfeasible in routine mycological laboratories with restricted budgets such as those in developing countries, phenotypic techniques have been encouraged in the development of differential media for the presumptive identification of these species. We examined the colony morphology and chlamydospore production of 30 C. dubliniensis isolates and 100 C. albicans isolates on two new proposed media: rosemary (Rosmarinus officinalis) extract agar (REA) and oregano (Origanum vulgare) extract agar (OEA). These substrates are traditionally used as spices and medicinal herbs. In both of these media, all C. dubliniensis isolates (100%) showed rough colonies with peripheral hyphal fringes and abundant chlamydospores after 24 to 48 hr of incubation at 25 degrees C. In contrast, under the same conditions, all isolates of C. albicans (100%) showed smooth colonies without hyphal fringes or chlamydospores. In conclusion, REA and OEA offer a simple, rapid, and inexpensive screening media for the differentiation of C. albicans and C. dubliniensis.

Candida dubliniensis is an opportunistic yeast that has been recovered from several body sites in many populations; it is most often recovered from the oral cavities of human immunodeficiency virus-infected patients. Although extensive studies on epidemiology and phylogeny of C. dubliniensis have been performed, little is known about virulence factors such as exoenzymatic and hemolytic activities. In this study we compared proteinase, hyaluronidase, chondroitin sulphatase and hemolytic activities in 18 C. dubliniensis and 30 C. albicans strains isolated from AIDS patients. C. albicans isolates produced higher amounts of proteinase than C. dubliniensis (p < 0.05). All the tested C. dubliniensis strains expressed hyaluronidase and chondroitin sulphatase activities, but none of them were significantly different from those observed with C. albicans (p > 0.05). Hemolytic activity was affected by CaCl2; when this component was absent, we did not notice any significant difference between C. albicans and C. dubliniensis hemolytic activities. On the contrary, when we added 2.5 g% CaCl2, the hemolytic activity was reduced on C. dubliniensis and stimulated on C. albicans tested strains (p < 0.05).

C. dubliniensis é uma levedura oportunista que, embora já tenha sido isolada de vários sítios anatômicos é, com maior frequência, encontrada na boca de pacientes infectados pelo HIV. Embora tenham sido realizados numerosos estudos sobre a epidemiologia e filogenia, seus fatores de virulência como atividade exoenzimática e atividade hemolítica, são, ainda, pouco conhecidos. Neste estudo comparou-se a atividade in vitro de proteinase, hialuronidase, condroitin sulfatase e atividade hemolítica de 18 cultivos de C. dubliniensis com 30 cultivos de C. albicans, todos isolados de pacientes com SIDA. Foi evidenciada maior atividade de proteinase em C. albicans em relação a C. dubliniensis (p < 0,05). Todos os isolados de C. dubliniensis evidenciaram atividade de hialuronidase e condroitin-sulfatase de forma similar ao observado com C. albicans (p > 0,05). Constatou-se que a atividade hemolítica foi influenciada pelo CaCl2; em sua ausência não foram observadas diferenças na atividade hemolítica das duas espécies; todavia, ao se agregar 2,5 por cento de CaCl2, a atividade hemolítica de C. dubliniensis foi reduzida enquanto a de C. albicans, estimulada (p < 0,05).

Avaliou-se a atividade antimicrobiana dos óleos essenciais de orégano (Origanum vulgare), tomilho (Thymus vulgaris) e canela (Cinnamomum zeylanicum) frente a 60 amostras de Salmonella enterica distribuídas entre 20 sorovares, todos isolados de carcaças de aves. A concentração inibitória mínima (CIM) e a concentração bactericida mínima (CBM) foram determinadas para cada isolado, utilizando-se a técnica de microdiluição em caldo. O óleo essencial de orégano evidenciou forte atividade antibacteriana (CIM média = 529mig ml-1 e CBM média= 661mig ml-1), seguido do tomilho (CIM média=961mig ml-1 e CBM média= 1074mig ml-1), com atividade moderada, enquanto que a menor atividade foi observada com o óleo essencial de canela (CIM média= 1335mig ml-1 e CBM média = 1979mig ml-1). Apesar deste claro padrão de atividade, os sorovares de Salmonella enterica evidenciaram significativas variações de suscetibilidade, embora nenhuma, em particular, pudesse ser classificada como totalmente sensível ou resistente aos três óleos essenciais avaliados. Assim, no momento em que os antibacterianos utilizados como promotores de crescimento estão sendo substituídos. E os óleos essenciais de temperos e condimentos se constituem numa alternativa importante, os resultados aqui apresentados sinalizam dois pontos: os óleos essenciais de orégano e tomilho são efetivos contra Salmonella, mas as variações de suscetibilidade entre os sorovares deverão também ser consideradas.


Essential oils from oregano (Origanum vulgare), thyme (Thymus vulgaris) and cinnamom (Cinnamomum zeylanicum) were tested for their inhibitory activity against sixty Salmonella sp. isolates. These microorganisms were distributed in 20 serovars, all of them isolated from broiler chickens. Based on microdilution technique, the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) were determined. The strongest antibacterial activity was obtained for oregano (MIC mean = 529mug ml-1 and MBCmean = 661mug ml-1), followed by a moderate activity for thyme (MICmean=961mug ml-1 and MBCmean=1074mug ml-1) and the lowest obtained activity observed for cinnamon (MICmean= 1335mug ml-1 and MBCmean= 1979mug ml-1). Despite of this activity pattern, the serovars of Salmonella enterica showed significant susceptibility variations. However none of them could be classified as sensitive or resistant to the three essential oils tested. At the moment, when the antibiotics, as growth promoters, are being replaced, the essential oils from spices could be a very reasonable choice and the present results show us two points: the essential oils from oregano and thyme are effective against Salmonella isolates but the variations among the serovars must be considered.
