RESUMO

Ammonia is a promising alternative hydrogen carrier that can be utilized for the solid-state reduction of iron oxides for sustainable ironmaking due to its easy transportation and high energy density. The main challenge for its utilization on an industrial scale is to understand the reaction kinetics under different process conditions and the associated nitrogen incorporation in the reduced material that originates from ammonia decomposition. In this work, the effect of temperature on the reduction efficiency and nitride formation is investigated through phase, local chemistry, and gas evolution analysis. The effects of inherent reactions and diffusion on phase formation and chemistry evolution are discussed in relation to the reduction temperature. The work also discusses nitrogen incorporation into the material through both spontaneous and in-process nitriding, which fundamentally affects the structure and chemistry of the reduced material. Finally, the effect of nitrogen incorporation on the reoxidation tendency of the ammonia-based reduced material is investigated and compared with that of the hydrogen-based reduced counterpart. The results provide a fundamental understanding of the reduction and nitriding for iron oxides exposed to ammonia at temperatures from 500 to 800 °C, serving as a basis for exploitation and upscaling of ammonia-based direct reduction for future green steel production.

The scaffolding protein, hematopoietic PBX-interacting protein (HPIP/PBXIP1), regulates cell migration necessary for cancer cell dissemination. However, the mechanism that governs this process remains unknown. We show here that HPIP expression is associated with stages of breast cancer where cell dissemination results in poor patient outcome. Our investigation finds a novel association of HPIP with focal adhesion kinase (FAK) regulating FA dynamics. Interestingly, this interaction that led to activation of FAK protein was mediated by the C-terminal domain of HPIP and not the typical integrin-binding motif. Further, short hairpin RNA-mediated knockdown of FAK expression significantly reduced HPIP-induced cell migration indicating participation of FAK pathway. Live-cell time-lapse imaging and biochemical analysis further established the role of HPIP in microtubule-induced FA disassembly. We also found that HPIP-mediated MAPK activation led to phosphorylation and subsequent activation of calpain2, and the activated calpain2 in turn proteolyses FA protein, talin. Interestingly, HPIP is also proteolysed by calpain2 in breast cancer cells. The proteolysis of HPIP and talin by calpain2, and the activation of calapin2 by HPIP-mediated MAPK phosphorylation, is a novel regulatory axis to modulate the cell migration signal. Together, we have determined HPIP as a novel activator of FAK and a new substrate of calpain2. These molecular interactions between HPIP and FAK, and HPIP and calpain2 regulate cell adhesion and migration through modulation of FA dynamics.

BACKGROUND: Streptococcus mutans is one of the most important cariogenic species of the human oral microbial flora. Biofilm style of microbial growth thought to resist the actions of antimicrobials. AIM: The purpose of this study was to evaluate the antimicrobial efficacy of Triphala, and 0.2% chlorhexidine against S. mutans biofilm formed on tooth substrate. SETTINGS AND DESIGN: Randomized control trial. METHODS: Extracted human mandibular premolars sectioned below the cemento-enamel junction were placed in the tissue culture wells exposing the crown surface to S. mutans to form a biofilm. At the end of 3 rd and 7 th day, all groups were treated for 10 min with the test solutions and control and were analyzed qualitatively and quantitatively. STATISTICAL ANALYSIS USED: One-way ANOVA. RESULTS: Qualitative assay with 3 days biofilm showed complete inhibition of bacterial growth with Triphala, but 0.2% chlorhexidine and saline showed the presence of bacterial growth. In quantitative analysis, 0.2% chlorhexidine and Saline treated tooth samples have shown 1052 Χ 10(4) ± 15.1 Χ 10(4) CFU/ml, 141.3 Χ 10(9) ± 2.1Χ10(9) CFU/ml, respectively. Qualitative assay with 7 days biofilm on crown portion showed dense growth when treated with 0.2% chlorhexidine and saline, whereas Triphala has shown minimal growth. In Quantitative analysis, Triphala showed statistically significant result when compared with 0.2% chlorhexidine and saline. CONCLUSION: Triphala showed statistically significant antibacterial activity against S. mutans biofilm formed on tooth substrate. The incorporation of Triphala in mouth rinse could prove to be effective in reducing S. mutans count in the oral cavity.

The aim of this study was to radiographically investigate the curve existing in slightly curved mesiobuccal canal of mandibular first molar by applying Piecewise straight line approximation. Extracted human mandibular molars were radiographed and one hundred radiographs were selected whose mesiobuccal canal showed slight curvature (10-20°) according to Schneider's method. The curves were traced and analyzed using Piecewise straight line method. Each curve was considered as a unit consisting of six different pieces of straight lines joining at seven specific points and the angle of curvature at these points was determined using the slope formula. All curves analyzed in this study had varying degrees of curvatures at different points on the curve. Maximum curvature (0.40°) was recorded at the middle third of the root canal. Within the limitation of the study, significant curvature occurs through out the curve existing in the mesiobuccal canal of mandibular first molar and the middle third of the curve showing greater degree of curvature. There is a possibility of greater curvature occurring in the coronal third of the curve. Though three dimensional studies would be more appropriate, Piecewise straight line approximation may be a better method than existing methods to simulate canal geometry.


El objetivo de este estudio fue investigar radiográficamente la curva existente en el canal radicular mesiobucal ligeramente curvado del primer molar inferior mediante la aplicación de aproximaciones a segmentos de línea recta. Se utilizaron molares inferiores humanos extraídos que fueron radiografiados. Fueron seleccionadas, según el método de Schneider, 100 radiografías cuyo canal mesiobucal mostró una ligera curvatura (10-20°). Las curvas fueron delineadas y analizadas mediante el método de aproximación a segmentos de línea recta. Cada curva se consideró como una unidad que consta de seis diferentes segmentos de línea recta que unen a siete puntos específicos y el ángulo de curvatura en estos puntos se determinó utilizando la fórmula de la pendiente. Todas las curvas analizadas en este estudio tenían diversos grados de curvaturas en diferentes puntos de la curva. La curvatura máxima (0,40°) se registró en el tercio medio del canal radicular. Dentro de la limitación del estudio, una curvatura significativa se produce a través de la curva existente en el canal mesiobucal del primer molar inferior y el tercio medio de la curva que muestra un mayor grado de curvatura. Existe la posibilidad que ocurra una mayor curvatura en el tercio coronal de la curva. Aunque un estudio tridimensional sería más apropiado, la aproximación a segmentos de línea recta puede ser un mejor método que los existentes para simular la geometría del canal.

INTRODUCTION: The purpose of this study was to evaluate the antimicrobial efficacy of Triphala, green tea polyphenols (GTP), MTAD, and 5% sodium hypochlorite against E. faecalis biofilm formed on tooth substrate. METHODS: Extracted human teeth were biomechanically prepared, vertically sectioned, placed in the tissue culture wells exposing the root canal surface to E. faecalis to form a biofilm. At the end of the 3rd and 6th weeks all groups were treated for 10 minutes with the test solutions and control and were analyzed qualitatively and quantitatively. RESULTS: Qualitative assay with 3-week biofilm showed complete inhibition of bacterial growth with Triphala, MTAD and NaOCl, except GTP and saline, which showed presence of bacterial growth. In quantitative analysis, GTP- and saline-treated tooth samples have shown 1516 +/- 17.2 CFU/mL, 156.4 x 10(9) +/- 3.1 x 10(9) CFU/mL respectively. Qualitative assay with 6-week biofilm showed growth when treated with Triphala, GTP and MTAD whereas NaOCl has shown complete inhibition. All groups except NaOCl showed eight log reduction when compared to control when analyzed quantitatively. CONCLUSIONS: 5% sodium hypochlorite showed maximum antibacterial activity against E. Faecalis biofilm formed on tooth substrate. Triphala, green tea polyphenols and MTAD showed statistically significant antibacterial activity. The use of herbal alternatives as a root canal irrigant might prove to be advantageous considering the several undesirable characteristics of NaOCl.

RESUMO

Pneumatosis intestinalis is a rare condition occurs in a wide variety of patients, some of whom require urgent surgery, while others can be observed with resolution of symptoms and radiographic findings. Most often the need for surgery is intestinal obstruction. We report a case of pneumatosis intestinalis with an abdominal mass causing obstruction. The histological surprise and the rarity of the obstruction is highlighted in this case report.

BACKGROUND: Thromboxane A2 (TXA2) is a positive feedback lipid mediator that is generated upon stimulation of platelets with various agonists. Aspirin works as an antithrombotic drug by blocking the generation of TXA2. The aim of this study was to evaluate the role of the purinergic P2Y receptors in thrombin-induced TXA2 generation. RESULTS: PAR1-activating peptide (SFLLRN), PAR4-activating peptide (AYPGKF), and thrombin, induced the activation of cytosolic phospholipase A2 (cPLA2), release of arachidonic acid (AA) from membrane-bound phospholipids, and subsequent TXA2 generation in human platelets. The actions of these agonists were significantly inhibited in the presence of the P2Y12 receptor antagonist, AR-C69931MX, but not the P2Y1 receptor antagonist, MRS2179. In addition, AYPGKF- and thrombin-induced TXA2 generation was significantly reduced in platelets from mice dosed with clopidogrel, confirming the results obtained with the human platelets. Also, Pearl mouse platelets that lack releasable nucleotides generated significantly less TXA2 when compared with the wild-type littermates in response to PAR stimulation. Inhibition of extracellular signal-regulated protein kinase 1/2 (Erk 1/2) activation using U0126, an inhibitor of MAP kinase kinase (MEK), suppressed PAR-mediated cPLA2 phosphorylation and TXA2 generation. Further, platelets that were pretreated with AR-C69931MX, as well as Pearl mouse platelets, displayed the reduced levels of Erk1/2 phosphorylation upon stimulation with the PAR agonists. CONCLUSIONS: Based on these findings, we conclude that thrombin-induced Erk1/2 activation is essential for PAR-mediated TXA2 generation, which is potentiated by the P2Y12 receptor-mediated signaling pathway but not the P2Y1 receptor-mediated signaling pathway. Finally, using selective inhibitors of Src kinases, we show that PAR-mediated Src activation precedes Erk1/2 activation.

The objectives of this study were to determine the relationship of age and body weight to testicular development and to establish norms for breeding soundness evaluations of Murrah buffalo bulls. Testicular measurements of 133 Murrah buffalo bulls of various ages were recorded with a caliper and a tape. Semen was collected twice a week for 5 weeks from groups of bulls which were 25-36 (n=17), 37-48 (n=16), 49-60 (n=14), of >60 (n=10) months of age. After examining volume, sperm concentration, and progressive motility semen was diluted in Tris-citric acid-egg yolk-fructose extender and frozen in 0.5 ml French straws. Testicular measurements of buffalo bulls were lower than those recorded for European breeds of cattle bulls. Nevertheless, like cattle bulls, scrotal circumference was highly correlated with other testicular measurements. Also, it had a significant positive relationship with semen volume and sperm concentration per ejaculate. Average sperm output per week in order of increasing age group was 15.3, 18.2, 19.8 and 23.6 x 10(9). Corresponding values for sperm output per week per gram of testis were 59.1, 45.8, 41.1, 36.2 x 10(6) indicating a reduction in spermatogenesis per unit of testis with advancing age. Compared to European breeds, daily sperm output in Murrah bulls was nearly 45% lower, presumably due to their nearly 40% lower scrotal circumference than Holstein bulls of the same age. These results indicate that in buffalo, as in cattle, scrotal circumference is a useful indicator of potential sperm output and may serve as an important criterion for selecting young bulls as AI sires.

Giant cell tumours of bones are uncommon and are usually found in the epiphyses of long bones. They are rare in flat bones (<10%). The tumour has to our knowledge never been described in the hyoid bone, although 18 cases have been reported in the cartilagenous laryngeal skeleton. We report a giant cell tumour of the hyoid bone in a 45-year-old man, which we excised along with the left half of the hyoid bone. It recurred locally one year later and was cured by excision and split course radiotherapy. The patient is disease-free after 30 months of follow-up.

Streptococcus pyogenes can be efficiently internalized by a variety of human epithelial cells. beta-lactam antibiotics, commonly used to treat S. pyogenes infections, do not readily permeate mammalian cells. There is growing evidence that the ability of streptococci to enter host cells contributes to the frequent failure of antibiotics to eradicate the organism from infected individuals. Recent studies have suggested that host cell entry requires the formation of a complex of a bacterial fibronectin (Fn) binding protein (e.g., M1 protein or protein F1/SfbI), human Fn, and the epithelial cell Fn receptor, integrin alpha5beta1. We report here that a low molecular weight, nonpeptide antagonist of integrin alpha5beta1, SJ755, can inhibit internalization of streptococci by primary human tonsillar epithelial cells and immortalized human epithelial (A549) cells, thus increasing the extent of bacterial killing by antibiotics. SJ755 blocked Fn binding by human tonsillar epithelial and A549 cells, suggesting that integrin alpha5beta1 is the major Fn receptor expressed by both cell types. SJ755 did not affect Fn binding by purified M1 protein or M1(+) bacteria. Purified M1 protein failed to associate with integrin alpha5beta1 unless the integrin had been prebound by Fn. Also, SJ755 blocked formation of alpha5beta1-Fn-M1 complexes in vitro. These results support the previous proposal that Fn functions as a molecular bridge between M1 protein and integrin alpha5beta1. Furthermore, these results suggest that integrin antagonists may enhance the efficacy of antibiotics in treatment of S. pyogenes infections.

Artemia functions as living filter for the secondary treatment of distillery effluent, which reduces up to 69% of the total solids and 33.34% of BOD in the saline medium of 60 ppt. Chemical analysis of diluted effluent after treatment with Artemia showed a significant (P less than 0.001) decrease in the electrical conductivity, calcium and potassium values. However, an increase in sodium concentration and pH values, was recorded during experiment.
