Background: Recognition of rare molecular subgroups is a challenge for precision oncology and may lead to tissue-agnostic approval of targeted agents. Here we aimed to comprehensively characterize the clinical, pathological and molecular landscape of RET rearranged metastatic colorectal cancer (mCRC). Patients and methods: In this case series, we compared clinical, pathological and molecular characteristics of 24 RET rearranged mCRC patients with those of a control group of 291 patients with RET negative tumors. RET rearranged and RET negative mCRCs were retrieved by systematic literature review and by taking advantage of three screening sources: (i) Ignyta's phase 1/1b study on RXDX-105 (NCT01877811), (ii) cohorts screened at two Italian and one South Korean Institutions and (iii) Foundation Medicine Inc. database. Next-generation sequencing data were analyzed for RET rearranged cases. Results: RET fusions were more frequent in older patients (median age of 66 versus 60 years, P = 0.052), with ECOG PS 1-2 (90% versus 50%, P = 0.02), right-sided (55% versus 32%, P = 0.013), previously unresected primary tumors (58% versus 21%, P < 0.001), RAS and BRAF wild-type (100% versus 40%, P < 0.001) and MSI-high (48% versus 7%, P < 0.001). Notably, 11 (26%) out of 43 patients with right-sided, RAS and BRAF wild-type tumors harbored a RET rearrangement. At a median follow-up of 45.8 months, patients with RET fusion-positive tumors showed a significantly worse OS when compared with RET-negative ones (median OS 14.0 versus 38.0 months, HR: 4.59; 95% CI, 3.64-32.66; P < 0.001). In the multivariable model, RET rearrangements were still associated with shorter OS (HR: 2.97; 95% CI, 1.25-7.07; P = 0.014), while primary tumor location, RAS and BRAF mutations and MSI status were not. Conclusions: Though very rare, RET rearrangements define a new subtype of mCRC that shows poor prognosis with conventional treatments and is therefore worth of a specific management.

UNLABELLED: An activated carbon commercially available named HGR, produced by Calgon-Carbon Group, was used to adsorbe metallic mercury. The work is part of a wider research activity by the same group focused on the removal of metallic and divalent mercury from combustion flue gas. With respect to previously published papers, this one is aimed at studying in depth thermodynamic equilibria of metallic mercury adsorption onto a commercial activated carbon. The innovativeness lies in the wider operative conditions explored (temperature and mercury concentrations) and in the evaluation of kinetic and thermodynamic data for a commercially available adsorbing material. In detail, experimental runs were carried out on a laboratory-scale plant, in which Hg° vapors were supplied in a nitrogen gas stream at different temperature and mercury concentration. The gas phase was flowed through a fixed bed of adsorbent material. Adsorbate loading curves for different Hg° concentrations together with adsorption isotherms were achieved as a function of temperature (120, 150, 200°C) and Hg° concentrations (1.0-7.0 mg/m(3)). Experimental runs demonstrated satisfying results of the adsorption process, while Langmuir parameters were evaluated with gas-solid equilibrium data. Especially, they confirmed that adsorption capacity is a favored process in case of lower temperature and they showed that the adsorption heat was -20 kJ/mol. Furthermore, a numerical integration of differential equations that model the adsorption process was proposed. Scanning electron microscopy (SEM) investigation was an useful tool to investigate about fresh and saturated carbon areas. The comparison between them allowed identification of surface sites where mercury is adsorbed; these spots correspond to carbon areas where sulfur concentration is greater. IMPLICATIONS: Mercury compounds can cause severe harm to human health and to the ecosystem. There are a lot of sources that emit mercury species to the atmosphere; the main ones are exhaust gases from coal combustion and municipal solid waste incineration. Furthermore, certain CO2 capture processes, particularly oxyfuel combustion in a pulverized fuel coal-fired power station, produce a raw CO2 product containing several contaminants, mainly water vapor, oxygen, and nitrogen but also mercury, that have to be almost completely removed; otherwise these would represent a strong drawback to the success of the process.

Alkaline waters produced by caprolactam plants polymerizing the fibres of nylon-6 are characterized by a very high alkalinity, salinity and COD values, in addition to the presence of recalcitrant organic molecules. These characteristics make alkaline wastewaters very difficult to treat; so the development of the suitable sequence to carry out in a depuration process appears of great interest. The proposed general process consists of three main steps: first, pre-treatment for the acidification of the polluted stream, second, a successive extraction of the bio-recalcitrant compound (noted as cycloexanecarboxysulphonic acid (CECS)) and a final biological treatment. In particular, this paper deals with the pre-treatment step: it consists of an acidification process by means of sulphuric acid with the concomitant precipitation of black slurries in the presence of different substances, such as solvents, CaCl2, bentonite, several flocculants and coagulants. The aim of this study is to set an experimental procedure, which could minimize fouling problems during sludge filtration. The use of additives like bentonite seems to give the best results, because it allows good COD reductions and a filterable precipitate, which avoids excessive fouling problems of the experimental apparatus.

A study on olive mill residues (OMR) as copper adsorbing material is reported in this work. A rough characterization of this waste material has been performed, by microanalysis and SEM pictures. Sorption tests with suspended OMR evidenced copper removal from solution, of about 60% in the investigated experimental conditions. The COD release in solution was also monitored during biosorption. Considering that it was significant, OMR washings with water were performed before biosorption. In this case the COD release in solution was reduced to less than 600 mg/L after two washings, while the OMR metal sorption properties did not change. Regenerated residues by acid solutions gave a copper removal of about 40%, in the same experimental conditions of the first adsorption test: regeneration with EDTA at different concentrations suggested that it presents a damage of adsorption active sites. On the other hand, the use of HCl and CaCl(2) led to completely regenerate the biosorbent material. Tests were also performed with a column filled with 80 g of OMR and the breakpoint was demonstrated to take place after that about 1L solution was treated in the investigated experimental conditions. Regeneration tests permitted to demonstrate that a concentration factor of about 2 can be obtained in no-optimized conditions, highlighting the possibility of using OMR for the treatment of metal bearing effluents. The main advantage of the process would be the "low cost" biosorbing material, considering that it represents a waste in the olive oil production.

RESUMO

In this work, a model for phosphorus crystallization in a fluidized bed reactor, able to describe the experimental results obtained during a semiscale pilot plant, is presented. In particular, the validity of the model proposed has been evenly extended with respect to a previous experiment, even at a lower contact time, and the length of each experiment has been increased, in order to verify the behaviour of the process for long term applications and to evaluate the maximum crystal growth of the system. Moreover, the state of the art of the available processes for phosphorus removal from wastewater is presented, together with a detailed review of the several models so far developed to describe the phosphorus crystallization mechanisms.

RESUMO

In this paper the phosphorus removal from a real anaerobic supernatant through the crystallization of struvite and or hydroxyapatite was investigated. A comparison between experimental results on phosphorus crystallization carried out in a fluidized-bed reactor (FBR) on a bench-scale and on a half-scale plant is presented, together with a double saturational model able to describe all experimental results, independent of the different geometry of the reactors, the distinct contact times and the unlike products obtained. Experimental results show that removal efficiencies are very satisfactory, and the maximum phosphorus removal is of 80%.

In this paper experimental results on the anaerobic digestion of sewage sludge and organic fraction of municipal solid waste (OFMSW) by using a double phase process are reported. The long-term experiment has been carried out on a pilot scale plant, performed in different sets of operative conditions, during which granulometric distributions of particles in sludges and rheological properties of sludges were monitored. A significant fluidification of sludge was evidenced in the meso-thermo process, especially taking into account the variation in sludge behaviour from the first to the second phase. In the thermo-thermo process a fluidification higher than that shown in meso-thermo conditions is not observed, this suggesting that better results in terms of sludge conditioning can be obtained in a long time spent in thermophilic anaerobic digestion. Total volatile solids (TVS) and total fixed solids (TFS) become the most important parameters when mathematical modelling is applied to these processes. In the acidogenic phase, hydraulic retention time (HRT) and temperature are used to determine rigidity coefficient (RC), while only temperature is needed for yield stress (YC). Organic loading rate (OLR) and specific gas production (SGP) exert an important role in methanogenic phase description.
