RESUMO

Background: The ethanolamine kinase 2 (ETNK2) gene is implicated in carcinogenesis, but its expression and involvement in kidney renal clear cell carcinoma (KIRC) remain unknown. Methods: Initially, we conducted a pan-cancer study in which we searched the Gene Expression Profiling Interactive Analysis, the UALCAN, and the Human Protein Atlas databases to determine the expression level of the ETNK2 gene in KIRC. The Kaplan-Meier curve was then used to calculate the overall survival (OS) of KIRC patients. We then used the differentially expressed genes (DEGs) and enrichment analysis to explain the mechanism of the ETNK2 gene. Finally, the immune cell infiltration analysis was performed. Results: Although the ETNK2 gene expression was lower in KIRC tissues, the findings illustrated a link between the ETNK2 gene expression and a shorter OS time for KIRC patients. DEGs and enrichment analysis revealed that the ETNK2 gene in KIRC involved multiple metabolic pathways. Finally, the ETNK2 gene expression has been linked to several immune cell infiltrations. Conclusions: According to the findings, the ETNK2 gene plays a crucial role in tumor growth. It can potentially serve as a negative prognostic biological marker for KIRC by modifying immune infiltrating cells.

Intestinal flora and its metabolites are closely related to the progression of type 2 diabetes mellitus(T2DM). Eubacterium is one of the dominant intestinal flora, and its metabolites short-chain fatty acids (SCFAs) play a leading role in regulating intestinal metabolic balance. It has been reported that SCFAs can regulate the secretion of glucagon-like peptide-1, improve the function of pancreatic β cells, participate in bile acids metabolism and regulate the production of inflammatory factors in T2DM. Based on the above research background, this article mainly reviews the relationship between Eubacterium and its metabolite SCFAs and T2DM and its regulatory mechanism.

Glucocorticoids (GCs) can effectively control airway inflammation, but can also cause airway epithelial injury. Tectorigenin, a type of isoflavone isolated from various medicinal plants, has hypolipidemic activity, hepatoprotective, and antioxidant effects. We aimed to investigate whether Tectorigenin can repair GCs-induced airway epithelial injury. Airway epithelial cell line (9HTE cells) were treated with dexamethasone (Dex), Tectorigenin, or further transfected, then cell viability, migration, and invasion were examined by Cell Counting Kit (CCK-8), wound healing, and Transwell assays. The expressions of potential miRNAs related to the effect of Tectorigenin were detected by quantitative polymerase chain reaction (qPCR). Expressions of poptosis-related proteins Bcl-2-associated protein-X (Bax), B-cell lymphoma-2 (Bcl-2), Cleaved Caspase-3, and related to Mitorgen-activated protein kinase (MAPK) signaling pathway serine/threonine kinase (Raf1), extracellular signal-regulated kinase kinase 1/2 (MEK1/2), and extracellular signal-regulated kinase 1/2 (ERK1/2) were detected by Western blot. Dex inhibited the cell viability, migration and invasion by promoting Bax and Cleaved Caspase-3 expressions (p <.001) and by inhibiting the expressions of Bcl-2 and miR-222-3p (p <.001). Then, 10 µmol/L Tectorigenin itself did not affect cell viability but could inhibit the effect of Dex on cell viability, migration, and invasion. Tectorigenin up-regulated the expressions of miR-222-3p, Bcl-2, p-Raf1, p-MEK1/2, and p-ERK1/2 (p <.01), but down-regulated the expressions of Bax and Cleaved Caspase-3 (p <.05) in Dex-induced cells. MiR-222-3p inhibitor reversed the antagonistic effect of Tectorigenin on Dex. The study demonstrates that Tectorigenin inhibits apoptosis of Dex-induced 9HTE cells by up-regulating the expression of miR-222-3p, which involves with the MAPK pathway.

OBJECTIVE@#To investigate the screening of β-thalassemia among newborns in Wuhan region, so as to explore the influencing factors of Hb A in dried blood spot.@*METHODS@#Concentrations of Hb A,Hb A2,Hb F in the dried blood spots collected from 99 275 neonates in Wuhan region were analyzed by Sebia capillary electrophoresis. The screening result of β-thalassemia was interpretated accroding to the ratio of each group, the suspicious β-thalassemia newborns were recalled and the gene of thalassemia in those newborns was checked.@*RESULTS@#Among 99 275 newborns, 1 408 positive patients were found, and the positive rate of screening was 1.41%. A total of 350 patients with gene mutation were found among 709 β-thalassemia suspicious patients. There were significantly statistical differences of positive predictive value among Hb A levels in different groups and there were also significantly statistical differences of positive predictive values among gestational weeks in different groups. No significantly statistical differences were observed among different genetic defects and phenotypes of heterozygous β-thalassemia in Hb A concentrations. Postnatal day and gestational age were significantly and positively associated with Hb A concentrations.@*CONCLUSION@#The capillary electrophoresis is an effective screening method for β-thalassemia of full-term neonate. Postnatal day and gestational age is associated with the pencentage of Hb A.

OBJECTIVE: The low number of oligodendrocytes (OLs) in the hippocampus of patients with schizophrenia suggests that hippocampal demyelination is changed in this condition. Sox10 is expressed throughout OL development. The effect of Sox10 on myelin regeneration is unknown. This study aimed to analyze changes in Sox10 expression in the hippocampus and its regulatory role in hippocampal myelin regeneration in a mouse model of demyelination. METHODS: Mice were fed 0.2% cuprizone (CPZ) for six weeks to establish the acute demyelinating model (CPZ mice). Behavioral changes of these mice were assessed via open field and tail suspension tests. The ultrastructure of the myelin sheaths in the hippocampus was observed by transmission electron microscopy. The expression levels of myelin sheath-related proteins and the transcription factor Sox10 were detected via immunohistochemistry and Western blots. Furthermore, Sox10-overexpressing adeno-associated virus was injected into the hippocampus after establishing the demyelinating model to investigate effects of Sox10 on remyelination. RESULTS: CPZ mice showed abnormal behavioral changes, a large number of pathological changes in the myelin sheaths, and significantly reduced protein expression of the myelin sheath markers myelin basic protein and proteolipid protein. This confirmed that the demyelinating model was successfully established. Meanwhile, the protein expression of the oligodendrocyte precursor cell marker neural/glial antigen 2 (NG2) increased, whereas Sox10 expression decreased. After Sox10 overexpression in the hippocampus, the abnormal behavior was improved, the ultrastructure of the myelin sheaths was restored, and the expression of myelin sheath protein was reversed. NG2 expression was upregulated. CONCLUSION: Overexpression of Sox10 promotes hippocampal remyelination after CPZ-induced acute demyelination.

OBJECTIVE: To investigate the relationship between Interleukin-1beta (IL-1beta) and diabetic peripheral neuropathy (DPN) using animal models. METHODS: The rat model of diabetic neuropathy was induced by intraperitoneal injection of a single dose of streptozotocin (STZ) at 65mg/kg. Diabetic rats were randomly divided into two groups (10 each), one treated with 0.9% saline (DMS group) and the other with interleukin-1 receptor antagonist (IL-1RA) at 50mg/kg (DMI group) twice a day for 5 weeks. Ten normal rats matched for weight, age and sex served as normal controls (Con group) and were treated with saline. Morphologic studies of sciatic nerves were achieved using light and transmission electron microscopy. RESULTS: Transmission electron microscopy of the sciatic nerve showed the ultrastructure of myelin and the axon in the IL-1RA group was highly protected compared to diabetic controls. CONCLUSION: High levels of circulating IL-1beta may be associated with the risk of DPN and anti-IL-1 treatment may provide a potential strategy for the prevention of diabetic neuropathy.

Ruthenium(II) polypyridyl complexes featuring peripheral quaternary ammonium structures were found to be able to selectively inactivate Gram-positive Staphylococcus aureus (S. aureus), including methicillin-resistant S. aureus (MRSA) upon visible light irradiation, but have low phototoxicity toward 293T cells, L02 cells and lack hemolysis toward rabbit red blood cells (RBC), exhibiting promising potential as a novel type of antimicrobial photodynamic therapy (aPDT) agents.

AIMS: To evaluate the effects of a novel, low-volume, high-intensity Progressive Resistance Training (PRT) technique on blood glucose control in elderly Chinese patients with Type 2 Diabetes. MATERIALS AND METHODS: The PReTTy2 trial enrolled 300 male and female patients with Type 2 Diabetes in a randomized resistance training program with the bioDensity™ technique. 100 were control patients with no training intervention and 200 had resistance training. Anthropometry, biochemical parameters, HbA1c and fasting plasma glucose (FPG) were measured at baseline, 3-month and 6-month intervals. RESULTS: 265 patients completed the study with no adverse events. There were no statistically significant differences in HbA1c for all patients, control and PRT groups, at baseline (p = 0.60), 3 months (p = 0.42) and 6-months (p = 0.45). Subgroup analysis with baseline HbA1c > 7.5% (58 mmol/mol), showed statistically significant differences in HbA1c and FPG between groups at 6 months (p < 0.05). All PRT group patients had statistically significant differences from baseline at 6 months for HDL (1.25 + 0.32 vs. 1.17 + 0.26 mmol/L, p < 0.001), LDL (3.23 ±â€¯0.89 vs. 2.93 ±â€¯0.80 mmol/L, p < 0.001) and total cholesterol (4.97 ±â€¯1.22 vs. 4.58 ±â€¯1.03 mmol/L, p < 0.001). CONCLUSIONS: PRT improves glycemic indices in elderly patients with Type 2 Diabetes with poor glucose control as an adjunct to diet and medication. Progressive Resistance Training with bioDensity™ is feasible, safe and effective in elderly patients with Type 2 Diabetes.

Gut microbiota plays an important role in maintaining intestinal barrier function and keeping body health. Changes of its structure and function are related to many common human diseases. As a class of non-coding single-stranded molecules, numerous studies have shown that the regulatory effect of microRNAs (miRNAs) at the gene level, can affect almost all biological processes in the body. In addition, gut microbiota can interact with miRNAs, and play a regulatory role in maintaining intestinal homeostasis and preventing metabolic diseases(diabetes) together. In this paper, we review the regulation of gut microbiota-miRNAs interaction, and how to regulate the occurrence and development of diabetes mellitus through this interaction.

OBJECTIVE@#To investigate the common genotypes of thalassemia of the pregnant woman in Wuhan area of China, and to make the prenantal gentic diagnosis for the fetus at high risk of thalassemia.@*METHODS@#A total of 357 pregnant woman with the primary positive screening in Wuhan area were included in this study. Genotypes were measured with PCR-flow cytometry, and fluorescence hybridization was used for detecting thalassmia gene. The husbands of the pregnant women with thalassmia were recalled for genetic analysis of thalassemia, and 9 cases of fetuses with high risk of thalassemia were detected by amniocontesis after genetic counseling.@*RESULTS@#In 357 cases of the pregnant women in Wuhan area, the 214 cases were diagnosed as thalassemia, 80 cases were diagnosed as alpha thalassemia (up to 90%), whose genotypes were determind as --/αα (78.75%) and -α/αα (15.00%), while 133 cases were determind with genotype of IVS-2-654/N (43.61%), CD41-42/N (20.30%) and CD17/N (19.55%) in beta thalassemia (up to 80%). 9 prenatal diagnosis continued pregnancy included 1case of -α/--, 1 case of -α/αα, 2 cases of --/αα, 2 cases of IVS-2-654/N and 3 cases of normal, however, the pregnancy in prenatal diagnosis of -α/-- voluntarily was terminated after genetic counseling. Follow-up results after delivery were consistent with prenatal diagnosis.@*CONCLUSION@#Minor and static thalassemia were very common in Wuhan area. Genetic detection after primary screening, genetic counseling and prenatal diagnosis in pregnant women could provide a theoretical basis for the development of regional specific prevention of intermedius and critical thalassemia which is meaning for rearing and bearing better children.

OBJECTIVE@#To investigate the β-thalassemia genotypes in neonates in Wuhan area of China and their characteristics of molecular epidemiology.@*METHODS@#A total of 2721 neonates in Wuhan who were positive in primary screening for β-thalassemia were included in this study. Genotypes of β-thalassemia gene were determined with PCR-flow cytometry and fluorescence hybridization assay.@*RESULTS@#There were 537 cases of β-thalassemia with over 15 kinds of genotypes, and 19 cases of α-composite β-thalassemia with 8 genotypes. Thalassemia minor appeared mostly in β-thalassemia, including 229 cases of IVS-2-654/N (42.64%), 121 cases of CD41-42/N (22.53%), 76 cases of CD17/N.(14.15%), 39 cases of CD26/N (7.26%) and 27 cases of CD27-28/N (5.03%) and the total ratio reached to 91.62%, however, 1 case of thalassemia intermediate was -29/IVS-2-654, and the genotype of 2 cases of thalassemia major was CD27-28/IVS-2-654 and CD41-42/IVS-2-654. The mutation frequency of IVS-2-654, CD41-42 and CD17 was higher in β-thalassemia, as follows: 42.93%, 22.36% and 14.13%, respectively.@*CONCLUSION@#β-Thalassemia minor is the majority of the neonants thalassemia in Wuhan area. The gene frequency of deletion type, such as IVS-2-654/N, CD41-42/N and CD17/N, is higher.

OBJECTIVE@#To explore the genetic basis for pedigree affected with hereditary nephrogenic diabetes insipidus (HNDI).@*METHODS@#Next generation sequencing (NGS) with an osteology system gene panel was carried out for the proband. Suspected mutation was validated by Sanger sequencing of two relatives with similar symptoms and two unaffected relatives from the pedigree.@*RESULTS@#The proband was found to carry a c.856C>T mutation of the AVPR2 gene. The same mutation was detected in the two relatives with similar symptoms and one unaffected healthy relative.@*CONCLUSION@#The HNDI in this pedigree may be attributed to the c.856C>T mutation of the AVPR2 gene.

@# To analyze the relationship between the degree of hepatic fibrosis, the inflammatory cell infiltrationandthepostoperativecholangitisafterKasaioperationinchildrenwithbiliaryatresia(BA).Methods Atotalof 92childrendiagnosedasBAinourhospitalfromJanuary2015toDecember2016wereselectedinthisstudy.Patientswere dividedintohepaticfibrosisgradeⅠgroup(n=8),hepaticfibrosisgradeⅡgroup(n=31),hepaticfibrosisgradeⅢgroup(n= 35)andhepaticfibrosisgradeⅣgroup(n=18),accordingtoJapan’sOhkuma’shepaticfibrosisgradingcriteria.Combined with Kasai age, the relationship between hepatic fibrosis and surgical age was analyzed. Samples of liver tissues were evaluatedbyhematoxylin/eosin(HE)andMassonstaining.Theexpressionsofspecificinflammatorycellmarkerantibody LCA,CD4,CD8andCD68inliverwereobservedbyIHCstaining.Combinedwiththepostoperativefollow-updataofKasai, the relationship between the degree of inflammatory infiltration of liver tissue, the degree of hepatic fibrosis and the postoperative cholangitis of Kasai in BA children was analyzed. Results Routine pathological staining showed that lymphocytesinfiltratedmainlyinlivertissue.TheexpressionlevelsofLCAandCD4werehigheringroupⅠandgroupⅣ comparedwiththoseofgroupⅡandgroupⅢ.TherewasnosignificantdifferenceintheCD8expressionbetweenfour groups.All92childrenwithBAwerefollowedup.Itwasfoundthatcholangitisoccurredin 50cases,earlycholangitis occurredin38cases,andfrequentcholangitisoccurredin23cases.Theincidenceofcholangitis,earlycholangitis,and frequentcholangitisafterKasaisurgerywasstatisticallysignificantinBApatientswithdifferentgradesofhepaticfibrosis (P<0.05). The incidence of cholangitis was higher in the groupⅠand group Ⅳ than that in groupⅡand group Ⅲ. Conclusion TheoccurrenceofcholangitisafterKasaioperationinchildrenwithBAiscorrelatedwiththehepaticfibrosis andliverinflammatorycells,especiallyTlymphocytesubgroups.

<p><b>OBJECTIVE</b>To investigate the genotypes of newborn α-thalassemia and to analyze its characteristics of molecular epidemiology in Wuhan area.</p><p><b>METHODS</b>The newborn α-thalassemia gene in 1376 cases with positive confirmed in the primary screening in Wuhan area was detected by PCR, flow cytometry and FISH.</p><p><b>RESULTS</b>The α-thalassemia in 436 newborns and α- β-composite thalassemia in 10 newborns were confirmed by detection in Wuhan area. The majority of thalassemia cases [up to 92.20% (402/436)] were found to be minor and static type thalassemia including 237 cases of -αα (54.36%), 135 cases of -α/ αα (30.96%) and 30 case of -α/αα (6.88%), however, the intermediate type -α/-was found in 1 newborn. The -/detetion, -α/detetion and -α/detetion were major in the detetion, the frequency was 54.12%, 32.29% and 7.13%, respectively; while the αmutation, αmutation and αmutation were less in the mutation, their frequency was 3.7%, 2.45% and 0.22% respectively.</p><p><b>CONCLUSION</b>The minor and static type newborn thalassemia is most common, the incidence of detetion type -/αα, -α/αα and α/αα is more high in Wuhan area of china.</p>

RESUMO

This article mainly elaborated around the four common categories of objective classification indicators, including constitution-gene and pathway, symptoms and signs, imaging examinations and biological indicators. We summarized advantages and limitations in all the objective classification categories, and put forward that both human secretory immunoglobulins A and salivary cortisol have the potential to be important classification indicators in constitution of traditional Chinese medicine, which might bring objective and quantitative criterion for constitution recognition and constitutional interventions in the future.

[Ru(bpy)2(py-SO3)](+) (3, bpy = 2,2'-bipyridine, py-SO3 = pyridine-2-sulfonate) was recently found to undergo py-SO3 ligand dissociation and py-SO3˙ radical generation under hypoxic conditions upon irradiation (Chem. Commun., 2015, 51, 428). To explore the substituent effect on the Ru-O homolysis by which the py-SO3˙ radical may be produced, [Ru(4,4'-(R)2-bpy)(py-SO3)](+), where R = OCH3 (1), CH3 (2), COOCH3 (4), were synthesized and their photochemical properties were investigated. The py-SO3˙ radical generation efficiencies followed the order of 4 > 3 > 2 > 1, and the radical generation efficiencies are wavelength dependent. As a result, 3 and 4 may lead to DNA covalent binding and DNA cleavage upon 355 nm irradiation, but merely DNA covalent binding upon 470 nm irradiation. In contrast, 1 and 2 can serve as DNA photo-binding agents only due to their less efficient Ru-O homolysis. The Ru-O homolysis via the (3)σ(Ru-O)π*(R-bpy) state is proposed to rationalize the substituent effect and the wavelength dependence, which is supported by TD-DFT calculations. This work gave insights into the mechanism of the Ru-O homolysis and provided guidelines for developing new [Ru(bpy)2(py-SO3)](+)-type complexes with higher Ru-O homolysis efficiency. Such complexes have dual activities of photoactivated chemotherapy (PACT) and photodynamic therapy (PDT) under hypoxic conditions and are therefore promising as a new class of antitumor drugs.

Stomach-Qi is an important concept in traditional Chinese medicine and widely accepted among many traditional Chinese medicine practitioners. However, the essence of stomach-Qi is unclear and not systematically reported. Thus, this article summarized some recent researches about stomach-Qi. It elaborated that the relationships among stomach-Qi and hemodynamic, immunity function and gastrointestinal function (gastrointestinal substance, gastrointestinal motility, gastrointestinal nutrition). With such review, TCM practitioners will well understand stomach-qi, emphasizeit, reinforce and protect it.

RESUMO

A novel azopyridine-based Ru(II) complex [Ru(bpy)2(L1)2](2+) (bpy = 2,2'-bipyridine, L1 = 4,4'-azopyridine) was designed and synthesized as a potential glutathione (GSH)-responsive photoactivated chemotherapy (PACT) agent, the DNA covalent binding capability of which can only be activated after GSH reduction and visible light irradiation.
