RESUMO

The clinical significance and comprehensive characteristics of chemokines and chemokine receptors in female patients with advanced colorectal adenocarcinoma have not ever been reported. Our study explored the expression profiles of chemokines and chemokine receptors and constructed a chemokine- and chemokine receptor-based signature in female patients with advanced colorectal adenocarcinoma. Four independent cohorts containing 1335 patients were enrolled in our study. Univariate Cox regression and least absolute shrinkage and selection operator (LASSO) analyses were performed to construct the signature. CIBERSORT was used to evaluate the landscape of immune cell infiltration. Thirty-two pairs of tissue specimens of female advanced colorectal cancer (CRC) patients and two CRC cell lines were used to validate the signature in vitro. Quantitative real-time PCR and western blotting were performed to validate the mRNA and protein expression levels of signature genes. EdU and colony formation assays were performed to examine proliferative ability. Transwell and wound healing assays were used to evaluate cell invasion and migration capacity. During the signature construction and validation process, we found that the signature was more applicable to female patients with advanced colorectal adenocarcinoma. Hence, the subsequent study mainly focused on the particular subgroup. Enrichment analyses revealed that the signature was closely related to immunity. The landscape of immune cell infiltration presented that the signature was significantly associated with T cells CD8 and neutrophils. Gene set enrichment analysis (GSEA) confirmed that the high-risk group was chiefly enriched in the tumor-promoting related pathways and biological processes, whereas the low-risk group was mainly enriched in anti-tumor immune response pathways and biological processes. The signature was closely correlated with CTLA4, PDL1, PDL2, TMB, MSI, and TIDE, indicating that our signature could serve as a robust biomarker for immunotherapy and chemotherapy response. ROC curves verified that our signature had more robust prognostic power than all immune checkpoints and immunotherapy-related biomarkers. Finally, we used 32 pairs of tissue specimens and 2 CRC cell lines to validate our signature in vitro. We first provided a robust prognostic chemokine- and chemokine receptor-based signature, which could serve as a novel biomarker for immunotherapy and chemotherapy response to guide individualized treatment for female patients with advanced colorectal adenocarcinoma.

BACKGROUND: The peach fruit moth, Carposina sasakii Matsumura (Lepidoptera: Carposinidae), is one of the most destructive pests of pome and stone fruits, while few studies of their molecular biology and physiology have been conducted. Research into CsasPBPs (Carposina sasakii pheromone binding proteins) and CsasGOBPs (Carposina sasakii general odorant binding proteins) may provide insights in to the mechanisms of olfaction in Carposina sasakii. RESULTS: In our study, results of real time quantitative polymerase chain reaction (qPCR) assays demonstrated that CsasPBP1-3 and CsasGOBP1-2 transcripts were abundantly expressed in the antennae of both sexes, suggesting they play a vital role in olfaction. In addition, to examine specific functional differences between pheromone binding proteins (PBPs) and general odorant binding proteins (GOBPs), fluorescence competitive binding assays were used to measured the binding affinities for the two sex pheromones and 18 apple plant volatiles. As a result, both PBPs and GOBPs showed stronger binding affinities to Z-7-eicosene-11-one than Z-7-nonadecene-11-one in two sex pheromones, whereas only PBP3 exhibited specific affinity towards both these two sex pheromone components, and PBP1 showed a high binding affinity to the sex pheromone components, and to other plant volatiles. In addition, GOBP1-2 displayed high binding affinity to general components of plant volatiles. CONCLUSION: Our study suggested CsasPBPs and CsasGOBPs play distinct physiological roles in the perception of sex pheromones and host plant volatiles. © 2018 Society of Chemical Industry.

BACKGROUND: The peach fruit moth, Carposina sasakii Matsumura (Lepidoptera: Carposinidae), poses a serious threat to a variety of fruits and causes significant economic loss owing to difficulties in its prevention and control. The olfactory sense is generally acknowledged to be a novel target for pest control. However, a systematic study of the olfactory genes expressed in C. sasakii has not been reported yet. Here, we reported the antennal transcriptome of C. sasakii using high-throughput sequencing and annotated the main chemosensory multi-gene families. RESULTS: In the chemosensory gene families, 29 odorant-binding proteins, 13 chemosensory proteins, 1 sensory neuron membrane protein, 52 odorant receptors, 8 ionotropic receptors and 11 gustatory receptors were annotated in the C. sasakii antennal transcriptome. The number of olfactory genes obtained in our transcriptome was consistent with that identified in other lepidopteran insects, confirming that we basically accomplished the annotation of the chemosensory genes of C. sasakii in the adult antennal transcriptome. All sequences were annotated and analyzed by BLAST (basic local alignment search tool), and some chemosensory genes with specific functions were named according to the BLAST results and phylogenetic trees. Based on the expression profile in the transcriptome and phylogenetic analysis, differentially expressed genes (DEGs) were analyzed in both male and female adults. Finally, fluorescence quantitative real-time PCR was used to identify the male-specific or female-specific chemosensory genes that were putatively related to odor detection and recognition. Moreover, expression levels of OR33 and PBP2 were significantly higher in males than in females, indicating that these genes may interact with sex pheromones. We found some conserved antennal IRs and GRs involved in detecting sugar compounds (GR2, GR5, GR6, GR8) and carbon dioxide (GR1), which were also identified based on phylogenetic analysis. CONCLUSIONS: There are 114 putative chemosensory proteins expressed in C. sasakii identified in this study. The identification of these proteins will make the molecular mechanism of odor recognition accessible.

The purpose of this study was to evaluate the sublethal effects of the beta-cypermethrin on calling behavior and fecundity of a major fruit-boring pest of apple, Carposina sasakii Matsumura. The mating rate, fertility (total number of eggs laid per female), and adult longevity of adults were remarkably decreased as compared with that in control when the adults were exposed to 10% lethal concentrations (LC10) of beta-cypermethrin (LC10♀ × LC10♂), and the age-specific survival rate (lx) was also negatively affected by sublethal beta-cypermethrin especially for the LC10â™€× LC10♂ mating combination. However, the age-specific fecundity (mx) was stimulated particularly in the combination of CKâ™€× LC10♂. Furthermore, the oviposition period was prolonged and the number of eggs was significantly increased for combinations of CKâ™€× LC10♂ and LC10â™€× CK♂. In the mating experiments, males in control or LC10-beta-cypermethrin treatments preferred to mate with females in control. It might be because of lower calling rate of female survivors treated with sublethal beta-cypermethrin. Our data indicate that treatment of beta-cypermethrin had a sublethal effect on the development and production of C. sasakii, and their mating behavior changes in surviving adults that may contribute to assortative mating.

Chlorantraniliprole is a novel diamide insecticide that targets the insect ryanodine receptor, a Ca(2+) release channel. Spodoptera exigua is a significant insect pest, and chlorantraniliprole is the most popular diamide insecticide used against this pest. To better understand the effects of diamides on RyR expression and [Ca(2+)], we isolated the SeRyR cDNA and investigated changes in SeRyR expression as a result of the application of chlorantraniliprole. The full-length cDNAs of SeRyR contain an open reading frame (ORF) of 15,357 bp with a predicted protein consisting of 5118 amino acids. SeRyR shares 77-92% identity with other insect RyR isoforms and 45-47% identity with vertebrate RyR isoforms. Furthermore, the relative expression abundances of RyR mRNA extracted from S. exigua fat body cells after 24 h of culture in 0.1, 1, 10, 100 nM, 1 µM and 100 µM of chlorantraniliprole changed 1.04-, 0.89-, 1.83-, 2.58-, 4.03- and 3.12-fold compared to blank control, respectively. The regression equation for the relative expression levels of SeRyR after 24 h as a function of the chlorantraniliprole concentration was Y = 0.6455 + 0.8188LgX, R(2) = 0.97093 for the cell line IOZCAS-Spex-II. These results outline the effects of chlorantraniliprole on the expression of SeRyR and provide a basis for the discovery of a compound that may exhibit selective insect activity.

In order to explore the effects of relative humidity (RH) on the population growth of summer fruit tortrix moth Adoxophyes orana, the development, survival, fecundity and longevity of A. orana were assessed at 6 different constant humidities (40%, 50%, 60%, 70%, 80% and 90%), (25 +/- 1) degrees C and a photoperiod of 12L:12D in the laboratory. The results showed that immature developmental time was gradually shortened with the increasing humidity at 40%-90% RH, ranged from 30.31 d at 40% RH to 25.87 d at 90% RH. However, the survival rate for immature moth was gradually increased from 40% RH to 90% RH. Hatchability was inhibited significantly by low humidity and the lowest was 39.3% at 40% RH. And at 80% RH, the longevity of adults was the longest, the oviposition was the most (110.8 eggs per female) , and the survival rates for eggs and larvae were the highest. It resulted in the highest net reproductive rate (R0=26.46) and intrinsic rate of increase (r(m) = 0.1018). In conclusion, the development and fecundity of A. orana were promoted and improved at the higher humidity.

To understand the influence of different apple varieties on the development and reproduction of the European red mite, Panonychus ulmi (Koch) (Acari: Tetranychidae), age-stage two-sex life tables of P. ulmi on 'Fuji,' 'Starkrimson Delicious,' 'Golden Delicious,' and 'Granny Smith' varieties were constructed under laboratory conditions at 23 +/- 1 degrees C, 75 +/- 5% relative humidity, and a photoperiod of 16:8 (L:D) h. Results showed that total development time of immature females was shorter on Fuji than on the other varieties, and this was because of its shorter egg duration. Immature survival of P. ulmi was 74.51-78.00% among four apple varieties, and no significant differences were found. The total fecundity per female was significantly higher on Golden Delicious (34.12 eggs per female) than that on Fuji (27.15 eggs per female), Starkrimson Delicious (25.15 eggs per female), and Granny Smith (20.62 eggs per female). Based on the intrinsic rate of population increase, Fuji and Golden Delicious were more suitable than Starkrimson Delicious and Granny Smith.

In order to understand the differences of Aphis citricola in its original and final selectivity to the apple leaves with different leaf age, and to ascertain the effects of mature and young apple leaves on the growth and development, survival, and fecundity of A. citricola, this paper studied the host preference of A. citricola to mature and young apple leaves, with the related age-stage two-type life tables constructed. A. citricola more preferred to oviposit on the young leaves. There existed differences in the behaviors between oriented selectivity and penetration selectivity, and the latter was the key factor determining whether A. citricola fed and oviposited on the host. The total development time of immature A. citricola on the mature leaves was significantly longer than that on the young leaves, and the survival rate was also lower, with the aphids died at the first and fourth instar stages and at mature stage. As compared with those on the young leaves, the apterae rate of A. citricola adults on the mature leaves was lower, and the longevity and fecundity were significantly shorter and lower. The population dynamic parameters suggested that the population growth of A. citricola was quicker on the young leaves than on the mature leaves, and it was difficult for A. citricola to establish a stable population on the mature leaves.

Panonychus ulmi Koch is one of the important pest insects of apple production in China. To clarify the spatiotemporal dynamics of P. ulmi on the apple tree crowns in an apple orchard of Liaoning, Northeast China, an investigation with random sampling was conducted on the pest mite number at each direction and each layer of the crowns in the whole growth season from May to November 2007. The spatial distribution pattern and time series dynamics of P. ulmi were analyzed by calculating the indices of aggregation and using the parameters of Iwao model. In the early and mid growth periods of apple tree, P. ulmi within whole crown fitted negative binomial distribution, presented an aggregated pattern, and its fundamental component was the group composed of several individuals that attracted each other. The aggregation intensity showed a negative fluctuation with population density, namely, high population density but low patchiness density, and low population density but high patchiness density, and there existed definite differences at different crown directions and layers, i. e., the patchiness density was the highest in south direction and the lowest in west direction, and was higher in mid and lower layers than in upper layer, and in inner layer than in outer layer.
