RESUMO

In the present study, we investigated the effect of infrasound on the expression of N-methyl-D-aspartate (NMDAR)1 as well as changes in intracellular calcium ion concentration ([Ca2+]i) in the hippocampus of rats. Sprague-Dawley (SD) rats were exposed for 2 h daily to infrasound at 8 Hz 90 dB or 130 dB, and NMDAR1 expression was examined on days 1, 7, 14, 21 and 28. The expression of NMDAR1 in the rat hippocampus upon exposure to infrasound at 8 Hz 90 dB sound pressure level (SPL) showed an initial decrease on day 1, an increase on days 7 and 14, a further decrease on day 21, and a return to normal levels on day 28. The peak level was observed on day 14 in every examined subregion of the hippocampus. By contrast, exposure to infrasound at 8 Hz 130 dB SPL had opposite effects, showing an increase on day 1, a decrease on day 7, a decrease to the lowest point on days 14, another increase on day 21 and a return to normal levels on day 28. The lowest expression of NMDAR1 was found in the CA1 and CA3 regions on day 14 and in the DG region on day 7 with exposure at 130 dB. There were significant differences in [Ca2+]i concentration on days 14 and 21 with infrasonic exposure at both 8 Hz 90 dB and 130 dB, but no significant differences in [Ca2+]i concentration on days 1, 7 and 28 compared to the control group. The highest [Ca2+]i level was noted on day 14 with infrasound exposure at 8 Hz 130 dB. These changes suggest that 8 Hz 90 dB/130 dB infrasound exposure induced certain reversible changes in NMDAR1 expression and [Ca2+]i concentration in hippocampal cells, which may influence mnemonic functions related to the hippocampus.

BACKGROUND/AIMS: Acupuncture has been reportedly used to treat gastrointestinal diseases, however, its precise mechanism remains unknown. METHODOLOGY: In our study, the effects and mechanism of electro-acupuncture (EA) at Tsusanli (ST 36), Shangchuhsu (ST 37) on regulation of gastric activity were observed. RESULTS: EA at Tsusanli showed that gastric electric change was the most obvious, with significantly higher frequency and wave amplitude compared with that of the Shangchuhsu group and other groups. EA at Shangchuhsu demonstrated that the change of gastric electric level was much higher than that of the non-acupoint group and control group. After bilateral vagotomy, Tsusanlis was electro-acupunctured, the changes of electro-gastric graph (EGG) weren't significant with the control group. The frequency of electro-physiological activity in nucleus of solitary tract (NTS) and dorsal motor nucleus of the vagus nerve (DMV) in the Tsusanli group was markedly increased compared with that in the other group. Fos and GFAP expression in NTS and DMV in the Tsusanli group was significantly higher than that in other groups and control group. The results have indicated that EA at Tsusanli and Shangchuhsu cannot only regulate gastric activity, but also activate neurons and astrocytes in NTS and DMV. The effects on regulation and activation of EA at Tsusanli were very obvious. CONCLUSIONS: Our study suggests that this electroacupuncture regulation of gastric activity may partially depend upon integrated nerve pathway and related central neurons and astrocytes in the vagus-solitary complex.

OBJECTIVE: To observe the expression of beta-endorphin and micro-opioid receptor (MOR) during wound healing process in rat with deep partial-thickness scald. METHODS: Thirty-six Wistar rats were randomly divided into control( n = 6, without treatment) , and scald ( n = 30, with 5% TBSA deep-partial thickness scald) groups. Skin specimens from wound were harvested immediately after scald and on 3, 7, 14, 21 post-scald days( PSD) for the determination of 1-endorphin and MOR expression with immunofluorescent staining. RESULTS: beta-endorphin and MOR were mainly distributed in nerve terminal at the border of dermis and epidermis , keratinocyte in some epidermis , in the fibroblast in dermis , with a weak expression The expression of beta-endorphin peaked in whole layer of skin on 3 PSD( 196 +/-16, P <0. 01) ,while that of MOR was concentrated in keratinocytes in the basal layer and the basement membrane. The expression of MOR was strengthened on 7 PSD with disarrangement of collagen , and it peaked on 14 PSD (306 +/- 23, P < 0.01) with epithelization in some wounds. There was still strong expression of beta-endorphin on 7 and 14 PSD. Complete epithelization was observed in scald group on 21 PSD, with nerve terminal approaching the boundary between the dermis and epidermis, and collagen began to arrange in good order. The expression of P-endorphin in scald group (31 +/-24)was similar to that in control group(30 +/- 18) on 21 PSD, but the expression of MOR (56 +/- 16) was still higher than that in control group (28 +/- 15 ). CONCLUSION: The expression of MOR and P-endorphin exhibits chronobiological nature during the process of wound healing,

Acupuncture at some specific acupoints of Foot Yangming can regulate gastric activity. However, its precise mechanism remains unknown. In our study, the effects and mechanism of electro-acupuncture (EA) at Tsusanli (ST 36), Shangchuhsu (ST 37) on the regulation of gastric activity were observed. EA at Tsusanli showed that gastric electric change had a significantly higher frequency and wave amplitude as compared to that of the Shangchuhsu group and other groups. EA at Shangchuhsu demonstrated the change of gastric electric was greater than that of the non-acupoint group and the control group. After bilateral vagotomy, the change of electro gastric graph (EGG) of EA at Tsusanlis was not significant compared to the control group. In the mean time, we have observed the electric discharge of the neurons in NTS and DMV. The frequency of electro-physiological activity in nucleus of solitary tract (NTS) and dorsal motor nucleus of the vagus nerve (DMV) in Tsusanli group and Shangchuhsu group were markedly increased compared with that in other groups. The results have indicated that EA at Tsusanli and Shangchuhsu not only regulate gastric activity, but also activate neurons in NTS and DMV significantly. Our study suggests that the effect of EA at Tsusanli and Shangchuhsu on the gastric activity may partially depend upon integrated nerve pathway and related central neurons in dorsal vagal complex.
