Herein, a highly efficient As(III) purifier Ce-Mn@N201 with excellent reusability was developed by stepwise precipitating hydrated cerium(IV) oxides (HCO) and hydrated manganese(IV) oxides (HMO) inside N201, a widely-used gel-type anion exchange resin. Owing to confinement of unique nanopores in N201, the in-situ generated nanoparticles (NPs) inside Ce-Mn@N201 were highly dispersed with ultra-small sizes of around 2.6 nm. Results demonstrated that HMO NPs effectively oxidized As(III) to As(V) with the conversion of Mn(IV) to Mn(II), while the generated Mn2+ was mostly re-adsorbed onto the negatively-charged surface of HMO NPs. During the regeneration process by simple alkaline treatment, the re-adsorbed Mn2+ was firstly precipitated as (hydr)oxides of Mn(II) and then oxidized to HMO NPs by dissolved oxygen to fully refresh its oxidation capacity. Though HCO NPs mainly served as adsorbent for arsenic, they could partially oxidize As(III) to As(V) at the beginning, while the oxidation capacities continuously diminished with the irreversible conversion of Ce(IV) to Ce(III). In 10 consecutive adsorption-regeneration cycle, Ce-Mn@N201 efficiently decontaminated As(III) from 500 µg/L to below 5 µg/L with Mn2+ leaching less than 0.3% per batch. During 3 cyclic fixed-bed adsorptions, Ce-Mn@N201 steadily produced 8500-9150 bed volume (BV) and 3150-3350 BV drinkable water from the synthesized and real groundwater, respectively, with Mn leaching in effluent constantly < 100 µg/L.

During coal combustion, the harmful element arsenic can be released into environment and cause potential significant harm to human beings. Therefore, it is very important to study the removal of arsenic from coal before combustion. In this work, simulated SO2-containing flue gas was used to leach arsenic from coal in a 1 L UV photoreactor. The effects of FeCl3, ultraviolet (UV), pH and the Cl-/Fe3+ molar ratio on arsenic leaching and SO2 removal were experimentally investigated and the enhancing mechanism was analysed. Experimental results demonstrated that FeCl3 and UV could efficiently increase iron and arsenic leaching percentages and SO2 removal efficiency. UV irradiation could induce the oxidation of most trivalent arsenic. The arsenic leaching percentage was significantly larger than that of iron. Low pH was favourable for iron and arsenic leaching. The optimal Cl-/Fe3+ molar ratio was determined to be 3:1. The introduced ferric chloride could not only increase the concentrations of free radicals and ferric iron oxidants, the chloride ion might also impede the formation of passive coatings, thus increasing the arsenic leaching percentage, intensifying the oxidation of trivalent arsenic and enhancing the removal of SO2.

BACKGROUD: The ECG profile of Hypertrophic Cardiomyopathy (HCM) includes ST-segment elevation (STE) that may lead to misdiagnosis of acute ST-segment elevation myocardial infarction (STEMI). This pseudo-STEMI may bring non-essential treatment. We aimed to confirm the ECG differences between HCM featured with pseudo-STEMI and acute STEMI. MATERIAL AND METHODS: We retrospectively enrolled 59 HCM cases (Group A) and 56 acute STEMI cases (Group B). Based on the locations of STE, all the patients were divided into four subgroups, including HCM with STE in anterior leads (Group A1), anterior STEMI (Group B1), HCM with STE in inferior leads (Group A2) and inferior STEMI (Group B2). Several ECG parameters were compared between these subgroups. RESULTS: ECG parameters significantly differed between these groups, especially the number of leads with TWI. We evaluated the diagnostic value of ECG profiles for those groups. ROC analysis showed that for Group A vs. Group B, number of leads with TWI showed the highest AUC value of 0.805 and its cutoff of 2.5, with 76.3% sensitivity and 76.8% specificity. For Group A1 vs. Group B1, it showed the highest AUC value of 0.801 and its cut-off point was 2.5, with 77.1% sensitivity and 79.1% specificity. For Group A2 vs. Group B2, it showed the highest AUC value of 0.822 and the cut-off value was 4.5, with 54.5% sensitivity and 92.3% specificity. CONCLUSION: ECG plays a valid tool to distinguish "Pseudo-STEMI" HCM from acute STEMI, especially number of leads with TWI.

Long period fiber gratings (LPFGs) have special advantages in the detection of salt concentrations due to small volume, corrosion resistance and immunity to electromagnetic interference. However, it is very difficult to distinguish low-concentration salt solutions with usual LPFGs owing to the poor sensitivity. In this paper, the detection capability of the LPFG to low-concentration salt solutions was significantly improved by assembling salt-containing poly (diallyldimethylammonium chloride) (PDDA) and salt-containing poly (sodium-p-styrenesulfonate) (PSS). Experimental results showed that, the responsive wavelength range of the LPFG was remarkably broadened in low-concentration salt solutions after assembling nanofilms. The suitable detection range of the PDDA/PSS films coated LPFG for salt concentrations was 0-3%. In such a range, the average refractive index sensitivity and the average salinity sensitivity of the LPFG was as high as 29545.9 nm/RIU and 52.2 nm/% respectively. Compared with the LPFG without nanofilms, the discrimination ability of the PDDA/PSS films coated LPFG to 0-3% salt solutions increased by 568 times. The analysis demonstrated that PDDA and salt in the assembly solutions played a pivotal role in the above effects. The proposed sensor has extensive application prospects in the monitoring of salt concentration in many fields such as seawater, food processing, fermentation process, etc.

Introduction: Cancer in patients of childbearing age continues to become increasingly common. The purpose of this study was to explore the impact of metastatic breast cancer (MBC) on overall survival (OS) and cancer-specifific survival (CSS) in patients of childbearing age and to construct prognostic nomograms to predict OS and CSS. Methods: Data from MBC patients of childbearing age were obtained from the Surveillance, Epidemiology, and End Results (SEER) database between 2010 and 2015, and the patients were randomly assigned into the training and validation cohorts. Univariate and multivariate Cox analyses were used to search for independent prognostic factors impacting OS and CSS, and these data were used to construct nomograms. The concordance index (C-index), area under the curve (AUC), and calibration curves were used to determine the predictive accuracy and discriminative ability of the nomograms. Additional data were obtained from patients at the Yunnan Cancer Hospital to further verify the accuracy of the nomograms. Results: A total of 1,700 MBC patients of childbearing age were identifified from the SEER database, and an additional 92 eligible patients were enrolled at the Yunnan Cancer Hospital. Multivariate Cox analyses identifified 10 prognostic factors for OS and CSS that were used to construct the nomograms. The calibration curve for the probabilities of OS and CSS showed good agreement between nomogram prediction and clinical observations. The C-index of the nomogram for OS was 0.735 (95% CI = 0.725-0.744); the AUC at 3 years was 0.806 and 0.794 at 5 years.The nomogram predicted that the C-index of the CSS was 0.740 (95% CI = 0.730- 0.750); the AUC at 3 years was 0.811 and 0.789 at 5 years. The same results were observed in the validation cohort. Kaplan- Meier curves comparing the low-,medium-, and high-risk groups showed strong prediction results for the prognostic nomogram. Conclusion: We identifified several independent prognostic factors and constructed nomograms to predict the OS and CSS for MBC patients of childbearing age.These prognostic models should be considered in clinical practice to individualize treatments for this group of patients.

An extremely sensitive multi-order mode refractive index (RI) sensor was fabricated by coupling titanium dioxide nanograss film coated FTO conductive glass with Kretschmann prism. Both calculation and experimental studies were carried out. Theoretical analysis by employing resonant waveguide modes indicated that the maximum sensitivity could be achieved when the mode worked at the weakly-bounded condition. The experimental results showed that for p-polarized and s-polarized light, the sensor exhibited a maximum RI sensitivity of 2938.21 nm/RI unit (RIU) and 1484.39 nm/RIU in the 1st order mode, respectively. Its maximum figure of merit was as high as 77.77. The proposed sensor is promising to be applied in environmental monitoring, immune analysis, nucleic acid test, etc.

OBJECTIVES: To identify key long non-coding (lnc)RNAs responsible for the epithelial-mesenchymal transition (EMT) of CNE1 nasopharyngeal carcinoma cells and to investigate possible regulatory mechanisms in EMT. METHODS: CNE1 cells were divided into transforming growth factor (TGF)-ß1-induced EMT and control groups. The mRNA and protein expression of EMT markers was determined by real-time quantitative PCR and western blotting. Differentially expressed genes (DEGs) between the two groups were identified by RNA sequencing analysis, and DEG functions were analyzed by gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses. EMT marker expression was re-evaluated by western blotting after knockdown of a selected lncRNA. RESULTS: TGF-ß1-induced EMT was characterized by decreased E-cadherin and increased vimentin, N-cadherin, and Twist expression at both mRNA and protein levels. Sixty lncRNA genes were clustered in a heatmap, and mRNA expression of 14 dysregulated lncRNAs was consistent with RNA sequencing. Knockdown of lnc-PNRC2-1 increased expression of its antisense gene MYOM3 and reduced expression of EMT markers, resembling treatment with the TGF-ß1 receptor inhibitor LY2109761. CONCLUSION: Various lncRNAs participated indirectly in the TGF-ß1-induced EMT of CNE1 cells. Lnc-PNRC2-1 may be a key regulator of this and is a potential target to alleviate CNE1 cell EMT.

Epstein-Barr virus (EBV) is positively related to the morbidity of nasopharyngeal carcinoma (NPC) in Asia. After infection, EBV can produce several proteins, including viral interleukin-10 (vIL-10). But the mechanism by which vIL-10 contributes to NPC cell proliferation and cell cycle progression is not well understood. In this study, EBV negative and positive cell lines, and the JAK2/STAT3 signal pathway inhibitor AG490 were used to illustrate the role of vIL-10 in NPC. Cell proliferation and cell cycle were measured by CCK-8 and flow cytometry. The expression levels of related protein were measured by Western blotting. High concentrations of vIL-10 and IL-6 were found in the EBV positive patients. The expression level of IL-6 was positively related to the presence of concentration of vIL-10. vIL-10 can promote cancer cell proliferation and G1 to S phase transmission via upregulating the IL-6 protein level by activating the JAK2/STAT3 signal pathway. Furthermore, EBV can induce the formation of cytotoxic T cells, whereas vIL-10 can block the function of cytotoxic T cells. Taken together, these results suggest that vIL-10 promotes cell proliferation and cell cycle progression via JAK2/STAT3 signaling pathway in NPC.

Drug safety is a severe clinical pharmacology and toxicology problem that has caused immense medical and social burdens every year. Regretfully, a reproducible method to assess drug safety systematically and quantitatively is still missing. In this study, we developed an advanced machine learning model for de novo drug safety assessment by solving the multilayer drug-gene-adverse drug reaction (ADR) interaction network. For the first time, the drug safety was assessed in a broad landscape of 1,156 distinct ADRs. We also designed a parameter ToxicityScore to quantify the overall drug safety. Moreover, we determined association strength for every 3,807,631 gene-ADR interactions, which clues mechanistic exploration of ADRs. For convenience, we deployed the model as a web service ADRAlert-gene at http://www.bio-add.org/ADRAlert/. In summary, this study offers insights into prioritizing safe drug therapy. It helps reduce the attrition rate of new drug discovery by providing a reliable ADR profile in the early preclinical stage.

Delivering safe and effective therapeutic treatment to patients is one of the grand challenges in modern medicine. However, drug safety research has been progressing slowly in recent years, compared to other fields such as biotechnologies and precision medicine, due to the mechanistic complexity of adverse drug reactions (ADRs). To fill up this gap, we develop a new database, the Adverse Drug Reaction Classification System-Target Profile (ADReCS-Target, http://bioinf.xmu.edu.cn/ADReCS-Target), which provides comprehensive information about ADRs caused by drug interaction with protein, gene and genetic variation. In total, ADReCS-Target includes 66,573 pairwise relations, among which 1710 are protein-ADR associations, 2613 are genetic variation-ADR associations, and 63,298 are gene-ADR associations. In a case study of exploring the mechanism of rash, we find that HLAs, C1QA and APOA1 are the key gene players and thus can be potential targets (or biomarkers) in monitoring or countermining rashes. In summary, ADReCS-Target can be a useful resource for the biomedical scientific community by serving researchers in the fields of drug development, clinical pharmacology, precision medicine, and from web lab to high-throughput computational platform. Particularly, it helps to identify drug with better ADR profile and design safer drug therapy regimen.

We propose a novel, highly sensitive refractive index (RI) sensor by means of combining the Kretschmann prism with a TiO2 nanowire array and do not use a metallic layer in the Kretschmann configuration. Its RI sensing performance was investigated through measuring different concentrations of sodium chloride solution. Experimental results showed that, with increasing RI of liquid, the resonant wavelength in the reflectance spectrum redshifted gradually in the visible light range. There was a very good linear relationship between resonant wavelength and RI in the range of 1.3330 to 1.3546. More importantly, in contrast to the surface plasmon resonance (SPR) sensor, the interferometric sensors showed higher sensitivity to the external RI. In the case of the transverse magnetic mode, the RI sensitivity is up to 320,700.93 a.u./RIU (refractive index unit) by expression of light intensity, which is 9.55 times that of the SPR sensor. As for the transverse electric mode, it achieves 4371.76 nm/RIU by expression of the resonant wavelength, which is increased by a factor of 1.4 in comparison with the SPR sensor. Moreover, the experimental results have favorable repeatability. A TiO2 nanowire array sensor has also other advantages, such as easy manufacturing, low cost, and in situ determination, etc. To our knowledge, this fact is reported for the first time. It has great potential applications in the field of biological and chemical sensing.

The response of a novel long-period fiber grating (LPFG) with a period of 180 µm to a surrounding refractive index (RI) was investigated. The results displayed that, with the increase in RI of the surrounding media of cladding glass in the grating region, the resonant peak located at 1336.4 nm in the transmission spectrum gradually shifts towards a shorter wavelength, while the resonant peak located at 1618 nm gradually shifted towards a longer wavelength. Moreover, the resonant peak at 1618 nm is much more sensitive to the surrounding RI than that of the one at 1336.4 nm. Compared with the conventional LPFG and other types of wavelength-interrogated RI sensors, such as ring resonators, surface plasmon resonance sensors, and Fabry-Perot interferometric sensors, this novel LPFG possesses a higher sensitivity, which achieved 10,792.45 nm/RIU (RI unit) over a RI range of 1.4436-1.4489.

OBJECTIVE: To investigate the effects of Bushen Huoxue Fang (BSHX) on the apoptosis of epithelial cells in the prostatic ductal system of rats with benign prostatic hyperplasia (BPH) and its possible action mechanism. METHODS: One hundred 3- month-old male Wistar rats were randomly divided into four groups of equal number (control, castrated, BPH model, and BSHX). BPH models were made by subcutaneous injection of testosterone following castration; the rats in the BSHX group were treated intragastrically with BSHX at 2.34 g/ml after modeling, while those in the other two groups with equal volume of saline, all for 37 days. On the 38th day, all the rats were sacrificed and their prostates harvested for detection of the distribution of TGF-beta1 and alpha-actin and the count of positive cells in the prostatic ductal system by immunohistochemical staining. The apoptosis rate of epithelial cells in the prostatic ductal system was determined by TUNEL assay. RESULTS: The expression of TGF-beta1 was significantly increased in the rats of the BSHX group as compared with the BPH models in both the proximal prostatic duct ([15.28 +/- 4.30]% vs [36.42 +/- 8.10]%, P < 0.01) and the distal prostatic duct ([4.42 +/- 2.07]% vs [8.71 +/- 2.28 ]%, P < 0.05), while the expression of alpha-actin in the proximal duct was remarkably higher in the BSHX-treated rats than in the models ([28.14 +/- 7.43]% vs [18.28 +/- 4.07]%, P < 0.01), but lower than in the control animals ([33.57 +/- 6.85]%, P < 0.05). Compared with the control group, the BPH models and BSHX-treated rats both exhibited markedly decreased apoptosis of epithelial cells in the proximal prostatic duct ([39.42 +/- 9.20]% vs [3.86 +/- 1.34]%, P < 0.01, and [31.14 +/- 5.64]%, P < 0.01) and distal prostatic duct ([17.60 +/- 4.86]% vs [3.07 +/- 1.14]%, P < 0.01, and [12.37 +/- 2.25]%, P < 0.05). The apoptosis rate of epithelial cells in the prostatic ductal system was significantly higher in the BSHX-treated rats than in the BPH models (P < 0.01). CONCLUSION: By upregulating the expression of TGF-beta, BSHX can suppress the reduction of smooth muscle cells in the proximal prostatic duct, promote the apoptosis of prostatic epithelial cells, and thus effectively inhibit benign prostatic hyperplasia.

An optical fiber Fabry-Perot (F-P) interferometer end surface was modified using layer-by-layer assembly and chemical covalent cross linking method, and the refractive index (RI) response characteristics of coated optical fiber F-P sensor were experimentally studied. Poly diallyldimethylammonium chloride (PDDA) and sodium polystyrene sulfonate (PSS) were chosen as nano-film materials. With the numbers of layers increasing, the reflection spectral contrast of optical fiber F-P sensor presents from high to low, then to high regularity. And the reflection spectral contrast has good temperature stability. The reflection spectra of the optical F-P sensor coated with 20 bilayers for a series of concentration of sucrose and inorganic solution were measured. Experimental results show that the inflection point extends from 1.457 to 1.462 3, and the reflection spectral contrast sensitivity to low RI material and high RI material is 24.53 and 3.60 dB x RI(-1), respectively, with good linearity. The results demonstrate that the functional coated optical F-P sensor provides a new method for biology and chemical material test.

In this paper, a novel TiO(2) nanoparticle thin film coated optical fiber Fabry-Perot (F-P) sensor had been developed for refractive index (RI) sensing by monitoring the shifts of the fringe contrast in the reflectance spectra. Using in situ liquid phase deposition approach, the TiO(2) nanoparticle thin film could be formed on the fiber surface in a controlled fashion. The optical properties of as-prepared F-P sensors were investigated both theoretically and experimentally. The results indicated that the RI sensitivity of F-P sensors could be effectively improved after the deposition of nanoparticle thin-films. It was about 69.38 dB/RIU, which was 2.6 times higher than that of uncoated one. The linear RI measurement range was also extended from 1.333~1.457 to 1.333~1.8423. More importantly, its optical properties exhibited the unique temperature-independent performance. Therefore, owing to these special optical properties, the TiO(2) nanoparticle thin film coated F-P sensors have great potentials in medical diagnostics, food quality testing, environmental monitoring, biohazard detection and homeland security, even at elevated temperature.
