RESUMO

Antimicrobial peptides (brevinin1 HYba1 and brevinin1 HYba2) identified from the skin secretion of an endemic frog species of Western Ghats were studied against fish pathogens. Post-translational modifications such as c-terminal amidation and cyclization of the peptides were enhanced on the activity against Aeromonas sobria. Based on the Minimum inhibitory concentration (3 µM), cyclic amidated brevinin Hyba2 was identified as the most promising antimicrobial agent against A. sobria and can be developed further as a lead drug molecule.

RESUMO

A cyclonic eddy (CE) in the southwestern Bay of Bengal (SWBoB; 10-15° N; 81-87° E) during winter monsoon 2005 and associated changes in the open ocean hydrography and productivity patterns were studied using satellite observations and in situ measurements. Analysis of the satellite-derived sea surface height anomaly (SSHA) indicated the existence of a large eddy (10-15° N; 81-87° E) from November to January, with its core centered at 13° N and 82° E. The large positive wind stress curl (~1.8 × 10(-7) N m(-2)) and resultant Ekman pumping (~3 × 10(-2) cm s(-1)) were identified as the prominent forcing mechanisms. In addition, the cyclonic storms and depressions experienced in the region during the study period seem to have served to maintain the strength of the CE through associated Ekman pumping. The cold (~26.6 °C), nutrient-enriched (NO3 > 2 µM, PO4 > 0.73 µM and SiO4 > 3 µM) upwelled waters in the upper layers of the CE enhanced the biological production (chl. a > 0.56 mg m(-3)). Dissolved oxygen in the surface waters was > 200 µM. The phytoplankton and zooplankton biomass recorded during the season was comparable or perhaps higher than the peak values reported from the northeastern Arabian Sea (chlorophyll a concentrations of 0.2 to 0.4 mg m(-3) and zooplankton biovolume 0.6 ml m(-3)) during winter. Occurrence of a higher mesozooplankton biovolume (0.8 ml m(-3)) and relatively low abundance of microzooplankton indicates the prevalence of a short food chain. In conclusion, high biological production, both at primary as well as secondary level, suggests the prevalence of an efficient food web as a result of physical forcing and subsequent ecological interactions evident up to tertiary level in an oligotrophic basin like BoB.

Characteristics of a cold-core eddy and its influence on the mesozooplankton community were studied along the central (87° E) Bay of Bengal during winter monsoon (November 2008) based on in situ data. The thermo-haline structure and the satellite-derived sea level anomaly maps showed the presence of a cyclonic eddy between 16° N and 20° N. The nutrient enhancement due to the eddy pumping in the euphotic column (∼ 50 m) had resulted in high chlorophyll a concentration, a factor of 8 times higher than that outside the eddy, which led to higher mesozooplankton biovolume (0.35 ± 0.36 ml m(-3)) and abundance (276 ± 184 ind m(-3)). The northern cyclonic eddy (NCE) seems to exist for approximately 6 months between July and January. During summer, the NCE is forced by local wind stress curl and the resultant Ekman pumping, whereas during fall and early phase of the winter, it is sustained by westward propagating semi-annual Rossby waves. The longer existence of NCE in the study region, which originated 6 months prior to the present observation, provides a favourable environment for the mesozooplankton community to grow and reproduce, resulting in noticeable increase in the biovolume. Hence, the persistent and longer existence of NCE significantly influences the biological production of the generally oligotrophic BoB, making it locally biologically 'active'.

All life forms are equipped with rapidly acting, evolutionally conserved components of an innate immune defense system that consists of a group of unique and diverse molecules known as host defense peptides (HDPs). A Systematic and Modular Modification and Deletion (SMMD) approach was followed to analyse the structural requirement of B1CTcu5, a brevinin antibacterial peptide amide identified from the skin secretion of frog Clinotarsus curtipes, India, to show antibacterial activity and to explore the active core region. Seventeen SMMD-B1CTcu5 analogs were designed and synthesised by C and N-terminal amino acid substitution or deletion. Enhancement in cationicity by N-terminal Lys/Arg substitution or hydrophobicity by Trp substitution produced no drastic change in bactericidal nature against selected bacterial strains except S. aureus. But the sequential removal of N-terminal amino acids had a negative effect on bactericidal potency. Analog B1CTcu5-LIAG obtained by the removal of four N-terminal amino acids displayed bactericidal effect comparable to, or in excess of, the parent peptide with reduced hemolytic character. Its higher activity was well correlated with the improved inner membrane permeabilisation capacity. This region may act as the active core of B1CTcu5. Presence of C-terminal disulphide bond was not a necessary condition to display antibacterial activity but helped to promote hemolytic nature. Removal of the C-terminal rana box region drastically reduced antibacterial and hemolytic activity of the peptide, showing that this region is important for membrane targeting. The bactericidal potency of the D-peptide (DB1CTcu5) helped to rule out the stereospecific interaction with the bacterial membrane. Our data suggests that both the C and N-terminal regions are necessary for bactericidal activity, even though the active core region is located near the N-terminal of B1CTcu5. A judicious modification at the N-terminal region may produce a short SMMD analog with enhanced bactericidal activity and low toxicity against eukaryotic cells.

AIM: The aim was to determine the chemical composition of solar dried blood and rumen content (DBRC) and further ascertain the concentration at which DBRC could be included in Japanese quail diets without any adverse effect on its performance. MATERIALS AND METHODS: Feeding trial on the effect of DBRC on performance of Japanese quails was studied up to 5 weeks. 252 numbers of day old (Nandanam Type III breed) Japanese quails were purchased from Poultry Research Station, Madhavaram and divided into 7 batches (control+ six treatments) each consisting of 36 birds. The DBRC was included at 0%, 5%, 10%, 15%, 20%, 25% and 30% in diets as control, treatment-1 (T1), treatment-2 (T2), treatment-3 (T3), treatment-4 (T4), treatment-5 (T5) and treatment-6 (T6) respectively in a completely randomized design to replace soybean meal in Japanese quail feed. The birds were provided with ad-labidum feed and drinking water ad-libitum during the entire experimental period. RESULTS: The crude protein (CP), crude fiber (CF), ether extract (EE) and ash contents of DBRC were 35.87%, 17.40%, 3.6% and 12.6%, respectively. The amount of essential amino acids and non-essential amino acid content were found to be 12.98 and 4.87 (g/100 g of feed) respectively in DBRC feed. Result showed that all birds fed DBRC diets performed better than the control group. Mortality was unaffected by dietary treatments. There was a significant difference (p<0.01) observed in weight gain in treatment groups compared to the control. CONCLUSION: Up to 30% DBRC could be incorporated in the diets of Japanese quails without any adverse effects on its performance.

BACKGROUND: Sural nerve is formed by communication of medial sural cutaneous nerve, that arise from tibial nerve in popliteal fossa and peroneal communicating nerve, a branch directly from common peroneal nerve or from lateral sural cutaneous nerve. The sural nerve is universally recognized by surgeons as a site for harvesting an autologous nerve graft and for nerve biopsies in case of neuropathies. SETTING AND DESIGN: Anatomy department and an observational study. STATISTICAL ANALYSIS: The Mean, Standard Deviation, relevant percentages and p value was calculated by Chi-square test. MATERIAL AND METHODS: The study consists of 50 lower limb specimens in 25 cadavers. All the male and female complete cadavers were included in the study. The data was collected by dissection method by using dissection instruments and parameters (measurements) were obtained by using measuring tape. RESULTS: The sural nerve complex was dissected and observed in 50 formalin fixed lower limb specimens. Study was done in 16 male and 9 female cadavers. Three types of formation of sural nerve were observed were - Type A was seen in 36 specimens (72%) of cases. Type B was seen in 14 specimens (28%) of cases. Type C sural nerve was not found in present study. Site of union of components of SN was seen in the upper 3(rd) of leg in 5.6%, in middle 3(rd) it was in 33.3% and in lower 3(rd) it was in 58.3% of cases. The length of the sural nerve shows a wide range from 2 cm to 32 cm. The symmetrical distribution was seen in 62.5% in males and 55.6% in female. CONCLUSION: The normal anatomy and variations may help to guide during surgeries in the region.
