The sipM gene of Bacillus megaterium encoding a type I signal peptidase (SPase) was isolated and structurally characterized. RNA analysis revealed a transcript size in accordance with a bicistronic operon comprising sipM and an adjacent open reading frame. Inactivation of sipM by targeted gene disruption could not be achieved, indicating its essential role for cell viability since there might be no other type I SPase of major importance present in B. megaterium. Plasmid-assisted amplification of the gene resulted in an increase in activity of the heterologous glucanase used as an extracellular reporter, suggesting a potential bottleneck for protein secretion within this species.

BACKGROUND: Gonadotropins and sex hormones are intimately related to the stability of affective states. Patients with affective disorders may demonstrate abnormal levels of sex hormones and gonadotropins. It is therefore possible that affective disorder patients may experience mood dysregulation by synthetic sex hormones and gonadotropins like lupron. METHODS: A case report of a young woman with a history of endometriosis and a past history of irritability and depression is described. Treatment of the endometriosis with lupron induced a manic episode. RESULTS: The lupron-induced mania was successfully treated with a mood-stabilizing agent, lithium carbonate. CONCLUSIONS: Patients with a history of affective disorder may develop manic episodes when treated with Lupron. Mood-stabilizing agents are helpful in ameliorating this unwanted effect.

We describe a UHF rewarming system which has been used to measure warming rates, and particularly the uniformity of warming on three orthogonal axes, in a rabbit kidney phantom 36 mm in diameter. The stabilizing effect is demonstrated of using an E-field directed along the temperature gradient (or normal to any surface of dielectric discontinuity). One hot spot remains unaccounted for. The average warming rates and power dissipation are related to the volumetric heat capacity of the phantom material at -30 degrees C.

The dielectric properties of cryopreserved biological tissue are discussed in relation to the problems which arise when EM fields are used for rapid rewarming. The UHF band is favored from two aspects: the avoidance of thermal runaway and the uniformity of heating inhomogeneous material. Various resonant cavity applicators are considered for efficient and uniform rewarming. The square-aspect TE 111 cylindrical applicator is favored principally because it allows variation of the E-field orientation as required during the warming profile. An appropriate kidney phantom organ is described. It is used to obtain measured values of the overall efficiency of a TE 111 applicator. The efficiency values are found to fall steadily with increasing temperature from 85% at -40 degrees C to 45% at the phase change, mainly due to the decreasing tan delta value of the phantom material.
