RESUMO

Optimization of antioxidants and angiotensin-converting enzyme (ACE) inhibitory potential gelatin hydrolysate production from Labeo rohita (rohu) swim bladder (SBGH) by alcalase using central composite design (CCD) of response surface methodology (RSM) was investigated. The maximum degree of hydrolysis (DH), 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), total antioxidants (TAO), and ACE inhibitory activity were achieved at 0.1:1.0 (w/w) enzyme to substrate ratio, 61 °C hydrolysis temperature, and 94-min hydrolysis time. The resulting SBGH obtained at 19.92% DH exhibited the DPPH (24.28 µM TE/mg protein), ABTS (34.47 µM TE/mg protein), TAO (12.01 µg AAE/mg protein), and ACE inhibitory (4.91 µg/mg protein) activity. Furthermore, SBGH at 100 µg/ml displayed osteogenic property without any toxic effects on MC3T3-E1 cells. Besides, the protein content of rohu swim bladder gelatin (SBG) and SBGH was 93.68% and 94.98%, respectively. Both SBG and SBGH were rich in glycine, proline, glutamic acid, alanine, arginine, and hydroxyproline amino acids. Therefore, SBGH could be an effective nutraceutical in functional food development.

Microwave (MW)-based dry blanching can inactivate oxidative enzymes like peroxidase (POD) and polyphenol oxidase (PPO) rapidly and retain a higher amount of water-soluble nutrients, like ascorbic acid. This study compared the MW-based dry blanching of potato slices of various thicknesses (5, 8, and 10 mm) with conventional methods (water and steam blanching). The time required for water and steam blanching was longer than that required for MW blanching. Potato slices of 10 mm thickness required a longer blanching duration compared with slices of a lesser thickness (5 and 8 mm). The MW-blanched samples (77.37-83.5%) retained a higher content of ascorbic acid, followed by steam-blanched (69.15-74.92%) and water-blanched (67.18-71.54%) samples. The Page, modified Page, Midilli-Kucuk, and Hii, Law, and Cloke models predicted the thin layer drying of potato slices (5 mm thickness) better with a higher coefficient of determination values (0.9607-0.9976) compared to Fick's and Exponential models (0.8942-0.9444).

Effect of four different cultivars and salt pretreatment on drying of Amla (Emblica officinalis) gratings were studied. Quality attributes namely, ascorbic acid, total phenolic content, antioxidant activity and colour were evaluated for dried (using Cabinet tray dryer at 55 ± 2 °C for 8 h) and stored (refrigerated, ambient and accelerated condition) samples of four cultivars (Krishna, Kanchan, NA-7 and Chakaiya). Salt pretreated dried samples showed better retention of nutrients and colour as compared to untreated. Retention of ascorbic acid, total phenolic content and antioxidant activity (IC50 value) in pretreated dried samples were in the range of 79.51-84.89%, 176.5-220.3 mg GAE/g db and 9.48 to 17.74 mg/µl, respectively. Colour retention was also found to be better in salt pretreated samples. Ambient storage condition resulted in higher retention of ascorbic acid and colour compared to accelerated condition. Taking into consideration, the nutritional value of the fresh Amla, ability to retain nutritional value and color during drying and storage, NA-7 cultivar Amla (pretreated with 1% salt, tray dried at 55 °C for 8 h) is the most suitable for preparation of Amla powder. The method developed in the present work is devoid of blanching step and found to be effective in retaining the nutrients during drying and storage and can be employed also for drying of similar fruits and vegetables.
