BACKGROUND: Anthrax is the most prioritized zoonotic disease in Kazakhstan due to its threatening potential to the public health and agricultural sector. Sporadic anthrax outbreaks are being reported annually among human and livestock populations throughout the country, with the highest frequency occurring in West Kazakhstan. METHODS: A cross-sectional study was conducted using a survey-based face-to-face interview. From January to May 2022, 489 randomly selected participants were surveyed in 6 districts of the Baiterek province in West Kazakhstan oblast to evaluate the knowledge, attitude and practice (KAP) regarding anthrax among community members. This is the first KAP study conducted relating to outbreaks of anthrax in Kazakhstan. RESULTS: In this study, most participants (74%) surveyed were males, and 40% of respondents had a secondary level education. Overall, 91% of the community respondents were engaged in agriculture and livestock rearing. Among these community members, cattle rearing was the most common (67%) occupation compared to other livestock species. Additionally, over a 50% of the population studied had no knowledge about the zoonotic nature of the disease, and about 82% and 87% of respondents were unaware of any animal and human anthrax symptoms, respectively. About 70% of the respondents were interested in vaccinating their livestock against anthrax. Individuals aged 45-54 displayed notably higher animal vaccination rates (45%; 95% CI: 38.4-52.0; p < 0.025) compared to those aged 25-34 and 65-74. Respondents residing in the Beles district (20%; 95% CI: 17.1-24.7; p < 0.005) exhibited a significantly higher level of awareness concerning the fatality of anthrax in contrast to participants from Bolashak. Roughly 61% of respondents held the belief that anthrax is a lethal disease. An overwhelming majority of the survey participants (99%) affirmed their non-participation in the slaughter of infected animals. CONCLUSION: The findings of this study indicate that KAP among community members relating to anthrax is low and requires swift implementation of education programmes in building awareness of anthrax under the One Health approach, especially in anthrax prone regions.

An outbreak of camelpox occurred in the Mangistau region of Kazakhstan in 2019. To control the outbreak of camelpox and to prevent its further spread to other regions, camels were vaccinated using live and inactivated camelpox vaccines produced in Kazakhstan. To evaluate the efficacy of these camelpox vaccines in the field, vaccine trials used 172 camels on camel farms in the Beineu district. Of these, 132 camels were vaccinated using a live attenuated camelpox vaccine and 40 camels were vaccinated using an inactivated vaccine to observe immunogenicity and safety. The live vaccine was inoculated into camels by scarification at a dose of 5 × 104 EID50, and the inactivated vaccine was injected intramuscularly at 5 mL twice, with an interval of 35 days. During the safety evaluation, camels administered either vaccine displayed no clinical signs of illness or any adverse effects. Post-vaccination seroconversion demonstrated that the live attenuated vaccine started to elicit antibody responses in some animals as early as day seven, while, by day 28, 99% of vaccinated camels responded. For camels immunized with the inactivated vaccine, seroconversion began on day 21 at low titers ranging from 1:2 to 1:4. Ninety days post vaccination, 77% of the camels demonstrated an immune response that was up to a titer of 1:16. The antibody response waned six months post vaccination in camels vaccinated with two types of vaccine. Nonetheless, both vaccines were 100% effective at preventing clinical disease in vaccinated camels during the camelpox outbreak. All unvaccinated camels became ill, with manifestations of clinical signs characteristic of camelpox. Following these successful field trials in Kazakhstan, a vaccination program for camels, to control camelpox using the domestically produced live attenuated camelpox vaccine, has started.

BACKGROUND: This study was performed to assess the knowledge, attitudes and practices (KAPs) of farmers and veterinary professionals towards foot-and-mouth disease (FMD) in the area studied. METHODS: The study was based on a comprehensive questionnaire administered through face-to-face interviews. Between January and May 2022, 543 households and 27 animal health practitioners (AHP) were visited in 4 provinces of the West Kazakhstan region to assess their KAPs towards FMD. RESULTS: A large proportion of herd owners (84%) had known the name of the disease, and nearly a half (48) of respondents had heard of FMD cases on farms in the neighbourhood. Oral mucosa lesions were the most consistent with clinical sign characteristic of FMD among farmers (31.4%), followed by hoof blisters (27.6%) and excessive salivation (18.6%). Farmers reported that new animal introduction was potentially the main factor associated with FMD occurrence in their herds. Over half of farmers (54%) interviewed prefer not to purchase livestock from unknown or potentially epidemiologically disadvantaged areas. CONCLUSION: All AHPs (27) reported that in their zone of veterinary responsibilities, vaccination against FMD is not practised because the area investigated possesses FMD-free status. However, in the past few years, numerous FMD outbreaks have been detected throughout the region. For this reason, immediate actions need to be taken to prevent further FMD occurrences by giving the region a status of an FMD-free zone with vaccination. The current study demonstrated that inadequate quarantine controls of imported animals, absence of regular vaccination and unrestricted animal movement within the country were the primary obstacles in controlling and preventing FMD in the investigated area.

The aim of this study was to determine the effect of two mineral-vitamin premixes on the health status (blood biochemical parameters), milk yield and composition of Holstein-Friesian (HF) cows in Kazakhstan. The study was performed on Holstein-Friesian cows kept on the Bek Plus dairy farm in the village of Korzhynkol, Fyodorovsky District, Kostanay Region. Forty primiparous cows, selected from the herd, were divided by the analogue method into two groups: a control group (C) and an experimental group (E) of 20 animals each. The diets fed to group E cows were supplemented with LI-R 18 PRO and PANTO® Mineral R-77 Premium mineral-vitamin premixes. The premixes positively affected cow productivity and blood biochemical parameters, whose values were higher in group E than in group C. Higher levels of glucose, albumins and globulins exerted a beneficial influence on the health status of cows. No cases of lameness or metabolic diseases were recorded in group E. The yields of milk, protein and fat were higher in group E than in group C. Dietary supplementation with two premixes had no significant effect on the chemical composition of milk, including the values of the following parameters: somatic cell count (SCC), dry matter (DM), lactose (Lse), casein (Cin), acidity (Aty), lactic acid (LA), density (Dty), free fatty acids (FFAs), glucose (Gse) and urea (Uea).

Lumpy skin disease is an important emerging disease posing a threat to the livestock industry worldwide. Moreover, factors involved in disease transmission in the field and at farm level remain unidentified. This research was based on a cross-sectional study using a questionnaire administered through face-to-face interviews with affected farmers. From January 2021 to July 2021, 543 households were visited in four provinces of the West Kazakhstan region to assess the prevalence of LSD and its associated risk factors. Animal and farm level risk factors were examined using univariable and multivariable mixed effect logistic regression. At animal level, the factors associated with LSD outbreaks include herd size Medium OR = 0.68, (95% CI: 0.54-0.84); large OR = 0.63, (95% CI: 0.49-0.81), purchasing animals OR = 11.67, (95% CI: 8.87-15.35), and selling animals during LSD outbreak OR = 1.24, (95% CI: 1.06-1.45). The overall animal level and herd level LSD prevalence were 10.2% (95% CI: 9.6 -0.10.9) and 49.2% (95% CI: 45.0 - 53.4) respectively. Our study demonstrates the dissemination of LSDV from primary outbreaks to new areas and risk factors associated with LSD in Kazakhstan. This finding will enhance knowledge on disease epidemiology and help develop coordinated actions in prevention and control of the possible LSD outbreaks.

This paper describes a preclinical study analyzing the immunogenicity and protective efficacy of Kazfluvac®, an adjuvant-based inactivated pandemic influenza A/H5N1 virus vaccine. In this study, laboratory animals (ferrets and mice) were vaccinated by the intramuscular or intraperitoneal route at an interval of 14 days with two doses of the vaccine containing different concentrations of influenza virus hemagglutinin (HA) protein. HA protein without adjuvant (aluminum hydroxide and Merthiolate) was used as a control. As a negative control, we utilized PBS. We assessed the protective efficacy of the candidate vaccine by analyzing the response to challenge with the influenza virus strain A/chicken/Astana/6/05 (H5N1). Our experimental results revealed substantially reduced clinical disease and an increased antibody response, as determined by hemagglutination-inhibition (HAI) test and microneutralization assay (MNA). This study showed that the candidate vaccine is safe and elicits an antigen-dose-dependent serum antibody response. In summary, we determined the optimum antigen dose in a Kazfluvac® adjuvant formulation required for induction of heightened immunogenicity and protective efficacy to mitigate H5N1 disease in experimental animals, suggesting its readiness for clinical studies in humans.

BACKGROUND: We developed a new oligonucleotide microarray comprising 16 identical subarrays for simultaneous rapid detection of avian viruses: avian influenza virus (AIV), Newcastle disease virus (NDV), infection bronchitis virus (IBV), and infectious bursal disease virus (IBDV) in single- and mixed-virus infections. The objective of the study was to develop an oligonucleotide microarray for rapid diagnosis of avian diseases that would be used in the course of mass analysis for routine epidemiological surveillance owing to its ability to test one specimen for several infections. METHODS AND RESULTS: The paper describes the technique for rapid and simultaneous diagnosis of avian diseases such as avian influenza, Newcastle disease, infectious bronchitis and infectious bursal disease with use of oligonucleotide microarray, conditions for hybridization of fluorescent-labelled viral cDNA on the microarray and its specificity tested with use of AIV, NDV, IBV, IBDV strains as well as biomaterials from poultry. Sensitivity and specificity of the developed microarray was evaluated with use of 122 specimens of biological material: 44 cloacal swabs from sick birds and 78 tissue specimens from dead wild and domestic birds, as well as with use of 15 AIV, NDV, IBV and IBDV strains, different in their origin, epidemiological and biological characteristics (RIBSP Microbial Collection). This microarray demonstrates high diagnostic sensitivity (99.16% within 95% CI limits 97.36-100%) and specificity (100%). Specificity of the developed technique was confirmed by direct sequencing of NP and M (AIV), VP2 (IBDV), S1 (IBV), NP (NDV) gene fragments. CONCLUSION: Diagnostic effectiveness of the developed DNA microarray is 99.18% and therefore it can be used in mass survey for specific detection of AIV, NDV, IBV and IBDV circulating in the region in the course of epidemiological surveillance. Rather simple method for rapid diagnosis of avian viral diseases that several times shortens duration of assay versus classical diagnostic methods is proposed.
