RESUMO

Sex role differentiation is a widespread phenomenon. Sex pheromones are often associated with sex roles and convey sex-specific information. In Lepidoptera, females release sex pheromones to attract males, which evolve sophisticated olfactory structures to relay pheromone signals. However, in some primitive moths, sex role differentiation becomes diverged. Here, we introduce the chromosome-level genome assembly from ancestral Himalaya ghost moths, revealing a unique olfactory evolution pattern and sex role parity among Lepidoptera. These olfactory structures of the ghost moths are characterized by a dense population of trichoid sensilla, both larger male and female antennal entry parts of brains, compared to the evolutionary later Lepidoptera. Furthermore, a unique tandem of 34 odorant receptor 19 homologs in Thitarodes xiaojinensis (TxiaOr19) has been identified, which presents overlapped motifs with pheromone receptors (PRs). Interestingly, the expanded TxiaOr19 was predicted to have unconventional tuning patterns compared to canonical PRs, with nonsexual dimorphic olfactory neuropils discovered, which contributes to the observed equal sex roles in Thitarodes adults. Additionally, transposable element activity bursts have provided traceable loci landscapes where parallel diversifications occurred between TxiaOr19 and PRs, indicating that the Or19 homolog expansions were diversified to PRs during evolution and thus established the classic sex roles in higher moths. This study elucidates an olfactory prototype of intermediate sex communication from Himalaya ghost moths.

A total of 300 pigs (241 × 600; DNA, Columbus, NE; initially 6.0 ±â€…0.01 kg) were used in a 42-d trial to determine the effects of vitamin E levels and partially replacing vitamin E with a polyphenol (Cabanin CSD, R2 Argo, Denmark) on growth performance, complete blood count, serum thiobarbituric acid reactive substances (TBARS), superoxide dismutase (SOD), and cytokine panel. Sixty pens of pigs were weighed and allotted to one of the five dietary treatments in a completely randomized design with 12 pens per treatment. A control treatment was formulated to provide 15 IU/kg of vitamin E equivalence from vitamin E. This control treatment was then used as a base for three replacement strategy diets to determine the effects of replacing an additional 60 IU/kg of vitamin E with polyphenol in diets containing a basal level of vitamin E requirement estimate (15 IU/kg). First, an additional 60 IU/kg of vitamin E was added for a total of 75 IU/kg of vitamin E equivalence. Second, 50% of the additional vitamin E (30 IU/kg) was replaced with the equivalency of polyphenol. Third, all 60 IU/kg of the additional vitamin E was replaced with the equivalency of polyphenol. To evaluate whether there are negative effects of feeding nursery pigs a high level of polyphenol, a fifth treatment was formulated to provide 575 IU/kg of vitamin E equivalence with 75 IU/kg from vitamin E and 500 IU/kg from polyphenol. Whole blood and serum samples were collected on days 10 and 42, and pig weights and feed disappearance were measured on days 10, 21, 31, 38, and 42. For growth performance, increasing vitamin E equivalence tended to improve (quadratic, P < 0.10) gain-to-feed ratio (G:F) from days 10 to 21, and tended to improve (linear, P < 0.10) G:F from days 21 to 42 and 0 to 42. There was a vitamin E equivalence × day interaction (P = 0.050) for serum SOD activity. Increasing vitamin E equivalence increased (linear, P < 0.05) serum SOD activity on day 42 but not on days 10 (P > 0.10). For serum cytokines, there was no evidence of differences (P > 0.10) between treatments and vitamin E equivalence. Moreover, there was no evidence of differences (P > 0.10) in all response variables between the three replacement strategies throughout the entire periods. In summary, increasing vitamin E equivalence tended to improve G:F, which may be related to the improved SOD activity. Furthermore, polyphenol can effectively replace vitamin E provided above the vitamin E requirement to provide similar benefits from increasing vitamin E equivalence.

A total of 3,888 pigs (337 × 1050, PIC, Hendersonville, TN; initially 6.0 ± 0.23 kg) were used in a 35-d study. At the time of placement, pens of pigs were weighed and allotted to one of three dietary treatments in a randomized complete block design with a blocking structure including sow farm origin, date of entry into the facility, and average pen body weight. A total of 144 pens were used with 72 double-sided 5-hole stainless steel fence line feeders, with one feeder serving as the experimental unit. For each feeder, 1 pen contained 27 gilts, and 1 pen contained 27 barrows. There were 24 replicates per dietary treatment. Diets were fed in three phases, and all contained 0.3 mg/kg added Se. A common phase 1 diet contained added Se from sodium selenite and was fed in pelleted form to all pigs from day 7 to approximately day 0. Three Se sources sodium selenite, Se yeast, and hydroxy-selenomethionine (OH-SeMet) were used to formulate three experimental diets in meal form for phase 2 (days 0 to 14) and phase 3 (days 14 to 35). During the pre-treatment period (days 7 to 0), there was a tendency (P = 0.097) of a difference in average daily feed intake between treatments, although no significant pairwise differences were observed (P > 0.05). There were no other differences in growth performance between treatments from days 7 to 0. Clinical disease attributed to Streptococcus suis was observed within the trial between days 0 and 14, and water-soluble antimicrobial therapy was administered to all treatment groups for 7 d. From days 0 to 35, pigs fed OH-SeMet tended to have decreased average daily gain (P < 0.10) and increased (P < 0.05) serum and tissue selenium concentration compared to other treatments. There was marginally significant evidence of a source × day interaction (P = 0.027) for total antioxidant capacity where the numerical increase over time was less for the OH-SeMet than sodium selenite or selenium yeast treatments. There was no difference (P > 0.05) in antioxidant status as measured by serum glutathione peroxidase or thiobarbituric acid reactive substances assay between treatments. In summary, compared to sodium selenite and selenium yeast, OH-SeMet may have a greater bioavailability as indicated by increased serum and tissue selenium concentration; however, antioxidant status was similar between treatments and OH-SeMet tended to reduce growth performance compared with pigs fed sodium selenite.

A total of 600 sows (line 3; PIC, Hendersonville, TN) were used to evaluate the effect of different lactation feeder types and drip cooling on sow farrowing performance and litter growth performance during the summer. For the feeder evaluation, the trial was conducted in two sequential groups with 300 sows per group. Five 60-farrowing-stall rooms with tunnel ventilation were used for each group. On approximately days 110 to 112 of gestation, sows were blocked by body condition score (BCS), parity, and offspring sire (lines 2 or 3 sires; PIC), then randomly allotted to one of three feeder types: 1) PVC tube feeder, 2) Rotecna feeder (Rotecna), or 3) SowMax feeder (Hog Slat). The three feeder types were placed in one of three stalls with the same sequence from the front to the end of all rooms to balance for environmental effects. For drip cooling evaluation, the trial was conducted during the 2nd group of 300 sows. Drippers were blocked in three of every six farrowing stalls to balance feeder type and environmental effects. After farrowing, sows had ad libitum access to feed. For litter performance data, only pigs from sows bred to line 2 sires were recorded. Line 3 sire pigs were not included in litter performance data, but sows of these pigs were included in sow body weight (BW) and feed disappearance data. After weaning, feeder cleaning time was recorded on a subsample of 67 feeders (19, 23, and 25 for PVC tube, Rotecna, and SowMax, respectively). There was no evidence of difference (P > 0.05) in sow entry BW, exit BW, BW change, and litter performance among the different feeder types. However, sows using the SowMax feeders had decreased (P < 0.05) total feed disappearance, average daily feed disappearance, and total feed cost compared to those fed with the PVC tube feeders. There was a marginal difference (P < 0.10) between feeder types in cleaning time, with PVC tube feeders requiring less time than the Rotecna feeders; however, cleaning time varied greatly between the personnel doing the cleaning. Sows with drip cooling had greater (P < 0.05) feed disappearance, litter growth performance, and subsequent total born, and reduced (P < 0.05) BW change. In conclusion, using a SowMax feeder reduced feed disappearance with no effects on sow and litter performance compared to a PVC tube feeder, and drip cooling improved sow and litter performance during summer.

Feed additives have shown benefits throughout the literature in improving grow−finish pigs' growth performance and carcass characteristics. However, the results have not been well summarized. Therefore, this review summarizes the available research (402 articles) on 14 feed additive categories fed to grow−finish pigs. The categories were acidifiers, betaine, Cr, conjugated linoleic acids, Cu, direct-fed microbials, carbohydrases, proteases, phytases, multi-enzymes, essential oils, L-carnitine, yeasts, and Zn. Qualified articles were collected and selected based on inclusion and exclusion criteria from online databases. The percentage difference for each response variable between the treatment and control group was calculated and summarized. Most results were positive for each feed additive; however, the magnitude of improvement varied, and most were not statistically significant. For ADG, DFM, Cu, L-carnitine, and multi-enzymes showed relatively large positive effects (>2.1% improvement) across a reasonable number of articles. Acidifiers, betaine, CLA, multi-enzymes, DFM, L-carnitine, and yeasts showed relatively large positive effects (>2.5% improvement) on improving G:F. Moreover, except for betaine, Cr, CLA, and L-carnitine, most feed additives showed little and non-significant effects on BF thickness (<1.7% improvement). This review provides a descriptive analysis for commonly used feed additives in the hope of better understanding feed additives' effects on grow−finish pigs.

Two 44-d experiments were conducted to evaluate nutritional strategies with different concentrations of dietary lysine (and other amino acids) on growth rate and subsequent compensatory gain of 90-kg finishing pigs. Three diets were formulated to contain 0.70 (control), 0.50% and 0.18% standardized ileal digestible (SID) Lys. In Exp. 1, 356 pigs (Line 241 × 600, DNA; initially 89.0 ± 1.10 kg) were used with four treatments. From d 0 to 28, pigs received either the control or the 0.50%-Lys diet. On d 28, pigs either remained on these diets or were switched the 0.18%-Lys diet until d 44. There were 18 pens per treatment from d 0 to 28 and 9 pens per treatment from d 28 to 44. From d 0 to 28, pigs fed the 0.50%-Lys diet had decreased (P < 0.001) ADG and G:F compared to those fed the control diet. From d 28 to 44, pigs switched to the 0.18%-Lys diet had decreased (P < 0.05) ADG and G:F compared to pigs that remained on the control or 0.50%-Lys diets. From d 0 to 44, pigs fed 0.50%-Lys diet for 44-d had decreased (P < 0.05) ADG, G:F, and percentage carcass lean compared to pigs fed the control diet. Pigs fed the 0.50%-Lys diet then the 0.18%-Lys diet had decreased (P < 0.05) ADG and G:F compared to other treatments. Pigs fed the 0.50%-Lys diet for 44-d and pigs fed the control diet then 0.18%-Lys diet had decreased (P < 0.05) ADG, G:F, and percentage carcass lean compared to control pigs. In Exp. 2, 346 pigs (Line 241 × 600, DNA; initially 88.6 ± 1.05 kg) were used to evaluate compensatory growth after varying durations of dietary lysine restriction. A total of four treatments were used including pigs fed the control diet for 44-d or fed the 0.18%-Lys diet for 14, 21, or 28-d and then fed the control diet until the conclusion of the experiment on d 44. There were nine pens per treatment. On average, pigs fed the 0.18%-Lys diet grew 49% slower than the control. Compared to the control, ADG of pigs previously fed the 0.18%-Lys diet increased (P < 0.05) 28% during the first week after switching to the control diet and 12% for the rest of the trial. Despite this improvement, overall ADG, G;F, final BW, and percentage carcass lean decreased (linear, P < 0.05) as the duration of Lys restriction increased. In summary, feeding Lys-restricted diets reduced the ADG and G:F of finishing pigs. Compensatory growth can be induced in Lys-restricted finishing pigs, but the duration of restriction and recovery influences the magnitude of compensatory growth.

A total of 1,890 growing-finishing pigs (PIC; 359 × 1,050; initially 27.1 kg) were used in a 124-d growth trial to compare the effects of high-protein distillers dried grains (HPDDG; 39% crude protein [CP]) or conventional distillers dried grains with solubles (DDGS; 29% CP) on growth performance and carcass characteristics. Treatments were arranged in a 2 × 2 + 1 factorial with main effects of distillers dried grains source (conventional DDGS or HPDDG) and level (15% or 30%). A corn-soybean meal-based diet served as the control and allowed linear and quadratic level effects to be determined within each distillers dried grains (DDG) source. All diets were formulated on an equal standardized ileal digestible (SID) Lys-basis with diets containing HPDDG having less soybean meal than diets with conventional DDGS. Pens were assigned to treatments in a randomized complete block design with initial weight as the blocking factor. There were 27 pigs per pen and 14 pens per treatment. Overall, increasing conventional DDGS decreased (linear, P < 0.04) final body weight (BW), whereas increasing HPDDG tended to decrease (linear, P = 0.065) final BW. The decreased final BW was a result of decreased (linear, P < 0.01) ADG in the grower phase of the study as either DDG source increased. However, there were no differences observed in the finisher phase or overall ADG between pigs fed either DDG source or either inclusion level. Pigs fed HPDDG had decreased (P < 0.001) ADFI and increased (P < 0.001) G:F compared with those fed conventional DDGS. For carcass traits, increasing either conventional DDGS or HPDDG decreased carcass yield and HCW (linear, P < 0.02); however, there were no differences between pigs fed HPDDG or conventional DDGS. Iodine value (IV) increased (linear, P < 0.02) with increasing DDG and was greater (P < 0.001) in pigs fed HPDDG than conventional DDGS. In summary, pigs fed HPDDG had no evidence of difference in overall ADG compared to pigs fed conventional DDGS, but had greater overall G:F. Carcass fat IV was also greater in pigs fed HPDDG compared with pigs fed conventional DDGS. These differences were probably due to the difference in oil content.

The Chinese cordyceps is a unique and valuable parasitic complex of Thitarodes/Hepialus ghost moths and the Ophiocordyceps sinensis fungus for medicine and health foods from the Tibetan Plateau. During artificial cultivation of Chinese cordyceps, the induction of blastospores into hyphae is a prerequisite for mummification of the infected Thitarodes larvae. To explore the microbial involvement in the induction of mycelia-blastospore transition, the microbiota of the hemolymph and gut from Thitarodes xiaojinensis larvae with or without injected O. sinensis blastospores were investigated by culture-dependent and -independent methods. Twenty-five culturable bacterial species and 14 fungal species, together with 537 bacterial operational taxonomic units (OTUs) and 218 fungal OTUs, were identified from the hemolymph and gut of samples from five stages including living larvae without injected fungi (A) or with high blastospore load (B), mummifying larvae without mycelia coating (C), freshly mummifying larvae coated with mycelia (D), and completely mummified larvae with mycelia (E). Two culturable bacterial species (Serratia plymuthica, Serratia proteamaculans), and 47 bacterial and 15 fungal OTUs were considered as shared species. The uninfected larval hemolymph contained 13 culturable bacterial species but no fungal species, together with 164 bacterial and 73 fungal OTUs. To our knowledge, this is the first study to detect large bacterial communities from the hemolymph of healthy insect larvae. When the living larvae contained high blastospore load, the culturable bacterial community was sharply inhibited in the hemolymph but the bacterial and fungal community greatly increased in the gut. In general, high blastospore load increased bacterial diversity but sharply decreased fungal diversity in the hemolymph and gut by OTUs. The bacterial loads of four culturable species (Chryseobacterium sp., Pseudomonas fragi, S. plymuthica, S. proteamaculans) increased significantly and O. sinensis and Pseudomonas spp. became dominant microbes, when the infected larvae became mummified, indicating their possible involvement in the larval mummification process. The discovery of many opportunistic pathogenic bacteria in the hemolymph of the healthy larvae, the larval microbial diversity influenced by O. sinensis challenge and the involvement of dominant bacteria during larval mummification process provide new insight into the infection and mummification mechanisms of O. sinensis in its Thitarodes hosts.

Fumonisin contamination in corn is an emerging issue in animal feed production. Fumonisin disrupts the metabolism of sphingolipids and reduces growth performance. This experiment was conducted to determine the effect of feeding fumonisin-contaminated corn on growth performance and sphinganine (SA) to sphingosine (SO) ratios of 9 to 28 kg pigs. A total of 350 pigs, were used with 5 pigs/pen and 14 pens/treatment. Dietary treatments contained fumonisin-contaminated corn (50 mg/kg of fumonisin B1 + B2) blended with low fumonisin corn (10 mg/kg of fumonisin B1 + B2) to provide dietary fumonisin concentrations of 7.2, 14.7, 21.9, 32.7, and 35.1 mg/kg. From day 0 to 28, increasing fumonisin concentration decreased (linear, p < 0.001) average daily gain, average daily feed intake (linear, p = 0.055), and gain:feed ratio (linear, p = 0.016). Although these response criteria tested linear, the greatest reduction in performance was in pigs fed with 32.7 and 35.1 mg/kg of fumonisin (B1 + B2). Increasing fumonisin concentration increased the serum SA:SO ratio (linear, p < 0.001) on day 14 and 28. In summary, for 9 to 28 kg nursery pigs, increasing fumonisin linearly decreased average daily gain and gain:feed ratio. However, despite the linear response, diets containing up to 21.9 mg/kg of fumonisin did not have as dramatic a decrease in growth performance as those fed more than 32.7 mg/kg. Further research is warranted to determine the effect of fumonisin concentrations between 21.9 and 32.7 mg/kg.

Two experiments were conducted to determine the efficacy of various commercial products on growth performance of nursery pigs fed diets high in fumonisin. In experiment 1, 350 pigs (241 × 600; DNA, Columbus, NE; initially 9.9 kg) were used with five pigs per pen and 14 replicates per treatment. After weaning, pigs were fed common diets for 21 d before the experiment started. The five dietary treatments consisted of a positive control (low fumonisin), a negative control (60 mg/kg of fumonisin B1 + B2 in complete diet), and the negative control with one of three products (0.3% of Kallsil Dry, Kemin Industries Inc., Des Moines, IA; 0.3% of Feed Aid Wide Spectrum, NutriQuest, Mason City, IA; 0.17% of Biofix Select Pro, Biomin America Inc., Overland Park, KS). Diets were fed in mash form for 14 d and followed with a low fumonisin diet for 13 d. For the 14-d treatment period, pigs fed the positive control diet and Biofix Select Pro had greater (P < 0.05) average daily gain (ADG), average daily feed intake (ADFI), and gain:feed (G:F) compared to those fed the high fumonisin negative control, or high fumonisin diets with Kallsil Dry or Feed Aid Wide Spectrum. Serum sphinganine to sphingosine ratios (SA:SO) were greater (P < 0.05) in all pigs fed high fumonisin diets compared to the positive control. In experiment 2, 300 pigs (241 × 600; DNA; initially 10.4 kg) were used. Procedures were similar to experiment 1 except there were 12 replicate pens per treatment, high fumonisin diets contained 30 mg/kg fumonisin, and experimental diets were fed for 28 d. Similar to experiment 1, pigs fed the positive control diet and treatment with Biofix Select Pro had greater (P < 0.05) ADG and G:F, and lower (P < 0.05) serum SA:SO compared to pigs fed the high fumonisin negative control, or high fumonisin diets with Kallsil Dry or Feed Aid Wide Spectrum. In summary, pigs fed diets containing 60 mg/kg of fumonisin for 14 d or 30 mg/kg of fumonisin for 28 d had poorer ADG and G:F and greater serum SA:SO compared to pigs fed a diet with less than 5 mg/kg of fumonisin. Adding Biofix Select Pro to diets appeared to mitigate the negative effects of high fumonisin concentrations, while Kallsil Dry and Feed Aid Wide Spectrum did not.

The research was conducted to investigate the accumulation, distribution and availability of Cd in paddy soil and their relation to Cd in rice plants under 30-year fertilization regimes. Six treatments were involved in the study: control without fertilization (CK), chemical fertilizer (NPK), high nitrogen chemical fertilizer (HN), rice straw incorporation (ST), low and high dosage of manure fertilizer (LM and HM). Total and DTPA extractable concentration of Cd (T-Cd and DTPA-Cd) in bulk soils (20 cm topsoil), profiles (0-60 cm) and aggregates (>2, 1-2, 0.5-1, 0.25-0.5, 0.053-0.25 and < 0.053 mm) were investigated. The Cd concentration in relevant rice plant (roots, stems, leaves, husks and grains) were also analyzed. Manure fertilizers caused T-Cd accumulation in bulk soil with a significant increase of 36.2% in LM and 81.2% in HM. Similar impacts of manure fertilizers were observed in DTPA-Cd in the bulk soil. Further, the HM generated a further accumulation in deeper soil layers, presenting a remarkable increase of T-Cd (28.3%-225%) in 10-40 cm and DTPA-Cd (116%-158%) in 10-30 cm profiles. Moreover, the continuous application of manure fertilizers enhanced the availability of Cd in all aggregate size classes with an increase of 17.3%-87.8% in DTPA-Cd. Organic fertilizers (LM, HM and ST) heightened the content of Cd (38.0%-152%) in all parts of rice plant. The accumulation of Cd in rice plants was directly affected by fertilization regimes and Cd availability in the 10-20 cm soil layers and 0.25-0.5 mm aggregates. In conclusion, long-term application of manures resulted in increasing availability of Cd in aggregates and in topsoil and subsoil layers, which accordingly enhanced the accumulation of Cd in rice plants.

A biofilm photobioreactor under unsaturated flow condition (BFPBR-U) is proposed using a polished optical fiber as the internal light source for photo-H2 production in continuous culture. The main chamber was filled with spherical glass beads to create the reaction bed and the cells were immobilized to form a biofilm under unsaturated flow condition obtained by pumping substrate solution over a packing bed at a rate to create a thin fluid film and injecting the argon to maintain the gas phase space. The effects of operational conditions, including flow rate and influent substrate concentration, on the photo-H2 production performance were investigated. The unsaturated flow conditions eliminated the inhibition caused by high organic loading rate and enhanced light transmission efficiency, leading to an improvement in the photo-H2 production performance.

When grown on Cd-contaminated soil, rice typically accumulates considerable Cd in straw, and which may return to the soil after harvest. This work was undertaken to assess the pollution risk of Cd associated to the Cd-contaminated rice straw after incorporating into an uncontaminated soil. With the Cd-contaminated rice straw added at 0, 1, 2, 3, 4 and 5 % (w/w), an incubation experiment (28 days) with non-planting and a followed pot experiment sequent with two planting (rice and Chinese cabbage, transplanted after 28-day incubation) were carried out to investigate the changes of soil Cd speciation and phytoavailability. The results indicated that the Cd-contaminated rice straw addition significantly increased soil pH and dissolved organic carbon during the 28-day incubation. For the high availability of Cd in contaminated rice straw, diethylenetriaminepentaacetic acid (DTPA) extractable Cd significantly increased, and the percentages of acetic acid extractable and reducible Cd in soil significantly enhanced after the addition of Cd-contaminated rice straw. However, the Cd-contaminated rice straw addition inhibited the rice growth and induced the decrease of Cd in rice grain and straw by 12.8 to 70.2 % and 39.3 to 57.3 %, respectively, whereas the Cd contents increased by 13.9 to 84.1 % in Chinese cabbage that planted after rice harvest. In conclusion, Cd associated with Cd-contaminated rice straw was highly available after incorporating into the soil, and thus the Cd pollution risk via the Cd-contaminated rice straw incorporation should be evaluated in the Cd-contaminated paddy region.

BACKGROUND: Out of total 3,081 assembled expressed sequence tags (ESTs) sequences representing 6,815 high-quality ESTs identified in three cDNA libraries constructed with RNA isolated from the midgut of Spodoptera litura, 1,039 ESTs showed significant hits and 1,107 ESTs did not show significant hits in BLAST searches. It is of interest to clarify whether or not these ESTs that did not show hits function in S. Litura. RESULTS: Twenty "no-hit" ESTs containing at least one putative open reading frame were selected for further expression analysis. The results from northern blot analysis showed that six of the selected ESTs are expressed in the larval midgut of this insect at different levels, suggesting that these ESTs represent true mRNA products, whereas the other 14 ESTs could not be detected. Homologues of the four larval midgut-predominant genes (Slmg2, Slmg7, Slmg9 and Slmg17) were detected in the genomes of other lepidopteran insects but not in Drosophila melanogaster. A novel gene, Slmg7, is expressed at a high level specifically in the midgut during each of the larval stages. Slmg7 is a single copy gene and encodes a 143-amino acids protein. The SLMG7 protein was localized to the cytoplasm of Spli-221 cells. CONCLUSIONS: Six ESTs from the no hit list are transcribed into mRNA and are mainly expressed in the midgut of S. litura. Slmg7 is a novel gene that is localized to the cytoplasm.

RESUMO

Aiming at raising awareness of preventing adverse effects of acupuncture in clinical practices, such cases were studied from the angle of therapy methods, the factors of patients constitution and practitioners. The findings showed that adverse physical and chemical effects of acupuncture increased in modern times because of multifactorial reasons, for instance, the introduction of modern medicine, the differences of individual constitution and the practitioners. This suggests that to decrease adverse cases acupuncture practitioners should be an expert not only in traditional Chinese medicine, but also in western medicine, such as anatomy, and pharmacology.

BACKGROUND: Due to the quick rhythm of life and work pressure, more and more people suffer from sleep quality problems. In this study, we investigated the effect of electroacupuncture on sleep quality of chronic insomniacs and the safety of electroacupuncture therapy. METHODS: Four courses of electroacupuncture treatment were applied to 47 patients. With pre-treatment and post-treatment self-control statistical method, Pittsburgh sleep quality index (PSQI) scores were used for evaluating sleep quality. Polysomnogram was used for detecting insomniacs' changes in sleep architecture. The safety of electroacupuncture was evaluated by monitoring the self-designed adverse events and side effects during treatment and post-treatment. RESULTS: Electroacupuncture considerably improved insomniacs' sleep quality and social function during the daytime. Electroacupuncture had certain repairing effect on the disruption in sleep architecture. At the same time, electroacupuncture prolonged slow wave sleep (SWS) time and relatively rapid eye movement sleep (REM sleep) time. There was no hangover, addiction or decrements in vigilance during the daytime (incidence rate was 0). However, insomnia rebound rate was about 23% within one month. CONCLUSIONS: These results suggest that electroacupuncture has beneficial effect on sleep quality improvement in the patients with chronic insomnia, which may be associated with repairing sleep architecture, reconstructing sleep continuity, as well as prolonging SWS time and REM sleep time. Electroacupuncture treatment for chronic insomnia is safe. Therefore, electroacupuncture therapy could be a promising avenue of treatment for chronic insomnia.

A nucleotides fragment 2020VP1 related to T-cell epitope (aa21-40) and B-cell epitope (aa141-160) of VP1 of Food-and-mouth disease virus (FMDV)type O was synthesized, and the recombined expression vector r2020-LTB-2020-STI was correctly constructed, which was used to express the fusion protein consisted of two copies of aa21-40aa141-160 of FMDV VP1 and LTB and STI Enterotoxins of Escherichia coli. The fusion protein with molecular weight of about 45kD was successfully expressed in E.coli BL21(DE3) RIL at high level and confirmed by SDS-PAGE. The purified fusion protein could be specifically recognized by CTB antibody. The purified protein was then used to vaccinate guinea pigs, rabbits and Balb/c mice at 6-8 week age, and immune responses were finally observed. The fusion protein could induce proliferation of spleen T cells in vaccinated guinea pigs and elicit a high level of neutralizing antibody in rabbits. It could be concluded that the fusion protein could activate FMDV-specific cellular immune-response and humoral immune-response simultaneously. At the same time, the vaccinated mice could survive challenge of Enterotoxigenic Escherichia coli C83902, and the sera of vaccinated rabbits could neutralize the STI toxin. Moreover, the fusion protein had no STI toxicity, indicating that the fusion protein had LTB and STI immunogenicities. Therefore, it can be concluded that the fusion protein could be explored as a potentially efficient FMDV and ETEC vaccine.

OBJECTIVE: To probe factors of influencing therapeutic effects of acupuncture in the patient of insomnia. METHODS: According to scores of degrees of anxiety and depression, 52 cases of insomnia were divided into 3 groups, group I (mild or less degree) and group II (moderate degree) and group II (serious degree). The Pittsburgh sleep quality index (PSQI) were compared before and after treatment in the 3 groups, and between two groups after treatment. Results There were significant differences in the therapeutic effect as the groups I, II compared with the group III (P < 0.01). The total sleep quality in the group I was better than that in the group II (P < 0.05). CONCLUSION: The degree of anxiety and depression in the patient of insomnia is one of important factors influencing therapeutic effect of acupuncture on insomnia.
