Introduction: Genetic engineering has revolutionized agriculture by transforming biotic and abiotic stress-resistance genes in plants. The biosafety of GM crops is a major concern for consumers and regulatory authorities. Methodology: A 14-week biosafety and toxicity analysis of transgenic cotton, containing 5 transgenes ((Cry1Ac, Cry2A, CP4 EPSPS, VIP3Aa, and ASAL)), was conducted on albino mice. Thirty mice were divided into three groups (Conventional, Non-transgenic, without Bt, and transgenic, containing targeted crop) according to the feed given, with 10 mice in each group, with 5 male and 5 female mice in each group. Results: During the study, no biologically significant changes were observed in the non-transgenic and transgenic groups compared to the control group in any of the study's parameters i.e. increase in weight of mice, physiological, pathological, and molecular analysis, irrespective of the gender of the mice. However, a statistically significant change was observed in the hematological parameters of the male mice, while no such change was observed in the female study group mice. The expression analysis, however, of the TNF gene increases many folds in the transgenic group as compared to the non-transgenic and conventional groups. Conclusion: Overall, no physiological, pathological, or molecular toxicity was observed in the mice fed with transgenic feed. Therefore, it can be speculated that the targeted transgenic crop is biologically safe. However, more study is required to confirm the biosafety of the product on the animal by expression profiling.

In order to investigate the effect of pre-loading damage on the structural performance of Carbon Fiber Reinforced Polymer (CFRP) strengthened Reinforced Concrete (R.C.) beams, experimental and Finite Element Modelling (FEM) investigation was carried out on six R.C. beams. Five of the R.C. beams were damaged up to different levels of strain in the main steel bars before Flexure CFRP strengthening. One of the R.C. beams loaded up to failure and was kept as a control beam for comparison. The experimental results showed that the failure mode of the CFRP strengthened specimen was controlled by CFRP debonding followed by concrete crushing; however, the control beam failed in concrete crushing after yielding the steel bars, which is a ductile failure. The CFRP sheet increases the strength and initial stiffness of the R.C. beams and reduces ductility and toughness. Also, CFRP application increases the first crack and yielding steel bars load by 87.4% and 34.4%, respectively. Furthermore, the pre-damage level does not influence the strength and ductility of the strengthened R.C. beams except for the highest damage levels, which experienced a slight decrease in load capacity and ductility. However, the initial stiffness decreases with increasing pre-damage levels by 40%. Design guideline ACI 440.2R (2004) predicts the ultimate load capacity marvelously for externally bonded Fiber-Reinforced Polymer (FRP) beams compared to the experimental maximum load capacity. The excellent agreement between experimental and FEM results indicates that the constitutive models used for concrete and reinforcement and the cohesive interface model can well capture fracture behavior. However, The FEM analysis predicts the beam to be slightly stiffer and more robust, probably because of the assumed perfect bond between concrete and reinforcement. The developed FEM can be used for further parametric study.

OBJECTIVE: To investigate the association of gold standard liver biomarkers with serum cytokeratin 18 (CK18), serum Alanine aminotransferase (ALT) and serum aspartate (AST). METHODS: This was cross sectional study. It was conducted at Mayo Hospital from January 2016 to December 2017. It comprised of 148 non-alcoholic fatty liver disease subjects of age 40-60 years. After written informed consent, study anthropometric measurements (age, height, waist circumference and hip circumference) were taken and serum AST, ALT and CK-18 were estimated by sandwiched ELISA technique. Data was analyzed using SPSS 21.0. Descriptive were presented as mean and standard deviation. Association between CK18, serum AST and ALT were analyzed by regression analysis and are presented as beta coefficient. P-value ≤ 0.05 was taken as significant. RESULTS: Study comprised of 148 subjects with mean age 44.81±6.2. Of total population 29.1% were male and 70.9% were female. Significant positive association of CK18 was found with serum ALT (P-value 0.005*). However, no association was found between AST and serum CK18. (P-value 0.29). CONCLUSION: Significant positive association was found between Serum CK18 and serum ALT.

BACKGROUND: In Pakistan thyroid cancer is responsible for 1.2% cases of all malignant tumors. Ultrasonography (US) is helpful in detecting cancerous thyroid nodules on basis of different features like echogenicity, margins, microcalcifications, size, shape and abnormal neck lymph nodes. We therefore aimed to calculate diagnostic accuracy of ultrasound in detection of carcinoma in thyroid nodules taking fine needle aspiration cytology as the reference standard. MATERIALS AND METHODS: A cross-sectional analytical study was designed to prospectively collect data from December 2010 till December 2012 from the Department of Radiology in Aga Khan University Hospital, Karachi, Pakistan. A total of 100 patients of both genders were enrolled after informed consent via applying non-probability consecutive sampling technique. Patients referred to Radiology department of Aga Khan University to perform thyroid ultrasound followed by fine-needle aspiration cytology of thyroid nodules were included. They were excluded if proven for thyroid malignancy or if their US or FNAC was conducted outside our institution. RESULTS: The subjects comprised 76 (76%) females and 24 males. Mean age was 41.8±SD 12.3 years. Sensitivity and specificity with 95%CI of ultrasound in differentiating malignant thyroid nodule from benign thyroid nodule calculated to be 91.7% (95%CI, 0.72-0.98) and 78.94% (0.68-0.87) respectively. Reported positive predictive value and negative PV were 57.9% (0.41-0.73) and 96.8% (0.88-0.99) and overall accuracy was 82%. Likelihood ratio (LR) positive was computed to be 4.3 and LR negative was 0.1. CONCLUSIONS: Ultrasonography has a high diagnostic accuracy in detecting malignancy in thyroid nodules on the basis of features like echogenicity, margins, micro calcifications and shape.
