It is proposed that prebiotic diet supplements improve intestinal function, in part by improving the barrier function of the intestinal epithelium with an associated increase in the expression of tight junction proteins, including occludin and zonula occludens-1 (ZO-1). We examined the expression of these proteins in two strains of laying hens (Lohman LSL-lite (White) and Lohman Brown-lite (Brown)) who were supplemented or not with 3% Chondrus crispus or 0.5% Ascophyllum nodosum seaweeds from 31 to 72 weeks of age. Occludin was localized to the lateral surfaces and across the intestinal epithelium in all animals. Reactivity for ZO-1 was concentrated at the apicolateral epithelial cell membrane border. Mood's median test indicated that White hens may express more occludin in villus epithelium (median intensity 3.5 vs. 2.5 in Brown hens, p = 0.06) but less ZO-1 in the deep cryptal epithelium (median intensity 1.5 vs. 2.5 in Brown hens, p = 0.06). Western blotting also showed higher levels of occludin in White than Brown hens (p < 0.05). A decrease in ZO-1 Western blot expression was associated with Chondrus crispus supplementation in comparison to controls (p < 0.05), but not with Ascophyllum nodosum supplementation (p > 0.05). In conclusion, genetic strain and dietary seaweed supplements affect tight junction regulatory protein expression levels but do not impact the anatomical distribution, as seen in cryosections.

The genome-wide analysis of runs of homozygosity (ROH) islands can be an effective strategy for identifying shared variants within a population and uncovering important genomic regions related to complex traits. The current study performed ROH analysis to characterize the genome-wide patterns of homozygosity, identify ROH islands and annotated genes within these candidate regions using whole-genome sequencing data from 100 American mink (Neogale vison). After sequence processing, variants were called using GATK and Samtools pipelines. Subsequent to quality control, 8,373,854 bi-allelic variants identified by both pipelines remained for further analysis. A total of 34,652 ROH segments were identified in all individuals, among which shorter segments (0.3-1 Mb) were abundant throughout the genome, approximately accounting for 84.39% of all ROH. Within these segments, we identified 63 ROH islands housing 156 annotated genes. The genes located in ROH islands were associated with fur quality (EDNRA, FGF2, FOXA2 and SLC24A4), body size/weight (MYLK4, PRIM2, FABP2, EYS and PHF3), immune capacity (IL2, IL21, PTP4A1, SEMA4C, JAK2, CCNA2 and TNIP3) and reproduction (ADAD1, KHDRBS2, INSL6, PGRMC2 and HSPA4L). Furthermore, Gene Ontology and KEGG pathway enrichment analyses revealed 56 and 9 significant terms (FDR-corrected p-value < 0.05), respectively, among which cGMP-PKG signalling pathway, regulation of actin cytoskeleton, and calcium signalling pathway were highlighted due to their functional roles in growth and fur characteristics. This is the first study to present ROH islands in American mink. The candidate genes from ROH islands and functional enrichment analysis suggest possible signatures of selection in response to the mink breeding targets, such as increased body length, reproductive performance and fur quality. These findings contribute to our understanding of genetic characteristics, and provide complementary information to assist with implementation of breeding strategies for genetic improvement in American mink.

Copy number variations (CNVs) are structural variants consisting of duplications and deletions of DNA segments, which are known to play important roles in the genetics of complex traits in livestock species. However, CNV-based genome-wide association studies (GWAS) have remained unexplored in American mink. Therefore, the purpose of the current study was to investigate the association between CNVs and complex traits in American mink. A CNV-based GWAS was performed with the ParseCNV2 software program using deregressed estimated breeding values of 27 traits as pseudophenotypes, categorized into traits of growth and feed efficiency, reproduction, pelt quality, and Aleutian disease tests. The study identified a total of 10,137 CNVs (6968 duplications and 3169 deletions) using the Affymetrix Mink 70K single nucleotide polymorphism (SNP) array in 2986 American mink. The association analyses identified 250 CNV regions (CNVRs) associated with at least one of the studied traits. These CNVRs overlapped with a total of 320 potential candidate genes, and among them, several genes have been known to be related to the traits such as ARID1B, APPL1, TOX, and GPC5 (growth and feed efficiency traits); GRM1, RNASE10, WNT3, WNT3A, and WNT9B (reproduction traits); MYO10, and LIMS1 (pelt quality traits); and IFNGR2, APEX1, UBE3A, and STX11 (Aleutian disease tests). Overall, the results of the study provide potential candidate genes that may regulate economically important traits and therefore may be used as genetic markers in mink genomic breeding programs.

Provision of light during incubation has shown the potential to enhance hatching traits and affect posthatch productivity, physiology, and behavior. In this study, 2 repeated trials were conducted to investigate the effect of photoperiod and strain on the embryo temperature, hatching traits and posthatch growth performance of 2 commercial strains of broilers (Ross 308 and Cobb 500). In each trial, hatching eggs were randomly distributed into 6 incubators with 3 photoperiod treatments: blue LED light for 12 h d-1 (12L:12D) or 18 h d-1 (18L:6D) during entire incubation were compared with no illumination condition (DARK). Data were analyzed as a 3 × 2 factorial arrangement with the trial as the blocking factor. Embryos incubated under 12L:12D and 18L:6D had lower air cell temperature (P < 0.05) than the DARK embryos from d 13 of incubation onward except on the day of candling. The response of air cell temperature to periodic illumination differed between 2 strains. Cobb embryos had lower air cell temperature in 12L:12D than those incubated with 18L:6D from d 16 of incubation onward, whereas lower air cell temperature was found in Ross embryos when illuminated with 18L:6D photoperiod compared to those under 12L:12D. The 12L:12D treatment was associated with improved (P < 0.05) navel closure condition of hatchlings. There were no differences in hatchability, embryo mortality, body weight, or length at hatch among photoperiod groups or its combination with strain. No differences in production parameters were found between DARK and illuminated groups. However, 12L:12D had heavier (P < 0.05) body weight on d 14 of age and higher (P < 0.05) body weight gain than 18L:6D from d 7 to 14 of age. The results of this study indicate that providing blue LED light up to 18 h d-1 has no detrimental effect on production of broilers, however, 12L:12D light regime improved chick quality at hatch compared to DARK and resulted in heavier birds by d 14 compared to 18L:6D.

BACKGROUND: Copy number variations (CNVs) represent a major source of genetic diversity and contribute to the phenotypic variation of economically important traits in livestock species. In this study, we report the first genome-wide CNV analysis of American mink using whole-genome sequence data from 100 individuals. The analyses were performed by three complementary software programs including CNVpytor, DELLY and Manta. RESULTS: A total of 164,733 CNVs (144,517 deletions and 20,216 duplications) were identified representing 5378 CNV regions (CNVR) after merging overlapping CNVs, covering 47.3 Mb (1.9%) of the mink autosomal genome. Gene Ontology and KEGG pathway enrichment analyses of 1391 genes that overlapped CNVR revealed potential role of CNVs in a wide range of biological, molecular and cellular functions, e.g., pathways related to growth (regulation of actin cytoskeleton, and cAMP signaling pathways), behavior (axon guidance, circadian entrainment, and glutamatergic synapse), lipid metabolism (phospholipid binding, sphingolipid metabolism and regulation of lipolysis in adipocytes), and immune response (Wnt signaling, Fc receptor signaling, and GTPase regulator activity pathways). Furthermore, several CNVR-harbored genes associated with fur characteristics and development (MYO5A, RAB27B, FGF12, SLC7A11, EXOC2), and immune system processes (SWAP70, FYN, ORAI1, TRPM2, and FOXO3). CONCLUSIONS: This study presents the first genome-wide CNV map of American mink. We identified 5378 CNVR in the mink genome and investigated genes that overlapped with CNVR. The results suggest potential links with mink behaviour as well as their possible impact on fur quality and immune response. Overall, the results provide new resources for mink genome analysis, serving as a guideline for future investigations in which genomic structural variations are present.

Feed cost is the largest expense of mink production systems, and, therefore, improvement of feed efficiency (FE) through selection for high feed-efficient mink is a practical way to increase the mink industry's sustainability. In this study, we estimated the heritability, phenotypic, and genetic correlations for different FE measures and component traits, including harvest weight (HW), harvest length (HL), final body length (FBL), final body weight (FBW), average daily gain (ADG), daily feed intake (DFI), feed conversion ratio (FCR), residual feed intake (RFI), residual gain (RG), residual intake and gain (RIG), and Kleiber ratio (KR), using data from 2,288 American mink (for HW and HL), and 1,038 to 1,906 American mink (for other traits). Significance (P < 0.05) of fixed effects (farm, sex, and color type), a covariate (age of animal), and random effects (additive genetic, maternal, and common litter) were evaluated through univariate models implemented in ASReml-R version 4. Genetic parameters were estimated via fitting a set of bivariate models using ASReml-R version 4. Estimates of heritabilities (±SE) were 0.28 ± 0.06, 0.23 ± 0.06, 0.28 ± 0.10, 0.27 ± 0.11, 0.25 ± 0.09, 0.26 ± 0.09, 0.20 ± 0.09, 0.23 ± 0.09, 0.21 ± 0.10, 0.25 ± 0.10, and 0.26 ± 0.10 for HW, HL, FBL, FBW, ADG, DFI, FCR, RFI, RG, RIG, and KR, respectively. RIG had favorable genetic correlations with DFI (-0.62 ± 0.24) and ADG (0.58 ± 0.21), and nonsignificant (P > 0.05) genetic correlations with FBW (0.14 ± 0.31) and FBL (-0.15 ± 0.31). These results revealed that RIG might be a superior trait as it guarantees reduced feed intake with faster-growing mink yet with no negative impacts on body weight and length. In addition, the strong positive genetic correlations (±SE) between KR with component traits (0.88 ± 0.11 with FBW, 0.68 ± 0.17 with FBL, and 0.97 ± 0.02 with ADG) suggested KR as an applicable indirect measure of FE for improvement of component traits as it did not require the individual feed intake to be measured. Overall, our results confirmed the possibility of including FE traits in mink breeding programs to effectively select feed-efficient animals.


Improvement of feed efficiency (FE) in American mink is highly beneficial, as feed costs comprise the largest expense of mink production systems. The present study estimated the heritability, phenotypic and genetic correlations for different FE measures and component traits in mink. The residual intake and gain can be applied as FE measurement in selection programs as it will guarantee faster-growing mink with reduced feed intake, yet without negative impacts on growth traits. In addition, Kleiber ratio had strong positive genetic correlations with component traits, which made this trait an appealing indirect FE trait for mink breeding programs, knowing the fact that this trait was not dependent on feed intake records. Overall, our results suggested that including FE traits can assist mink breeding programs to develop an index for the selection of feed-efficient mink and, therefore, reduce the cost of mink production.

RESUMO

Despite the significant improvement of feed efficiency (FE) in pigs over the past decades, feed costs remain a major challenge for producers profitability. Improving FE is a top priority for the global swine industry. A deeper understanding of the biology underlying FE is crucial for making progress in genetic improvement of FE traits. This review comprehensively discusses the topics related to the FE in pigs including: measurements, genetics, genomics, biological pathways and the advanced technologies and methods involved in FE improvement. We first provide an update of heritability for different FE indicators and then characterize the correlations of FE traits with other economically important traits. Moreover, we present the quantitative trait loci (QTL) and possible candidate genes associated with FE in pigs and outline the most important biological pathways related to the FE traits in pigs. Finally, we present possible ways to improve FE in swine including the implementation of genomic selection, new technologies for measuring the FE traits, and the potential use of genome editing and omics technologies.

The search for alternatives to antibiotics in poultry production is still on-going and has been directed towards investigation of the efficacy of different potential alternatives. However, it is important that the sought alternatives are cost-efficient and have no negative impact on meat quality, for ease of adoption and profit maximization. This study aimed at exploiting an agro-industrial waste, grape pomace (GP) as an alternative to in-feed antibiotics and assessing the effects on growth, intestinal morphology, ceca microbiota, ceca short-chain fatty acid (SCFA) concentration, blood biochemical parameters, and breast muscle myopathies of broiler chickens. A total of 576 one-day-old Cobb-500 broiler chicks were randomly allotted to 3 dietary treatments - Negative control (NC, a corn-wheat soybean-based diet), NC + 0.05% bacitracin methylene disalicylate (BMD), and NC + 2.5% GP. Each treatment was assigned to 8 replicate pens with 25 birds per pen. Body weight (BW), feed intake (FI), and feed conversion ratio (FCR) were determined weekly. On d 36, 2 chickens/pen were euthanized for measuring blood biochemical parameters, ceca SCFA, and ceca microbiota. White striping (WS) and wooden breast (WB) incidence were assessed in 4 chickens/pen on d 42. The GP diet increased (P < 0.05) average FI throughout the feeding phases compared to the other treatments, but overall FCR was similar. Birds in the GP treatment had higher (P < 0.05) villus height (VH) and increased VH:crypt depth ratio in the duodenum and jejunum compared to other treatments. The level of ceca SCFA and the incidence of WS and WB was the same for all treatments. Plasma Ca and P were significantly higher (P < 0.05) in birds fed GP and BMD, compared to the NC. Birds in the GP treatment had significantly reduced (P < 0.05) plasma aspartate transaminase than other treatments. Birds receiving GP had a higher (P < 0.05) relative abundance of the phylum Bacteroidetes and reduced (P < 0.05) Firmicutes compared to other treatments. The relative abundance of Bacteroides and Lactobacillus genera were higher (P < 0.05) among birds fed GP compared to other treatments. Inclusion of 2.5% GP in broiler chicken diets improved gut morphology and modified the cecal bacterial community and blood biochemical profiles with no adverse effect on growth performance and meat quality.

This study evaluated the effect of an essential oil blend and its delivery routes on broiler chicken growth performance, blood biochemistry, intestinal morphology, and immune and antioxidant status. Eggs were incubated and allotted to 3 groups: non-injected group, in ovo saline group, and in ovo essential oil group. On day 18 of incubation, essential oil in saline or saline alone was injected into the amnion. At hatch, chicks were assigned to post-hatch treatment combinations (1) in ovo essential oil + in-water essential oil (in ovo + in-water EO); (2) in ovo essential oil (in ovo EO); (3) in ovo saline; (4) in-water essential oil; (5) in-feed antibiotics (Bacitracin methylene disalicylate) and (6) a negative control (NC; corn-wheat-soybean diet) in 8 replicate cages (6 birds/cage) and raised for 28 day. The in ovo EO group reduced (p < 0.05) chick length and hatchability, all groups recorded no difference in growth performance at 0-28 day. The in ovo + in-water EO treatment reduced (p < 0.05) blood creatine kinase and aspartate aminotransferase levels whilst increasing (p < 0.05) total antioxidant capacity in birds. The in ovo + in-water delivery of EO might represent a potential antibiotic reduction strategy for the poultry industry but more research is needed to address the concern of reduced hatchability.

Previous findings have reported that providing light during incubation can affect hatchability and chick quality. This study was conducted to investigate the effects of providing light during incubation on posthatch broiler production parameters, thermoregulation and immune response. Lights with different wavelengths were used over the course of four separate hatches. Ross 308 broiler hatching eggs were randomly distributed into 4 lighting treatments for each hatch. The incubation lighting treatments included: dark as control, white, red, or blue lights for 12 h d-1 (200 lux at egg level). Broilers hatched from each incubator with the same gender were placed into one of 8 sets of pens (3 pens/set) and raised under 18 h d-1 photoperiod. Six birds per pen were immunized intraocularly with AviPro ND-IB Polybanco vaccine on d 10 and 21 posthatch. Chicks hatched under white and blue lights had heavier (P < 0.05) body weight and higher (P < 0.05) feed consumption than the control group during the first 6 h postplacement. No differences in vent temperature were found among treatments at 24 h posthatch (P > 0.05). Chicks hatched with light stimulation however had more stable (P < 0.05) cloaca temperature at 36 h posthatch. No differences in average body weight gain, feed consumption or feed conversion ratio were found among lighting treatments between d 7 and d 35. On d 14 of age, birds hatched from red light had higher (P < 0.05) total IgG concentration than those hatched under dark, blue or white light. These results indicated that in ovo light stimulation with different wavelengths did not affect growth parameters of broilers at market age. Providing photoperiodic blue and white light during incubation improved the production parameters of broilers during the first week posthatch.

Providing lighting schedule during incubation has been shown to improve chick quality and reduce stress posthatch. This study was conducted to evaluate the effects of providing light of different colors during incubation on embryo development, air cell temperature, the spread of hatch, and hatching performance. Four batches of eggs (n = 2,176, 1,664, 1,696 and 1,600) from Ross 308 broiler breeders were used in the experiment. In each trial, eggs were randomly distributed into 4 lighting treatments. The incubation lighting treatments included: incubated under dark as control, illuminated with white, red or blue lights for 12 h daily. There were no incubation lighting treatment differences in embryo development, the spread of hatch, hatchability, embryo mortality, hatch weight, chick length, navel closure quality, yolk-free body weight, or relative spleen weight. However, embryos incubated under red light had lower average air cell temperature than those in dark, white or blue light treatments. This finding may suggest higher melatonin secretion during the scotophase when illuminated with red light. Male chicks incubated under dark had a higher bursa of Fabricius weight than males illuminated with blue light. In conclusion, these results suggest that the red, white and blue light stimulation during incubation had no negative effects on hatchability, embryo mortality, spread of hatch or day-old chick quality, but may have potential impacts on immunity and energy metabolism in broiler embryos.

This study was planned to investigate the effects of seaweed supplementation, genetic strain, heat stress and their interactions on laying hen performances, blood chemistry and hematology. In a short-term trial, laying hens of the two genetic lines Lohman LSL-Lite (White) and Lohman Brown-Lite (Brown) were supplemented with Chondrus crispus (CC) at 3% for 21 days, while a control group was not. In a long-term trial, the same two strains were assigned to control (0%), 3% red seaweed Chondrus crispus (CC) or 0.5% brown seaweed Ascophyllum nodosum (AN)-supplemented diets for 41 weeks, concluding with a four-week control or heat-stress period. The White hens displayed higher egg production and a lower feed/egg ratio. The short-term inclusion of CC significantly reduced the feed intake, weight gain and feed/egg ratio. The long-term seaweed intake affected the plasma albumin and gamma-glutamyl transferase (GGT) (p < 0.05), and there were significant strain-heat stress interactions; heat stress in the Brown birds was associated with reduced protein, globulin and glucose and increased cholesterol and GGT levels and higher heterophil-to-lymphocyte (H/L) ratios (p < 0.05) in response to heat stress (p < 0.05). In conclusion, a long-term seaweed supplementation affected the plasma protein and enzyme profiles, yet had little effect on hen leukocyte counts and the overall performance.

Experiments were conducted to evaluate the effectiveness of a lysozyme product (InovapureTM) (LP) against E. coli penetrating eggshells. In the first microbiological experiment, 60 agar-filled eggs were inoculated with E. coli suspension, then fumigated with distilled water, 1.5% or 3.0% LP or a quaternary ammonium product (QA) at 0.125% for 10 min. In the second microbiological experiment, another 60 agar-filled eggs were fumigated with the same sanitizer treatments first, then inoculated with the E. coli suspension. Eggshells were candled and visual colonies were counted after 48 h incubation. An animal experiment was conducted to evaluate LP applied to the surface of 2080 broiler hatching eggs on hatching and growth performance. Hatching eggs were submerged in an E. coli suspension. After drip drying, eggs were randomly divided into four fumigation treatments, each with four subsets of 150 eggs. Fumigation treatments were the same as in the microbiological experiments. Eggs were incubated in 8 incubators (2 replicate incubators per treatment) and the broilers were grown to 33 d of age. In the microbiological experiments, inoculated eggs fumigated with 3.0% LP and 0.125% QA reduced (P < 0.05) the total amount of E. coli to 11 cfu/egg and 10 cfu/egg, respectively. When eggs were sanitized prior to inoculation, 3.0% LP demonstrated (P < 0.05) ongoing bactericidal action to prevent E. coli penetration. No differences in hatchability, fertility rate or egg weight loss percent were found among sanitation treatments. At hatch, body weight or the ratio of yolk sac weight to yolk-free body weight were not affected by the sanitation treatments. However, the application of sanitizers decreased (P < 0.05) the presence of E. coli in the yolk sac of newly hatched chicks. Feed consumption, body weight and feed conversion ratio were not affected by sanitation treatments. However, average daily body weight gain was lower (P < 0.05) following QA. Overall, 3.0% LP demonstrated acceptable activity against E. coli on eggshells, and provided ongoing bactericidal action to prevent E. coli penetration without negatively affecting growth performance.

Salmonella Enteritidis is vertically transmitted to eggs from laying hens through infected ovaries and oviducts. S. Enteritidis can also penetrate the eggshell from contaminated feces. Reducing S. Enteritidis in laying hens is vital to provide safer eggs and minimize the spread of salmonellosis to humans. Antibiotics have been widely used to control bacterial diseases in broilers and laying hens. However, there is a major concern that the use of antibiotics leads to the development of antibiotic resistance and adverse effects on microbiota of the treated birds. Thus, there is an interest in developing alternatives to antibiotics, such as dietary prebiotics. In the present study, feed supplemented with the red seaweeds: Chondrus crispus (CC) or Sarcodiotheca gaudichaudii (SG), was offered to laying hens late in production to control S. Enteritidis. Diets contained one of the following; 2% or 4% Chondrus crispus (CC2, and CC4, respectively) or Sarcodiotheca gaudichaudii (SG2 and SG4, respectively). Chlortetracycline was used in the positive control diet. During week-4, 48 birds were orally challenged with 2 × 109 CFU/mL of S. Enteritidis. Eggs and fecal samples were collected 1, 3, 5, and 7 days' post inoculation. Birds were euthanized and organs (ceca, ovary, liver, and spleen) were sampled and analyzed for the presence of S. Enteritidis, 7 days' post inoculation. Results showed that seaweed reduced the negative effect on body weight and egg production in S. Enteritidis-challenged laying hens. Analysis of fecal samples showed that the antibiotic (CTC) reduced S. Enteritidis in the intestinal tract and fecal samples, 3 days' post inoculation. Fecal samples from Chlortetracycline and CC4 supplemented birds tested negative for S. Enteritidis on days 5 and 7 post inoculation (lowest detection limit = 10-1). S. Enteritidis colonization in the ceca was also significantly reduced in birds fed CC (4%) and Chlortetracycline. Blood serum profiles revealed that there were no significant differences in serum aspartate aminotransferase (AST) and sodium. However, the level of serum immunoglobulin (IgA) was higher in the CC4 treatment. The relative abundance of Lactobacillus acidophilus was significantly higher in CC4 while, the abundance of the pathogenic bacteria, Clostridium perfringens and Salmonella Enteritidis were reduced compared to control. Results indicate that feed supplemented with 4% CC is effective in providing protection against Salmonella Enteritidis colonization in laying hens.

Red seaweeds are a rich source of unique bioactive compounds and secondary metabolites that are known to improve human and animal health. S. Enteritidis is a broad range host pathogen, which contaminates chicken and poultry products that end into the human food chain. Worldwide, Salmonella outbreaks have become an important economic and public health concern. Moreover, the development of resistance in Salmonella serovars toward multiple drugs highlights the need for alternative control strategies. This study evaluated the antimicrobial property of red seaweeds extracts against Salmonella Enteritidis using the Caenorhabditis elegans infection model. Six red seaweed species were tested for their antimicrobial activity against S. Enteritidis and two, Sarcodiotheca gaudichaudii (SG) and Chondrus crispus (CC), were found to exhibit such properties. Spread plate assay revealed that SG and CC (1%, w/v) significantly reduced the growth of S. Enteritidis. Seaweed water extracts (SWE) of SG and CC, at concentrations from 0.4 to 2 mg/ml, significantly reduced the growth of S. Enteritidis (log CFU 4.5-5.3 and log 5.7-6.0, respectively). However, methanolic extracts of CC and SG did not affect the growth of S. Enteritidis. Addition of SWE (0.2 mg/ml, CC and SG) significantly decreased biofilm formation and reduced the motility of S. Enteritidis. Quantitative real-time PCR analyses showed that SWE (CC and SG) suppressed the expression of quorum sensing gene sdiA and of Salmonella Pathogenesis Island-1 (SPI-1) associated genes sipA and invF, indicating that SWE might reduce the invasion of S. Enteritidis in the host by attenuating virulence factors. Furthermore, CC and SG water extracts significantly improved the survival of infected C. elegans by impairing the ability of S. Enteritidis to colonize the digestive tract of the nematode and by enhancing the expression of C. elegans immune responsive genes. As the innate immune response pathways of C. elegans and mammals show a high degree of conservation, these results suggest that these SWE may also impart beneficial effects on animal and human health.

The aim of this study was to evaluate the effect of the inclusion of red seaweed supplementation to standard poultry diets on production performance, egg quality, intestinal histology, and cecal short-chain fatty acids in Lohmann Brown Classic laying hens. A total of 160 birds were randomly assigned to 8 treatment groups. Control hens were fed a basal layer diet; positive control hens were fed a diet containing 2% inulin; and 6 treatment groups were fed a diet containing one of the following; 0.5, 1, or 2% Chondrus crispus (CC0.5, CC1, and CC2, respectively) and one of the same 3 levels of Sarcodiotheca gaudichaudii (SG0.5, SG1, and SG2, respectively). Dietary supplementation had no significant effect on the feed intake, BW, egg production, fecal moisture content, and blood serum profile of the birds. The feed conversion ratio per gram of egg was significantly more efficient (P = 0.001) for CC2 and SG2 treatments. Moreover, SG1 supplementation increased egg yolk weight (P = 0.0035) and birds with CC1 supplementation had higher egg weight (P = 0.0006). The SG2 and CC2 groups had greater (P < 0.05) villus height and villus surface area compared with the control birds. Seaweed supplementation increased the abundance of beneficial bacteria [e.g., Bifidobacterium longum (4- to 14-fold), Streptococcus salivarius (4- to 15-fold)] and importantly reduced the prevalence of Clostridium perfringens in the gut of the chicken. Additionally, the concentrations of short-chain fatty acids, including acetic acid, propionic acid, n-butyric acid, and i-butyric acid, were significantly higher (P < 0.05) in CC and SG treatments than in the control. In conclusion, dietary supplementation using red seaweed inclusions can act as a potential prebiotic to improve performance, egg quality, and overall gut health in layer hens.

The aim of this study was to evaluate the quality of eggs from 2 selected commercial strains of laying hens and 2 unselected lines of chickens fed diets with different combinations of Ca and vitamin D and relate it to the profile of uterine proteins and ultrastructure of the shell. A group of 4 chickens was housed in each of 24 cages. The group consisted of one representative from each of the following breeds: Lohmann LSL- Lite, Lohmann Classic-Brown, Fayoumi, and Light Sussex. Six dietary combinations of Ca and vitamin D(3) (3.35%, 2,500 IU; 4.10%, 2,500 IU (control); 4.85%, 2,500 IU; 3.35%, 200 IU; 4.10%, 200 IU; and 4.85%, 200 IU) were randomly assigned to 4 replicate cages for 2 treatment periods (26-29 and 56-59 wk of age). Data were analyzed as a split-plot design with cage as the main plot and hen as the subplot. Egg quality traits were different (P < 0.0001) between commercial and heritage breeds. Lohmann Brown had stronger shells with higher specific gravity compared with other breeds. Both commercial and heritage birds responded to a drop in vitamin D3 level by marked reduction in shell thickness. The SDS-PAGE profiles of uterine fluid samples revealed a decrease (P < 0.05) in 200-, 150-, 116-, and ≤6.5-kDa proteins, whereas proteins with molecular weight (MW) of 80, 55, 52, 45, 42, and 28 kDa increased with bird age. A 36- and 52-kDa protein band was most intense for Fayoumi compared with other breeds. Ultrastructural characteristics showed flattened and deeply etched mammillary caps for Lohmann Brown and the presence of type A and type B bodies between mammillary cones in eggshells from Fayoumi and Lohmann Lite. The negative correlation between ultrastructural characteristics, which decrease with bird age, and the 116-kDa uterine protein band could provide insight into reduced eggshell quality as hens age.

Eggs have been identified as a source of salmonellosis, making the transmission of Salmonella to eggs of great concern to the poultry industry. The goal of this experiment was to determine the ability of Salmonella to penetrate the eggshell of 5 different breeds of noncommercial chicken, Barred Plymouth Rock, White Leghorn, Brown Leghorn, Fayoumi, and Light Sussex, and 1 commercial Lohmann LSL-Lite. Egg weight, breaking force, shell weight, and shell thickness measurements were taken for 30 eggs per breed. A 1 cm in diameter hole was cut out from the narrow end of 30 additional eggs per breed. The shells were filled with plate count agar containing tetracycline and 0.1% 2,3,5-triphenyl terazolium chloride and sealed with paraffin wax. Agar-filled eggs were submerged for 1 min in an overnight culture of tetracycline-resistant Salmonella Heidelberg and incubated at 37°C for 40 h. Eggs were candled and visual colonies were counted and reported as cfu per egg and cfu per gram of shell. The SAS mixed model was used to evaluate differences between breeds for egg quality characteristics and the number of cfu per egg and per gram of shell. Commercial layers (62.6 g) and Barred Plymouth Rock (61.5 g) produced the largest eggs, whereas Fayoumi (47.1 g) produced the smallest (P < 0.05). Force to break the shell was lowest (P < 0.05) for Barred Plymouth Rock (3.6 kg) and greatest for the commercial (4.4 kg), White Leghorn (4.4 kg), and Fayoumi (4.2 kg). Bacteria penetrating the shell was lowest (P < 0.05) for Barred Plymouth Rock (10.7 cfu/g) and highest for Light Sussex (27.7 cfu/g) and Brown Leghorn (27.2 cfu/g), with other breeds intermediate. These results indicate that there are breed-specific influences on the ability of an egg to resist Salmonella, which cannot be explained by shell quality measurements. Further investigations are warranted to determine the contributing factors to shell penetration by bacteria. This study highlights the value in maintaining heritage chicken breeds as a genetic resource for the future.

BACKGROUND: Apple skins are a rich source of flavonols, in particular quercetin (Q) glycosides. The objective of the present study was to investigate the presence of Q metabolites in plasma, various tissues, and excreta when the commercial broiler chicken's diet was supplemented with Q (0, 50, 150, 300, or 600 mg kg(-1) body weight per day), an apple skin extract (ASE; 50, 150 mg total phenolics kg(-1) body weight per day), or a dried apple skin powder (ASP; 50 mg total phenolics kg(-1) body weight per day). RESULTS: When Q was supplemented for 3 days, Q sulfate, Q glucuronide, Q glucoside glucuronide, Q glucoside sulfate, and isorhamnetin glucoside were detected by liquid chromatography-tandem mass spectrometry in the liver and duodenum. Deconjugated Q was also detected in the breast and thigh tissues of ASE- and ASP-supplemented broilers. Regardless of the source or concentration of Q, the antioxidant capacity measured by ferric reducing ability of plasma (FRAP) assay in the plasma and tissues of the broilers did not change significantly. CONCLUSIONS: As far as is known, this is the first report to demonstrate that Q and its glycosides can be absorbed and metabolized by broiler chickens.

Bactericidal activity of Brifisol K™ (a commercial blend of sodium acid pyrophosphate and orthophosphoric acid) (BK Ladenburg, Ladenburg, Germany) was evaluated during chilling of broiler carcasses. Brifisol K™ (1.5% at 1°C for 60 min) significantly reduced Escherichia coli , coliforms and aerobic plate counts (APC) on postchill broilers and increased shelflife by 1 to 2 days when stored at 4.4°C. Reductions in incidence and levels of Salmonella were directly related to successful neutralization of the carcass rinse solutions. Effective neutralization of all phosphate applications, whether the treatment is alkaline or acidic based, is extremely important for accurate quantification of bactericidal efficacy.
