RESUMO

Marked variability of the ceftazidime pharmacokinetics (Cmax and T1/2) was observed in 3 newborns and 2 infants with purulent septic infections. The patients were under complex treatment in a reanimation unit (artificial pulmonary ventilation, infusions). It was recommended to perform the treatment with monitoring the antibiotic plasma concentrations to prevent the drug failure because of the changes in the distribution and excretion patterns. The use of HPLC for the purpose is advisable.

RESUMO

The functional activity of erythrocytes and platelets was studied in 50 patients with various types of coronary heart disease. The patients' plasma exhibited higher ADP levels due to erythrocyte microhemolysis and higher platelet malonic dialdehyde and intracellular calcium, which led to enhanced platelet functional activity. Treating the patients with the calcium antagonist diltiazem was found to result in lower ADP and platelet calcium levels whereas the concentrations of malonic dialdehyde remained unchanged.

RESUMO

A study of 20 anginal patients demonstrated signs of platelet hyperreactivity: increased platelet aggregation, increased mobilization of Ca2+ from platelet membranes, and increased production of malonic dialdehyde. A short-term treatment with isoptin-120 (360 mg daily) produced a considerable drop in those parameters. It is suggested that the drug's basic effect is due to the inhibition of intracellular calcium redistribution in the platelets of coronary patients.

RESUMO

Erythrocyte filterability and its relation to erythrocyte volume, hemoglobin level and cholesterol/phospholipid ratio were studied in anginal patients. A 25% decrease in filterability and elevated erythrocyte cholesterol/phospholipid ratios were found in anginal patients, as compared to normal subjects. A close inverse correlation (r = -0.623) was demonstrated between erythrocyte filterability and cholesterol/phospholipid ratio. Mean erythrocyte volume and Hb levels were similar in anginal patients to those of normal subjects.

RESUMO

Increased molar cholesterol/phospholipid ratios, significantly increased membrane microviscosity, reduced filterability through a 3 micron-pore filter, increased aggregation and longer disaggregation time, and occasional abnormally large cells were found in erythrocyte assays of 80 coronary patients, as compared to normal subjects. It is suggested that erythrocyte microrheologic disorders are related to excessive cellular cholesterol content in coronary patients.

RESUMO

A study of 80 anginal patients showed their platelet molar cholesterol/phospholipid ratios, platelet membrane microviscosity, membrane release of calcium and fatty acids, and platelet aggregation capacity to be elevated. It is assumed that excessive platelet cholesterol has a significant effect on platelet aggregation, that is mediated by the intraplatelet processes examined, in anginal patients.

RESUMO

Electronic paramagnetic resonance was used to study the structural and functional properties of platelets from patients with coronary heart disease. It was established with the use of the spin probe 5-doxylstearate that cytoplasmic platelet membranes of coronary heart disease patients are marked by a higher molecular packing as compared to normal. The rate of platelet aggregation studied independently of plasma factors appeared also higher, with a direct correlation being discovered between the aggregation properties and viscosity of platelet membranes. Such a modification of the physical and functional properties might be accounted for by cholesterol accumulation which is evidenced by an increase in the molar content of cholesterol in the cells. It is suggested that platelets from coronary heart disease patients, which have more rigid membranes, have a greater ability to adhere to other, particularly to endothelial cells, thereby giving rise to the atherosclerosis of the vascular wall.

RESUMO

Blood rheology was studied in 92 patients with ischemic heart disease having cardiac rhythm disorders. The syndrome of high blood viscosity and increased blood platelets aggregation were observed in paroxysmal tachycardia. In patients suffering from myocardial infarction with arrhythmia there was noted higher blood viscosity and more intensive blood platelet aggregation than in atherosclerotic cardiosclerosis. Treatment of paroxysmal tachycardia by means of electric impulse and agents blocking beta-adrenergic receptors results in a decrease of blood viscosity, the blood platelets aggregation intensity being unchanged.

RESUMO

The examination was conducted in 77 patients with a varying course of acute myocardial infarction, in 10 normal individuals, 10 patients with angina pectoris, and 20 patients with post-infarction cardiosclerosis. The patients with acute myocardial infarction demonstrated a higher, than in the control group, blood viscosity, fibrinogen, hematocrit level and erythrocytes aggregation rate. The most significant changes in the rheologic properties of blood were observed in the cases of complicated myocardial infarction. Patients with severe coronary shock displayed both high and low levels of blood viscosity, fibrinogen and hematocrit, which may be due to the different stages of the coronary shock.
