A longitudinal study was conducted in a single dairy goat herd to investigate the relationship between subclinical small ruminant lentivirus (SRLV) infection in does and litter size (LS) or birth body weight of kids (BW). Each year kids born to seropositive and seronegative does were weighed before the first consumption of colostrum. LS and BW of each kid were recorded. BW was significantly negatively linked to LS (p = 0.006) - singletons weighed (mean ± SD) 4.20 ± 0.67 kg, twins - 3.75 ± 0.62 kg, and triplets and quadruplets - 3.38 ± 0.47 kg. Male kids were significantly heavier than female kids in twin litters (3.97 ± 0.53 kg vs. 3.52 ± 0.60 kg; p < 0.001) and triplet or quadruplet litters (3.62 ± 0.40 kg vs. 3.17 ± 0.43 kg; p < 0.001). However, BW of male and female kids from singleton litters did not differ (4.31 ± 0.71 kg vs. 4.07 ± 0.65 kg; p = 0.154). Then, two mixed models were developed to assess the relationship between LS (mixed Poisson log linear regression model) or BW (mixed linear model) and SRLV infection in the doe, controlling for potential confounders such as the effect of an individual doe, year in which the parturition took place, parity and kid's sex. Neither LS nor BW proved to be significantly associated with SRLV infection (p = 0.788 and p = 0.214, respectively). On this basis it was concluded that LS and BW were not affected by the subclinical SRLV infection of a doe.

The aim of this study was to determine the effect of diet supplemented with selenized yeast (Se-yeast) on milk yield and milk composition of goats and expression of casein and mammary-gland-immune system genes in milk somatic cells (MSC). Twenty-four dairy goats in their second to fourth lactations were divided into control and experimental groups, balanced according to lactation number and breed (Polish White or Fawn Improved). Morning milk and blood samples were collected four times during lactation (on the 21st, 70th, 120th, 180th day after kidding). The control and experimental groups were fed diets with 0.7 mg inorganic Se/goat/day (sodium selenite) or 0.6 mg organic Se/goat/day (selenized yeast), respectively. Milk, fat and protein yields during lactation as well as average somatic cell count, fat, protein and lactose contents in milk were evaluated. Microelements in milk and blood serum and biochemical parameters in blood serum were determined at the beginning and the end of the experiment. The expression levels of the genes encoding αS1-casein (CSN1S1), αS2-casein (CSN1S2), κ-casein (CSN3), interleukin 8 (IL-8), serum amyloid A3 (SAA3), interleukin 1ß (IL-1ß), bactenecin 7.5 (BAC7.5), bactenecin 5 (BAC5), ß2-defensin (GBD2), hepcidin (HAMP), chemokine 4 (CCL4), tumour necrosis factor α (TNFα), toll-like receptor 2 (TLR2), cathelicidin-7 (MAP34) and cathelicidin-6 (MAP28) were determined in MSC. Milk, fat, and protein yields were higher and somatic cell count (SCC expressed as natural logarithm) was lower in the milk of goats fed organic Se. The Se concentration in milk was twice as high in the organic vs. inorganic treatment groups at the end of the experiment, while there were no differences in studied biochemical parameters between groups. The transcript levels of CSN1S2 and BAC7.5 were higher and IL-8 was lower in MSC of Se-yeast treated groups. Such results may indicate better health status of mammary glands of goats treated with organic Se as well as positive impact of selenized yeast on the goat's milk composition. Differences in the IL-1ß and IL-8 transcript levels were also noted between the stages of lactation, with the highest expression at the peak of lactation (day 70), highlighting the metabolic burden at this time. We concluded that the Se-yeast supplementation improved the productivity and health status of goats and could have significant economic impact on farmer's income.

The aim of the study was to analyze acute phase protein and cathelicidin gene responses to small ruminant lentivirus (SRLV) infection in goats. In uninfected goats, we found higher Cp and lower Fbγ mRNA levels in blood leucocytes (BL) than in milk somatic cells (MSC), as well as lower SAA, Hp, and CRP and higher Cp and AGP concentrations in blood serum than in milk. In SRLV-infected goats, we found higher Fbγ and MAP28 and lower Cp expression in MSC than in BL, and higher SAA, Hp, Fb, and MAP28 and lower AGP concentrations in milk than in blood serum. Higher SAA and Hp expressions in BL and Hp expression in MSC were found in SRLV-infected goats. In SRLV-infected goats, we observed a higher concentration of SAA in blood serum, while in milk, lower SAA, Cp, and MAP28 and higher MAP34 concentrations were observed. The expression profiles of the studied genes differed between BL/serum and MSC/milk. The elevated SAA concentration in blood serum was accompanied by a decreased concentration of SAA and Cp in the milk of infected goats. No differences in the expression of the other studied genes may mean that the SRLV has the ability to evade the immune system, continuing to replicate. The elevated concentration of SAA in blood serum may promote viral multiplication. This higher concentration of SAA in blood serum and simultaneous reduced concentration of SAA and Cp in milk may be additive indicators of this infection.

We examined acute phase protein (APP) concentrations in viral infections of dairy ruminants and assessed the potential role of characteristic patterns of APP changes in auxiliary diagnosing viral diseases. All viruses reviewed are common causes of farm animal diseases. APPs are among the first agents of immunity, and their concentrations could be diagnostically relevant. In the most common ruminant viral diseases, elevated serum amyloid A (SAA) and haptoglobin (Hp) levels in blood serum have been observed. However, since these proteins are the main APPs in many viral infections, it is impossible to use their levels for diagnosing particular infections. Decreased Cp and albumin expression could help differentiate the bluetongue virus infection from other diseases. Lastly, analysis of SAA levels in blood serum and milk could be helpful in diagnosing small ruminant lentivirus infection. While promising, APP levels can only be considered as an auxiliary tool in diagnosing viral diseases in ruminants.

We carried out this study to determine for how long small ruminant lentivirus (SRLV)-specific antibodies can be detected by three commercial ELISA kits in goat kids after suckling infected does in field conditions. Forty-one kids born to SRLV-seropositive asymptomatic does were blood sampled prior to colostrum consumption, and then weekly for 6 months in total. The sera were screened with three commercial ELISA kits: whole-virus ELISA (wELISA), recombinant transmembrane and capsid antigen ELISA (TM/CA-ELISA), and surface antigen ELISA (SU-ELISA). All but one kid were seronegative in all three ELISAs right after birth. At the age of 1 week all kids turned seropositive in wELISA, 39 kids (95%) in TM/CA-ELISA, and 35 kids (85%) in SU-ELISA. All seropositive kids turned seronegative in wELISA by the 15th week, and in SU-ELISA by the 19th week (median of 8 weeks in both ELISA), whereas in TM/CA-ELISA five kids (13% of 39 initially seropositive) were still seropositive at the age of 6 months (median of 11 weeks). Antibody levels at the age of 1 week proved significantly linked to the duration of maternal antibodies in all three ELISAs and could be employed to predict for how long maternal antibodies would remain detectable.

Haptoglobin (Hp) and serum amyloid A (SAA) are considered as the major acute phase proteins (APPs) in goats. These APPs have been investigated in several studies during the last decade. In most studies, a colorimetric assay for Hp and a solid phase sandwich ELISA for SAA have been used for quantification. In 2015, reference intervals for APPs were determined using a new type of assay, the competitive ELISA (cELISA). Results obtained by the cELISA differed significantly from results obtained by previously used assays. The present study aimed to assess the agreement between so far used assays and cELISAs. Sera of 152 female dairy goats of two Polish national breeds were analysed. The concentration of Hp was determined using a colorimetric assay (Hp-CA) and the cELISA (Hp-cELISA), while a solid phase sandwich ELISA (SAA-sELISA) and the cELISA (SAA-cELISA) were used to measure SAA. Agreement between test results was assessed by preparing Bland-Altman plots, and analyzing 95% limits of agreement (LoA). Finally, the assays for Hp and SAA were compared using 147 and 138 serum samples, respectively, as 5 and 14 paired measurements, respectively, were excluded from agreement analyses to avoid extrapolation of Hp and SAA concentration. Measurements obtained by the Hp-CA and Hp-cELISA showed weak positive correlation (r = 0.24, P = 0.003). Limits of agreement (LoA) ranged from + 1.6 (95% CI 1.4 to 1.8) g/L to - 1.5 (95% CI - 1.7 to - 1.3) g/L. Measurements yielded by the SAA-sELISA and SAA-cELISA did not correlate (r = - 0.01, P = 0.855). LoA ranged from + 14.5 mg/L (95% CI 12.9 to 16.1) to - 8.5 mg/L (95% CI - 10.1 to - 6.9). Agreement between the two types of commercial assays for determination of Hp and SAA concentrations in goats is poor and cELISAs tend to underrate both Hp and SAA concentrations.

We documented changes in serum amyloid A (SAA) and haptoglobin (Hp) concentration in goats during pregnancy, as measured by competitive ELISAs. Fifteen does (pregnant group) and 20 castrated males (control group) were enrolled in the study. Blood samples were collected on the same day from all 35 goats, 7 times throughout the study period: at mating, then once every month, during the week preceding kidding, and 1 mo after kidding. Baseline SAA and Hp concentrations at mating were identical in the 2 groups. In the pregnant group, SAA concentration rose significantly in the second month and remained elevated until the end of pregnancy, with the peak concentration at kidding. In the control group, SAA concentration remained unchanged compared to the baseline concentration throughout the study. SAA concentration was significantly higher in the pregnant than control group only at the end of the fourth month of pregnancy and at kidding. Hp concentration did not change during pregnancy or between groups. SAA concentration at kidding was affected only by parity of does - it was highest in does in the third and fourth pregnancy and gradually lower in older does.

Knowing about the genes involved in immunity, and being able to identify the factors influencing their expressions, helps in gaining awareness of the immune processes. The qPCR method is a useful gene expression analysis tool, but studies on immune system genes are still limited, especially on the caprine immune system. Caprine arthritis encephalitis, a disease caused by small ruminant lentivirus (SRLV), causes economic losses in goat breeding, and there is no therapy against SRLV. The results of studies on vaccines against other viruses are promising. Moreover, the Marker-Assisted Selection strategy against SRLV is possible, as has been shown in sheep breeding. However, there are still many gaps in our knowledge on the caprine immune response to infection. All types of cytokines play pivotal roles in immunity, and SRLV infection influences the expression of many cytokines in different types of cells. This information encouraged the authors to examine the results of studies conducted on SRLV and other viral infections, with an emphasis on the expression of cytokine genes. This review attempts to summarize the results of studies on the expression of cytokines in the context of the SRLV infection.
