BACKGROUND: Cysteine (Cys), glutathione (GSH), and homocysteine (Hcy), as three major biothiols are involved in a variety of physiological processes and play a crucial role in plant growth. Abnormal levels of Cys can cause plants to fail to grow properly. To date, although a very large number of fluorescent probes have been reported for the detection of biothiols, very few of them can be used for the selective discrimination of Cys from GSH and Hcy due to their structural similarity, and only a few of them can be used for plant imaging. RESULTS: Here, three fluorescent probes (o-/m-/p-TMA) based on TMN fluorophore and the ortho-/meta-/para-substituted maleimide recognition groups were constructed to investigate the selective response effect of Cys. Compared to the o-/m-TMA, p-TMA can selectively detect Cys over GSH and Hcy with a rapid response time (10 min) and a low detection limit (0.26 µM). The theoretical calculation confirmed that the intermediate p-TMA-Cys-int has shorter interatomic reaction distances (3.827 Å) compared to o-/m-TMA-Cys (5.533/5.287 Å), making it more suitable for further transcyclization reactions. Additionally, p-TMA has been employed for selective tracking of exogenous and endogenous Cys in Arabidopsis thaliana using both single-/two-photon fluorescence imaging. Furthermore, single cell walls produced obvious two-photon fluorescence signals, indicating that p-TMA can be used for high-concentration Cys analysis in single cells. Surprisingly, p-TMA can be used as a fluorescent dye for protein staining in SDS-PAGE with higher sensitivity (7.49 µg/mL) than classical Coomassie brilliant blue (14.11 µg/mL). SIGNIFICANCE: The outstanding properties of p-TMA make it a promising multifunctional molecular tool for the highly selective detection of Cys over GSH and Hcy in various complex environments, including water solutions, zebrafish, and plants. Additionally, it has the potential to be developed as a fluorescent dye for a simple and fast SDS-PAGE fluorescence staining method.

Electroactive hydrogels have garnered extensive interest as a promising approach to myocardial tissue engineering. However, the challenges of spatiotemporal-specific modulation of individual pathological processes and achieving nontoxic bioresorption still remain. Herein, inspired by the entire postinfarct pathological processes, an injectable conductive bioresorbable black phosphorus nanosheets (BPNSs)-loaded hydrogel (BHGD) was developed via reactive oxide species (ROS)-sensitive disulfide-bridge and photomediated cross-linking reaction. Significantly, the chronologically programmed BHGD hydrogel can achieve graded modulation during the inflammatory, proliferative, and maturation phases of myocardial infarction (MI). More details, during early infarction, the BHGD hydrogel can effectively reduce ROS levels in the MI area, inhibit cellular oxidative stress damage, and promote macrophage M2 polarization, creating a favorable environment for damaged myocardium repair. Meanwhile, the ROS-responsive structure can protect BPNSs from degradation and maintain good conductivity under MI microenvironments. Therefore, the BHGD hydrogel possesses tissue-matched modulus and conductivity in the MI area, facilitating cardiomyocyte maturation and electrical signal exchange, compensating for impaired electrical signaling, and promoting vascularization in infarcted areas in the maturation phase. More importantly, all components of the hydrogel degrade into nontoxic substances without adverse effects on vital organs. Overall, the presented BPNS-loaded hydrogel offers an expandable and safe option for clinical treatment of MI.

A method to use a pulsed solid-state laser to create structural color modulation on metal-coated colloidal crystal surfaces by changing the scanning speed has been proposed. Vivid colors as cyan, orange, yellow, and magenta are obtained with different predefined stringent geometrical and structural parameters. The effect of laser scanning speeds and polystyrene (PS) particle sizes on the optical properties is studied, and the angle-dependent property of the samples is also discussed. As a result, the reflectance peak is progressively red shifted along with increasing the scanning speed from 4 mm/s to 200 mm/s with 300 nm PS microspheres. Moreover, the influence of the microsphere particle sizes and incident angle are also experimentally investigated. For 420 and 600 nm PS colloidal crystals, along with a gradual decrease in the scanning speed of the laser pulse from 100 mm/s to 10 mm/s and an increase in the incident angle from 15° to 45°, there was a blue shift for two reflection peak positions. This research is a key, low-cost step toward applications in green printing, anti-counterfeiting, and other related fields.

This paper examines the impact of using narratives to communicate a controversial health issue, gaming disorder, on adolescents' issue attitudes. In a between-subjects experiment, 115 adolescent participants read either a narrative or an informational message on gaming disorder. Results indicated that compared to the informational message, the narrative health message generated more positive attitudes toward the medical view of problematic gaming and greater attitude certainty. Transportation mediated the narrative's effect on attitudes. Behavioral involvement moderated the narrative's effect on attitudes and attitude certainty, such that the positive effects of the narrative on attitudes and attitude certainty were more pronounced for high-involvement adolescents than for low-involvement adolescents. In addition, behavioral involvement also enhanced the effect of message absorption on attitudes. By extending our research on narrative effects to the adolescent population, this study presents findings with both theoretical and practical implications.
