Vegetated land surfaces play an important role in determining the fate of carbon in the global carbon cycle. However, our understanding of the terrestrial biosphere on a global scale is subject to considerable uncertainty, especially concerning the impacts of climatic variables on the carbon cycle. Soil is a source and also a sink of CO2 exchange and helps in carbon sequestration. Agricultural management practices influence soil water dynamics, as well as carbon cycling by changing soil CO2 emission and uptake rates. The rate of soil CO2 emission varies for different crops and different organic amendments. The major goal of this study was to assess the impacts of the type and rate of organic amendment on soil CO2 emission in a collard greens crop grown in the southeast Texas environment. Thirty-six plots were developed to grow collard greens on Prairie View A&M University's Research Farm. Three types of organic amendments (Chicken manure, Dairy manure, and Milorganite), at four levels of application (0, 168, 336, and 672 kg N/ha) were used and replicated three times. Each organic amendment type was applied to nine randomly selected plots. Three random plots were used as a control in each row. We measured daily soil CO2 emission for the first two weeks and every other day in a week during the experiment. We evaluated the effects of organic amendments and the application rates on soil CO2 emission for collard greens during two growing seasons. The results showed higher the application rates for each organic amendment, higher the CO2 emissions from the soil. The results also showed higher cumulative CO2 emissions for the soils amended with chicken manure and milorganite, but lowest for the soils amended with dairy manure. This field experiment and analyses help better understand the temporal and spatial variations of soil CO2 emission, and also help to develop best management practices to maximize carbon sequestration and to minimize soil CO2 emissions during the growth periods of collard greens under changing temperatures using different organic amendments, and application rates.

AIM: To investigate the association between NSUN2/IGF-II signature and ovarian cancer survival. METHODS: Using a publicly accessible dataset of RNA sequencing and clinical follow-up data, we performed Classification and Regression Tree and survival analyses. RESULTS: Patients with NSUN2 high IGF-II low had significantly superior overall and disease progression-free survival, followed by NSUN2 low IGF-II low, NSUN2 high IGF-II high and NSUN2 low IGF-II high (p < 0.0001 for overall, p = 0.0024 for progression-free survival, respectively). The associations of NSUN2/IGF-II signature with the risks of death and relapse remained significant in multivariate Cox regression models. Random-effects meta-analyses show the upregulated NSUN2 and IGF-II expression in ovarian cancer versus normal tissues. CONCLUSION: The NSUN2/IGF-II signature associates with heterogeneous outcome and may have clinical implications in managing ovarian cancer.

Cytotoxic T lymphocytes (CTLs) are a key player in cancer immunotherapies, and MHC class I molecules on the cell surface are crucial for cellular recognition. However, the aberrant expression of MHC class I molecules is frequently found in various malignancies. IFNγ has dual functions in cancer progression, and its effect on tumor immunity is controversial. To investigate whether IFNγ can enhance cytotoxic efficiency of the tumor antigen-specific CTLs, we generated the CTLs using modified human dendritic cells as antigen presenting cells, then studied the activities of CTLs on human leukocyte antigen (HLA)-A2 positive glioma cells treated with, or without IFNγ. The results from both ELISpot and cytotoxicity assays demonstrated that the CTLs recognized and eliminated the HLA-A2 positive glioma cells treated with IFNγ more effectively when compared to the glioma cells deprived of IFNγ treatment. In addition, in vitro experiments showed that the levels of MHC class I molecules were upregulated in all of the HLA-A2 positive glioma cells. Using the publicly accessed TCGA data of low-grade glioma, we found significantly positive associations between IFNγ and both MHC class I molecules and CD8+ T cell activation score (p<0.0001). Furthermore, we found a significantly reduced risk of death in the glioma patients with high T cell activation score in comparison to those with low score (p=0.022). These findings suggest that a clinical application of IFNγ treatment may have potential benefits.
